Stress has been found an important role in modulating behaviors. It is believed that acute and chronic stress have positive and negative effects on learning, respectively, through non-genetic and genetic pathways. However, the effect and mechanism of stress between several hours(acute) and several weeks(chronic) are not very clear. Here, we reported a special kind of stress, repeat swimming stress(RSS), which lasted for 5 days and had unique effects on learning and synaptic transmission. We found that after RSS associative learning in mice was impaired but their spatial learning and associative memory were intact. Pain threshold and locomotion activity were also changed by RSS. Staining results showed that various areas like m PFC, PVN and LC were activated during RSS. Our intraperitoneal injection experiments revealed that norepinephrine receptor antagonist could rescue learning impairment following RSS. Optogenetics results showed that repeat light stimulation on terminal of LC neurons that projected to mPFC could also block associative learning. Slice recording results showed t-LTP in m PFC was impaired following RSS. These findings suggested that RSS might become a potential model to study stress-induced learning disorder which might be caused through hyperactivity and alterative neural transmission of mPFC.

Alzheimer's disease(AD) is an age-related neurodegenerative disorder, and characterized by chronic progressive deterioration of cognition, learning and memory. The distinctive pathological changes in AD are aggregation and deposition of amyloid β-protein(Aβ) and tau protein. Various surveys have found that AD patients suffered from circadian rhythm disorders, concluding irregular sleep-wake rhythm, excessive daytime sleepiness, nighttime restlessness, difficulty falling asleep, fragmented sleep, which affects patient's quality of life more seriously. In addition, the circadian rhythm disorder also aggravate the deteriorating effects of AD on cognition, learning and memory. Unfortunately, there is lack of effective therapeutic regimen for circadian rhythm disorder in AD patients. Several studies have found some astonishing similarities between T2 DM and AD. Exendin-4 is a new listed therapeutic strategy for T2 DM, and it may have great potential for AD. The purpose of the study is to explore and describe the disruptions in circadian rhythm induced by Aβ31-35 which can be ameliorated by Exendin-4, and investigate the possible molecular mechanism. Using Vital View software to record the locomotor activity of C57BL/6J mouse, the results showed that intra-hippocampal injection of Aβ31-35 led to the circadian rhythm abnormalities in C57BL/6J mouse, and the abnormalities can partially recover by pretreatment with Exendin-4. Furthermore, the results indicated that CLOCK and BMAL1, the core clock protein, express rhythmically in hippocampal region of normal mice, nevertheless, the expressions of CLOCK and BMAL1 are arrhythmic due to the hippocampal injection of Aβ31-35, which are partially corrected by Exendin-4. In summary, Exendin-4 can exert neuroprotective effect and recovery rhythmic expression of circadianclock protein in hippocampus of Aβ mouse models, which provide experimental evidence for novel therapeutic strategies for AD.

The parieto-insular vestibular cortex(PIVC) shows directionally-selective responses to vestibular stimuli. To explore whether PIVC plays a role in heading perception based on vestibular signals, we trained macaques to report their perceived direction of self-motion in a forced-choice heading discrimination task, and quantified neuronal sensitivity with ROC analysis based on vestibular signals, and compared with previous studies in area MSTd and VIP. Spike counts were computed in two time windows, roughly corresponding to the times of peak acceleration/deceleration. The most sensitive PIVC neurons had thresholds comparable to behavior, but the modal neuron was much less sensitive than the monkey. The average neuronal threshold in PIVC was not significantly different from that for area MSTd and VIP. PIVC neurons also exhibited significant choice probabilities(CPs), which characterize the trial-to-trial covariation between neural responses and the animal's choices. Early in the trial(acceleration phase), average CP in PIVC was smaller than in MSTd and VIP. However, CP was substantially larger in PIVC than MSTd but smaller than VIP late in the trial. Most l Larger CPs were observed in more anterior portions of PIVC. To test for a causal link between PIVC and vestibular heading discrimination, muscimol injections were used to reversibly inactivate either anterior or posterior PIVC. Bilateral inactivation of anterior PIVC resulted in substantial deficits, whereas inactivation of posterior PIVC had weaker effects. Together, our findings suggest that PIVC(especially the anterior portion) plays a functional role in heading perception based on vestibular signals.

Objective. The enteric nervous system(ENS) control the function of gastrointestinal tract, such as gastrointestinal motility. In Parkinson's disease(PD), in addition to the typical motor symptoms, attentions have been drawn to a number of non-motor symptoms, such as constipation, implying dysfunction of the ENS. Studies have shown that Lewy bodies(LBs), the neuropathological hallmarks of PD, in which α-synuclein is present as its primary protein component, appear in ENS prior to the CNS. It has been suggested that α-synuclein aggregates are generated in the ENS and then retrogradely transported to the CNS via a prion-like mechanism. In the present study, we aim to characterize the age-dependent morphological alterations and aggregation of α-synuclein in the ENS in the gastrointestinal tract of a α-synuclein transgenic mouse model. Materials and Methods We collected gastrointestinal tracts from the stomach to the colon at ages of one week, 1, 3, 6, 9, 18 and 24 months of BAC-α-synuclein-GFP mice after transcardial perfusion with 4% paraformaldehyde in 0.1 M PBS. The small intestines were first studied. Transverse sections were cut with a cryostat(14 μm thick), The native GFP(reflecting α-synuclein transgene expression) and immunohistochemically stained samples were examined with a confocal microscope. Results 1) With age, expression and accumulation of α-synuclein–GFP increase gradually in the submucosal plexus and myenteric plexus. 2) α-synuclein appears highly overlapped with different neuropeptides, such as VIP, n NOS, and Calretinin. n NOS and Calretinin levels increased with age, but not to VIP. 3) The S129-phospho-α-synuclein level was increased with age in transgenic mice, in contrast tothe wildtype mice, reflecting pathological changes of α-synuclein. Conclusion The age-dependent changes and phosphorylation of α-synuclein in the small intestine of BAC-α-synuclein-GFP transgenic mice can provide us a unique model to study the relationship between ENS and PD pa

A network perspective to understanding the relationship between human brain and behavior has attracted more and more attention recently. There are accumulated evidences to show that the human brain is organized of functional specialized networks. Different networks may be related with special cognitive function and human behavior. Using the networks perspective to describe the functional diversity of human brain could offer a different aspect about the distribution structure of human brain in large scale. Resting-state fMRI is a popular approach used to detect the intrinsic connectivity networks which are also rhythmically showed up in the load state. The independent component analysis is widely adopted to extract the various intrinsic connectivity networks in resting-state. Using ICA approach, we separated twenty components based on resting-state f MRI data which were from Human Connectome Project(100 subjects). After manually choosing the noise components, eighteen components was recognized as significant networks. Based on the participation in different intrinsic connectivity networks, we firstly defined the brain regions which homogeneously participated in different networks using the Components Homogeneity(Co Ho) index. Then, we used the functional diversity index, which measured the anisotropic participation, in the brain regions which possessed the high Co Ho value. High Co Ho value in brain region determined that this brain region homogeneously participated in different networks, which could be viewed as a functional consistent area. Further, using the functional diversity index, we found that the primary cortex including visual cortex and motor area obtained low diversity. Meanwhile, some association cortex significantly participated in different networks, which included the precuneus, inferior parietal lobule, superior frontal gyrus medial orbital. Postpartum depression(PPD) is one of the mental disorders that affect mothers and their babies. Currently the mechanism of postpartum depression remains unknown. The Diagnostic and Statistical Manual of Mental Disorders Fifth Edition(DSM-5) defines PPD as the depressive episode that begins 4 weeks after delivery. The effect factors may include the sudden drop of estrogen, dysfunction of HPA or HPT axis and so on. So far, the estrogen withdraw theory is the most generally accepted one. Clinical data has shown that there's a strong correlation between estrogen level and depressive mood during postpartum period. It has been reported that estrogen can regulate the expression of neuropeptide galanin. Meanwhile, changes in expression of galanin and its receptors in depression models have been demonstrated, indicating that galanin may be involved in depression pathogenesis. The aim of the present study is to replicate the rat ovarian-steroid withdrawal PPD model to obtain a more thorough understanding of PPD pathogenesis by analyzing the plasma galanin level and expression of galanin and its receptors in different brain regions in rat postpartum depression model at both m RNA and protein level. The results showed that the plasma galanin level was increased in estrogen-treated group. Q-PCR results showed that galanin m RNA level was increased in pre-frontal cortex(PFC) and adrenal pituitary in estrogen-treated group. Besides, PPD group had higher Gal R1 m RNA level in pre-frontal cortex than other groups. Then si RNA-Gal R1 interference was used to further study the effect of Gal R1 in the PFC in this model. The anti-depressive effect was observed after the administration of si RNA-Gal R1 in PFC in PPD group. These results indicated that the galanin expression changed in PPD and Gal R1 in PFC might be involved in the pathogenesis of PPD.

Learning from others in a group of individual has a distinct evolutionary advantage. Many fish, such as zebra fish, live in groups of individuals and are likely to have evolved means to communicate knowledge between the individuals of the group. However, how learned knowledge is communicated and transferred behaviorally between members of a group is still largely unknown. Here we take the first steps to investigate the learning of visual stimuli and transfer of knowledge in small groups of adult zebra fish. To this end, we have developed a novel tracking system to accurately track every individual fish over a long period of time in an online manner. Similar to the tracking method of [1], our method ensures that individual fish tracks do not intermix individuals by including an fish-recognition system. The newly implemented fish recognition system, however, uses different features than [1]-namely rotated and de-bended color-images of individual fish refined by finding local MSER regions(Maximal stable extremal regions) –and a multivariate Gaussian approach for classification. Moreover, the classifier is updated in anon-line manner, and does not need multiple swipes through a recorded video as in[1], so that online feedback experiments are possible in principle. Here, we first evaluate our new tracking system and compare its accuracy to the method of [1] and found that our implementation was at least as accurate as the approach of[1] in our experiments using a small group of zebra fish. Using this tracking system, we further investigated the adaptation behavior of a group of 3 individuals towards are petitive visual stimulus as a first step towards studying group learning. Testing various measures for describing the behavior, such as relative speed and acceleration, we found that visual flashes indeed changed the statistics of the movement tracks of the individual fish during and short after the flash stimuli in respect to a controlperiod, indicating successful behavi

Objective: Deep brain stimulation(DBS) is a neural modulation therapy which uses electrodes to deliver electrical stimulation onto specific nuclei or brain areas in human deep brain with the purpose of treating or relieving disorders in the central neural systems. Subthalamic nucleus(STN) DBS has been successfully introduced in remedy of movement disorders such as Parkinson's disease(PD), dystonia, tremor, and also psychiatric disease such as obsessive-compulsive disorders, etc.. Increasing evidence suggests that the neural oscillations with different oscillatory frequency bands and patterns in local field potentials(LFPs), which recorded from the STN of patients with PD, have characteristic distributions in and around the STN and related to different pathological symptoms, therapies, and functional events and tasks. However, the distributions of subthalamic neural oscillations and the modulations by movement and medication is less clear. This study aims to develop an intraoperative targeting method using movement related LFP oscillations with the 3-D visualized MRI image of the STN and quantify the oscillatory and dynamic characteristics of LFPs inside STN under the conditions of ON and OFF medication in PD. Methods: STN LFPs were recorded during auditory cued clicking motor task in twelve patients with Parkinson's disease. The movement related neural oscillations were extracted using time-frequency analysis and statistical analysis. Subsequently, the spatial distribution of the dynamic synchronization or desynchronization features was mapped onto the 3-D visualized MRI images which immerged and reconstructed with pre-operative and post-operative MRI images of each relevant patient. In this case, contacts located inside STN were confirmed. Furthermore, resting LFPs on the states of ON/OFF medication were respectively recorded from STN and analyzed with power spectral density, short-time Fourier transform and coefficient of variation(CV) to differentiate the oscilla

Background: Electroconvulsive therapy(ECT) is an important treatment option for depression in clinical practice. Compared to antidepressant drugs, ECT has a more rapid onset and is also effective for treatment-resistant depression patients. But ECT could cause cognitive impairment and other side effects, which limit its promotion and application. However, little is known about the underlying therapeutic mechanisms of ECT, even though ECT has been applied in clinical practice for over 70 years. Objective: A lot of studies highlights the role of amygdala-prefrontal circuit in emotion regulation, and current opinion holds that amygdala-prefrontal circuit underlying emotion processing are abnormal in depression. The aim of this study was to observe the change of functional connectivity between amygdala and prefrontal cortex in depression patients before and after ECT, and explore the role of amygdala-prefrontal circuit in therapeutic mechanism of ECT, with the method of resting-state functional magnetic resonance imaging(rs-f MRI). Methods: We set the bilateral amygdale as the seeds respectively, and observe the change of functional connectivity between amygdala and prefrontal cortex in 18 depression patients before and after ECT, compared with 20 healthy controls. Results: The results showed that, compared with depression patients before ECT, the functional connectivity between left amgydala and left lateral frontopolar cortex(BA 10), left amgydala and right lateral frontopolar cortex(BA 10), right amygdala and right medial prefrontal cortex(BA 10) were significantly increased in depression patients after ECT. Compared with healthy controls, the functional connectivity between left amgydala and left lateral frontopolar cortex(BA 10), left amgydala and right lateral frontopolar cortex(BA 10), right amygdala and right medial prefrontal cortex(BA 10) were significantly lower in depression patients before ECT. Objective Empathy is an ability to share the feelings of others and plays a fundamental role in the social life of humans. Balanced activation of brain areas that mediate empathy is required for motivating appropriate social behaviors. Our previous research has found that the medial prefrontal cortex(mP FC) played a pivotal role in the empathic process in rats. The present study aims to investigate the roles of the m PFC in pro-social ***(1) A rat was allowed to interact freely within a transparent plastic box for 30 minutes with a cagemate in pain induced by s.c. bee venom(BV) injection and was designated as cagemate observer(CO). 1) Mechanical pain sensitivity of the CO rats that received sham or lesion surgery of the mP FC was measured in isolation and compared to baseline mechanical pain sensitivity measured before the treatments;2) The effects of mP FC lesion on the emotional alterations of CO rats were measured with the open-field test.(2) The effects of m PFC lesion on the social care or social approach to pain were measured with a modified three chamber paradigm. A pair of cagemates was put into cages placed at the two ends of the three-chambered box;one of the rats received a s.c. BV injection before being put into the cage and the other rat was untreated. The social care behavior was presumed if the test rat spent more time in the chamber with its cagemate suffering from pain induced by the BV injection.(3)The effects of mPFC lesion on sociabilityamong cagemates were measured withthe three chamber test. Results The present study showedthat m PFC lesions diminished the bilateral mechanical hypersensitivity in the CO rats. Bilateral lesions of the m PFC eliminated both the self-experienced pain inducedanxiety-like behaviors, which are induced by BV injection, and the vicarious experienced anxiety-like emotional responses in the COrats, which are induced by social interaction with a cagemate in pain.
