T cell non-Hodgkin's lymphoma (T-NHL) is an uncommon lymphoma in human This type of lymphoma demonstrated evident heterogeneity. Currently, the domestic and foreign studies on pathomorphology, immunology, molecular genetics, or clinical therapy of T-NHL are not enough. The pathogenesis of human tumor is a multistep process involving abnormal accumulation of many genes, including oncogene activation, tumor-suppressor gene (TSG) inactivation. It is known that there are two primary types of gene or genomic instability, one is mircosatellite instability (MSI), which is characterized by defect of DNA mismatch repair gene;the other one is loss of heterozygosity (LOH), which is characterized by inactivation of TSG LOH involves the mutation of one allele together with the loss of the second allele in a biallelic locus. LOH of TSG may result in the development of human cancers, and LOH analysis with high polymorphic microsatellite markers is a powerful technique for identifying assumed or foregone TSG in genetic map.

The clonal origin of single neuroendocrine cell in adenocarcinoma has not been confirmed by molecular methods yet. We used Laser Capture Microdissection, whole genome amplification, polymerase chain reaction and sequencing to detect and compare the genomic microsatellite abnormalities including loss of heterozygosity, microsatellite instability and p53 mutations in individual cells taken from either gastric adenocarcinoma or matched neuroendocrine cells of six filtered samples. According to microsatellite analysis, 1 case showed highest discordance and almost no concordance suggesting that NE cells and adenocarcinoma cells have different origins. 5 cases had concordant microsatellite abnormalities in different degree amongst which 4 had p53 gene exon 7 or intron 7 concordant mutations, 1 only had exon 8 mutation in adenocarcinoma components. Therefore, we drawed the conclusion that the genetic origin of gastric adenocarcinoma and matched neuroendocrine cells is same in 4 cases but different in 2 cases. The genetic heterogenesis of neuroendocrine cells reflects complexity of progression of canceration.

背景:Licl是糖原合成酶-3β(glycogen synthase kinase-3β,GSK-3β)的非ATP竞争性抑制剂。研究发现,LiCl可以降低癌发生率、通过抑制GSK-3β的活性抑制肝细胞癌的生长。最新的研究提示,GSK-3β活性与前列腺癌进展相关,抑制GSK-3β能够增强肿瘤坏死因子相关凋亡诱导配体(Tumor necrosis factor-related apoptosis-inducing ligand,TRAIL)介导的前列腺癌细胞凋亡。LiCl 能否抑制前列腺癌细胞的生长尚未见报道。目的研究氯化锂(LiCl)是否影响前列腺癌的生长并初步探讨其作用机制。方法体外培养人前列腺癌PC-3细胞,绘制时间及剂量生长曲线、计算对数生长期细胞倍增时间;同细胞的裸鼠背部皮下移植瘤,腹腔注射LiCl或PBS,测量、计算移植瘤体积, 绘制各组移植瘤生长曲线,八周后,处死动物测量肿瘤湿重,检测LiCl对培养细胞的生长及移植瘤的发生发展状态的影响。同时MTT法检测细胞增殖活性、流式细胞术检测细胞周期分布、BrdU掺入实验检测移植瘤细胞DNA合成状态以及western blot检测DNA复制相关基因的变化．结果细胞实验表明:当LiCl浓度达到10mM时,培养第三天与对照组相比PC-3细胞生长可见明显抑制。在该浓度下,细胞对数生长期倍增时间对照组为21．5小时,LiCl组为30．2小时,在LiCl作用下细胞增殖活性呈现明显时间和剂量依赖性抑制,S期细胞比例增加、DNA复制相关基因cdc6。cdc25C, cyclinA和E蛋白水平下调、CDK inhibitor p21CIP1蛋白水平上调。动物实验结果显示:当肿瘤移植与LiCl注射同步进行时,对照组移植后4周左右开始出现移植瘤,LiCl注射组从6-7周开始出现;无论是同步注射实验还是肿瘤发生后再行注射的实验,LiCl注射组肿瘤的体积和重量都显著地小于对照组,显示LiCl抑制移植瘤的发生发展。移植瘤BrdU掺入的免疫组化染色结果显示,LiCl注射组皮下移植瘤的BrdU标记明显低于对照组,提示LiCl引发DNA合成阻滞。结论 LiCl抑制PC-3前列腺癌生长,其机制可能与干扰细胞DNA合成,导致S期静止有关。

Purpose To study the clinical pathological characteristics of Osteoclastoma-like giant cell tumor of the liver (OGC). Methods The clinical data, histological architecture and immunohistochemistical staining of 2 cases of OGC were analyzed. Other 11 cases OGCs reported on Chinese (lease) and English Literature (10 cases) were reviewed. Results Histological features of OGC in my cases were composed of Osteoclastoma-like giant cells admixed with noncohesive pleomorphic atypical mononuclear cells. There was no obvious typical figure of undifferentiated liver cell carcinoma or cholangiocarcinoma founded after closely examined all slides of the 2 cases. Immunohistochemically, the epithelial markers CK and EMA were focally positive and K167 was strong positive (>40%) on atypical mononuclear cells. The giant cells were strongly positive with the histiocytic marker CD68 and negative with AFP. Follow-up showed the two male patients still lived (12 months and 4 months) without recurrence. In previous reports, most of OGC were considered to be primitive undifferentiated carcinomas;despite the lack of epithelial differentiation,most of them have displayed phenotypic evidence of an epithelial lineage. Conclusions The OGC might be further divided into epithelial origin and stromal origin. With a worse prognosis, the OGC histological pattern in the formal origin is a portion of undifferentiated liver cell carcinoma or cholangiocarcinoma. There might be a real stromaly original OGC in liver which can be classificated as soft tissue giant tumor with good prognosis.

AIM: To observe the effects of berbenne on glucose and lipid metabolism and the liver and its mechanism in type 2 diabetic rats . METHODS : Fifty-four male Sprague-Dauley rats were divided into four groups : normal control group, high-fat diet group , type 2 diabetic group and berberine treatment group. Type 2 diabetic rats were established by feeding with high fat laboratory chow and injection of streptozotocin intraperitoneally. Berberine treatment rats were given berberine at the dosage of 100mg/kg/d for 12 weeks. The indices of blood glocuse, insulin, cholesterol (TC), triglycerides (TG), low-density lipoprotein-cholesterol (LDL-c) and high-density lipoprotein-cholesterol (HDL-c) were measured. Morphological changes of the hepatic tissue in each group were observed with light microscopy. The activity of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH- Px) and the contents of malondialdehyde (MDA) in hepatic homogenate were measured biochemically. In addition, the activity of nitric oxide synthase(NOS)、inducible nitric oxide synthase (iNOS) and the contents of nitric oxide (NO) were determined. RESULTS: Compared with the normal group, in type 2 diabetic group, it was manifested as liver fatty degeneration, the concentration of the blood glucose, TC, TG, LDL-c ,serum insulin and hepatic MDA were increased obviously ,but the concentration of HDL-c decreased, the activity of hepatic SOD、GSH-PX、CAT decreased significantly ,the activity of hepatic iNOS、NOS and the contents of NO increased. Compared with diabetic rats, the hepatic pathologic changes in berberine treated group were improved. the contents of serum glucose and insulin, TC, TG, LDL-c and hepatic MDA in berberine treated group were significantly reduced, HDL-c increases but not significant,the activity of hepatic SOD、GSH-PX、CAT was increased markedly, while the activity of hepatic iNOS、NOS and the contents of NO were descended markedly. CONCLUSION: Berberine is effective

Lmplantation of Leydig cells either in suspension or encapsulated in alginatepoly-Llysine microspheres has been proposed as a possible treatment of hypogonadism. A protocol for effective cryopreservation of these cells pror to implantation has been developed. Progenitor(PLC), immature(lLc)and adult rat Leydig cells (ALC)were purified from 21-35-and 90-dayold rats by Percoll separation. The cells were re-suspended in a culture medium supplemented with 1% bovine serum albumin (S-DMEM/F12)in a final concentration of 2×106 cells/mL, and the effects of varying concentrations of dimethylsulfoxide(DMSO,10,15or20%)were assessed after freezing at -70℃ and then liquid nitrogen After 12 months of frozen storage, these cells were thawed rapidly at 37℃ and trypan blue exclusion staining and attachment to culture dishes were assessed as measures of viability. The viabilities for PLC,ILC, and ALC were over85% in the presence of 15% DMSO. Increasing the concentration of fetal calf serum (FCS)in the DMSO-based medium to 20% had no effect on survival after freezing and thawing. In consideration of possible effects of FCS components on steroidogenic function, a serum-free cryopreservative medium was adopted. S-DMEM/F12+15%DMSO. After 12 months in this medium at liquid nitrogen, the Leydig cell steroidogenic capacity in response to maximal challenge doses of luteinizing hormone(LH, 100ng/mL) was unchanged compared to freshly isolated control cells (e.g.,ILC freshly isolated 27.36±4.74 vs. frozen 30.14±1.43ng/106cell ·3hr). Furthermore, the steady state messenger RNA levels for Leydig cell specific transcripts including LH receptor, steroidoenic acute regulatory protein(StAR), cytochrome P450 side chain cleavage enzyme, 17α-hydroxylase/20-lysase,3β-hydroxysteroid dehydrogenase 1 and 2 and 17β-hydroxysteroid dehydrogenase 3, were maintained. Immunoglobulin (Ig) is a very important molecule produced by B cell. The Immunoglobulin heavy chain variable region (IgVH) is formed during B-cell ontogeny by an ordered process of Ig gene rearrangement leading to the assembly of distinct variable(V), diversity(D), and joining(J) gene segments. An additional process of sequence diversification by somatic hyper mutation following antigen encounter occurs in B cells proliferating within the microenvironment of the germinal center (GC) and it increases the immunoglobulin affinity for antigen. Nowadays it is widely accepted that B cell can be divided in to three types according to their SHM states: (1)Pre-germinal center B cells usually exhibit unmutated germinal receptors;(2)Antigen-activated proliferating germinal center B cells show high mutation rates and evidence of ongoing mutations;(3)Post-germinal center memory B cells show mutated but stable receptors. Correspondingly, when B cellsneoplastic transformation has occurred, the tumor cells have the same characteristics of IgVH gene in the development of lymphomas is worth while. Objective: Our study was aimed at acquiring insight into the nature of the immunoglobulin heavy chain variable region sequences in diffuse large B cell lymphomas (DLBCL), to get the relationship between the VH gene structure and development of these lymphomas. Material and Methods: CD20 (L26) and CD45RA (4KB5) were used to mark the 27 DLBCL for definite diagionse. A seminested stragegy was used for the PCR amplification of the VH genes using the FR2A consensus primer, complementary to the conserved framework-2(FR2) of the variable region, and the LJH and VLJH consensus primers for the J region. All those samples amplified successfully were directly sequenced in both directions. Cloning of monoclonal cases was performed with the pGEM T-Easy vector (promega) using DNA excised and purified from monoclonal bands in the polyacrylamide gels. ＜Abstract＞Aim Backgound: Normal numbers of adult Leydig cells per testis are is maintained primarily from the normal proliferation of its progenitors. Postnatal development of Leydig cells in rodents can be divided into four distinct stages of differentiation: initially fibroblast-like stem Leydig cells （SLC） proliferates by postnatal days （PND） 7 to 14, and then they commit to fibroblast-like progenitors （PLC） by 14-21;subsequently, as immature Leydig cells （ILC） by day 21-35, they acquire steroidogenic organelle structure and enzyme activities but metabolize most of the testosterone they produce;finally, as adult Leydig cells （ALC） by day 90 they actively produce testosterone. Targeted gene deletion of insulin-like growth factor-I （IGF-I） results in diminished numbers of Leydig cells （LCs） and lower circulating testosterone levels in adult males. Aim: The aims of the present study were to determine dynamic proliferating and differentiating profiles of adult Leydig cell population during postnatal period in IGF-I null mice. Methods: Testicular cells in mice were labeled by injection of BrDU and testes were collected by PND14, 21, 28, 56 and 90. Leydig cells were immunohistochemically stained using 3β-hydroxysteroid dehydrogenase antibody and analyzed using a imaging analytical software. Results: SLC become proliferative at PND 14 in wild-typed testis with labeling index of 10.8±4.6 （Mean±SEM）, LI of SLC greatly decreased through PND 21-56, and LI peaked for PLC at PND 21 and LI for LC peaked at PND 35. LI of SLC did not change in IGF-I null mice （7.4±3.3） compared to wild-type mice, while PLC and ILC did not proliferate through PND 14 to PND90. The Leydig cell number and cell size were significantly reduced in IGF-I mutant adult testis, e.g. at PND90, 980±155 cells/mm2 and 132.6±6.4μm2 compared to 1876±116 cells/mm2 and 280.9±10.6 μm2 in wild-type testis, respectively. It has been reported that neuronal signaling directly controls Leydig cell steroidogenic function. Neuronal pathways to the testis and to the vicinity of Leydig cells have been demonstrated, but neurotransmitter senstitivity in these cells has remained unclear. The objectives of the present study were to determine if acetylcholine receptors (AchRs) are expressed in Leydig cells. Progenitor, immature and adult Leydig cells were purified from 21-, 35-and 90-day-old male Sprague Dawley rats, respectively, and AchR messenger RNA levels were measured by DNA macro- arrays. The nicotinic-AchR subunits α1,α2,α3,α4,α5,α6,α7,β1,β2,Δ,γ,and σ,and muscarinic AchR M2 through M5 were all included in the analysis. Subunits α1,α5, α4, α6,β1, β2, and σ were detected above significant signal threshold levels. None of the muscarinic receptor forms were detected. Subunit α4 had the highest signal level: it was undetectable in progenitor Leydig cells and increased sharply during the transitions to immature and adult Leydig cell by up to 100 fold. Transcript levels for the enzyme acetylcholinseterase, which metabolizes Ach, were also abundant. Adult Leydig cells were cytotoxically eliminated from adult testes by administration of the drug ethylene dimethanesulfonate. This induced Leydig cell regeneration, and it was noted that a4 subunit expression was eliminated along with the Leydig cells and reappeared only after 35 days with the restoration of normal serum testosterone concentrations. Purified adult Leydig cells were treated with different concentrations of nicotine (10-7 to 10-3M) for 3 hours. Nicotine exerted a biphasic dose response for LH stimulated T production. Lower doses, 10-5M, increased T levels by 20% (control 141±5 vs.171±9ng/106cell·3hr), whereas higher doses,e.g., 10-3M,lowered T production by 50% (71±1ng/106cells·3hr).
