Ru（Ⅱ） complexes have long been extensively studied as potential inhibitors against the proliferation and migration of tumor ***,a polypyridine ruthenium（Ⅱ） complexes have been ***'s results that compound exhibited promising observably inhibitory activity against the growth of MDA-MB-231 breast cancer *** further studies showed that compound can inhibit the migration and invasion of MDA-MB-231 cells,as well as the formation of angiogenesis in ***,it's revealed that compound can inhibit the formation of invadopodia of MDA-MB-231 cells through apoptosis via DNA-damage ***,In the tumor zebrafish model,the compounds significantly inhibited the metastasis of MDA-MB-231 cells in *** summary,compound may be developed as a potent anticancer in the near future.

G proteins act as molecular switches in a number of G protein-coupled receptor signaling pathways,and are key mediators for numerous of important physiological *** cyclic depsipeptide YM-254890,together with its structural similar depsipeptide FR900359,are the only known selective inhibitors of the Gq subfamily of G *** have recently reported the first total synthesis of YM-254890 and FR900359,followed by preparing analogues to perform the structure-activity relationship *** we reported the first systematic structure-activity relationship study towards the N-methyldehydroalanine motif in YM-254890,by designing and synthesizing new *** characterization of the seven compounds at Gq-,Giand Gs-mediated signaling showedsimplified analogue YM-19 was the most potent Gq inhibitor,while the other analogues had no obvious inhibition on Gq *** study provides information for future designing of potent and simplified YM-254890 analogues.

A series of Meridianin derivatives with glycogen synthase kinase-3（GSK-3） inhibitors against type Ⅱ diabetes was subjected to three-dimensional quantitative structure-activity relationship（3 D-QSAR） studies using the comparative molecular field analysis（CoMFA） approach（Figure 1）.The most potent compound 1 was used as the template to align all the *** a result,the best predictive model was obtained with CoMFA including both the steric and electrostatic force fields（q=0.532,r=0.972）.This model was validated by predicting the training set of 41 compounds with the satisfactory predictive r value of *** model could provide the guide for further designing of the potential inhibitors against glycogen synthase kinase-3 for treating type Ⅱ diabetes.

Recently,high-throughput phenotypic screen in zebrafish embryos provides unique advantages in the early drug discovery,because zebrafish embryos are genetically tractable,easy to manipulate,and can be generated in large *** addition,zebrafish as a complex vertebrate with high genetic homology to mammals have also gained increasing acceptance as a preliminary/complementary approach to lead compound discovery via phenotypic *** screens have expanded beyond the use of developmental phenotypes to include behavioral,cardiac,metabolic,proliferative and regenerative ***,many creative strategies have been used to discover the mechanisms of action of new small molecules including chemical phenocopy,genetic phenocopy,mutant rescue,and spatial localization ***,zebrafish emerged as a leading model of in vivo phenotypic screens,and efficient method to identify lead compounds that modulate specific biological processes.

Matrix metalloproteinases（MMPs） have been implicated in virtually all aspects of cancer *** particular,MMP-2 and MMP-9 have been reported to be crucial for tumor angiogenesis and the formation of metastasis,thus becoming attractive targets for cancer *** evidence suggests that the upregulation of nitric oxide（NO） formation from NO donors can decrease MMPs levels. Our group have recently endeavored to identify arylsulfonamide pyrrolidine hydroxamates as potent MMPs inhibitors. Inspired by the theory of "multifunctional drugs",we thereof designed and synthesized a novel series of MMPs inhibitors containing furoxan as the NO donor, which is integrated at the C-4 position of the pyrrolidine *** these hybrid compounds exhibit additive antitumor activity via MMP inhibition and NO release and might offer a prospective avenue to discover novel anti-cancer drugs.

Inflammatory cells can be accumulated to local tissues to cause inflammation by C-X-C chemokine receptor type 4(CXCR4)/C-X-C chemokine ligand 12(CXCL12) ***,developing CXCR4 inhibitors is a desirable strategy to control and treat *** shown in Fig.1,the key pharmacophores of compounds AMD3100,Q-122,and amidesulfamide derivatives were incorporated into a single structure to develop CXCR4 inhibitors with more potent anti-inflammatory activity and improved pharmacokinetic *** 7 a demonstrated the best anti-inflammatory activity,effectively blocking the aggregation of inflammatory cells to the site of *** results confirmed that developing CXCR4 modulators to control and reduce inflammation is a feasible and effective strategy for the treatment of inflammation.

Efficacy and safety of drug therapy are two fundamental issues of great *** known,the impurities are studied to contribute to the improvement of drug safety.(S)-Pantoprazole as one of the chiral proton pump inhibitors with high biological activity and uses in the clinical treatment of gastric ulcer,duodenal ulcer,reflux esophagitis and peptic system *** to the literatures,more than six impurities in(S)-pantoprazole were identified,only synthesis of impurities A,B,C,F and G were *** this article,nine target impurities including impurities A,B,C,F and G were synthesized,and the detailed synthetic procedures of impurities D,E,H and I have not been reported *** the target impurities were characterized by IR,MS,NMR and the corresponding purities have been measured by HPLC.

As a gene associated with anemia,the erythropoiesis gene is physiologically expressed under hypoxia regulated by hypoxia-inducing factor-α（HIF-α）.Thus,stabilizing HIF-α is a potent strategy to stimulate the expression and secretion of *** this study we applied click chemistry to the discovery of HIF prolyl hydroxylase 2（HIF-PHD2） inhibitors for the first time and a series of triazole compounds showed preferable inhibitory activity in fluorescence polarization *** particular note was the orally active MF-PHD inhibitor DDO-3136(IC = 62.23 nM),which was almost ten times more active than the phase III drug FG-4592(IC = 591.4 nM).Furthermore,it can upregulate the hemoglobin of cisplatin induced anemia mice（120 g/L） to normal levels（160 g/L） with no apparent toxicity observed in *** results confirm that triazole compound DDO-3136 is a promising candidate for the treatment of renal anemia.

Osteoarthritis(OA) accounts for more than 50%of elder people,the estimated incidence of OA is67 million which will be the most common form of arthritis in United States in *** pathophysiologic progression of OA remains *** analyzed the grycosaminoglycan(GAG)and chondroitin sulfate(CS) composition between remote and lesion *** GAG quantity in lesion group was lower than in the remote *** possessed the major content among the GAG ***,only CS-A and total CS showed significantly decrease in the di-saccharide analysis of CS by *** had a significant inverse correlation with the CS-A concentration in lesion cartilage,but CS-0,CS-C and total CS not The microstructure analysis of cartilages indicated that CS-A,CS-C and total CS gradually increased from superficial to deep *** the lesion cartilages,the concentration of CS-A and total CS were significantly reduced,which can be benchmarks of degraded cartilage for cartilage regeneration *** expression of CS catabolic and anabolic enzyme may clarify the key point of OA progression.

Histone deacetylase（HDAC） inhibitors have emerged as a potential therapeutic approach for the treatment of various human diseases including *** on our previous work we designed and synthesized 33 hydantoin derivatives as HDAC *** activity evaluation results suggest that some compounds showed good inhibitory activity of HDACs and that compounds 13 a and 13 c exhibited an obvious advantage in the inhibitory activity of HDACs,an antiproliferative activity in vitro compared to the vorinostat（SAHA） positive ***,target compounds 13 a and 13 c also showed a strong ability to induce apoptosis at 0.8 μ***,target compound 13 a exhibited preferable inhibition for HDAC6 compared to HDAC1,4 and 8.
