Extracellular vesicles (EVs) are potential biomarkers due to their important roles in intercellular communication. Current size exclusion chromatography (SEC) used in EVs isolation requires sophisticated chromatography systems for process monitoring which is difficult to deploy in point-of-care application. Herein, we report a low-cost microfluidic SEC device (µSEC) for integrated EVs isolation and monitoring using commercial off-the-shelf UV glue. Its modified T-junction injector with tunable resistance enables nanoliter sample injection (600 nL) within 1.5 s. Its built-in tubing connectors also allow flexible coupling with upstream/downstream modules to develop a real time process monitoring platform for EVs manufacturing or EVs-based clinical diagnostics. © 2021 MicroTAS 2021 - 25th International conference on Miniaturized Systems for Chemistry and Life Sciences. Photonics-based sensing in the mid-IR has been proven to be the key technology for in a plethora of applications ranging from environmental monitoring to industrial process control, medical diagnostics, water quality, medical and more. Compared to other sensing approaches, mid-IR spectroscopy-based sensing enables fast, reliable, and consumables-free operation for the detection of trace amounts to high concentration of the targeted analytes. The interest in the technology has been significantly increased due to the maturing of the Quantum Cascade Lasers (QCLs). QCLs offer an up to 2-orders of magnitude enhancement in the signal-to-noise ratio while enabling the direct access to the characteristic molecular fingerprint region of the targeted analytes. The Horizon Europe project M3NIR develops a modular sensing platform for the mid-IR, to significantly boost the technology in terms of performance (low limit of detection, multiple-species detection), footprint (co-integrating of lasers and components) reduction of energy consumption and cost. M3NIR will demonstrate its novel approaches for the monitoring of GHG and ships emission, detection of phosphates and nitrates in water and the breath analysis for health and well-being related applications.

Reasons for issues caused by latency spikes are described. Method of self-diagnostics of Internet Protocol (IP) audio codec software that is suitable for development, quality control, and operation and allows detecting these issues without external equipment and a special signal is demonstrated. The method of manual signal quality testing is mentioned. Based on that a method of automated testing using a second IP audio codec with specially modified software that allows detecting signal distortion in realistic network conditions is described. These methods simplify software quality control and accelerate the development of new versions of software, allowing detection of issues that are hard to detect with other methods.

The article gives the basic concepts of reliability, performance, durability, considers the issues of changing the technical state of the machine during operation. The models under consideration have the ability to introduce various perturbations in order to simulate physical defects, and numerical simulation on a computer made it possible to obtain solutions in the form of temporary realizations of oscillations for different states of the simulation object. The cooperative use of the full-scale method and mathematical modeling made it possible to reduce the volume of full-scale experiments and apply statistical methods of analysis in the problem of classifying the technical condition by constructing mathematical models of the probability of failure and uptime during their operating time. The content of the main types of machine states is revealed, considerable attention is paid to the issues of identifying and influencing failures and the causes of failures in the machine, the reasons for the loss of performance during operation.

The article proposes to use the methods of applied optical spectroscopy for solving the problem of combustion processes control. Spectrometer is used as a control device, which examines optical radiation as a signal carrying spectroscopic information about the combustion process. The authors review spectroscopic methods used for controlling combustion processes and informative parameters characterizing combustion for each of these methods. The results of an experimental study on measuring the spectrum of radiation arising from the combustion of gaseous hydrocarbon fuel are presented. The spectroscopic informative parameters characterizing the combustion process of hydrocarbon fuel are determined. © 2021 IEEE.

The article is devoted to the current and promising direction of the implementation of the Industry 4.0 program based on the introduction of artificial intelligence (AI) approaches in the industrial production of the oil and gas industry. A review of the application of modern bioinspired AI approaches for solving problems of control and diagnostics of industrial equipment is carried out. The application of the approach of artificial immune systems (AIS) and various modified AIS algorithms is substantiated in the creation of innovative and competitive applications for solving various specific engineering problems and creating a modern highly efficient technology for automating production processes at oil refineries. The theoretical foundations for the creation of an intelligent innovative technology based on a unified artificial immune system (UAIS) for distributed systems for complex objects control using modern computer technology have been developed. Definitions of UAIS and immunological homeostasis are given. Unification is carried out with the aim of reducing the variety of modified AIS algorithms and their optimal use for solving the assigned tasks for the technological processes control. The classification and assessment of the effectiveness of the application of modified AIS algorithms is carried out. The paper considers the application of the principles of self-organization for the automation of the business process of industrial safety inspection (ISI). Self-organization refers to the process of ordering elements (components) in an information system due to internal factors, without specific external influence. Self-organization is parametric and structural in nature. The architecture of the system as a whole and the main aspects of the main module, namely, scheduler, which provides self-organization of an open multicomponent information system of the ISI, are described. An ontological model of the object of expertise, the task, the method, and the operation that implements it, as the basis of self-organization, is proposed. The model and algorithms for implementing the scheduler are detailed. In particular, the algorithms of the main operations are described: the formation of the task description, the methodology for solving the problem, and the "intelligent" task execution. It also lists the principles of formation and describes the main local rules of the knowledge base of the scheduler, which are responsible for describing the methodology, the decision process, the coordination of expert opinions, and self-learning. A conceptual description of the implementation of the scheduler based on the components of the software platform is given. Some results of application of the considered approach for the problem of technical diagnostics of ISI are given: fragments of the generated software, forms of the user interface demonstrating the description of the object of expertise, local rules, methods and results of self-organization.

Surveillance testing is a key strategy to control the spread of COVID-19. Unlike the gold standard testing method, quantitative reverse transcription polymerase chain reaction (RT-qPCR), CRISPR diagnostics have recently become a more appealing alternative as they are proven to be faster, simpler, and more affordable. However, the current CRISPR diagnostic readouts are typically non-quantitative, making them error-prone and lacking crucial information of viral load. To further improve the CRISPR diagnostic method, we have developed a custom computer vision algorithm that works in complement to common transilluminators to process fluorescence images of the diagnostic samples, quantify their fluorescence signals, and assign the test results. Our analysis showed that the quantified fluorescence intensity was directly correlated to the sample viral load, useful information for transmissibility and disease severity. Verified through laboratory and clinical samples, our algorithm accurately discriminated the samples with the viral RNA as low as 6.25 copies/uL, and correctly classified nasopharyngeal swab (NP swab) samples with 100% accuracy. Our work serves as a potential technique to improve the accuracy of CRISPR diagnostics of COVID-19 and promote rapid testing vital to the containment of the ongoing pandemic. © 2021 IEEE.
