目的:建立甲基苯丙胺依赖大鼠戒断模型,观察甲基苯丙胺戒断大鼠前额叶皮层(PFC)、伏隔核(NAc)、杏仁核(Amy)及海马(Hip)等4个脑区的PER1及多巴胺受体2(DRD2)的表达情况。方法:28只200-250 g Spraguea-Dawley雄性大鼠,在12h/12h昼夜环境下饲养1周后随机分成两组,甲基苯丙胺戒断组大鼠隔天(ZT4)交替腹腔给予甲基苯丙胺(3 mg/kg)和生理盐水(1 ml/kg),生理盐水组大鼠连续腹腔给予生理盐水(1 ml/kg),注射后立即将大鼠置于CPP箱中45分钟进行训练,共14天。第14天进行CPP测试,选择依赖的大鼠戒断10天,戒断第10天ZT4-ZT5时间段取相关脑区,用蛋白质免疫印迹技术测定PER1和DRD2。结果:甲基苯丙胺组大鼠条件位置偏爱值(CPP)明显高于生理盐水组(P<0.05);与对照组相比,甲基苯丙胺组大鼠中的PER1在前额叶皮层、伏隔核的表达量显著升高,在杏仁核和海马中表达无显著变化;甲基苯丙胺戒断大鼠DRD2在前额叶皮层、伏隔核、杏仁核以及海马表达量较对照组表达量均无明显变化。结论:(1)采用多次腹腔注射甲基苯丙胺训练CPP,成功建立甲基苯丙胺依赖大鼠模型;(2)甲基苯丙胺依赖大鼠戒断后PFC和NAc的PER1表达升高,DRD2无变化;提示PER1可能参与甲基苯丙胺依赖大鼠戒断行为,而DRD2不起关键作用。

In humans,substances use during adolescence increases the risk of psychiatric disorders and drug addiction in *** represents a critical period for maturation of the ***,adolescent brain is more sensitive to neurobiological changes induced by internal or external stimuli,such as drug *** cortex(PFC) and hippocampus(HP) play important roles in cognition,emotion and decision making,and have been highly implicated in development of addiction and mental *** present study is to investigate the morphological,neurochemical and functional changes in adult PFC and HP that may be induced by adolescent cocaine ***,the impaired responses to stress in adult animals by an adolescent cocaine-exposed history have been also examined using behavioral tests and *** preliminary results suggest that substance use during early life may lead to deficits in the development and function of brain,and ultimately contribute to the abnormal behaviors in the later life.
