Through innovating pretreatment procedure and gas-liquid separator（GLS）,as well as optimizing of some factors,the developed procedure offered an accurate alternative to those based on a previous complete digestion of samples for the determination of lead in ***, some certified reference cattle bloods and some human whole bloods were accurately determined by this *** 20 shares of human whole blood samples determined by graphite furnace atomic absorption spectrometry（GFAAS） and this method,there were no significant difference between value found by this method and *** analytical performances of this method were listed below:the limit of detect（LOD） of this method was less than 0.6μg L（3σ）;the limit of quantification（LOQ） of this method was less than 2.0μg L（10σ）;the relative standard deviation（RSD） of this method was less than 5.0%(100μg L).The method based on the excellent sensitivity attainable by hydride generation-atomic fluorescence spectrometry（HGAFS）, on a non-digestion soft room-temperature treatment of the blood samples and on the decomposition of organic complex by UV irradiation,was a safe and comfortable methodology for operators,which could reduce the reagent consumption and time of analysis.

目的建立及评价离体兔眼(isolated rabbit eye,IRE)试验的试验方法,评价IRE试验的预测能力、重复性和对不同刺激性的受试物的预测能力。方法选取26种参照化学物和26种化妆品,参照欧洲替代方法验证中心(European Centrefor the Validation of Alternative Methods,ECVAM)推荐的IRE试验方法进行眼刺激性检测,首先将健康家兔的离体眼球置于离体眼球表面灌流系统中,维持其正常形态和功能,受试物接触离体眼球10s后以20ml生理盐水冲洗,观察离体眼球接触受试物后不同时间点角膜混浊度、荧光素滞留程度和角膜水肿率的改变,并根据标准进行评分分级。每种受试物的IRE试验均重复三次。同步采用我国卫生部《化妆品卫生规范》(2007年版)进行眼刺激性检测。对IRE试验和Draize试验分级结果进行Pearson x 2检验、McNemar-Bowker、Gamma和Kappa统计学分析,以Draize试验分级结果作为"金标准",分别计算IRE试验检测不同刺激能力受试物的灵敏度、特异度和ROC曲线下面积。结果对于26种参照化学物,IRE试验与Draize试验结果具有较高的等级相关性(SpearmanR=O.943,P=O.000;Gamma=1.000,P=O.000)及分级一致性(Kappa=0.799,P=O.000),两种试验方法分级的差别无统计学意义(McNemar Bowker W=5.00,P=O.172);三次IRE试验结果重复性良好(F=0.439,P=O.646);IRE试验对强刺激性参照化学物的预测能力较强,灵敏度为81.8%、特异度为100%,ROC=0.909(P=O.000)。对于26种化妆品,IRE试验与Draize试验结果也具有较高的等级相关性(Spearman R=O.764,P=O.000;Gamma=O.861,P=O.000)及分级一致性(Kappa=0.581,P=O.000),两种试验方法分级的差别无统计学意义(McNemar-Bowker W=3.20,P=O.525);IRE试验对无刺激化妆品的预测能力较强,灵敏度为100%、特异度为93.9%,ROC=0.970(P=O.028),IRE试验对刺激性化妆品的预测能力较强,灵敏度为100%、特异度为95.5.9%,ROC=O.977(P=O.003)。结论在本实验室成功建立了IRE试验的试验方法。IRE试验可以作为Draize试验的一种替代方法,尤其适用于无刺激性和强刺激性受试物的检测。
