Gene delivery vectors based on adeno-associated virus (AAv) have been utilized in a large number of gene therapy clinical trials, which have demonstrated their strong safety profile and increasingly their therapeutic efficacy for treating monogenic diseases. For cancer applications, AAv vectors have been harnessed for delivery of an extensive repertoire of transgenes to preclinical models and, more recently, clinical trials involving certain cancers. This review describes the applications of AAv vectors to cancer models and presents developments in vector engineering and payload design aimed at tailoring AAv vectors for transduction and treatment of cancer cells. We also discuss the current status of AAv clinical development in oncology and future directions for AAv in this field. (C) 2016 Elsevier B.v. All rights reserved.

Multilayered TiAlN films on Ti6Al4v alloy for biomedical applications by closed field unbalanced magnetron sputter ion plating process

Ti6Al4v alloy has been widely used as a suitable material for surgical implants such as artificial hip joints. In this. study, a series of multilayered gradient TiAlN coatings were deposited on Ti6Al4v substrate using closed field unbalanced magnetron sputter ion plating (CFUBMSIP) process. Taguchi design of experiment approach was used to reveal the influence of depositing parameters to the film composition and performance of TiAlN coatings. The phase structure and chemical composition of the TiAlN films were characterized by X-ray diffractometry (XRD) and X-ray photoelectron spectroscopy (XPS). Mechanical properties, including hardness, Young's modulus, friction coefficient, wear rate and adhesion strength were systematically evaluated. Potentiodynamic tests were conducted to evaluate the corrosion resistance of the coated samples in Ringer's solution at 37 degrees C to simulate human body environment Comprehensive performance of TiAlN films was evaluated by assigning different weight according to the application environment S8, deposited by Ti target current of 8 A, Al target current of 6 A, bias voltage of -60 v and nitrogen content with OEM (optical emission monitor) value of 45%, was found to achieve best performance in orthogonal experiments. Depositing parameters of S8 might be practically applied for commercialization of surgical implants. (C) 2015 Elsevier B.v. All rights reserved.

We present the handheld scanning probes that we have recently developed in our current project for biomedical imaging in general and for optical Coherence Tomography (OCT) in particular. OCT is an established, but dynamic imagistic technique based on laser interferometry, which offers micrometer resolutions and millimeters penetration depths. With regard to existing devices, the newly developed handheld probes are simple, light and relatively low cost. Their design is described in detail to allow for the reproduction in any lab, including for educational purposes. Two probes are constructed almost entirely from off-the-shelf components, while a third, final variant is constructed with dedicated components, in an ergonomic design. The handheld probes have uni-dimensional (1D) galvanometer scanners therefore they achieve transversal sections through the biological sample investigated - in contrast to handheld probes equipped with bi-dimensional (2D) scanners that can also achieve volumetric (3D) reconstructions of the samples. These latter handheld probes are therefore also discussed, as well as the possibility to equip them with galvanometer 2D scanners or with Risley prisms. For galvanometer scanners the optimal scanning functions studied in a series of previous works are pointed out;these functions offer a higher temporal efficiency/duty cycle of the scanning process, as well as artifact-free OCT images. The testing of the handheld scanning probes in dental applications is presented, for metal ceramic prosthesis and for teeth.

The origami-based folded core has a broad prospect in applications. However, the fabrication difficulties limit its developments due to the complicate structures with multiple zigzags and fluctuations. In this study, several feasible methods to fabricate polymer origami-based folded core were proposed. Based on the analysis of folded structural features, the vacuum forming process of polymer v-type folded core was deeply investigated. Wall thickness distributions of the folded structure on polyvinyl chloride (PvC) and polyethylene terephthalate (PET) sheets were measured. The results showed that the wall thickness along the straight crease line presents a single peak distribution, while that along the zigzag one shows a dual peak distribution, and the wall thickness of the top zigzag crease line is more uniform and large than that of the bottom one. The mold temperature, polymer sheet temperature and material properties of polymer have great influence on product thickness. vacuum forming is a feasible way to fabricate polymer origami-based folded core, and the high-quality fabrication of folded structure may be accomplished by designing more reasonable forming mold and selecting optical process parameters.

为了满足POCT仪器体积小、操作简单、携带方便、结果报告及时化的检验模式新要求,设计并开发一款基于Y形光纤的便携式免疫荧光定量检测仪。该设备以恒流源驱动激光二极管作为激发光源,Y形光纤作为光信号传输介质,光电二极管作为荧光信号接收传感器,替代复杂庞大的传统共聚焦光学系统,使光路设计及信号采集模块小型化;设计Sallen-Key拓扑结构的二阶有源低通滤波器对信号进行放大滤波处理,以减少杂波干扰,提高系统灵敏度;采用STM32F103微处理器为主控芯片,控制步进电机驱动机械扫描模块、光电信息采集模块、数据处理及显示模块完成免疫层析试纸条的荧光值定量检测。AD采集精度实验显示,该系统的AD采样偏差为10μv,标准差为1.91E-06;实测荧光微球实验表明,荧光微球稀释倍数和荧光强度信号之间的乘幂关系良好,相关系数为0.99,该系统检测灵敏度高,重复性好。

CONTROLLER design is more troublesome in aerospace systems due to, inter alia, diversity of mission platforms, convoluted nonlinear dynamics, predominantly strict mission and resource constraints, and demands for guaranteed operability within a wide range of operating conditions that can undergo structural or unexpected changes. Most of the space systems (e.g., planetary observers, rovers, space telescopes, spacecraft optical systems, etc.) require studious design, production, and testing processes. Indeed, space systemsare required to endure a wide spectrumof environmental changes with limited resources and are typically subject to partial, highly expensive, or even nonexistent service or repair. Clearly, aerospace missions induce high cost, require long development times as well as long mission lifespan, and demand high-fidelity operation so that, not surprisingly, related control tasks are significantly more demanding in aerospace compared to many other industries. CHEOPS (CHaracterizing ExOPlanets Satellite) is the first ESA Small Mission as part of the ESA Cosmic vision program 2015-2025. The mission was formally adopted in early February 2014 with a planned launch readiness end of 2017. The mission lead is performed in a partnership between Switzerland, led by the University of Bern, and the European Space Agency with important contributions from Austria, Belgium, France, Germany, Hungary, Italy, Portugal, Spain, Sweden, and the United Kingdom. The mission is dedicated to searching for exoplanetary transits by performing ultrahigh precision photometry on bright starts already known to host planets whose mass has been already estimated through ground based observations. The instrument is an optical Ritchey-Chretien telescope of 30 cm clear aperture using a single CCD detector. The optical system is designed to image a de-focused PSF onto the focal plane with very stringent stability and straylight rejection requirements providing a Fov of 0.32 degrees full cone. The system design is adapted to meet the top-level science requirements, which ask for a photometric precision of 20ppm, in 6 hours integration time, on transit measurements of G5 dwarf stars with v-band magnitudes in the range 6 <= v <= 9 mag. Additionally they ask for a photometric precision of 85 ppm in 3 hours integration time of Neptune-size planets transiting K-type dwarf stars with v-band magnitudes as faint as v=12 mag. Given the demanding schedule and cost constrains, the mission relies mostly on components with flight heritage for the platform as well as for the payload components. Nevertheless, several new developments are integrated into the design as for example the telescope structure and the very low noise, high stability CCD front end electronics. The instrument and mission have gone through critical design review in fall 2015 / spring 2016. This paper describes the current instrument and mission design with a focus on the instrument. A very low peak voltage GaAs/AlAs resonant tunnelling diode (RTD) grown by molecular beam epitaxy (MBE) has been studied in detail. Excellent growth control with atomic-layer precision resulted in a peak voltage of merely 0.28 v (0.53 v) in forward (reverse) direction. The peak current density in forward bias is around 15.4 kA/cm2(± 7%). As for reverse bias, the peak current density is around 22.8 kA/cm2(± 4%) which implies excellent scalability. In this work, we have successfully demonstrated the design and fabrication of a GaAs/AlAs RTD by using a conventional optical lithography and chemical wet-etching with very low peak voltage suitable for application in low dc input power RTD-based oscillators. © 2016 IEEE.

TAIPAN will conduct a stellar and galaxy survey of the Southern sky. The TAIPAN positioner is being developed as a prototype for the MANIFEST instrument on the GMT. The design for TAIPAN incorporates 150 optical fibres (with an upgrade path to 300) situated within independently controlled robotic positioners known as Starbugs. Starbugs allow precise parallel positioning of individual fibres, thus significantly reducing instrument configuration time and increasing the amount of observing time. Presented is an engineering overview of the UKST upgrade of the completely new Instrument Spider Assembly utilized to support the Starbug Fibre Positioning Robot and current status of the Starbug itself.
