Systems biology is an approach to the analysis and prediction of the dynamic behaviour of biological networks through mathematicalmodelling based on experimental data. The current lack of reliable quantitative data, especially in the field of signal transduction, means that new methodologies in data acquisition and processing are needed. Here, we present methods to advance the established techniques of immunoprecipitation and immunoblotting to more accurate and quantitative procedures. We propose randomisation of sample loading to disrupt lane correlations and the use of normalisers and calibrators for data correction. To predict the impact of each method on improving the data quality we used simulations. These studies showed that randomisation reduces the standard deviation of a smoothed signal by 55% +/- 10%. independently from most experimental settings. Normalisation with appropriate endogenous or external proteins further reduces the deviation from the true values. As the improvement strongly depends on the quality of the normaliser measurement, a criteria-based normalisation procedure was developed. Our approach was experimentally verified by application of the proposed methods to time course data obtained by the immunoblotting technique. This analysis showed that the procedure is robust and can significantly improve the quality of experimental data.

In this paper a modem methodology of biplots is introduced as a means for monitoring the behaviour of process systems. This sophisticated methodology allows for the projection of high-dimensional data to a low-dimensional subspace that can be visualised by a human operator. The projections are highly graphical in nature, and rich in information regarding variation in process variables, correlations among these variables, as well as class separation, taking into account the multivariate character of the data. Moreover, as is shown by way of two case studies, process disturbances can be visualised and explored quantitatively by superimposing alpha-bags on biplots. (c) 2005 Elsevier Ltd. In this paper, we investigate human perception of the difference tone by two ultrasonic sounds. Although it has been generally agreed that the difference tone is a sound which does not exist physically but only in human subjective feelings, we doubt its truth. We therefore derive the human perception model for such tone and verify the model by physiological experiments using the magnetoencephalography (MEG).

The paper presents a critical appraisal of existing methods for modelling and simulation of CNC machine tool feed drives. Their goal is the development of mathematical models which adequately replicate the dynamic behaviour of the actual machines. The advantages and disadvantages of lumped parameter models and modular approach are discussed and the suitability of mixed, hybrid, distributed-lumped modelling is emphasised. Some applications of other methods to CNC machine tool feed drives are examined. The importance of a thorough study regarding the factors influencing the machine dynamic performance and the strategies for error compensation and correction is underlined. The achieved accurate simulations for multi-axis machine tool applications will be used for automatic tuning and condition monitoring purposes.

We argue that traditional systems engineering (TSE) is inadequate for engineering complex systems. This inadequacy becomes particularly problematic when the system in question is a System of Systems (SoS) whose behaviour depends on how human users of the component systems, anticipating the consequences of their own behaviour, interact with those systems and with each other. Considering its users to be part of the SoS renders such a system anticipative, and therefore necessarily complex. To be effective, the designer of an SoS must incorporate an understanding of its users' models of their contexts of use. However, the process of design then becomes reflexive, since it must include within itself models of its users' anticipations of its relation to the SoS's environment, including those of its designers. As a result, complex systems engineering (CSE) has to become a distinct discipline, involving explicit processes that occur at the time of use of an SoS, not only for negotiating shared meaning between component systems within peer-to-peer relationships, but also for considering the dynamic effects of differing uses of component systems on the behaviour of the SoS as a whole. By modeling users' relations to demand as well as to the behaviour and organisation of component systems, triply articulated modeling supports the negotiation of shared meaning and accommodates the reflexivity required of a CSE regimen.
