Polymorphisms of porcine ob intron 2 and exon 3 among different breeds including Min pig, Wuzhishan pig, Zang pig, E'xi Black pig, Licha Black pig, Yangxin pig, Laiwu pig, were analyzed by PCR-SSCP in the study. G1109T, G1112A, G1184A mutation in intron 2 and G3717T, G3787T, G3862A mutation in exon 3 were found in this study. Min pig, Licha black pig, Wuzhishan pig and Zang pig with high PIC(0.63,0.58,0.56,0.54) in intron 2 and (0.37,0.38,0.37,0.35) in exon 3 should be reserved.

MC5R plays an important role on energy balance regulation. There is an A/G mutation resulting in Ala 109 Thr variation. BsaH I-PCR-RFLP was used to to analyze genotypes of MC5R gene on Large White, Landerance, Large White×Landerance (LW×L) and Landerance×Large White (L×LW). e genetic diversity and genetic structure of 30 Chinese sheep breeds were investigated by using 27 microsatellite markers. Average expected heterozygosity and observed heterozygosiy of all breeds were 0.799 and 0.531 irrespectively. An overall genetic differentiation (Fst) across all loci was 0.079 (P < 0.001), which indicated roughly 7.9% of genetic diversity could be explained by genetic variation between populations. The neighbour-jioning phylogenetic tree constructed based on DA genetic distance indicated 30 Chinese sheep breeds were clustered into 5 main groups, which throw new sight on Chinese sheep classification. Scatter plots of estimates from principal component analysis showed support the result of phylogenetic cluster partly. Weitzman's approach was applied to make the conservation priority, and the result suggested that the optimum conservation strategy is to give priority to those breeds with the highest conservation *** than to the most endangered breeds; Tengchong (TC) sheep and Jiaxian (JX) sheep were considered to be given the higher priority.
