Tylosin is a macrolide antibiotic which was widely used in the prevention and treatment of livestock and poultry respiratory tract,intestinal,reproductive tract infection caused by mycoplasma,gram positive bacteria,spirochetes and other *** tylosin is obtained by acetylation of C-3 and isovalerylation of C-4" in tylosin through microbial transformation,which is known to be effective for preventing the growth of tylosin-resistant *** this paper,a series of aminated derivatives were prepared by reductive amination of C-20-aldehyde group of acetylisovalentyl tylosin,and the synthesis route for the target compound was depicted in Scheme *** the synthesized compounds were confirmed by H,C-NMR and *** activity were also preliminary evaluation,and compounds 18 i and 18 j with MIC of 0.146 μg/ml and 0.175 μg/ml,respectively.

Current enzymatically activated nitric oxide（NO） donors could inevitably produce off-target effects because those enzymes are overexpressed rather than exclusively expressed in cancer cells. Inspired by the bioorthogonal chemistry-based prodrug strategy, a class of O-alky1 caged diazeniumdiolates were designed and synthesized as new bioorthogonal NO precursors（Figure 1）,which are metabolically stable in plasma while can be effectively activated in the presence of the biocompatible Pd（dba）2 catalyst via bioorthogonal chemistry reactions to liberate NO,exhibiting potent anti-proliferative activity against cancer ***,the Pd-labile NO precursors would serve to expand the therapeutic potential of NO-based anticancer agents and the scope of bioorthogonal chemistry utility in the near future.

Sulfonamides have a series of biological activities such as antibacterial,antiviral,anti-inflammatory,antitumor and so *** protein inhibitor 1 developed by Eisai Corporation of Japan showed potent antitumor activity against a variety of transplantations tumors including colon cancer,non-small cell lung cancer,breast cancer,and pancreatic *** 1,2,3-triazole ring has a large dipole distance and hydrogen bonding ability,linking to other active fragments as a Linker group to synthesize novel biologically active *** our group,a series of sulfonamide-1,2,3-triazole derivatives were synthesized through click *** chemical strructures were identified by H-NMR,C-NMR,and mass *** antiproliferative activity of PC-3,MGC-803,and EC-109 cells was evaluated by MTT assay.

The natural product artemisinin have opened a new paradigm for antimalarial drug discovery,and an increasing number of artemisinin analogues（artemalogues） have been explored for activities of inducing ***,we infused the skeleton of artemisinin to venetoclax（2）,an inhibitor of antiapoptotic protein Bcl-2 approved by FDA,purposing for synergetic apoptosis effect（Figure 1）.A series of novel artemalogues were prepared for replacing the hydrophobic fragment l-chloro-4-（2,4,4-trimethylcyclohexyl）benzene of 2,and all compounds hold improved selectivity for Bcl-2/Bcl-xl comparing to *** compounds showed elevated activity against Bcl-2 protein and moderate potency against Bcl-2-dependent acute lymphoblastic leukemia（ALL） cell line RS4;11 with no hERG ***,this work provides a series of novel selective Bcl-2 inhibitors by infusing the skeleton of artemisinin.

Since 1960 s,[1,2]researchers have been working on research coelenterazine,novel substrates and luciferases are gradually *** them,Renilla luciferase system is widely *** to firefly luciferin luciferase and the bioluminescent bacterial system,coelenterazine-luciferase system is more simple and without other cofactor except for oxygen.[3]Coelenterazine is combined to O at the C-2 position of the imidazopyrazinone core,then peroxide is formed ***,peroxide forms a four-membered ring,at the same time,the amide anion of coelenteramide in its excited state,releasing *** emission of light is occurred during coelenteramide transition from excited state to ground ***,Hydrogen sulfide participates in various physiological and pathological process as a signal molecular.[4]Thence,developing efficient,accurate and convenient tool for understanding molecular processes is ***,we design and synthesize a new type of H S probes based on Renilla luciferase *** have assessed their sensitivity and selectivity in vitro and in *** probes show good biocompatibility and have potential for development.

[Pt（L）(DMSO)Cl]（1） and[Pt（L2）(DMSO)Cl]（2） with 9-[3-pyrrolidinopropionamido]/[3-（4-methyl）-pyrrolidinopropionamido]-oxoisoaporphine（H-L） were synthesised,respectively,a nd the two new platinum（II） complexes were more selective for MGC80-3 tumor cells versus normal HL-7702 cells,experiments on the MGC80-3 cells showed that comple×1 caused cell cycle arrest at the S ***,cytotoxicity mechanismstudi es suggested that complex 1-induced apoptosis was achieved via disruption of the mit ochondrial function,which led to a significant loss of the △ψ1,an increase in the cell ular levels of reactive oxygen species,and a fluctuation of the intracellular Ca conce *** altogether,complex 1 can trigger cancer cell death by inducing apopto sis through a mitochondrial dysfunction pathway.

It has been reported that 2-ME plays an anti-angiogenesis role via inhibiting the proliferation of vascular endothelial *** was selected from a series of derivatives of2-methoxyestradiol（2-ME）.In this study,we evaluated both its activity in anti-angiogenesis and effect on tumor xenografts in nude *** anti-angiogenesis induced by GHY was determined using chicken embryo membrane assay,wound healing assay and transwell assay in HUVEC and MCF-7 ***-angiogenesis via VEGF pathways were confirmed by detecting the regulation of VEGFR,SHC,and *** angiogenesis is critical to the growth and reproduction of cancer cells,we considered that GHY may play an important role in anti-tumor *** further demonstrated that GHY suppression the migration of MCF-7 via affecting the EMT *** injection of GHY showed obvious inhibition of tumor growth in the xenograft *** serum analysis and Histological analysis demonstrated no adverse effects on major organs in nude *** studies provide support for new drug development These studies provide support for new drug development.

As an important pattern recognition receptor of human innate immune,cyclic guanosine adenosine monophosphate synthase(cGAS) can recognize the abnormal change of cytosol nucleic acids,synthesize cyclic guanosine adenosine monophosphate(cGAMP).cOAMP binds with stimulator of interferon genes(STING) to activate STING-TBK1-IRF3 signal pathway,induce IFN I production and prime adapted immune *** in vivo instability and low transmembrane activity resulted from the phosphodiester bonds in cGAMP are the obstacles for its clinical *** structure-activity relationship(SAR) of cGAMP is not available in *** this report,cGAMP analogues were designed and synthesized with the help of molecular docking and molecular mechanic *** bioactivity of the analogues was evaluated using hSTING protein,human and mouse immune cells in order to establish SAR,and find compounds with good in vivo stability,transmembrane ability and antitumor immunotherapeutic *** compounds could be used as the molecular probes for the research of human innate immune mechanism,and as lead of new antitumor immunotherapeutic drugs.

Compared with the parent β-cyclodextrin（β-CD）,the synergistic interaction of double β-CD cavities have a better chiral chromatographic performance owing to the expanded spatial recognition domain,wider analysis range,higher resolution and shorter analysis time.A novel β-CD derivative — 2-biselenium bridged bis（β-CD）s（2-diSeCD） was synthesized and chemically immobilized onto the surface of silica *** evaluate the enantioseparation ability of the column,investigate the enantiomers of three drugs（guanoxan sulfate,metoprolol and hydroxyzine） and optimize the chromatographic conditions（including the proportion of methanol,flow rate and column temperature）.The results demonstrated that three of thess racemates were all successfully baseline enantioseparated（Rs>1.5）.Therefore,the chiral stationary phase（CSP） of 2-diSeCD bonded on silica gel exhibited excellent separation capability for several chiral compounds in high-performance liquid chromatography（HPLC）.It is expected that the β-CD phase will be widely used in the field of chiral compounds' separation.

Influenza virus infection is still a major threat to public *** the emergence of drug-resistant of influenza viruses highlight the urgent need of new antiviral drugs with a different mode of action by targeting a critical step in the viral life *** acid（GA）,a well-known pentacyclic triterpene obtained from the hydrolysis of glycyrrhizin,has been shown to have good anti-influenza virus *** continue our studies towards the development of novel antiviral inhibitors based on natural pentacyclic triterpenes, a series of water-soluble β-cyclodextrin（CD）-GA conjugates with a flexible linker at the primary face of β-CD have been designed and *** primary antiviral activity screening showed that one compound has high inhibition rate at concentration of 50μM based on a cytopathic effect（CPE） reduction assay while no apparent cytotoxicity against uninfected MDCK cells was observed at a concentration of 100μ*** study supports that these conjugates are potential candidates for further evaluation as anti-influenza agents.
