Objective:Polymeric micelles self-assembled by biodegradable amphiphilic block copolymers have been widely investigated as controlled drug delivery ***-linking micelle has been recognized as a promising approach to stabilizing micelles,which can hold self-assembled *** addition,redox-sensitivity allows the facilitated release of loaded drugs in the cytoplasm in response to the intracellular reductive *** to its enhanced stability during systemic circulation and accelerated intracellular drug release intracellular,the biocompatible and biodegradable cross-linking polymeric micelle is a promising drug vehicle for cancer therapy. Methods and Results:Chitosan-g-octadecylamine（Chi-g-ODA）was first synthesized by amide bonding between 3,3'-dithiodipropionic acid and octadecylamine,which using dicyclohexyl-carbodiimide（DCC）as coupling *** chitosan （1.5×10～4 Da）was conjugated to the residual carboxyl groups of the intermediates to form *** spectrum and H NMR was used to confirm the structure and substituted degree,which showed the micelles contained about 5%of ODA in molar *** critical micelle concentration of Chi-g-ODA was about 73.6μg/*** light scattering showed that the micelle had high stability with a size of about *** bioreducible property of cross-linked micelles in 10 mM 1,4-dithiothreitol（DTT）solution was monitored by in vitro release:DOX released from micelles showed significantly improved compared with micelles without DTT, presenting a redox-response drug-release profile at 37℃.To investigate the intracellular redox-sensitivity,cellular uptakes and cytotoxicity assay were performed on human alveolar adenocarcinoma cell line（A549）.ICvalue of Chi-g-ODA was about 300μg/mL,which indicated the micelle to be a safe drug delivery *** addition,Chi-g-ODA/DOX showed similar cytotoxicity with free DOX against A549 cell line within 72 hours. [PURPOSE]Poly（amidoamine）(PAMAM)dendrimers are widely used as drug delivery *** large hydrophobic cavities on these hyper-branched molecules could encapsulate and improve the water solubility of the hydrophobic *** this study,two generations（G5 or G7）of PAMAM dendrimers,with positive-charged surface groups（-NH with or without PEGylation）,were applied to investigate their effects on the water solubility of probucol,a hydrophobic ***,the transepithelial transports of probucol-liposomes across Caco-2 monolayers,an intestinal epithelial model,were explored in the presence of these four kinds of PAMAM dendrimers（G5-NH,G5-NH-PEG,G7-NH,and G7-NH-PEG）. [METHODS]The influence of the PAMAM dendrimers to the water solubility of probucol was investigated by using the different generations and surface groups of the dendrimers and at the different concentrations and incubation *** saturated solubility of probucol was then determined by *** probucol-liposomes were prepared by film dispersion method,and modified by the dendrimers through ionic ***-2 cells were seeded in transwells and cultured to form the *** transports of probucol-liposomes from the apical-to-basolateral side（A→B）of the Caco-2 monolayers with or without the presence of the dendrimers were explored over time and determined by HPLC. [RESULTS]The dendrimers significantly increased the solubility of probucol in a concentration-dependent manner,and the maximum enhancement of the solubility of probucol was obtained at 24 hours after incubation of the drug with the dendrimers. Objective:Polymersomes(POS)is a novel drug delivery *** block copolymers can self assemble to form supram olecular aggregates with specific shapes in a soluble ***,the chemical and physical properties of POS,in cluding vesicle size,vesicle surface modification,drug loading,even in vitro and in vivo behavior may be adjusted due to the rich diversity of block copolymer *** this study,we present a self-assembly POS modified by transferrin c ombined Tet-1 peptide,to obtain a dual-brain targeting system(Tf/Tet-1-POS).The system will across the blood-brain barri er(BBB)through transferrin receptor mediated transcytosis,then target the neurocyte using the Tet-1 's high affinity to the GT1B receptor on the nerve cells. Methods:The diblock copolymer of NHS-PEG-PLGA and maleimide-PEG-PLGA were synthesized by the ring-opening polymerization of dl-lactide and *** Tf/Tet-1-POS were prepared by self-assemble method and characterized using dynamic light scattering to give size and zeta *** morphological appearance of POS was observed by transmission electron microscope(TEM).Chemical composition of the surface layer was determined using X-ray photoelectron spectroscopy(XPS).The distribution of the formulations in mouse brain tissue were determined with 6-coumarin as fluorescence probe loaded in the system.A plasmid,pGPU6/GFP/Neo(PGN),was incorporated into Tf/Tet-1-POS,and a transfection for neuro-2A cells was *** the distributions of Tf/Tet-1-POS loaded DIR fluorescence probe were observed by in vivo imaging method in animals. Aim:Study on the effect of miRNAs targeted delivery system to prostate ***:This study designed polyamidoamine (PAMAM)nanoparticle as drug carriers,miR-15a/miR-16-l as model drug,aptamer(A10-3.2)as prostate-targeting ligand, prostate-specific membrane antigen(PSMA)as target,and all these co-constructed miRNAs targeted nanoparticle mediated by aptamer(PAMAM-PEG-A10/miRNA).The effect of PAMAM-PEG-A10/miRNA on prostate cancer was investigated in the transplanted tumor model of nude ***:PAMAM-PEG-A10 was applied as macromolecular polymer to encapsulate DNA to synthesize PAM AM-PEG-A10/DNA nanoparticle which has a particle size around 180nm and zeta potential around 18 *** is spherical and has regular shape and high stability under the examination of electron *** PAMAM-PEG-A10/miRNA inhibited the growth of androgen-dependant prostate tumor which is on the basis of gene pharmaceuticals microRNA-15a and microRNA-16-1 that can inhibit the growth of androgen-dependant prostate tumor cell ***-PEG-A10/miRNA has better targeting ability,better prevention from proliferation of prostate tumor cells which has significant differences in contrast to the other three control groups(The tumor inhibition rates compared with NaCl group were PAMAM-PEG-A10/NC-miRNA 2.45%, PMAMA-PEG/miRNA 44.39%,PAMAM-PEG-AlO/miRNA 69.17%,).The results of the detection of hepatic and renal function demonstrated that there was no significant differences between these four group(P>0.05).Conclusion:PAMAM-PEG-A10/miRNA can enhance the targeting ability and has higher activity in killing tumor,meaning that PAMAM-PEG-A10/miRNA is a new safer gene therapeutic system.

Objective:Polymeric micelles have proven to be a promising nanoscale material for drug delivery ***（PTX）is known to be a wide-spectrum anticancerogen,while the therapeutic effect of PTX is often limited by the poor solubility,the absence of selective cytotoxicity,and the appearance of *** aim of this study was to design a dextran-based micelle system for the PTX delivery. Methods:The copolymers poly（lactic-co-glycolic acid）-g-dextrans（Dex-PLGAs）were successfully synthesized via an esterification *** physicochemical properties of Dex-PLGA such as particle size,morphology,encapsulation efficiency,drug loading and in vitro release profile were *** addition,the in vitro and in vivo antitumor activities of PTX-Dex-PLGA micelles were tested against human breast carcinoma cells,their drug resistant variants and human ovarian cancer,and against human ovarian cancer cells bearing nude mice. OBJECTIVE:The purpose of our study was to comprehensively compare in vivo performances of discoidal and spherical recombinant high density lipoproteins loaded with cardiovascular drug tanshinoneⅡA（TA-d-rHDL and TA-s-rHDL）,in order to provide some references for rHDL as cardiovascular drugs delivery systems. METHODS:TA-d-rHDL and TA-s-rHDL were successfully constructed,*** next their in vitro physiochemical characterizations and biomimetic properties were ***,their pharmacokinetic behaviors,atherosclerotic lesions targeting effects and antiatherogenic efficacies were elaborately performed and compared in atherosclerotic New Zealand White （NZW）rabbits. RESULTS:In vitro characterizations studies showed that TA-d-rHDL had a mean diameter of 138.2±5.1 nm with high entrapment efficiency（EE）of 93.3±1.5%and drug-loading coefficient（DL）of 4.6±0.2%,and TA-s-rHDL 132.6±4.8 nm with EE of 98.5±1.2%and DL of 6.3±0.3%,and their micromorphologies were proven by transmission electron microscopy（TEM）.Additionally, the TA-d-rHDL maintained remodeling behaviors induced by lecithin cholesterol acyltransferase（LCAT）,and the in vitro release tests showed that the TA loaded in the TA-d-rHDL leaked during the remodeling *** studies indicated that the plasma concentration-time curve(AUC)of TA-d-rHDL and TA-s-rHDL were 5.62 and 6.75-fold bigger than TA suspensions （TA-S）,*** the total body clearance（CL）were 6.18 and 7.08-fold lower,and mean residence time（MRT）15.07 and 16.72-fold longer than TA suspensions,***-vivo imaging using near-infrared（NIR）fluorophore DiR demonstrated that rHDL bound more avidly to atherosclerotic lesions than to normal vessel walls,and s-rHDL had the best atherosclerotic lesions targeting effect than the *** pharmacodynamics showed that both TA-d-rHDL and TA-s-rHDL had much stronger antiatherogenic efficacies,which were characterized as healthier serum lipid,smaller lesions size,hi

Purpose:The concept of co-delivery of drugs and siRNAs has been proposed by single delivery system to accomplish the combined effect of chemo-and *** delivery of both drugs can be accomplished simultaneously,and it is powerful to home them specifically to tumor tissue,and control the release of drugs,thus it can decrease the toxic effects on normal *** purpose of our research was to develop a co-delivery *** formulation,PEI as the carrier,was contained conjugated DOX and siRNA for treatment *** active targeted was achieved by the folate conjugated to PEI with the PEG as the linker. Methods:DOX was combined to the PEI by the pH-sensitive linkage（hydrazone bond）at the 13-C carbonyl group of DOX（named PEI-MPH-DOX）.Another part of PEI conjugated PEG-folate was synthetized（named PEI-PEG-FOL）.The DOX release from PEI-MPH-DOX was ***-MPH-DOX/PEI-PEG-FOL/siRNA complexes was prepared and characterization by dynamic light scattering and agarose gel *** cellular uptake of complexes was studies by two-color flow cytometry and confocal laser scanning microscopy（CLSM）.Anti-proliferative activity of the PEI-MPH-DOX conjugate against cancer cells was investigate in *** siRNA-mediated reduction of target gene expression was tested using PEI-MPH/PEI-PEG-FOL/siGFP （without DOX）at a stable green fluorescent protein（GFP）-expressing HeLa cell line in vitro and in vivo. The aim of this study was to investigate the antitumor effect of docetaxel (DTX) loaded Pluronic P105/F127 mixed micelles (PF-DTX) against drug-resistant Lung Adenocarcinoma Cell Line A549/*** loaded Pluronic P105/F127 mixed micelles was prepared by thin-film hydration method and optimized using central composite *** of multidrug resistance (MDR) by Pluronic mixed micelles was evaluated in Taxol-resistant Human lung adenocarcinoma cell line A549 (A549/Taxol).In vitro cytotoxicity was determined by CCK8 assay,while cellular apoptosis was detected by cell nuclei staining and Annexin V-FITC apoptosis detection *** cycle arrest was also confirmed by flow ***,in vivo antitumor efficacy of PF-DTX was extensively evaluated in comparison with *** in vitro release of DTX from the mixed micelles presented a sustained-release *** in vitro cytotoxicity assay showed that on the sensitive A549 cells,the IC50 values for Taxotere and PF-DTX were similar,while on A549/Taxol cells,IC50 values ofPF-DTX (0.059μg/ml) was much lower than those of Taxotere injection (0.593μg/ml) and the flee DTX (0.857μtg/ml),which showed superior hypersensitizing effect on MDR *** enhanced anti-cancer efficacy of PF-DTX in vitro was associated with DTX-induced apoptosis and cell arrest in the G2/M *** BALB/c mice bearing A549/Taxol tumor xenografts,stronger antitumor efficacy was shown in PF-DTX group,with good correlation between in vitro and in ***,it was concluded that Docetaxel loaded Pluronic P105/F127 mixed micelles significantly enhanced the anti-cancer activity of DTX and might be considered a promising drug delivery system to overcome MDR in lung cancer.

Objective:To prepare CsA solid self-microemulsion,and determining the particle size,zeta potential of CsA self-microemulsion before the adsorption and CsA self-microemulsion after the *** to evaluate the powder properties of CsA solid self-microemulsion,and determining its content. Methods:Using spherical crystallization technique prepare porous microspheres of ethyl cellulose;in conjunction with other carrier treating liquid CsA microemulsion;determining the change of particle size,zeta potential of CsA self-microemulsion before the adsorption and after the adsorption by Malvern,investigate powder properties and content determination. Results:*** Solid self-microemulsion shape,mobility,drug loading to cure（drug loading liquid microemulsion completely adsorption minimum amount of calculation）as the main evaluation,placed in non-imprinted on the filter paper completely cured after ***,microcrystalline cellulose,mannitol,lactose,PEG 4000,homemade EC microspheres of one or several,on 1.0gCsA microemulsion curing,results in Table *** table 1,we can see Using EC microspheres,in conjunction with SiO treating liquid CsA microemulsion,wo can get higher drug loading and better powder properties. *** electron microphotographs of CsA solid *** 1 shows the more adsorption former EC microspheres empty,larger and deeper,EC microspheres after adsorption,less empty,smaller and *** preliminary part of the CsA microemulsion has entered the hollow of the EC microspheres EC porous microspheres can adsorb CsA microemulsion,and improve the liquidity of the common carrier adsorption preparation of solid microemulsion. Self-restraint CsA solid self-microemulsion has better mobility,compressibility,filling from table2. *** particle size,zeta potential of CsA self-microemulsion before the adsorption are 27.19±0.54nm、-5.71±0.73mV and the particle size,zeta potential of CsA self-microemulsion after the adsorption are 28.82±0.68
