OBJECTIVE—Lower quantity and quality of high density lipoprotein(HDL) are important characteristics of type 2 diabetes *** HDL infusion results in obvious fall of plasma glucose in *** study aims to investigate the underlying mechanisms mediating HDL actions on glucose metabolism,and the therapeutic effects of long-term HDL infusion on diabetic db/db mice. REASEARCH DESIGN AND METHODS—Actions of HDL on glucose uptake and glucose transporter 4(GLUT4) translocation were assessed with l-[H]-2-deoxyglucose and plasma membrane lawn,respectively,in 3T3-L1 *** analysis was performed with amyloglucosidase both in vitro and in *** phenotypes of db/db mice with long-term HDL infusion were monitored by analyzing metabolism parameters,adipose tissue function,inflammatory cytokines and oxidative-stress indexes. Corresponding signaling molecules were detected by western blot. RESULTS—HDL had a great influence on glucose *** stimulated glucose uptake in 3T3-L1 adipocytes in a time- and concentration-dependent manner with maximal effects at about 20 min (50μg protein/ml),and apoA-I is one of the main components to mediate this action through PI3K/Akt and eNOS associated ***4 translocation was also significantly increased,the net amount of GLUT4 in plasma membrane were significantly resulted from increase in exocytosis through AKT and AMPKa pathway,and also decrease in endocytosis mediated by *** deposition got enhanced by HDL in L6 cells and gastrocnemius muscle,paralleling with elevated GSK3 phosphorylation. Meanwhile,increased Akt-Ser473 and AMPKa phosphorylation were detected in HDL-treated adipocytes and gastrocnemius ***,HDL was found to change the phenotypes of db/db mice. Long-term HDL infusion could pronouncedly reduced blood glucose,and improve insulin sensitivity and inhibit pancreatic islets proliferation with interesting insulin secretion enhancement in rodent models with obesity a

Objective TanshinoneⅡ-A(Tan),a bioactive diterpene isolated from Salvia miltiorrhiza Bunge (Danshen),possesses anti-oxidant and anti-inflammatory *** present study investigated whether Tan can reduce and stabilize atherosclerotic plaques in Apolipoprotein-E knockout(ApoE KO) mice maintained on a high cholesterol diet(HCD). Methods and Results Six week-old mice challenged with a HCD were randomly assigned to 4 groups:(a) C57BL/6J;(b) ApoE KO;(c) Tan-30(30mg/kg/d);(d) Tan-10(10mg/kg/d). After 16 weeks of intervention,Tan treated mice showed decreased atherosclerotic lesion size in the aortic sinus and en face ***,immunohistochemical analysis revealed that Tan rendered the lesion composition a more stable phenotype as evidenced by decreased macrophage infiltration and increased smooth muscle cell and collagen *** also significantly reduced in situ superoxide anion production, aortic expression of NF-KappaB,and matrix metalloproteinase-9(MMP-9).In vitro treatment of murine macrophages with Tan significantly suppressed oxidized LDL induced reactive oxygen species(ROS) production and pro-inflammatory cytokines(IL-6,TNF-a,MCP-1) expression.
