目的 Knowledge of the molecular mechanisms on malignant tumors is very critical for the development of new treatmentstrategies like molecularly targeted *** last five years,many investigations suggest that stathmin is over-expressed in avariety of human malignant tumors,and potentially promote the occurrence and development of ***,downregulationof stathmin can reduce cell proliferation,motility and metastasis and induce apoptosis of malignant ***,anstathmin antagonist,such as a specific inhibitor(antibody,small molecule compound,peptide,or siRNA and so on),may be anovel strategy of molecular targeted *** review summarizes the research progress of latest 5 years on the role ofstathmin in tumorigenesis,the molecular mechanisms of stathmin on malignant tumors and the development of stathminantagonists as candidate anti-cancer *** suggests that continued investigations into the function of stathmin in theneoplasia could lead to more rationally designed therapeutics targeting stathmin for treating human malignant tumors.

目的 The purpose of the study is to observe the capacity of progesterone(PROG)to reverse the corticosteroid insensitivity and todetect its synergistic effects with glucocorticoids in protecting against the inflammation.方法 male C57/BL6 mice are divided into 4 parts as follows：O3+normal saline,O3+BUD,O3+BUD+low PROG and O3+BUD+high *** were killed for the mean linear intercept and airway inflammatory cells infiltration density and measurement ofthe numbers of inflammation cells and the inflammatory cytokines of bronchoalveolar lavage fluid(BALF),serum and lung tissuecollection to examine the expression of the inflammatory cytokines,NF-κB,glucocorticoids receptors(GR)and p38MAPK.

目的使用主要代表性过敏原诊断的方式排除因交叉反应及其他各种原因对结果的干扰,建立更简便快捷更精确的诊断方法,以便进行更有针对性更有效的治疗。方法使用粉尘螨主要代表性过敏原Der f 1,Der f 2以及泛过敏原/交叉过敏反应组分tropomyosin包板,依据UniCAP诊断的血清过敏级别及检测诸多影响参数来设计正交试验,并使不同级别的血清在96孔板上能均匀分布,设置阴性和阳性对照,依据ELISA读板的吸光值来判断病人血清阳性与否,并进行相应分级,使用统计学方法比较传统诊断方法与主泛过敏原联合诊断方法的准确性、灵敏度、特异性及MCC值。同时结合临床脱敏治疗病人随访,分析疗效与主-泛过敏原阳性与否的对应关系。结果使用ELISA进行组分诊断的结果与常规诊断存在一致性,但有部分病人对尘螨主要过敏原并不显示阳性;而部分病人对交叉反应过敏原反应阳性且对尘螨主要过敏组分反应性较低,这一部分病人的脱敏治疗疗效欠佳。结论主要代表性过敏原与泛过敏原联合诊断可以反映传统诊断方法的结果,同时能鉴别并排除传统方法中因交叉反应及其他因素的干扰,提高诊断准确率,为后续开展准确而高效的治疗提供重要依据。

目的通过心肺运动试验研究特发性肺纤维化(IPF)对患者通气有效性及其运动耐力的影响.方法对IPF患者和对照)进行常规肺功能检测、动脉血气分析和心肺运动试验.结果 IPF组FVC、FEV1、MVV、TLC、RV、DLCO均显著低于正常对照组,IPF组PaO2、SaO2低于正常对照组,而IPF组的肺泡动脉氧分压差则明显高于正常对照组.心肺运动试验结果显示,IPF组每分钟通气量与CO2排出量(VE/VCO2)的线性斜率、VE/VCO2最小值均显著高于正常对照组;IPF组4个时期的VE/VCO2和死腔气量与潮气量比均显著高于正常对照组(均P＜0.01);IPF组峰值运动负荷占预计值百分比、峰值摄氧量占预计值百分比显著低于正常对照组.IPF患者VE/VCO2@AT、VE/VCO2线性斜率及VE/VCO2最小值与静息时的肺CO弥散量占预计值百分比呈负相关,其峰值摄氧量占预计值百分比与VE/VCO2@AT、VE/VCO2线性斜率、VE/VC02最小值呈负相关. 结论特发性肺纤维化使患者的通气有效性明显降低,严重影响患者的运动耐力.结论特发性肺纤维化使患者的通气有效性明显降低,严重影响患者的运动耐力.

目的心血管系统损害是阻塞性睡眠呼吸暂停综合征的严重并发症,慢性间歇缺氧(Chronic intermittent hypoxia, CIH)是阻塞性睡眠呼吸暂停综合征的重要病理生理学机制,很多动物试验发现CIH可引起心肌肥大、心肌细胞凋亡、心肌纤维化等多种心血管系统损害,但目前其机制尚不清楚.Wnt/β-catenin信号通路在心血管系统发生及心血管疾病发病过程中发挥重要作用,在多种心血管系统疾病中可观察到此信号通路的异常.此项研究拟探讨Wnt/β-catenin信号通路在慢性间歇缺氧(Chronic intermittent hypoxia, CIH)诱导的小鼠心肌细胞凋亡中所发挥的作用.方法将32只四周龄雄性C57/BL小鼠随机分配为4组(每组8只)：CIH+生理盐水(normal saline ,NS)组, CIH+XAV-939组, sham CIH+NS组, 和sham CIH+ XAV-939组.缺氧暴露结束后用蛋白免疫印迹(Western blotting,WB)来检测小鼠心肌中GSK-3β和β-catenin蛋白的表达,用原位末端转移酶标记(Terminal deoxynucleotidyl transferase mediated dUTP-biotin nick end labeling,TUNEL)法检测心肌细胞凋亡情况,并用Masson染色检测心肌细胞纤维化程度.结果 CIH组小鼠心肌中GSK-3β活性降低而β-catenin的表达量增加;此外,CIH组小鼠心肌细胞凋亡显著增加.而Wnt/β-catenin通路抑制剂XAV-939的应用可显著改善CIH诱导的心肌细胞凋亡,减轻CIH诱导的GSK-3β活性抑制,抑制CIH诱导的β-catenin表达量增加.结论我们的研究提示Wnt/β-catenin信号通路可能在CIH诱导的小鼠心肌细胞凋亡中发挥重要作用,XAV-939可能通过干预Wnt/β-catenin信号通路而减轻CIH诱导的心肌细胞凋亡.

目的 Calponin,VEGF and IL-17 are functional proteins associated with asthma ***,whether calponin changes with asthma,and whether these proteins cross-link to each other,remain *** we try to find the connection between calponin,VEGF and t IL-17 to provide a new clue for understanding of the asthma pathogenesis.方法 This study employed a rat model of asthma by intraperitoneal injection of ovalbumin and aluminum *** volume,airway pressure,airway resistance,and bronchial smooth muscle layer thickness were evaluated for model ***,immunohistochemical,and western blot analyses were performed to determine the expressions of calponin,VEGF and IL-17 on rat smooth muscle cells.结果 The results showed that the animal model of asthma was successfully established,and the calponin expression was significantly inhibited and the VEGF and IL-17 expressions increased in both acute asthma and chronic asthma rats.结论 In asthma model,there is the abnormal calponin expression,providing a new method for the treatment of asthma.

目的 Cyclin dependent kinase 1 (CDK1) is an essential kinase that involves in a wide range of biological processes which are key to cell survival. It has been reported that CDK1 participates in homologous recombination repair induced by DNA double strand break (DSB) through functional phosphorylation of BRCA1 , and therefore cancer cells with compromised CDK1 activity shows hypersensitivity to inhibiton of PARP1,a bypath DSB repair protein . However,due to the complicated role of CDK1 in this regulatory nexus, impaired BRCA1 phosphorylation per se is not sufficient to explain the remarkable synergistc effect displayed by simutaneous inhibiton of CDK1 and *** incorporation of CDK1 inhibitors into cancer treatment remians controversary. Here We sought to find a supplemtary mechanism which backs potential usage of CDK1 inhibitors in cancer therapy.

目的 Pulmonary alveolar proteinosis(PAP) is a rare pulmonary disease. There are three clinically distinct forms of PAP： congenital, secondary, and *** PAP is associated with cancers, mostly with hematologic malignancies.方法 We report the first case of PAP that occured 3 months before a diagnosis of chronic myelomonocytic leukemia(CMML) and review advances in treatment of SPAP. 结果 A 66-year-old Chinese man presented with recurrent coughand progressive dyspnea was admitted to our *** chest computed tomographyscan disclosed bilateral ground-glass opacities with interlobular septal thickening in lower *** lung biopsy(TBLB) revealed PAS-positive panlobular diffuse areas of intra-alveolar granular material consistent with *** studies revealed peripheral blood monocytes＞3.6×103cells/uL and bone marrow aspirates showed monocytes 30％.He was dignosed with CMML and treated with γ-interferon and *** whole lung lavage, his symptoms relieved gradually and the radiological manifestation absorbed 3 months later. 结论 So for PAP patients, underlying disease including hematologic disorders should be considered.

目的 To investigate the role of natural killer T cell(NKT)in asthma of different severity.方法 In this study,we have investigated the functions of NKT cells in murine model of asthma induced by 1％ovalbumin(OVA)and 5％OVA.结果 Our results demonstrate that murine model of asthma induced by 5％OVA was more severe compared with that induced with 1％OVA.α-Galactosylceramide(α-GalCer)administration activated NKT *** the 1％OVA-induced asthma model,α-GalCer administration significantly reduced eosinophil proportions in bronchoalveolar lavage fluid(BALF);decreaed levels of IL-4,IL-5 in the BALF;reduced pathologic scores of bronchiole,perivascular and pulmonary interstitial inflammation through histopathological analysis of lung tissue section;and reduced airway ***,the above significance did not occur in the 5％OVA-induced asthma model administrated by α-GalCer.结论 These results suggest that NKT cells may serve as an adjuvant to reduce inflammatory response in 1％,but not 5％OVA-induced murine model of asthma.
