Nanosecond pulsed electric fields (nsPEFs) with their wide frequency content, are recently known to be able to interact with cellular and subcellular membranes inducing reversible membrane permeabilization for biomedical applications. To improve treatments efficacy, lipid vesicles are studied and designed as smart delivery system able to respond to nsPEFs. In this work, authors report the study of an experimental bench suitable for in vitro exposure of cells and liposome nanocarriers to nsPEFs, compliant with the wideband requirements for nanosecond pulses. A multiphysics modelling able to predict cell and liposome nanoelectroporation is proposed to define the characteristics of signals to be used in experiments. © 2021 URSI.

Patient-specific lung-mechanics during mechanical ventilation (MV) can be modelled via using fully ventilated/controlled waveforms. However, patient asynchrony due to spontaneous breathing (SB) effort commonly exists in patients on full MV support, leading to variability in breathing waveforms and reducing the accuracy of identified, model-based, and patient-specific lung mechanics. This study aims to extract ventilated breathing waveforms from affected asynchronous breathing cycles using an automated virtual patient model-based approach. In particular, change of lung elastance over a pressure-volume (PV) loop is identified using hysteresis loop analysis (HLA) to detect the occurrence of asynchrony, as well as its type and pattern. The identified HLA parameters are then combined with a nonlinear mechanics hysteresis loop model (HLM) to extract and replicate the ventilated waveforms from the coupled asynchronous breaths. The magnitude of asynchrony can then be quantified using an energy dissipation metric, E-asyn, comparing the area difference of PV loops between model-reconstructed and original breathing cycles. A proof-of-concept study is conducted using clinical data from 2700 breathing cycles of two patients exhibiting asynchrony during MV. The reconstruction root mean square errors are within 5-10% of the clinical data for 90% of the cycles, indicating good and robust reconstruction accuracy. Estimation of E-asyn shows significant asynchrony magnitude for Patient 1 with E-asyn greater than 10% for over 50% breaths, while asynchrony occurrence for Patient 2 is lower with 90% breaths at E-asyn < 10%, which is a minimal asynchrony magnitude. These results match direct observation, thus validating the ability of the virtual patient model and methods presented to be used for a real-time monitoring of asynchrony with different types and magnitudes, which in turn would justify prospective clinical tests. A nonlinear time-delay realization for gastroparesis in patients with diabetes

The realization problem is fundamental in control systems theory and is well understood for linear systems. The situation is quite different for nonlinear systems with or without time delays. New results are given herein either as an abstract characterization of the class of delay-free nonlinear systems which are known to admit a realization, or by characterizing the existence of a realization for a wider class of retarded type or neutral type time delay nonlinear systems. The problem is motivated by the very practical modelling and realization problem of diabetes with or without gastroparesis. Up to 4% to 12% patients with diabetes are affected by the so-called gastroparesis which delays the digestion process. Several concurrent state space mathematical models do exist for diabetes and require a re-evaluation based on physiologic facts. These are reviewed herein to derive a (minimal) realization. The hypothalamic-pituitary-adrenal (HPA) axis orchestrates the physiological response stress. Moreover, the HPA axis exhibits prominent circadian activity and synchronizes peripheral circadian clocks to daily environmental cycles, thereby promoting homeostasis. Persistent disruption of homeostatic glucocorticoid circadian rhythmicity due to chronic stress exposure is correlated with the incidence of various pathological conditions including depression, diabetes and cancer. Allostatic habituation of the HPA axis can therefore confer fitness advantages by preventing the sustained dysregulation of glucocorticoid-responsive signaling pathways. However, such allostatic adaptation results in a physiological cost (allostatic load) that might impair the homeostatic stress-responsive and synchronizing functions of the HPA axis. We use mathematical modeling to characterize specific chronic stress-induced allostatic adaptations in the HPA network. We predict the existence of multiple personalized regulatory strategies enabling the maintenance of homeostatic glucocorticoid rhythms, while allowing for flexible HPA response characteristics. We show that this regulatory variability produces a trade-off between the stress-responsive and time-keeping properties of the HPA axis. Finally, allostatic regulatory adaptations are predicted to cause a time-of-day dependent sensitization of the acute stress response and impair the entrainability of the HPA axis Copyright (C) 2020 The Authors.

Traffic congestion on motorways is a serious threat for the economic and social life of modern society as well as for the environment, which calls for drastic and radical solutions. Conventional traffic management measures, currently applied, are valuable, but face limitations. During the last decades, a variety of vehicle automation and communication systems (VACS) have been developed and deployed, and many more are expected to appear in the near future. These systems provide a novel basis for a new generation of traffic management, which exploits emerging vehicle automation functions and connectivity channels to enable sensible traffic flow improvements in terms of efficiency and safety. A number of innovative concepts, tools and results that open up new horizons for traffic management research and practice in presence of VACS have been produced recently in the frame of TRAMAN21, an ERC Advanced Grant. This paper presents a collection of novel problems as well as of related traffic flow modelling, estimation and control developments for motorway traffic that can be used in the evolving traffic environment with VACS. Traffic control for freeway networks with sustainability-related objectives: Review and future challenges

Sustainability is a key point in the design and management of mobility and traffic systems, which affects also the development of traffic control strategies for freeway networks. According to sustainability-related concepts, freeway traffic controllers should be devised not only for maximally exploiting the road capacity and decreasing vehicles travel delays, but also for reducing pollutant emissions, fuel consumptions, accidents, noise, and so on. This paper analyses the state of the art of freeway traffic control strategies characterised by sustainability-related objectives, in particular referred to traffic emissions and traffic safety, by providing a classification of the research papers in this area. The final part of the paper highlights the main research challenges for modelling and control techniques brought by the introduction of emerging information and communication technologies, that are becoming more and more widespread and are transforming the concept of vehicles in intelligent and connected agents, able to measure the traffic state and to implement specific traffic control policies. This paper deals with the modelling, validation and control of the ESA space manipulator DELIAN. A numerical model accounting for joints and links flexibility is developed, and structural parameters are identified through experiments on the real robot. Then, the model is validated comparing the behavior of the simulated system with the real one. Moreover, an analysis aiming to understand the contribution of each elastic element to the overall flexibility is carried out. Finally, wave-based control technique is combined to the well-known P/PI scheme in order to reduce system vibrations. Especially, a velocity-based implementation of the WBC is introduced which provides superior performance with respect to the position-based one. Chemical-looping Combustion (CLC) has recently emerged as a promising technology to curb CO2 emissions. The novelty of CLC resides on its inherent ability of avoid direct contact between the fuel and the air, while producing a highly concentrated CO2 stream. This study presents a dynamic modelling and controllability study that demonstrates the technical feasibility of a fixed bed CLC reactor to produce a constant high temperature air stream during the oxidation stage. The heterogeneous model, which considers mass and heat transport resistances in the oxygen carrier particle and the bulk fluid phase, was validated using data reported in the literature. Also, a sensitivity analysis was conducted to gain insight on system's behaviour. Furthermore, an optimal control problem was formulated to identify optimal control profiles that can improve the efficiency of this process.
