Immune escape has become a leading challenge in the immunotherapy,with the phenomenon of immune system lacking ability to eliminate transformed cells prior to and after tumor *** 2,3-dioxygenase 1（IDO1） plays an important role in the immune escape,which can catalyze L-tryptophan to N-formylkynurenine through the kynurenine ***,the discovery of more potent and specific IDOl inhibitors has become a research hotspot4-Aryl-1,2,3-triazoles were discovered in 2010,with parent compound displaying an enzymatic IC value of 60 μM and passing all structural *** on that,we designed and synthesised of novel 1,2,3-triazoles and *** all compounds,the compounds with chlorine and/or piperazinyl have shown the most potent inhibiting activity of IDO1,one of which has displayed enzymatic and cellular IC values of 1.7 μM and 5 μ*** novel scaffolds exhibit better specificity,which could be used as a starting point for further development of IDO1 inhibitors discovery with improved potency and selectivity.

How cells communicate during development and regeneration is a critical *** mechanism of intercellular communication is via exosomes,Exosomes are small vesicles derived from the intraluminal membrane budding of multivesicular bodies with a size ranging from 40 to 150 run and are constitutively released into the extracellular environment through fusion with the plasma *** have reported that LSD1 can promote the development of gastric *** this study,we are interested in whether LSD1 can be transfered between gastric cancer cells through exosomes,and whether transferred LSD1 will affect target ***,we do detecte LSDl in the exosomes come from MGC-803 by *** means LSD1 can be secreted into *** addition,exosomes can carry LSD1 to the target cells（MKN-45）,accompanied with corresponding increase of sternness-related markers in the target cell,while there is no increase when MKN-45 cultured with LSD1-knockout cell's ***,compared with control group and LSD1-knockout group,recipient cells,MKN-45,treated with exosomes from MGC-803-cells showed more spheres grown in 3 D cell culture ***,our data supported the view that exosomal LSD1 mediates sternness in gastric cancer.

Histone lysine specific demethylase 1（LSD1/KDM1 A） is a highly conserved flavin adenine dinucleotide（FAD） dependent amino oxidase,which plays an important role in maintaining normal physiological *** recent years,the biologic roles of LSD1 have been partly characterized,such as p53 demethylation,***1 is often associated with the occurrence and development of varies diseases,particularly the cancers（AML & SCLC）,virus infection and neurodegenerative ***,LSD1 has been becoming an important target for disease *** LSD1 inhibitors including reversible and irreversible ones have been disclosed,some of them such as ORY-1001,*** presently being assessed in clinical trials for cancer ***,we found an indoline-based hit compound MQS-001(LSD1 IC= 803 nm) by high-throughput virtual screening of Enamine compound library（～2 million compounds）,which was then employed as a template to obtain more potent and selective LSD1 inhibitors through the structure-based drug design（SBDD）*** these compounds,the shortlisted compound MQS-113 inhibited LSD1 potently with an IC value of 276 *** extensive SARs studies and mechanistic investigation are currently ongoing in our lab.

Lung cancer has the highest rate of morbidily and mortality among various cancer types and is responsible for over 1.69 million *** 80-85%of lung cancer patients are diagnosed belong to non-small cell lung cancer（NSCLC）.Most often,the epidermal growth factor receptor（EGFR） overexpression has been observed in 40-85%of patients with NSCLC *** on our achievement,we conducted a focused library of furanopyrimidine which was rapidly synthesized using a variety of primary and secondary amines as nucleophile in parallel reactors and in situ screened for EGFR kinase activity to identified the initial 1 K317 as hit compound.1 K317 possess（S）-2-phenyIglycinol functional group on the furanopyrimidine core as key fragment for the *** further introduced of an acrylamide into phenyl group at meto-position on the 3-position of the fused furan ring led to 3 K007 as a *** structure modification based on scaffold hopping strategy and changing the pharmacophore moiety had led us to discover DBPR112 as a potent EGFR-TKI clinical candidate showing excellent inhibitory ability against EGFR mutation cell lines.

In the current study,a novel series of N-hydroxy-3-sulfamoylbenzamide-based HDAC8 selective inhibitors（12 a-12 p） were designed and synthesized,among which compounds 12 a,12 b and 12 c（Figure A） exhibited potent HDAC8 inhibition with two-digit nanomolar ICvalues,and considerable selectivity over HDAC2（> 180-fold） and HDAC6（0-fold） which was confirmed by western blot *** is worth noting that 12 a,12 b and 12 c displayed highly selective anti-proliferative activity to T-cell leukemia cell lines Jurkat,Molt-4 and neuroblastoma cell line SK-N-BE-（2）.Such selective cytotoxicity was also observed in the well-known HDAC8 selective inhibitor PCI-34051（Figure A） but not in the pan-HDAC inhibitors SAHA and PXD101,indicating that HDAC8 selective inhibitor should have preferable benefit-risk profile in comparison with pan-HDAC ***,the HDAC8 selectivity of 12 a,12 b and 12 c was rationalized by molecular docking study（Figure B）.

Early weaning commonly results in gastrointestinal disorders,inflammation and diarrhea in infants and young animals.A large number of intestinal bacteria and endotoxin LPS invade circulation and intestinal tissue,triggering bacterial translocation and intestinal endotoxemia,which further aggravate intestinal epithelial cell *** polysaccharides（APS） can improve the body's immunity and intestinal ***,the mechanism of action of APS is ***,we investigated mRNA expression profile by transcriptome analysis in porcine intestinal epithelial cells（IPEC-J2）.RNA-seq analysis identified and matched DEGs to the KEGG *** Control vs LPS,the results showed that the signal transduction played an important role in the process of APS regulating the expression of inflammatory factors and chemokines,and three pathways including the TNF,NF-kB or MAPK signaling pathway were *** were 34 up-regulating genes and only 7 down-regulating genes（IL-6,IL-8,TNF-α,CXCL2,CXCL8 etc.） enriched on these pathways in LPS,compared to the control group,indicating that these pathways were activated by LPS ***,mere was almost no difference between the groups that were added to APS and the control *** together,these findings suggest that APS improved intestinal immune function by regulating the expressions of inflammatory mediators via the inhibition of NF-kB and MAPK signal pathway.

Pin1 is a member of peptidyl-prolyl isomerases（PPIases） family that recognize and catalyze isomerization of pSer/Thr-Pro motifs in many kinases and *** a specific enzyme of regulating Pro-directed phosphorylation in numerous signaling pathways,Pin1 has pivotal role in diverse cancer-related cellular processes,including cell-cycle progression,apoptosis,and immune ***,the overexpression and overactivation of Pinl have been associated with many cancers,especially for prostate cancer,breast cancer and lung ***,the inhibition of Pin1 presents a promising option for cancer *** manuscript reported the design,synthesis,and structure-activity relationship study of novel ring A modified 11-keto-boswellic acid（AKBA） derivatives as Pinl *** compounds showed superior Pinl inhibitory activities to *** of the most promising compounds,10 a,potently inhibited Pinl with IC value of 0.46 μM,while it displayed excellent anti-proliferative effect against prostate cancer cells PC-3 with GIso value of 1.82 ***-activity relationship indicated that reasonable structural modifications in ring A had significant impact on improving *** mechanism research revealed that 10 a decreased the level of Cyclin Dl and caused cell cycle arrest at G0/G1 phase in PC-3 cancer ***,compound 10 a may serve as potential anti-prostate cancer agent for further investigation through Pin1 inhibition.

ERK1/2 and ERK5 are proposed to have their pivotal roles in several cancer hallmarks' 11,and ERK5 provides a common bypass route in certain cancer cells,which rescues cell proliferation upon abrogation of ***,we develop a co-targeting ERK1/2 and ERK5 inhibitory strategy to overcome the above-mentioned compensatory *** first selective dual-target inhibitor LYN-1712 has been discovered by using structure-based drug design,which shows highly affinity towards ERK1/2 and ERK5 and good anti-proliferative effects in several breast cancer cell *** addition,we found LYN-1712 had more potent anti-tumor efficacy than reported ERK1/2 inhibitor and ERK5 inhibitor,as well as induced autophagy/autophagic cell death of breast ***,LYN-1712 displayed a therapeutic potential on breast cancer xenograft mouse models without apparent *** together,these results demonstrate that this selective dual-target inhibitor of ERK1/2 and EKR5 induces autophagy to overcome compensatory mechanism in breast cancer,which may provide a candidate drug for future cancer therapy.

Lung cancer treatment has evolved significantly during the past decade but this disease remains the leading cause of cancer-associated mortality ***-small cell lung cancer（NSCLC）accounts for almost 85%of all lung cancer *** growth factor receptor（EGFR）activating mutations and anaplastic lymphoma kinase（ALK） gene rearrangement in advanced non-small cell lung EGFR mutations and ALK rearrangement are observed frofn the cancer（NSCLC） represent the two oncogenic events with an impact on current clinical ***,the two molecular alterations have been viewed as mutually ***,recently,more and more concomitant clinic,which still lacks effective single-agent ***,it is urgent to develop a dual inhibitor of EGFR and *** have discovered a highly potent dual kinase inhibitor JTS-1-39,which displayed strong antiproliferative effect against H197 S and H3122 cancer cell ***-1-39 significantly suppressed the tumor growth in xenograft *** dual inhibitor JTS-1-39 might provide a novel therapeutic opportunity to treat EGFR-and ALK-codriven malignancies in NSCLC.

Doxorubicin has shown good anticancer activity but the therapy is limited by *** combination with a safer drug could be a strategy in reducing dosage and toxicity while increasing anticancer ***,a phosphodiesterase type 5(PDES) inhibitor,sensitized doxorubicin-induced apoptosis associated with a decrease in S and G2/M phases in human hormone refractory metastatic prostate cancer cells(HRMPC).Sildenafil exaggerated the down-regulation of anti-apoptotic Bcl-2 family members and the activation of caspase *** and localization of several DNA repair proteins related to homologous recombination(HR) and non-homologous end joining(NHEJ) systems,including γ-H2 AX,RPA32,Rad51 and Ku protein,were altered in sildenafil-mediated sensitization *** of PDES did not mimic sildenafil-induced *** contrast,vardenafil but not tadalafil(structure-related and-unrelated to sildenafil,respectively) reproduced sildenafil-mediated ***,sildenafil combined with other inhibitors of topoisomerase Ⅱ but not topoisomerase Ⅰ also triggered apoptotic *** conclusion,the data suggest that doxorubicin combined with sildenafil induces a PDES-independent apoptotic *** causes cell apoptosis that is associated with DNA repair processing composed of both HR and NHEJ repair *** improves apoptosis through the impairment of both repair systems that are evident by Rad51 down-regulation and its decreased foci formation in the nucleus,and by a decrease of the DNA end-binding of *** combinatory treatment may enable an important strategy for anti-HRMPC development.
