Exploring technical improvements for environmental nucleic acids-based biodiversity assessment and management in coastal ecosystems

Assessing and conserving marine biodiversity remain critical global challenges, particularly in highly disturbed coastal regions. The use of environmental DNA (eDNA)-metabarcoding has revolutionized biodiversity assessment and management;however, the prevalence of both false positives and negatives continues to be a significant concern. To address these technical errors, we tested two potential methodological improvements in the highly disturbed Guangdong-Hong Kong-Macao Greater Bay Area: (1) the use of random whole-genome amplification (WGA) to reduce false negatives derived from low eDNA concentration, and (2) the application of environmental RNA (eRNA)-metabarcoding to mitigate false positives arising from eDNA contamination by human activities. Using fish communities as our target, we found that WGA enhanced downstream PCR amplification for metabarcoding but significantly reduced the detection of rare taxa, altered community structure, and increased false negatives (p < 0.001 for all tests). Interestingly, WGA led to higher levels of false negatives in more biodiverse communities. eDNA-metabarcoding revealed that 20.9%-23.6% of detected taxa were pure freshwater species (false positives) incapable of surviving in estuarine and coastal regions, highlighting the often-overlooked eDNA contamination in disturbed coastal ecosystems. In contrast, eRNA-metabarcoding significantly reduced false positives (p < 0.001), with error taxa accounting for only 2.5%-6.3% of all detections. Comparisons between eDNA and eRNA metabarcoding further revealed differences in their rare taxa recovery capacity. The findings provide critical insights into method selection for biodiversity assessment and management in highly disturbed coastal regions and highlight the need for further technical improvement of eDNA and eRNA-based biodiversity monitoring and conservation in aquatic ecosystems.

Amphiphilic aminated fullerenes have a broad margin of safety and significant antitumor ***,we develop a simple and versatile synthesis strategy for tetraamino-[60]fullerene epoxide(C_(60)(NR^(1)R^(2))_(4)O)using C_(60)Cl_(6)as a precursor,which notably reduces the reaction time to less than 1 h while retaining a high yield of over 80%with both cyclic and linear secondary amine substrates even at the gram *** molecular structure of C_(60)(NR^(1)R^(2))_(4)O is first validated by single-crystal diffraction,and a two-step reaction mechanism comprising nucleophilic substitution of Cl and the oxidative elimination of Cl_(2)is proposed based on experimental verification and density functional theory simulation.A set of water-soluble aminated C_(60)(NR^(1)R^(2))_(4)O was prepared in large quantities,and in vitro antitumor evaluation unveiled the critical role that terminal primary amino moieties of C_(60)(NR^(1)R^(2))_(4)O play in their antineoplastic *** work provides an effective synthesis method for aminated C_(60)(NR^(1)R^(2))_(4)O,facilitating the development of fullerene-derived tumor-targeted drugs.

中国兽类(即哺乳动物)种类繁多,对维持生态平衡发挥着重要的作用。自John ***于1829—1834年在我国广东开展兽类调查以来,近200年我国兽类分类及系统学研究取得了令世人瞩目的进步和发展。目前中国已知的兽类物种数已达686种,约占全世界兽类种数的10%,是世界上兽类物种多样性最丰富的国家之一。随着我国对生态环境保护的重视,生态环境日益改善,但全球气候变化、生境破碎化、人类活动增加及人兽共患重大疫情涌现等问题仍十分突出,兽类多样性调查及分类学研究的必要性越发明显。同时,兽类分类学这门古老而传统的学科也在不断引入各种新方法与技术,如整合分类学、标本数字化、模式标本测序、便携式测序技术及基于深度学习技术的物种识别鉴定等,分类学研究的成果及应用在近年得到了飞速发展。动物分类学作为传统的基础学科,是遗传学、生理学、生态学、医学、药学等现代生物学的基石。然而,由于学科特征和差异等原因,该学科近年来没有得到足够的重视,导致出现了学科萎缩和分类学人才后继无人的危机。因此,从国家层面对分类学、形态学等基础学科的人才培养、课题设置和资金投入等,予以特殊的政策支持,十分必要,也亟待解决。

Understanding the interaction between life history, demography and population genetics in threatened species is critical for the conservations of viable populations. In the context of habitat loss and fragmentation, identifying the factors that underpin the structuring of genetic variation within populations can allow conservationists to evaluate habitat quality and connectivity and help to design dispersal corridors effectively. In this study, we carried out a detailed, fine-scale landscape genetic investigation of a giant panda population from the Qinling Mountains for the first time. With a large microsatellite data set and complementary analysis methods, we examined the role of isolation-by-barriers (IBB), isolation-by-distance (IBD) and isolation-by-resistance (IBR) in shaping the pattern of genetic variation in this giant panda population. We found that the Qinling population comprises one continuous genetic cluster, and among the landscape hypotheses tested, gene flow was found to be correlated with resistance gradients for two topographic factors, slope aspect and topographic complexity, rather than geographical distance or barriers. Gene flow was inferred to be facilitated by easterly slope aspect and to be constrained by topographically complex landscapes. These factors are related to benign microclimatic conditions for both the pandas and the food resources they rely on and more accessible topographic conditions for movement, respectively. We identified optimal corridors based on these results, aiming to promote gene flow between human-induced habitat fragments. These findings provide insight into the permeability and affinities of giant panda habitats and offer important reference for the conservation of the giant panda and its habitat.

21世纪以来,中国电影在日本市场票房历经浮沉,在"洋邦之争"中偶为翘楚。因文化不兼容,并涉及域外传播中的情感自尊因素,导致了"文化刻奇"的出现;亦因东亚共同体的历史基因及生态文明下的文化贴合,产生出了胜于东瀛的效果。基于此,本文提出了强化中日文化IP锻造,产出记忆黏合效应;提升中国电影类型片的"海外增益",促进合拍片质量;强化电影节聚媒效应,培植国际主流电影;增进互联网聚合效应和跨国粉丝培养维系等举措,提升中国电影在日本的传播力,最终实现文化记忆视阈下的中日影像传播的"和合之美"。

Diacylglycerol kinase epsilon protects against renal ischemia/reperfusion injury in mice through Kruppel-like factor 15/klotho pathway

Although recent studies have indicated that mutations in the gene encoding diacylglycerol kinase epsilon (DGKE) result in some proteinuria related hereditary kidney diseases, the DGKE expression pattern in the kidney and its contribution to acute kidney injury (AKI) remain unknown. Therefore, the present study was designed to detect the role of DGKE in mice with AKI. DGKE expression was time-dependently altered in the kidneys of mice with renal ischemia/reperfusion injury (IRI). Compared with wild-type (WT) mice, DGKE- overexpressing mice (Rosa26-Dgke(+/+) ) exhibited protective effects against renal IRI, including reduced serum creatinine, blood urea concentration, tubular cell death and inflammatory responses as well as improved morphological injuries. Consistently, in vitro, DGKE overexpression in human renal proximal tubule (HK-2) cells also protected against oxygen-glucose deprivation (OGD)/reoxygenation-induced cell death. Mechanistically, DGKE regulated Klotho expression, at least partly via the transcription factor Kruppel-like factor (KLF) 15. Moreover, a significant reduction in DGKE was also found in kidneys from patients with ischemia-associated acute tubular necrosis (ATN). Collectively, our studies demonstrate that DGKE protects against AKI in mice at least partly through KLF15/Klotho signaling pathway, indicating that DGKE may present an innovative therapeutic strategy for treating patients with AKI.

Carriers Transport Behavior and Path for Hfox -Based Rram Devices With Improved Performance Bynitrogen Annealing

The nitrogen annealing can effectively improve the performance of the HfOx-based resistive random access memory (RRAM) devices. However, few more profound research explore the carriers transport behavior and path on nitrogen annealing improving the resistive switching (RS) properties of HfOx-based RRAM. In this work, we systematically investigated the effect of nitrogen annealing on RS performance in Ti/HfOx/Pt RRAM structure by I−V characteristics and the impedance spectroscopy (IS) under different temperatures. The results indicated that nitrogen annealed HfOx films could effectively decrease the operate voltage and enhance RS uniform and stability. The I−V characteristics showed that charge carriers were trapped and released through oxygen vacancy (VO) traps, and hopped conduction along these Vo traps when a Vo conductive filament (CF) was formed, which realized RS. The IS analysis exhibited that the carrier transport was along the VO CF at the grain boundary (GB). Moreover, appropriate GB resistance and capacitance relaxation time were crucial to obtain high-performance RRAM. Further, the first-principles simulation manifested that the VO formation energy at the GB decreased after annealing, which was the microscopic nature for improved device performance. © 2023, The Authors. 目的探讨老年股骨转子间骨折术后1年死亡风险的危险因素,构建生存预测模型。方法回顾性分析2020年1月至2022年9月新疆维吾尔自治区人民医院和信阳市人民医院收治的532例老年股骨转子间骨折患者的临床资料,记录患者的人口特征、实验室指标和手术变量,研究结局为术后1年死亡率。通过单因素和多因素Cox回归分析,筛选危险因素并依此构建预后模型。通过一致性指数(C-Index)、时间依赖性受试者工作特征(ROC)曲线、校准曲线、决策曲线(DCA)来评价模型的预测效能。结果多因素Cox回归分析显示:改良5项衰弱指数(mFi-5)(OR=1.338,95%CI:1.147~1.561,P<0.001)、是否入住重症监护室(ICU)(OR=1.694,95%CI:1.230~2.333,P=0.001)、术前血红蛋白(OR=1.281,95%CI:1.016~1.616,P=0.036)、手术等待时间(OR=1.570,95%CI:1.063~2.319,P=0.023)、年龄(OR=2.196,95%CI:1.712~2.816,P<0.001)是老年股骨转子间骨折术后1年死亡的独立危险因素,依此建立的预测模型C-Index:建模组为0.769(95%CI:0.723~0.818),验证组为0.715(95%CI:0.612~0.750)。生存预测模型建模组和验证组的时间依赖性ROC曲线下面积分别为0.802(95%CI:0.722~0.850)和0.718(95%CI:0.640~0.808)。建模组和验证组校准曲线显示模型拟合度良好。DCA显示:该模型具有明显的正向获益,临床适用性良好。结论mFi-5、是否入住ICU、术前血红蛋白、手术等待时间、年龄是老年股骨转子间骨折术后1年死亡的独立预测因子,依此建立的预后模型,预测预后价值准确,有助于临床医师快速个体化评估,促进合理预期。

Federated Learning (FL) since proposed has been applied in many fields, such as credit assessment, medical, etc. Because of the difference in the network or computing resource, the clients may not update their gradients at the same time that may take a lot of time to wait or idle. That's why Asynchronous Federated Learning (AFL) method is needed. The main bottleneck in AFL is communication. How to find a balance between the model performance and the communication cost is a challenge in AFL. This paper proposed a novel AFL framework VAFL. And we verified the performance of the algorithm through sufficient experiments. The experiments show that VAFL can reduce the communication times about 51.02% with 48.23% average communication compression rate and allow the model to be converged faster.
