In order to visualize sugar content of a melon, the relationship between sugar content and absorption spectra was investigated using a near-infrared (NIR) spectrometer. The absorbance at 676nm, which is close to the chlorophyll absorption band, had a high inverse correlation with sugar content. A high-resolution cooled CCD imaging camera with the band-pass filter of 676nm was used to capture the spectral absorption image. The calibration method for converting the absorbance on the image into Brix sugar content was developed in accordance with MR techniques. Applying this method to each pixel of the absorption image, a color distribution map of the sugar content was constructed. In addition, a special slicing device that can cut a melon in each 5mm thickness was developed in order to create a 3-D image of sugar content distribution.

A lens-less holographic microscopy is developed for observation of high-resolution 4-D (spatio-temporal) images. A complex-amplitude in-line hologram with a large numerical aperture is extracted from a large off-axis hologram by applying one-shot digital holography. An angular spectrum method is developed for fast numerical reconstruction of precise 3-D image from the in-line hologram with a large numerical aperture. 3-D intensity and phase images with no distortion, with a resolution higher than 1 mu m, and with a depth larger than 1mm are recorded and reconstructed by the present holographic microscopy. Possibility is demonstrated to realize 4-D measurement of microorganisms swimming in water.

Using an acousto-optic beam deflector and a line scan camera, we constructed a non-mechanical, slit-scanning confocal microscope. It generates two-dimensional 512x512 images with speed of 60 frames/sec, which can be easily expanded, to 100 frames/sec. The measured axial and lateral resolutions of the system are 3.3 mu m and < 1 mu m, respectively, with a 50X objective. This simple design can produce frames rates faster than other commercial products with comparable resolution, which may be useful for analysis of rapid interactions in various biological applications.

Structured Illumination microscopy is a simple and effective method to remove out-of-focus light in widefield fluorescence microscopy. Neil et al. originally proposed a simple square-law method for calculating the optically sectioned image from the three raw images with the structured illumination pattern super-imposed. However, the Neil method does not make the most efficient use of the three raw images. The three structured illumination images can also be used to separate three copies of the image covering shifted regions of frequency space in a similar manner to that developed by Gustafsson et al. These can then be combined using a generalized Wiener filter to create an image with a well-behaved optical transfer function in which the missing cone has been filled in, providing optical sectioning. Here, we compare the Neil and Gustafsson methods and show that the Gustafsson method provides an image with higher fidelity and a better Signal to Noise Ratio (SNR) at low photon counts. We apply the two methods to images of fluorescent beads and GFP labeled septins in Aspergillus nidulans.

We use a new illumination technique that let us image bio-molecules down to 50nm with optical microscopy. A combination of e-beam lithography (EBL), laser ablation, reactive ion beam etching (RIBE) and different wet etching techniques was used to fabricate different transparent three-dimensional structures in a three-layer system (Si3N4, SiO2, Si3N4) on quartz specially sized for the trapping of living neural cells under physiological conditions.
