In the middle 1990's the U.S. National Library sponsored the acquisition and development of the Visible Human Project ® data base. This image database contains anatomical cross-sectional images which allow the reconstruction of three dimensional male and female anatomy to an accuracy of less than 1.0 mm. The male anatomy is contained in a 15 gigabyte database, the female in a 39 gigabyte database. This talk will describe why and how this project was accomplished and demonstrate some of the products which the Visible Human dataset has made possible. I will conclude by describing how the Visible Human Project, completed over 20 years ago, has led the National Library of Medicine to a series of image research projects including an open source image processing toolkit which is included in several commercial products.

We describe an iterative reconstruction algorithm (HYPR-OSEM) which improves the signal-to-noise ratio (SNR) in single frame static imaging by incorporating the HighlY constrained back-PRojection (HYPR) de-noising directly within the ordered subsets expectation maximization (OSEM) algorithm. The proposed HYPR operator in this work operates on the target image(s) from each subset of OSEM and uses the sum of the preceding subset images as the composite which was updated every main iteration. 3 strategies were used to apply the HYPR operator in OSEM: i) within the image space modeling component of the system matrix for forward-projection only (HYPR-F-OSEM), ii) within the image space modeling component for both forward-projection and back-projection (HYPR-FB-OSEM), and iii) on the new image estimate after each OSEM subset update (HYPR-AU-OSEM). Resolution and contrast phantom simulations with various sizes of hot and cold regions were used to evaluate the performances of various forms of HYPR-OSEM with respect to OSEM and OSEM with a post reconstruction filter. Although the convergence in contrast recovery coefficients (CRC) obtained from all forms of HYPR-OSEM was observed to be slower than that from OSEM (with AU being the fastest among the 3 forms), HYPR-OSEM achieved up to 2-3 times lower noise level for a fixed CRC as compared to OSEM in the simulations conducted. When comparing to the filtered OSEM, HYPR-OSEM images contained similar noise level but with higher CRC due to the preservation of resolution in the HYPR-OSEM images. In summary, HYPR-OSEM can improve the SNR without degrading the resolution or contrast. In general, it can achieve: better accuracy in CRC at equivalent noise level than OSEM, better precision than OSEM, and better accuracy than filtered OSEM. In addition to static imaging, the proposed method can be applied to improve the image quality of any frame-based iterative reconstruction for both SPECT and PET.

Serial femtosecond nanocrystallography (SFX) is a form of x-ray coherent diffraction imaging that utilises a stream of tiny nanocrystals of the biological assembly under study, in contrast to the larger crystals used in conventional x-ray crystallography using conventional x-ray synchrotron x-ray sources. Nanocrystallography utilises the extremely brief and intense x-ray pulses that are obtained from an x-ray free-electron laser (XFEL). A key advantage is that some biological macromolecules, such as membrane proteins for example, do not easily form large crystals, but spontaneously form nanocrystals. There is therefore an opportunity for structure determination for biological molecules that are inaccessible using conventional x-ray crystallography. Nanocrystallography introduces a number of interesting image reconstruction problems. Weak diffraction patterns are recorded from hundreds of thousands of nancocrystals in unknown orientations, and these data have to be assembled and merged into a 3D intensity dataset. The diffracted intensities can also be affected by the surface structure of the crystals that can contain incomplete unit cells. Furthermore, the small crystal size means that there is potentially access to diffraction information between the crystalline Bragg peaks. With this information, phase retrieval is possible without resorting to the collection of additional experimental data as is necessary in conventional protein crystallography. We report recent work on the diffraction characteristics of nanocrystals and the resulting reconstruction algorithms.

Neuro-scientific studies are often aimed at imaging brain activity, which is time-locked to external stimuli. This provides the possibility to use statistical methods to extract even weak signal components, which occur with each stimulus. For electroencephalographic recordings this concept is limited by inevitable time jitter, which cannot be controlled in all cases. Our study is based on a cross-correlation analysis of trials to alignment trials based on the recorded data. This is demonstrated both with simulated signals and with clinical EEG data, which were recorded intracranially. Special attention is given to the evaluation of the time-frequency resolved phase-locking across multiple trails.

We present a method which utilizes advantages of fuzzy object representations and image processing techniques adjusted to them, to further increase efficient utilization of image information. Starting from a number of low-resolution images of affine transformations of an object, we create its suitably defuzzified high-resolution reconstruction. We evaluate the proposed method on synthetic data, observing its performance w.r.t. noise sensitivity, influence of the number of used low-resolution images, sensitivity to object variation and to inaccurate registration. Our aim is to explore applicability of the method to real image data acquired by Transmission Electron Microscopy, in a biomedical application we are currently working on.

Iterative algorithm has good performance as it does not need complete projection data in 3D image reconstruction area. It is possible to be applied in BGA based solder joints inspection but with low convergence speed which usually acts with x-ray Laminography that has a worse reconstruction image compared to the former one. This paper explores to apply one projection classification based method which tries to separate the object to three parts, i.e. solute, solution and air, and suppose that the reconstruction speed decrease from solution to two other parts on both side lineally. And then SART and CAV algorithms are improved under the proposed idea. Simulation experiment result with incomplete projection images indicates the fast convergence speed of the improved iterative algorithms and the effectiveness of the proposed method. Less the projection images, more the superiority is also founded.

In this paper, submillimeter three dimensional tomography imaging of paramagnetic contaminants flow rate in multiphase flow pipelines is presented. The device, which is based on Magnetic Particle Imaging (MPI), consists of an array of twelve coils and a pair of permanent magnets and is not influenced with the other phases that constitute the crude oil (e.g. oil, water, sand, and gas) and which are mainly diamagnetic materials. The concentration of the paramagnetic particles can be measured in a three dimensional volumetric space with high spatial and temporal sensitivities which are proportional to the strength of the applied magnetic field. This is also influenced by the size and distribution of the particles and the anisotropy of the permanent magnet. To increase the sensitivity and improve the spatio-encoding field, a two dimensional Linear Field Scanning (LFS) technique coupled with a two dimensional excitation field is proposed. The results demonstrate that the technique would constitute a breakthrough in the area of solid flow measurements and imaging.

In Region-Of-Interest(ROI) tomography, image reconstruction is an ill-conditioned inverse problem due to the presence of additive noise and incomplete projection data. To improve reconstructed image quality, this paper is aimed at reconstructing a small portion of an object from noisy observations of its projections sampled in and near the ROI. The proposed method can divided into two steps to reconstruct ROI of an object from a set of its noisy projection lines that passed through ROI. First, we introduce filter matrices to filter noisy projection data in multiresolution local tomography. Second, we apply total variation(TV) methods to make reconstructed image deblurring. Experimental results show that the proposed method outperforms state-of-the-art method. © 2015 Taylor & Francis Group, London.

We present a novel method for fast reconstruction of dynamic MRI from undersampled k-space data, thus enabling highly accelerated acquisition. The method is based on kernel regression along the manifold structure of the sequence derived directly from k-space data. Unlike compressed sensing techniques which require solving a complex optimisation problem, our reconstruction is fast, taking under 5 seconds for a 30 frame sequence on conventional hardware. We demonstrate our method on 10 retrospectively undersampled cardiac cine MR sequences, showing improved performance over state-of-the-art compressed sensing.

We present a new compressed sensing framework for reconstruction of incomplete and possibly noisy images and their higher dimensional variants, e.g. animations and light-fields. The algorithm relies on a learning-based basis representation. We train an ensemble of intrinsically two-dimensional (2D) dictionaries that operate locally on a set of 2D patches extracted from the input data. We show that one can convert the problem of 2D sparse signal recovery to an equivalent 1D form, enabling us to utilize a large family of sparse solvers. The proposed framework represents the input signals in a reduced union of subspaces model, while allowing sparsity in each subspace. Such a model leads to a much more sparse representation than widely used methods such as K-SVD. To evaluate our method, we apply it to three different scenarios where the signal dimensionality varies from 2D (images) to 3D (animations) and 4D (light-fields). We show that our method outperforms state-of-the-art algorithms in computer graphics and image processing literature.
