chemical threat detection has long been of interest to military, law enforcement, environmental agencies, and forensic investigators. Recently, as the propensity for both foreign and homegrown terrorism, illegal drug manufacture, and concern for environmental regulation continues to grow, the demand for rapid, portable chemical threat detection capabilities has increased dramatically. In particular, the ability to identify chemical threats (explosives, narcotics, toxic industrial chemicals, etc.) at a distance (standoff) is of special interest as it increases the safety of the end user during interrogation. Traditional analytical laboratory techniques such as high-performance liquid chromatography or gas chromatography coupled with mass spectrometry offer excellent sensitivity for detection and identification of trace amounts of threatening material. However, these techniques often lack the portability necessary for remote on-site interrogation as samples must be physically collected and brought to a laboratory for analysis. Vibrational spectroscopic techniques offer both the chemical identification and miniaturization capabilities required for portable, on-site chemical threat detection. Most importantly, spectroscopic techniques are inherently and uniquely standoff, where emitted or scattered photons are collected at some distance from the sample. The challenge then becomes miniaturizing the instrumentation while maximizing the distance at which accurate chemical detection can be made. Here we report on portable chemical threat detection instrumentation developed by Alakai Defense Systems, which employs deep ultra-violet Raman spectroscopy. We discuss the general system aspects such as basic optical design and ambient light rejection techniques. We also present data on the performance capabilities using several substances including actual narcotics and other compounds commonly used as cutting agents. remote detection of toxic chemicals of very low vapour pressure deposited on surfaces in form of liquid films, droplets or powder is a capability that is needed to protect operators and equipment in chemical warfare scenarios and in industrial environments. Infrared spectroscopy is a suitable means to support this requirement. Available instruments based on passive emission spectroscopy have difficulties in discriminating the infrared emission spectrum of the surface background from that of the contamination. Separation of background and contamination is eased by illuminating the surface with a spectrally tuneable light source and by analyzing the reflectivity spectrum. The project AMURFOCAL (Active Multispectral Reflection Fingerprinting of Persistent chemical Agents) has the research topic of stand-off detection and identification of chemical warfare agents (CWAs) with amplified quantum cascade laser technology in the long-wave infrared spectral range. The project was conducted under the Joint Investment Programme (JIP) on CBRN protection funded through the European Defence Agency (EDA). The AMURFOCAL instrument comprises a spectrally narrow tuneable light source with a broadband infrared detector and chemometric data analysis software. The light source combines an external cavity quantum cascade laser (EC-QCL) with an optical parametric amplifier (OPA) to boost the peak output power of a short laser pulse tuneable over the infrared fingerprint region. The laser beam is focused onto a target at a distance between 10 and 20 m. A 3D data cube is registered by tuning the wavelength of the laser emission while recording the received signal scattered off the target using a multi-element infrared detector. A particular chemical is identified through the extraction of its characteristic spectral fingerprint out of the measured data. The paper describes the AMURFOCAL instrument, its functional units, and its principles of operation.

Nanostructures of silicon are becoming increasingly important as they may combine new opto-electronic features or devices with CMOS technology. This enables the integration of e.g. nanowire sensors with readout electronics or nanoscale LEDs grown directly on Si wafers. A problem here is that nanostructures are often fabricated using a bottom-up approach necessitating a second deposition step of random nature or involving a tedious alignment procedure. This is because current lithography techniques do not allow sub 10 nm precision for direct top-down fabrication. In this presentation I will show examples of fabricating some generic silicon structures such as nanowires, nanopores and quantum dots using optical or e-beam lithography where a second process step is used to shrink feature sizes in a controlled way. Thus, the lithography step for positioning of nanostructures and the size determining process step become decoupled, as shown by three examples from our work: (i) Silicon nanowires were fabricated using a top-down approach using optical lithography. Thinning of the device layer to ∼20 nm enabled picomolar sensitivity for detection of biomolecules, avoiding traditional CVD growth of nanowires. These “nanoribbons” allows multiple parallel detection and direct electrical readout. (ii) Nanopores are interesting for single molecule detection and DNA sequencing. We have fabricated silicon membranes with arrays of nanopores down to sub 10 nm diameter. electro-chemical etching was used as the final size control. This allows multiple optical detection of DNA translocation. (iii) Finally, individual Si quantum dots have been fabricated using e-beam lithography and a final tuning of the core by oxidation in a “self-limiting” regime. This has allowed single-dot spectroscopy revealing quantum confined exciton recombination with ultra-sharp emission lines. Many other features such as blinking and spectral diffusion have been observed, similar to direct bandgap quantum dots.

Recent progress in fiber technology has enabled new laser designs along with all fiber lidar architectures. Their asset is to avoid free-space optics, sparing lengthy alignment procedures and yielding compact setups that are well adapted for field operations and on board applications thanks to their intrinsic vibration-resistant architectures. We present results in remote sensing for disaster management recently achieved with fiber laser systems. Field trials of a 3-paths lidar vibrometer for the remote study of modal parameters of buildings has shown that application-related constraints were fulfilled and that the obtained results are consistent with simultaneous in situ seismic sensors measurements. remote multi-gas detection can be obtained using broadband infrared spectroscopy. Results obtained on methane concentration measurement using an infrared supercontinuum fiber laser and analysis in the 3-4 mu m band are reported. For gas flux retrieval, air velocity measurement is also required. Long range scanning all-fiber wind lidars are now available thanks to innovative laser architectures. High peak power highly coherent pulses can be extracted from Er3+:Yb3+ and Tm3+ active fibers using methods described in the paper. The additional laser power provides increased coherent lidar capability in range and scanning of large areas but also better system resistance to adverse weather conditions. Wind sensing at ranges beyond 10 km have been achieved and on-going tests of a scanning system dedicated to airport safety is reported.

For years, scientists have used thermal broadband cameras to perform target characterization in the longwave (LWIR) and midwave (MWIR) infrared spectral bands. The analysis of broadband imaging sequences typically provides energy, morphological and/or spatiotemporal information. However, there is very little information about the chemical nature of the investigated targets when using such systems due to the lack of spectral content in the images. In order to improve the outcomes of these studies, Telops has developed dynamic multispectral imaging systems which allow synchronized acquisition on 8 channels, at a high frame rate, using a motorized filter wheel. An overview of the technology is presented in this work as well as results from measurements of solvent vapors and minerals. Time-resolved multispectral imaging carried out with the Telops system illustrates the benefits of spectral information obtained at a high frame rate when facing situations involving dynamic events such as gas cloud dispersion. Comparison of the results obtained using the information from the different acquisition channels with the corresponding broadband infrared images illustrates the selectivity enabled by multispectral imaging for characterization of gas and solid targets.

A second-generation long-wave hyperspectral imager based on micro-electro-mechanical systems (MEMS) technology is in development. Spectral and spatial encoding using a MEMS digital micro-mirror device enables fast, multiplexed data acquisition with arbitrary spectral response functions. The imager may be programmed to acquire spectrally selective contrast imagery, replacing more time-consuming hyperspectral data collection. A single-element detector collects encoded data and embedded real-time hardware generates imagery. An internal scanning mechanism enables rapid retrieval of full hyperspectral imagery. The resulting rugged, low-cost sensor will provide chemically specific imagery for applications in gaseous and surface contaminant detection, surveillance, remote sensing, and process control.

Persistent surveillance and collection of airborne intelligence, surveillance and reconnaissance information is critical in today's warfare against terrorism. High resolution imagery in visible and infrared bands provides valuable detection capabilities based on target shapes and temperatures. However, the spectral resolution provided by a hyperspectral imager adds a spectral dimension to the measurements, leading to additional tools for detection and identification of targets, based on their spectral signature. The Telops Hyper-Cam sensor is an interferometer-based imaging system that enables the spatial and spectral analysis of targets using a single sensor. It is based on the Fourier-transform technology yielding high spectral resolution and enabling high accuracy radiometric calibration. It provides datacubes of up to 320x256 pixels at spectral resolutions as fine as 0.25 cm(-1). The LWIR version covers the 8.0 to 11.8 mu m spectral range. The Hyper-Cam has been recently used for the first time in two compact airborne platforms: a belly-mounted gyro-stabilized platform and a gyro-stabilized gimbal ball. Both platforms are described in this paper, and successful results of high-altitude detection and identification of targets, including industrial plumes, and chemical spills are presented.

Traditionally, human toxicant bioavailability screening has been forced to proceed in either a high throughput fashion using prokaryotic or lower eukaryotic targets with minimal applicability to humans, or in a more expensive, lower throughput manner that uses fluorescent or bioluminescent human cells to directly provide human bioavailability data. While these efforts are often sufficient for basic scientific research, they prevent the rapid and remote identification of potentially toxic chemicals required for modern biosecurity applications. To merge the advantages of high throughput, low cost screening regimens with the direct bioavailability assessment of human cell line use, we re-engineered the bioluminescent bacterial luciferase gene cassette to function autonomously (without exogenous stimulation) within human cells. Optimized cassette expression provides for fully endogenous bioluminescent production, allowing continuous, real time monitoring of the bioavailability and toxicology of various compounds in an automated fashion. To access the functionality of this system, two sets of bioluminescent human cells were developed. The first was programed to suspend bioluminescent production upon toxicological challenge to mimic the non-specific detection of a toxicant. The second induced bioluminescence upon detection of a specific compound to demonstrate autonomous remote target identification. These cells were capable of responding to mu M concentrations of the toxicant n-decanal, and allowed for continuous monitoring of cellular health throughout the treatment process. Induced bioluminescence was generated through treatment with doxycycline and was detectable upon dosage at a 100 ng/ml concentration. These results demonstrate that leveraging autonomous bioluminescence allows for low-cost, high throughput direct assessment of toxicant bioavailability.
