Background:The normal expression of Ca2+-regulatory protein is critical for efficient myocardial function. Aims:The present study tested the hypothesis that rosuvastatin treatment may attenuate left ventricular(LV) remodeling and dysfunction in the failing heart,which may be associated with alterations of Ca2+-regulatory protein. Methods:We investigated the change of LV remodeling and function in a rat model of heart failure due to myocardial infarction(MI) with or without rosuvastatin(10 mg/kg/day) treatment for 10 *** protein expression of sarcoplasmic reticulum Ca2+ ATPase(SERCA)2a,phospholamban(PLB) and phospho-PLB at serine-16(pSer16-PLB),as well as SERCA activity,interleukin(IL)-6 and IL-10 levels were evaluated. Results:After rosuvastatin treatment,LV remodeling and dysfunction were *** partially restored the low expression of SERCA2a and pSer16-PLB,increased SERCA activity,but showed no effect on PLB ***,rosuvastatin reduced the increased IL-6 level and further elevated IL-10 level in the peri-infarct and remote zones of *** lipid levels remained unchanged. Conclusion:Rosuvastatin is effective in reducing LV remodeling and dysfunction in the failing heart. The molecular mechanism may be related to normalization of SERCA2a expression,SERCA activity,and pSer16-PLB levels,as well as through cytokine alterations independent of its lipid-lowering effect.

Objective To assess the values of electrocardiogram in predicting the culprit artery in patients with inferior acute myocardial infarction(AMI).Methods 78 patients with inferior AMI underwent selective coronary angiography within 1 month of hospitalization and were devided into two groups,Right coronary artery(RCA) group and Left circumflex coronary artery(LCx) group,according to the infarct-related *** admission 12-lead electrocardiograms were evaluated for ST-segment deviation amplitude at 0.08 second after the J-point and R/S ratio in leadsⅠand ***(1) The incidence of ST-segment depression≥1mm in lead aVL was significantly higher in RCA group patients than that in LCx group patients(92%vs 24%,P<0.01);(2) R/S ratio in lead aVL was significantly lower in RCA group patients than that in LCx group patients(2.7±0.4 vs 3.4±0.6,P<0.01);(3) The sensitivity, specificity,positive predictive values and negative predictive values of ST-segment depression≥1mm or R/S ratio≤3 for predicting RCA culprit lesion were 92%、90%、96%、77%and 86%、95%、98%、70%, *** It is suggests that ST-segment depression or decreased R/S ratio during inferior AMI confers a greater likelihood of RCA disease.

It is well known that the density of cardiac stronger inward rectifying potassium current(Ik1) in atrium is smaller than that of *** the amphotericine B perforated patch recordings;we verified that the density of the atrium is smaller than that in the ventricle in *** the heart,Ik1 belongs to the Kir 2 family composed by the Kir2.1;Kir2.2,and Kir2.3 *** contribution of the hetermoer kir2.x to the difference of Ik1 between the atrium and ventricle was studied by examining the property of co-expression gpKir2.1 and gpKir2.3 *** co-expression DNA ratio of Kir2.3 in the co-expression is 80%,50%,and 20%,*** parallel,the DNA ratio of Kir2.1 in the co-expression is 20%,50%, and 80%,*** conventional whole cell recordings,the currents of homogenous Kir2.3 and co-expressional channels showed the different property from the currents of homogenous Kir2.1,Kir 2.2, and isolated cardiac IK1 of guinea-pig in their external pH sensitivity and the time constant of the activation phase of *** results illustrated that the component of Kir2.3 in both of atrial and ventricular cells of guinea-pig is *** distribution of the Kir 2 subunits can not explain the density of Ik1 in atrium is smaller than that in ventricle in guinea-pig.

Objective:Matrix metalloproteinases(MMPs) have been implicated in the initiation,progression,and rupture of abdominal aortic aneurysms(AAAs).Doxycycline,an MMP inhibitor,has been demonstrated to prevent the formation of angiotensinⅡ(AngⅡ)-induced *** purpose of this study was to determine whether doxycycline influenced aneurysmal progression and rupture when administered after the initiation of AAAs. Methods and Results:LDL receptor-/- male mice were fed a high-fat diet and infused with AngⅡ(1,000 ng/kg/min) via subcutaneous osmotic pumps for a 28-day *** with AAAs were implanted sequentially with Alzet pumps every 28 days to permit continuous delivery of AngⅡfor another 56 days,and simultaneously given doxycycline(100 mg/kg/day) or vehicle *** dose causes a therapeutic serum drug concentrations of 2.4±0.5μg/*** revealed the progressive dilation of suprarenal aortas during continuous AngⅡ***,doxycycline did not retard the expansion of AAAs or modify the cellularity and matrix remodeling of AAA ***, doxycycline did not affect serum MMP-2 or *** with our previous reports, doxycycline did not affect the development of atherosclerosis in mice with 3 months AngⅡinfusion. Neither body weight,systolic blood pressure,nor serum cholesterol concentrations were affected by doxycycline. Conclusion:Continuous infusion of AngⅡpromotes AAA progression that differ from the process involved the ***,doxycycline at therapeutic concentrations did not retard AAA expansion and ***-induced MMP inhibition does not influence the progression and rupture of AAAs.

Farnesyl pyrophosphate synthase(FPPS),an essential enzyme in the mevalonate pathway,was reported to be upregulated in young spontaneously hypertensive rats(SHR) when compared with Wistar-Kyoto(WKY) rats, and this was accompanied by development of left ventricular ***-week-old rats were daily gavaged with vehicle or an FPPS inhibitor(alendronate,1 or 10 mg/kg) and blood pressures was monitored by the tail-cuff method every other ***-weeks of alendronate treatment attenuated the left ventricular weight to body weight ratio(LVW/BW),hydroxyproline content,collagen deposition in the interstitia,and gene expression of atrial natriuretic peptide,B-type natriuretic peptide,and procollagen typeⅠ/Ⅲin the SHR left ventricle,all of which were significantly higher in SHRs than in WKY ***,long-term treatment with an FPPS inhibitor significantly reduced RhoA activation,ERK phosphorylation,and TGF-β1 expression in the SHR left ventricle,all of which were upregulated more in SHRs than in WKY *** conclusion, chronic treatment with an FPPS inhibitor attenuates the development of cardiac hypertrophy and fibrosis,and the suppression of ERK1/2 phosphorylation and TGF-β1 expression by inhibiting RhoA activation may be an important mechanism.

Background and purpose:Cholesterol is an integral component of all eukaryotic cells. Spontaneously hypertensive rat(SHR) is accompanied by cardiovascular remodeling,but its serum cholesterol level is ***,it is possible that the expression and activity of HMG-CoA reductase in SHR tissues might be *** purposes of this study were to study the expression and activity of HMG-CoA reductase both in liver and in extrahepatic tissues(heart,aorta and kidney) of SHR,and to investigate the possibility that atorvastatin may reverse the adverse alteration of the enzyme in SHR. Experimental approach:SHR and normotensive Wistar-Kyoto rat(WKY) were treated daily with atorvastatin(50 mg kg-1) from 8 to 18 weeks of *** levels of serum and tissues(liver,heart, aorta and kidney) were determined by commercial enzymatic *** blot analysis and high performance liquid chromatogram(HPLC) were used to assay the expression and activity of enzyme respectively. Key results:The expression and activity of HMG-CoA reductase both in hepatic and extrahepatic tissues of SHR were higher than those in WKY,although serum cholesterol levels in SHR were much *** atorvastatin treatment,both the expression and the activity of HMG-CoA reductase were remarkably declined in hepatic and extrahepatic tissues of *** in WKY,they were altered only in liver where the enzyme expression upregulated but the activity decreased. Conclusions and implications:HMG-CoA reductase both in liver and in extrahepatic tissues of SHR is abnormally upregulated,and these adverse changes could be reversed by atorvastatin *** data are useful to identify or understand the pathological implication of abnormal cholesterol metabolism in SHR.

Objectives:The effect of low dose of cyclophosphamide protecting cardiac function from ischemia-reperfusion injury was studied on *** premise is that CP inhibits immune and inflammatory process,thereby limits I/R injury and improves myocardial ***:Open chest rats were submitted to 30 min of ischemia followed by 3 h,12 h or 24 h of *** were divided into sham group,I/R group and CP group,and each group included 3 time-point subgroups(3 h,12 h and 24 h;n=8 for each subgroup).750mg/m2 cyclophosphamide was intraperitoneally administrated in CP group and saline was given to I/R group.A polyethylene tube was inserted into the left ventricular cavity to detect left ventricular systolic pressure(LVSP) and±dp/dt *** the end of experiment,hearts were harvested for histopathological assessment and infarct size ***:Compared with I/R group,rats treated with low dose CP showed a significant recovery in myocardial function with improved LVSP(88±11 vs. 69±11 mmHg of 3 h;92±11 vs.64±14 mmHg of 12 h;90±11 vs.64±14 mmHg of 24 h;P<0.01 respectively).The±dp/dt max also had the similar *** myocardial infarct size was reduced in CP group compared to that in I/R group;the infiltration of polymorph nuclear leukocytes(PMNs) in myocardium was decreased in CP *** histopathological damage score was also attenuated. Conclusions:These findings suggest that low dose CP rescue cardiac function from ischemia-reperfusion injury by alleviating histopathological damage in rat myocardium.

BACKGROUND: Calcific aortic valve stenosis is a complex disease,resembling both atheroinflammatory processes and skeletal *** cathelicidin antimicrobial peptide LL-37 is a multifunctional mediator in the complex network of immune responses and inflammatory *** objective of this study was to explore the potential role of LL-37 in the pathogenesis of calcific aortic valve stenosis. METHODS: We investigated the presence and cell origin of LL-37 in human calcified and noncalcified aortic valves using *** pro-apoptosis effects of LL-37 on cultured pig aortic valve interstitial cells were examined by cell viability with MTT assay,annexin V expression by FACS analysis. The bone differentiation effects of LL-37 on cultured PAVICs were also examined,using nodule formation and ALP activity analysis. RESULTS: LL-37 was present in calcified human aortic valves,and was expressed by CD68 positive multinuclear giant ***-37 did not increase ALP activity,or nodule formation,indicating that LL 37 did not cause differentiation of PAVICs into oesteobalst-like ***,LL37 inhibited the viability of PAVICs by MTT assay and increased the expression of annexin-V on the cells,suggesting LL37 induced apoptosis in PAVICs at higher concentrations. CONCLUSIONS: Our findings suggest that human cathelicidin antimicrobial peptide LL-37 is involved in the pathogenesis of calcific aortic stenosis,possiblely by inducing VIC *** effects may represent an early event in the calcification processes that may subsequently be followed by mineralisation of the apoptotic bodies.

Background:Heat shock protein90(hsp90) is an ubiquitously expressed molecular chaperone that controls the folding,assembly,intracellular disposition and proteolytic turnover of many proteins,most of which are involved insignal transduction processes,such as Akt,Raf-1,ErbB,which regulate cell growth, differentiation and *** mesenchymal stem cells are multipotent stem cell and able to differentiate into endothelial cells,vascular smooth muscle cells,and even cardiac-like myocytes cells when transplanted into the ischemic *** can improve cardiac function through possible myogenesis and angiogenesis after myocardial *** had demonstrated that hsp90 could promote angiogenesis in cancer cells via Akt and eNOS pathway. Objective:We hypothesized that MSCs could express hsp90 in normoxia culture conditions. Materials and Methods:Bone marrow mononuclear cells were isolated and harvested from Male Sprague-Dawley *** were cultured ex vivo in the normoxia conditions and passages 3-5 were enrolled in this *** were lysised and total protein concentration was determined with BCA protein assay *** was run on a 8-12%gradient gel by SDS-PAGE,then transferred to PVDF *** were blocked with 5%fat free milk in TBS-T at room temperature for 2 hrs,and incubated with anti-hsp90 overnight at 4℃.The membrane was washed and incubated with secondary *** were developed on films using the ECL technique.
