videomicroscopy is a promising tool combined with machine learning for studying the early development of in vitro fertilized bovine embryos and assessing its transferability as soon as possible. We aim to predict the embryo transferability within four days at most, taking 2D time-lapse microscopy videos as input. We formulate this problem as a supervised binary classification problem for the classes transferable and not transferable. The challenges are three-fold: 1) poorly discriminating appearance and motion, 2) class ambiguity, 3) small amount of annotated data. We propose a 3D convolutional neural network involving three pathways, which makes it multi-scale in time and able to handle appearance and motion in different ways. For training, we retain the focal loss. Our model, named SFR, compares favorably to other methods. Experiments demonstrate its effectiveness and accuracy for our challenging biological task.

Digital Holographic microscopy (DHM) can obtain three-dimensional (3D) information about the fine structure of an object by utilizing the phase information of coherent light. In DHM, during the process of reconstructing the shape of an object, phase information is extracted by windowing the sideband containing the phase information. The 3D information of the object is then obtained from the acquired phase information. When windowing the sideband in a narrow range, it can remove noise caused by the DC spectrum, but detailed shape information of the object is lost. On the other hand, when windowing in a wide range, detailed shape information of the object can be obtained, but noise from the DC spectrum increases. To solve this trade-off relationship, we propose a method of determining the range for windowing the sideband using a function and then averaging the windowed sidebands. This proposed method can reduce the impact of noise from the DC spectrum and enables the acquisition of the detailed shape information. In this paper, we explain the reconstruction algorithm of this method and compare it with conventional methods and the previous study, phase differences averaging (PDA).

Gabor domain optical coherence microscopy (GD-OCM) is an emerging tomographic imaging technique that overcome the depth of focus (DOF) limit of high numerical objective lens by acquiring multiple cross-section images corresponded with different focal planes of the objective along an axial direction and combining them to form an invariant high resolution cross-section image. We previously reported an alternative processing method of GD-OCM called spectral fusing GD-OCM (SF-GD-OCM). SF-GD-OCM extracts in-focus information directly from acquired spectral interference signals. The advantage of SF-GD-OCM over the original GD-OCM is that the Gabor filtering and fusing can be implemented in an acquisition hardware instead of a host computer. Here, we have investigated the implementation of SF-GD-OCM by using FPGA frame grabber. The experimental setup of the acquisition system as well as the implementation of the spectral Gabor fusion in FPGA device have been developed and verified. Results show that the data processing of SF-GD-OCM in FPGA enabled acquisition device is possible and could lead to improvement in overall imaging speed of GD-OCM.

Optical microscope image quality is directly dependent on the objective lenses selected. In this paper we investigate a method of improving image quality by applying subpixel shifts, i.e. displacements smaller than the resolving power of the objective lens. The increase in spatial resolution is achieved by a mathematical calculation based on the expression of the spectrum of discrete signals, which uses the apparatus of generalized functions. To produce high-resolution elements, it is required to divide the Fourier spectrum generated from a few subpixel shifted images by a coefficient termed the aperture function. The aperture function depends on the kind of lens and can be its characteristic stated in the microscope passport. This paper presents a technique for calibrating a low-resolution lens based on the analysis of images taken with higher quality lenses. After determining the aperture function, you can apply a low-resolution lens to obtain higher quality images similar to the original. Structurally, as the resolution of a micro lens increases, its field of view decreases. At the same time, lenses with lower magnification provide a wider field of view. From a manufacturing point of view, lenses with lower resolution are easier to build, allowing them to exhibit higher metrological performance. The paper also discusses an analytical expression for the image spectrum that can be used for image reconstruction in the field of optical microscopy. Application of the inverse Fourier transform to this expression allows to reconstruct the original image with improved resolution.

In recent years, research on remote sensing object detection has primarily focused on oriented box representation and optimization, while overlooking the importance of studying feature extraction networks, leading to inaccurate identification of some targets. The conventional feature extraction networks employ fixed-size convolutional kernels, which fail to capture different levels of visual features for targets of varying sizes. In this paper, we propose a mechanism called the 3D-Weight Guide Selective Kernel (3D-WGSK) to jointly utilize spatial and channel information of input features to derive three-dimensional weights. These weights are then employed for convolutional kernel selection, enabling adaptive receptive field acquisition for capturing features of targets with different sizes. We devise a network architecture named the 3D-WGSK Net for feature extraction in remote sensing object detection. It possesses the capability of multi-scale information fusion and adaptive kernel adjustment, effectively extracting crucial features of targets with varying sizes. Experimental results on the publicly available DOTA-v1.5 dataset demonstrate the superior detection performance of our method.

We have proposed virtual-phase-conjugation-based optical tomography (vPC-OT) using a virtual phase conjugation technology for single-shot and three-dimensional optical tomography. In vPC-OT, a random-spatial-phase-modulated probe beam is irradiated to the sample to be measured, and the complex amplitude of the signal composed of a superimposition of light reflected from each layer of the sample is measured. A three-dimensional tomogram of intensity and phase is obtained by reproducing the measured complex amplitude using a phase conjugate wave in a virtual optical system built in a computer. At this time, by changing the parameters of the virtual optical system, it becomes possible to obtain information of various tomographic planes from the data obtained with a single measurement. In the ideal virtual phase conjugate reproduction process, free space propagation can be assumed;however, in the actual measurement, due to the distortion of the waves and the surroundings of the sample to be measured, a mismatch will occur in modulation and demodulation, and the separation accuracy between different tomographic planes would be degraded. We perform an experiment to clarify the characteristics of vPC-OT in this situation. In this experiment, three-dimensional optical tomography is performed using an etching glass having a periodic structure of 30 mu m as a sample, and the phase distribution is measured quantitatively. Furthermore, by placing a cover glass in front of the object and performing the same measurement, we discuss the characteristics and performance of vPC-OT when there is an optical distortion around the sample to be measured.

The EU Horizon 2020 project»JointPromise« implies the conception and implementation of an end-to-end automated production platform for three-dimensional joint implants, paving the way for tissue-engineered implants able to regenerate deep osteochondral defects. Spheroid-based implants provide a novel approach in tissue engineering by aggregating progenitor cells into potent microtissues. After the differentiation of cartilaginous microtissues, functional joint implants are assembled via 3D bioprinting to match the complex structural organization of native cartilage tissue. As the automation approach of the project aims to overcome bottlenecks in manual production such as product variability, lack of scalability and high personnel costs, a high-throughput quality control system is crucial for the production of reliable Advanced Therapy Medicinal Products (ATMPs). By establishing not only a technical solution for the full digitization of the cell culture plates but also an intelligent image processing algorithm for the detection of the cell spheroids, relevant process parameters like size distribution and growth curves can be detected. Critical thresholds in spheroid growth are evaluated to minimize risks of carcinogenic tissue formation in vivo as well as to define harvest criteria to prevent inhomogeneous bioprinting results. In order to calculate the required throughput and elaborate optimization potentials of the automated spheroid production, voids in the cultivation vessel or disrupted aggregates due to media changes or transportation are detected. Ultimately, the high-speed-microscopy complies with the requirements of a high-throughput automated cell production platform to meet the rising demand for alternative therapeutic approaches in regenerative medicine. © 2023 The Authors. A compact, prismbased spectrograph was designed for 2-photon light sheet microscopy based on broadband, ultrashort optical pulses that corrects for chromatic aberrations and distortion. Gaussian or Bessel beams of broadband optical pulses may be used to create narrow cylindrical, nonlinear excitation volumes from which fluorophores emit characteristic spectra. In practice, a slit aperture is often used to optically section the excitation volume before being imaged onto the camera chip. Optical dispersion of the fluorescence in the transverse direction of the imaging slit allows for hyperspectral image acquisition. Hyperspectral imaging systems may be used to simultaneously image and segment multiple fluorescent reporters in biological tissue. However, current systems have properties which are undesirable for low-light microscopy including chromatic aberrations, distortion, low optical transmission, and large footprint which consumes precious laboratory real estate. Here, we present a novel spectrograph that has sufficient optical transmission, achromaticity, and distortion correction for microscopy of fluorescent reporters spanning the visible spectrum (400 650 nm). Across the spatial dimension of the excitation volume, the spectrograph has uniform separation of the spectral bands while maintaining a compact size and profile. A second order deconvolution algorithm is used to spectrally deconvolve overlapping fluorophores.

We have recently developed quantitative oblique back-illumination microscopy (qOBM), which enables full-field quantitative phase imaging (QPI) of objects embedded in a thick scattering medium. This epi-mode technique makes use of multiple scattering as a source of transmissive illumination from within, allowing for rich structural detail based on optical path delay. We now produce quantitative 3D renderings of index of refraction with subcellular detail by computing a 3D transfer function of the entire optical system, including the multiply scattered illumination, and deconvolving it from a vertical stack of phase gradient contrast images. This approach allows truly non-invasive, label-free, tomographic quantitative reconstructions if index of refraction in thick scattering samples including thick tissue samples, at low cost and with simple operation, bringing QPI's unparalleled access to sub-cellular structural detail to previously unavailable domains of investigation.
