Tourette syndrome (TS) is a childhood-onset neurobehavioral disorder and its pathophysiological mechanism remains elusive. At present, TS-related abnormalities in either structural connectivity (SC) or functional connectivity (FC) have extensively been described, and discrepancies were apparent between the SC and FC studies. However, abnormalities in the SC-FC correlation for early TS children remain poorly understood. In our study, we used probabilistic diffusion tractography and resting-state FC to construct large-scale structural and functional brain networks for 34 drug-naive TS children and 42 healthy children. Graph theoretical approaches were employed to divide the group-averaged FC networks into functional modules. The Pearson correlation between the entries of SC and FC were estimated as SC-FC coupling within whole-brain and each module. Although five common functional modules (including the sensorimotor, default-mode, fronto-parietal, temporo-occipital and subcortical modules) were identified in both groups, we found SC-FC coupling in TS exhibited increased at the whole-brain and functional modular level, especially within sensorimotor and subcortical modules. The increased SC-FC coupling may suggest that TS pathology leads to functional interactions that are more directly related to the underlying SC of the brain and may be indicative of more stringent and less dynamic brain function in TS children. Together, our study demonstrated that altered whole-brain and module-dependent SC-FC couplings may underlie abnormal brain function in TS, and highlighted the potential for using multimodal neuroimaging biomarkers for TS diagnosis as well as understanding the pathophysiologic mechanisms of TS.

Normal and aberrant cognitive functions are the result of the dynamic interplay between large-scale neural circuits. Describing the nature of these interactions has been a challenging task yet important for neurodegenerative disease evolution. The origin of Alzheimer's disease lies in the hippocampus and subsequently diffuses to the temporal, parietal and prefrontal cortices. Determining the sources of dementia is crucial to the prediction of the disease evolution and choice of treatment. State-of-the-art method for determining dementia progression are network diffusion models derived from the heat equation without diffusion sources. We propose a different research avenue based on epidemic modeling to localize the disease sources. These models may better characterize the empirical spread of dementia through brain regions. We explore an estimation algorithm based on a susceptible-infected (SI) epidemic algorithm and a network diffusion model for comparison purposes emulating the disease evolution from sources (susceptible) to non-recovered (atrophy, infected) areas. The goal is to identify the probable disease diffusion sources, which we accomplish via a ranking heuristic based upon steady-state convergence times. Graph centrality measures are employed to provide a baseline for further comparison. Our results applied on structural brain networks in dementia suggest that epidemic models are able to accurately describe the different node roles in controlling trajectories of brain networks comparably to the existing diffusion approach.

Accurate and in vivo characterization of structural, functional, and molecular characteristics of biological tissue will facilitate quantitative diagnosis, therapeutic guidance, and outcome assessment in many clinical applications, such as wound healing, cancer surgery, and organ transplantation. However, many clinical imaging systems have limitations and fail to provide noninvasive, real time, and quantitative assessment of biological tissue in an operation room. To overcome these limitations, we developed and tested a multiview hyperspectral imaging system. The multiview hyperspectral imaging system integrated the multiview and the hyperspectral imaging techniques in a single portable unit. Four plane mirrors are cohered together as a multiview reflective mirror set with a rectangular cross section. The multiview reflective mirror set was placed between a hyperspectral camera and the measured biological tissue. For a single image acquisition task, a hyperspectral data cube with five views was obtained. The five-view hyperspectral image consisted of a main objective image and four reflective images. Three-dimensional topography of the scene was achieved by correlating the matching pixels between the objective image and the reflective images. Three-dimensional mapping of tissue oxygenation was achieved using a hyperspectral oxygenation algorithm. The multiview hyperspectral imaging technique is currently under quantitative validation in a wound model, a tissue-simulating blood phantom, and an in vivo biological tissue model. The preliminary results have demonstrated the technical feasibility of using multiview hyperspectral imaging for three-dimensional topography of tissue functional properties.

It's widely known that HIV infection would cause white matter integrity impairments. Nevertheless, it is still unclear that how the white matter anatomical structural connections are affected by HIV infection. In the current study, we employed a multivariate pattern analysis to explore the HIV-related white matter connections alterations. Forty antiretroviral-therapy-naive HIV patients and thirty healthy controls were enrolled. Firstly, an Automatic Anatomical Label (AAL) atlas based white matter structural network, a 90 x 90 FA-weighted matrix, was constructed for each subject. Then, the white matter connections deprived from the structural network were entered into a lasso-logistic regression model to perform HIV-control group classification. Using leave one out cross validation, a classification accuracy (ACC) of 90% (P=0.002) and areas under the receiver operating characteristic curve (AUC) of 0.96 was obtained by the classification model. This result indicated that the white matter anatomical structural connections contributed greatly to HIV-control group classification, providing solid evidence that the white matter connections were affected by HIV infection. Specially, 11 white matter connections were selected in the classification model, mainly crossing the regions of frontal lobe, Cingulum, Hippocampus, and Thalamus, which were reported to be damaged in previous HIV studies. This might suggest that the white matter connections adjacent to the HIV-related impaired regions were prone to be damaged.

Tourette syndrome (TS) is a childhood-onset neurobehavioral disorder. At present, the topological disruptions of the whole brain white matter (WM) structural networks remain poorly understood in TS children. Considering the unique position of the topologically central role of densely interconnected brain hubs, namely the rich club regions, therefore, we aimed to investigate whether the rich club regions and their related connections would be particularly vulnerable in early TS children. In our study, we used diffusion tractography and graph theoretical analyses to explore the rich club structures in 44 TS children and 48 healthy children. The structural networks of TS children exhibited significantly increased normalized rich club coefficient, suggesting that TS is characterized by increased structural integrity of this centrally embedded rich club backbone, potentially resulting in increased global communication capacity. In addition, TS children showed a reorganization of rich club regions, as well as significantly increased density and decreased number in feeder connections. Furthermore, the increased rich club coefficients and feeder connections density of TS children were significantly positively correlated to tic severity, indicating that TS may be characterized by a selective alteration of the structural connectivity of the rich club regions, tending to have higher bridging with non-rich club regions, which may increase the integration among tic-related brain circuits with more excitability but less inhibition for information exchanges between highly centered brain regions and peripheral areas. In all, our results suggest the disrupted rich club organization in early TS children and provide structural insights into the brain networks.

We are developing and evaluating choline molecular imaging with positron emission tomography (PET) for monitoring tumor response to photodynamic therapy (PDT) in animal models. Human prostate cancer (PC-3) was studied in athymic nude mice. A second-generation photosensitizer Pc 4 was used for PDT in tumor-bearing mice. MicroPET images with 11C-choline were acquired before PDT and 48 h after PDT. Time-activity curves of 11C-choline uptake were analyzed before and after PDT. The dynamics of large-scale neural circuits is known to play an important role in both aberrant and normal cognitive functioning. Describing these phenomena is extremely important when we want to get an understanding of the aging processes and for neurodegenerative disease evolution. Modern systems and control theory offers a wealth of methods and concepts that can be easily applied to facilitate an insight into the dynamic processes governing disease evolution at the patient level, treatment response evaluation and revealing some central mechanism in a network that drives alterations in these diseases. Past research has shown that two types of controllability - the modal and average controllability - are key components when it comes to the mechanistic explanation of how the brain operates in different cognitive states. The average controllability describes the role of a brain network's node in driving the system to many easily reachable states. On the other hand, the modal controllability is employed to identify the states that are difficult to control. The first controllability type favors highly connected areas while the latter weakly connected areas of the brain. Certain areas of the brain or nodes in the connectivity graph (structural or functional) can act as drivers and move the system (brain) into specific states of action. To determine these areas we apply the novel concept of exact controllability and determine the minimum set and the location of driver nodes for dementia networks. Our results applied on structural brain networks in dementia suggest that this novel technique can accurately describe the different node roles in controlling trajectories of brain networks, and show the transition of some driver nodes and the conservation of others in the course of this disease. © 2020 spie
