Hydroxysafflor yellow A(HSYA),the main active pharmaceutical ingredient of the saffiower plant(Carthamus tinctorius L.),is a hydrophihc drug with taw oral bioavailability(BA).The objective of the present study was to improve the oral BA of HSYA by formulation *** main reason for the taw oral BA of HSYA is its low liposolubility and high water-solubility,which can hinder its penetration across the epithelial *** pharmaceutical excipients have been shown to enhance the permeability of BCS III drugs,such as sodium caprate(SC),sodium deoxycholate,P-cyclodextrin(p-CD) and Potaxamer 188(PI88).Therefore,the above five excipients were selected to investigate their ability to enhance the oral BA of HSYA through animal ***,chitosan is the only basic polysaccharide in nature that is positive charged,the properties of its bioadhesion and absorption enhancement make it a suitable carrier in mucosal drug delivery *** special properties could be utilized to prepare a HSYA-CS *** optimized the preparation process,the binding rate reached 99.4%.Then,HSYA granules,which contain the HSYA-CS complex and a penetration enhancer,were *** these granules have been administered,a portion of the amino groups of each CS molecule bound with HSYA,and another portion of the amino groups,could adhere to the negatively charged intestinal mucosa by electrostatic ***,many drug release cores will be formed and distributed on the surface of intestinal ***,the action time and intensity between HSYA and the intestinal epithelia would be extended and *** the same time,the tight junctions of the intestinal cell monolayer would be opened or the permeability of intestinal cell would be increased due to the action of the absorption enhancer;thus,HSYA could permeate into the cell membrane,which could significantly enhance the absorption of *** results of the pharmacokinetics in Sprague-Dawley rats sho

Objective Folate receptors(FRs),over-expressed on the surface of a variety of tumor cells,are potential targets for tumor targeting *** is to continue our earlier work on the folate conjugated pullulan acetate nanoparticle(FPA NP),which were further used as an Epirubicin(EPI) carrier to enhance cancer targeting *** aim of the present work was to clarify the mechanism of FPA NP internalized by cancer cells,which provides the basis for developing more safe and effective nano-sized drug *** FPA NP and FPA/EPI NP were prepared by dialysis *** cytotoxicity of FPA/EPI NP against HepG2 cells and He la cells was evaluated by MTS *** cellular uptake of the FPA/EPI NP was systematically evaluated by fluorescence intensity which detected by flow ***,several selective endocytosis inhibitors(chlorpromazine,flipin,chloroquineto) and folate were selected to study the potential endocytosis *** FPA/EPI NP was successfully formulated into nanoparticles(mean particle size 268.5±12.0nm) by dialysis with an almost spherical *** assay revealed that FPA/EPI NP showed cytotoxicity against HepG2 and Hela cells,and at72 h the half maximal inhibitory concentration(IC50) of FPA/EPI NP were 168 μg/mL and 105 ng/mL,*** vitro experiments with endocytic inhibitors revealed that clathrin-mediated endocytosis and macropinocytosis were involved in the internalization of FPA/EPI NP on the HepG2 *** clathrin- mediated endocytosis and FRs-mediated endocytosis were involved in the internalization of FPA/EPI NP on the Hela *** Our FPA NP may serve as a drug carrier for tumor target therapy.

The central dogma of drug delivery is to steer therapeutic cargos to target tissues and/or cells to achieve maximal therapeutic efficacy with minimal toxic *** of biomimetic designs as promising sustained drug release and site-targeted delivery systems for a variety of therapeutic agents from different fields of cancer therapy has gained a remarkable degree of interest in recent ***,we introduce a robust dual-functional cell-penetrating peptide(CPP)/recombinanted high-density lipoproteins(cp-rHDL)-based delivery system that affords high payload and efficient delivery of a natural compound gambogic acid(GA),which could enhance proteasome inhibitor-induced anticancer activity due to pleiotropic apoptotic effects through a complex mechanism involving the participation of multi-molecular ***-encapsulated cp-rHDL nanoparticles(cp-rHDL/GA) consists of(1)hydrophobic core for modulation of the anticancer therapeutic GA,(2) apolipoprotein(apoA-I) with attractive integrating and packaging ability,and(3) stearyl CPP anchored into phospholipid monolayer due to hydrophobic *** tailored nanosystem cp-rHDL/GA presents desirable particle size(128.3 ± 5.3) nm,excellent entrapment efficiency(95%for GA),superior biostability,and favorable buffering capacity leading to proton sponge *** stably assembled cp-rHDL/GA was fabricated by introduce of apoA-I,which enabled targeted connection to scavenger receptor BI(SR-BI) resulting in directly cytosolic GA delivery,and tumor acidity-responsive CPP(R6H4) responsible for controllable permeability through an endocytotic pathway with ability of rapid endo-lysosomal escape(designated SR-BI "channel" /endo-lysosome co-mediated intracellular mechanism).Upon stepwise incubation with apoA-I and R6H4,cp-rHDL/GA enhanced cell interaction of 96.1%positive human hepatic carcinoma(HepG2) cells atpH6.4,and remarkable cytotoxicity and apoptotic effect via triggering p53 pathway,approximately 4.5-fold increa

Nowadays,a novel thiolated chitosan and its S-protected derivative have attracted broad attention for their permeation enhancing and efflux pump inhibitory *** to chitosan,the permeation enhancing properties of thiolated chitosan is related to its effect on tight junctions,and the mechanism has been ***,the efflux pump inhibitory mechanism that is related to its effect on P-glycoprotein(P-gp) remains *** present study aimed to investigate the effect of the thiolated chitosan,chitosan-thioglycolic acid(CS-TGA),and its S-protected derivative(CS-TGA-6MNA) on the activity of P-glycoprotein(P-gp) and unravel the action *** the synthetic process,thioglycolic acid(TGA) were first covalently bound to chitosan(CS),then thiol groups of thiolated chitosan were protected by disulfide bond formation with the thiolated aromatic residue6-mercap to nicotinamide(6-MNA).P-gp inhibition effects were evaluated ex vivo using rat intestinal mucosa and in vitro via Caco-2 cells using the known P-gp substrate rhodamine-123(Rho-123) as the model drug.S-protected thiolated chitosan exhibited pronounced P-gp inhibition effects in contrast to chitosaa The fluorescence polarization measurement results suggested the plasma membrane fluidization was a contributor for inhibition of P-gp by S-protected thiolated chitosaa At the same time,S-protected thiolated chitosan inhibited P-gp ATPase *** evident change in P-gp express level was detected by flow cytometry after exposure of the cells to S-protected thiolated chitosaa The depletion of intracellular ATP,which was likely to be a result of partial inhibitionof cellular metabolism,was directly correlated with inhibitory potential for P-gp mediated efflux by S-protected thiolated chitosaa Thus,the efflux pump inhibitory properties appeared to the integrated results of plasma membrane fluidization alteration,intracellular ATP-depletion and P-gp ATPase activity *** features approve S-p

Objective:To enhance the effective diagnosis of tumor by magnetic resonance imaging（MRI）,aptamer-targeted liposomes combined with gadolinium（Gd）were designed and constructed as a novel MRI contrast *** aptamer used in this study was a modified GBI-10,which has high affinity to tenascin C（TN-C）overexpressed in various ***:Non-targeted liposomes（NLs） were first prepared by lipid film hydration and extrusion *** aptamer-targeted liposomes（TLs） were obtained by incubating GBI-10-Chol with ***-6 was loaded into NLs and TLs respectively to compare their interaction with tumor cells by confocal laser scanning microscopy and flow *** particle size and zeta potential of NLs and TLs were *** clinical 3.0 Tesla MR scanner was used to assess the imaging efficiency（indicated as Ti） of NLs and TLs,which were compared to that of commercially available contrast agents,Gd-DTPA（magnevist）.The relaxation rate（r） was obtained by the linear fitting curve of concentration gradient and 1/T).Results:The particle size ofNLs and TLs were 126.4±2.4 nm and 157.2±1.9nm,while zeta potential were-3.94±0.53 mV and-12.85±0.35 mV,*** images of confocal laser scanning microscopy（Figure 1 A） for NLs and TLs shows the amount of NLs entered in cytoplasm was much less compared to TLs at 37℃.The fluorescence intensity of TLs increased to about 3 times than that of NLs（Figure 1B）.The MR images for NLs,TLs and Gd-DTPA were shown in Figure *** values of r calculated for NLs,TLs and Gd-DTPA were 4.7±0.3 mMs,4.4±0.2mMs and3.9±0.1 mMs,respectively（n=3）.Conclusion:The results suggested that GBI-10-targeted paramagnetic liposomes prepared in this study were potential contrast agent for MRI diagnosis of tumor with the high expression of TN-C.

The therapeutic efficacy of doxorubicin(DOX) in cancer treatment is limited by two main factors,systemic toxicity resulted from the nonspecific transportation and the emergence of tumor *** overcome these hurdles,we used a peptide called pH low insertion peptide(pHLIP) to couple with DOX through disulfide bond and developed a pHLIP-SS-DOX *** reports have demonstrated that pHLIP has three states:soluble in water near neutral pH(state I),bound to the surface of a membrane near neutral pH(state H),and inserted across the membrane as an a-helix at low pH(state 111).In addition,the insertion process is unidirectional(C terminus in).The conjugate pHLIP-SS-DOX is expected to insert across the tumor cell membrane because of the acidic extracellular microenvironment of tumor,and then the intracellular reductive environment will break the disulfide bond to release DOX to cytosoL Furthermore,some reports show that attachment of a transporter to small molecule drugs can dramatically change their physical properties and,therefore,mode of cell entry,thereby avoiding Pgp-based *** we expected that the conjugate synthesized here might reverse multi-drug *** successfully synthesized the conjugate through a three-step synthetic reaction Flow cytometry and laser confocal microscope studies revealed that the uptake of the conjugate at low pH was more than that at neutral pH in both MCF-7 and MCF-7a *** the uptake of the conjugate in MCF-7 and MCF-7a cells had no significant differences while the uptake of free DOX in MCF-7a cells was dramatically decreased compared with the uptake in MCF-7 *** summary,experiments in vitro demonstrated that pHLIP-SS-DOX can translocate small molecules into cells and reverse multi-drug resistance.

Pure nanodrug,i.e.,nanocrystal（NC）,is a promising drug delivery system and has been used in formulating of water-insoluble drugs for oral drug *** significantly improved drug dissolution in GI endowed by NCs accounts for the enhanced oral absorption of water insoluble ***,efficient drug absorption is still difficult to achieve when NCs encounter epithelium barrier in ***,NCs may not completely dissolve in the GI after administration,and the undissolved NCs are difficult to uptake by the gastrointestinal epithelia cells;Secondly,some drugs have poor membrane permeability due to drug efflux pump transporters,such as ***,we are trying to formulate a functional Lipo-membrane encapsulated nanocrystals（Lipo-NC） aiming to enhance the transport of a BCS class Ⅳ drug（poor solubility,poor membrane permeability）,saquinavir,across intestinal epithelial *** saquinavir Lipo-NCs were prepared by hydration of lipid membrane with saquinavir nanocrystal *** particle size and Zeta-potential of Lipo-NC were 241.4±15.2 nm and 2.6±0.9 mV,*** Lipo-NCs showed a distinct core-shell structure observed by transmission electron microscope（TEM） and confocal laser scanning microscope（CLSM）.The release of drug was greatly restricted by the encapsulation of NC with lipid membrane,however,the lipid membrane encapsulation significantly enhanced the cellular uptake and transport of drug,which were 18.93-folds and 3.59-folds compared to coarse crystals and saquinavir NCs,*** Lipo-NCs circumvented P-gp efflux and used both caveolae- and clathrin-mediated transcytosis *** ex vivo experiments carried out in isolated rat intestine demonstrated that Lipo-NCs could be well absorbed by epitheliums and were found in lamina propria of intestinal villi,showing good transcytosis *** pharmacokinetic study in rats showed that the AUC of Lipo-NCs were 4.29-times and 1.77-times higher than that of coarse crys

The failure of sufficient accumulation of nanoparticles in tumor tissues is a primary problem to tumor therapy,which was because the extracellular matrix(ECM)produced by tumor and stromal cells and the poor tumor-target ability of *** ECM,containing collagen,hyaluronan and proteoglycans,can compress the tumor vessels and leads to impaired drug *** the angiotensin inhibitor,losartan,was pre-injected to increase the vessel permeability in advance by down-regulate the collagen I level,aiming at more paclitaxel loaded pH-sensitive liposome was distributed in tumor *** pH-sensitive liposome contained PEG5K-Hydrazone-PE and R8 peptide,PEG5K-Hydrazone-PE was synthesized via pH-sensitive hydrazone bond and used to shield R8 in blood circulation and expose it in tumor tissues(Fig.1).The cleavage ration of PEG5 K derivative reached to 23.81%and 17.65%in pH 7.4 and pH 8.0 after 4 h respectively,while the cleavages increased to 97.82%and 91.95%after 4 h incubation,demonstrating the PEG5 K derivative could satisfy the requirements of extracellular dePEGlyation of liposomes in vitro *** in vivo imaging confirmed that pre-injection of losartan could lead to more Cl-Lip distribute in tumor tissues,which was 1.22 times as the group only in treated with ***,the tumor inhibition rate of PTX-Cl-Lip in 4T1 bearing mice was 37.79%,after pre-injection of losartan,the inhibition rate increased to57.84%,meanwhile,the losartan didn't affect the tumor growth rate(Fig.2).All data show pre-injection of losartan could improve the distribution of Cl-Lip in tumor tissues and enhance antitumor ***,this could be a promising drug delivery strategy in treating tumors.

Purpose:To prepare docetaxel-loaded and chitosan-coated solid lipid nanoparticles(SLNs),and to study the oral absorption mechanism of ***:SLNs were prepared by an emulsion-solvent evaporation method with docetaxel(DTX) as a model drug and further modified with chitosan(CS) or chitosan quaternary ammonium salt(HACC) by electrostatic *** physical properties of SLNs evaluated and the stability of SLNs were also studied in simulated gastric and intestinal fluids(SGF and SIF).The in vitro release study was carried out in phosphate-buffered saline(pH 7.4)using a dialysis bag *** cellular uptakes of SLNs in Caco-2 and FAM monolayer models were analyzed using flow cytometry,fluorescence microscope and confocal microscope,*** cellular transport of SLNs in caco-2 and FAE monolayers were also quantitatively analyzed by a HPLC *** oral absorption mechanism of SLNs in Caco-2 monolayer was preliminarily studied by measuring c lauding-1,occluding and ZO-1 proteins using a western blot ***:The particle size of DTX-SLNs,CS-DTX-SLNs and HACC-DTX-SLNs were 160.5±1.2,318.1±4.7 and 311.6±22.8 nm,the zeta potential were-20.24±0.50,8.83±0.12 and23.10±0.20 mV,the drug encapsulation efficiency were all over 80%,the drug loading efficiency were 3.54±0.03,2.54±0.01 and 2.55±0.01%,*** SLNs showed uniform and round shape,and the surface became rough after modified with CS or ***-DTX-SLNs were stable in both SGF and SIF,while CS-DTX-SLNs were unstable in *** drug release from various SLNs was almost same with no significant difference between each *** uptake of SLNs was time dependent,and M cell tended to uptake SLNs through *** results of quantitative analysis showed that the percentage of cellular transport was HACC-DTX-SLNs>CS-DTX-SLNs>DTX-SLNs>DTX,and the transport amount in FAE monolayer was higher than in Caco-2 *** results of western blot showed that the expression of claudin-1
