Stem-Cell Derived Exosomal microRNAs as Biomarkers and Therapeutics for Pediatric Cardiovascular Disease

Purpose of ReviewIn recent years, several pre-clinical studies have demonstrated the therapeutic potential of stem cell-derived exosomes in the treatment of cardiovascular disease (CVD). Here, we evaluate their potential as biomarkers for the detection and monitoring of CVD, with a particular focus on pediatric heart *** FindingsExosomes isolated from stem cell sources, including mesenchymal stem cells (MSCs) and pluripotent stem cells (PSCs), benefit cardiovascular function, inflammatory responses, and angiogenesis in injured and diseased hearts. These exosomes carry a variety of cargo, such as proteins, lipids, and nucleic acids. However, the majority contain non-coding RNA *** of the existing literature for several non-coding RNAs and their relationship to CVD suggests that exosomes containing microRNAs (miRNAs) can serve as promising biomarkers for CVD due to their presence in circulation, ease of isolation, and therapeutic potential. These biomarkers are especially promising as screening and diagnostic tools for the early detection of pediatric and congenital heart disease.

Atrazine is a pesticide of the triazine class and is one of the most used pesticides in North America. We are investigating the toxicological effects of atrazine exposure on the crayfish species, Faxonius virilis. The effects of atrazine exposure on crayfish are of interest because crayfish are a keystone species. Atrazine has been known to negatively impact many aspects of crayfish health; atrazine has been demonstrated to diminish the ability for crayfish to locate food and find a mate, it affects the expression and activity of detoxification enzymes, and it induces histological changes that demonstrate the toxicity of the pesticide. We aim to quantitate atrazine and its metabolites in crayfish hepatopancreas tissue to assess correlation between atrazine levels and the previously observed behavioral and physiological effects of atrazine. A method for extracting atrazine and its metabolites from the crayfish hepatopancreas tissue was developed and validated. The method involves liquid chromatograph-mass spectrometry (LC-MS)-based quantitation using isotopic internal standards for each analyte. Spike recovery tests resulted in recoveries over 90% for all analytes. To further validate the revised method, the limit of detection, the limit of quantitation, and intermediate precision were also determined. The validated method was used to investigate accumulation of atrazine and its metabolites in crayfish exposed to 100 ppb atrazine for four days compared to an unexposed control group. Additionally, an untargeted LC-MS metabolomics analysis was performed on the control and exposed groups to explore potential metabolic changes following atrazine exposure.

Attention deficit/hyperactivity disorder (ADHD) is a neurodevelopmental condition affecting cognitive and social functions all over childhood. Monosodium glutamate (MSG) is a common food additive associated with ADHD-like symptoms in children. Nutraceuticals, like sesamol (SE) and astaxanthin (AST), or physical activity (PHA) were reported to possess beneficial effects on human health. Meanwhile, still their neuroprotective effect against ADHD has been poorly investigated. This study aimed to investigate the impact of SE, AST and PHA either separately or combined on ADHD-like behaviors induced by MSG in rat pups. Eighty-four male Sprague Dawley rat pups were randomly allocated into seven groups;control, MSG, (PHA + MSG), (SE + MSG), (AST + MSG), (SE + AST + MSG), and (COMB [PHA + SE + AST] + MSG) and treated for eight weeks. MSG-induced ADHD-like behavior was evaluated, via assessing behavioral outcomes;neurotransmitters' levels;five pathway biomarkers, coupled with histopathological and immunohistochemical studies. Rats exposed to PHA or treated with SE or AST either separately or combined exhibited enhanced attention, locomotor, and cognitive abilities, compared to MSG-intoxicated group. All treatments remarkably improved MSG-induced abnormalities in neurotransmitters' levels;biochemical markers;along with histological findings, via modulating HMGB1/RAGE/JAK-2/STAT-3, PI3K/AKT/CREB/BDNF, AMPK/SIRT-1 and PERK/CHOP pathways. Nevertheless, the combination of PHA with nutraceuticals (SE and AST) elicited more favorable effects in all measured parameters and histological findings, compared to other treated groups. In conclusion, this study revealed the superiority of the combination of nutraceuticals with PHA, over other standalone treatments, in amelioration of MSG-induced ADHD-like behaviors in rat pups, via fine-tuning of HMGB1/RAGE, PI3K/AKT/CREB/BDNF, AMPK/SIRT-1 and PERK/CHOP pathways.

We interviewed 12 Jordanian women who had experienced domestic violence (DV) and were receiving assistance at the Jordanian Women's Union (JWU). Our aim was to explore the history and factors supporting attainment of freedom from DV. Narratives revealed themes of DV toward girls;forced marriage;physical, psychological, or sexual abuse before and during marriage;and escalation and enduring DV. Escaping from DV required family and JWU support. In the context of a strongly patriarchal, religious society, we observed a process of resolution by shifting cultural values and themes of empowerment, with an undercurrent of suffering blamed on inequalities in the legal process.

Effects of different diets on glycemic control among patients with type 2 diabetes: A literature review

Background: Globally, type 2 diabetes mellitus (T2DM) is one of the most common diseases, and the incidence is gradually increasing. Diet management could improve the elevated HbA1c levels. Aim: This review aims to examine the effects of different types of diets on glycemic control in patients with T2DM. Methods: PubMed and Google scholar databases were searched. Reports published from the years 1991 to 2021 that focused on the effect of different diets on diabetes control were selected. Results: All the studies reported that the type of diet can affect insulin secretion and glycemic control in patients with diabetes. The recommended macronutrient distribution for patients with diabetes is 30% of fat, 45-60% of carbohydrates, and 15-35% of protein. Consuming more or less of these macronutrients may lead to changes in glucose metabolism and may affect insulin secretion. In fact, following alow-fat diet improves glycemic control and decreases HbA1c levels. Studies reported that a low-carbohydrate diet had the greatest effect on improving glycemic control and insulin parameters. A low-calorie diet reduced fasting plasma glucose, while a very low-calorie diet resulted in a long-term decrease in HbA1c level. A healthy diet free of processed foods and sugar, and rich in nutrients such as fiber, vitamins, and minerals contributed to maintaining controlled blood sugar and lipid plasma levels. Although studies show a low-salt diet improves blood pressure, which is common inT2DM, other findings show that restricting salt intake is associated with increased sugar consumption. Conclusion: Healthy diet with adequate intake of energy and low-fat and low-sugar foods can enhance the glycemic control and reduce T2DM complications.

Physician Attitude toward Their Attires and Laundering Habit Changes during the COVID-19: A Cross-Sectional Survey in a Tertiary Care Center

Patient safety is of utmost importance and every effort is to be made to reduce the risk of hospital-acquired infection. Contaminated attire is proposed as a mode of hospital infections spread. This study aims to assess the laundering habits, the perception of healthcare workers toward the contamination of their attire, and the effect of coronavirus disease 2019 (COVID-19) pandemic on their cleaning practices in non-operative settings. This is a cross-sectional study conducted using a self-administered questionnaire which was distributed among physicians at King Abdul-Aziz Medical city, Riyadh. The questionnaire queried the physicians about their laundering habits, knowledge toward their attire, and the difference in cleaning practices after the emergence of COVID-19. Out of 220 questionnaires distributed, 192 physicians responded. Majority of physicians were male (54%) and were in the 20 to 30 age group. Female gender was significantly associated with the frequency of uniform washing ( value < 0.0001) and place of cleaning (home vs. outside home) ( value <0.0001). Physicians in intensive care were more likely to take off their uniforms daily before leaving the hospital compared to others ( value of 0.018). Most physicians did not prefer to use the hospital laundry system for cleaning their uniforms but consultants were the most to use it. COVID-19 pandemic led to changes in washing habits in 108 physicians (60%). Majority of physicians accepted washing their uniforms multiple times per week and their washing habits increased during the COVID-19 pandemic. Female gender and younger physician both were associated with increasing washing habits.

Health status of working children in Jordan: Comparison between working and nonworking children at schools and industrial sites

The purpose of this study is to explore the psychosocial and physical health impact of child labor and in Jordan and compare between working and nonworking children at schools and industrial sites. Methods: A sample of 4008 children at schools and industrial sites answered the self-report questionnaires that include physical and psychosocial health concerns, health screening survey, anger expression and child abuse survey. The results showed that both nonschool and school working children have various physical and psychosocial health problems. Physical complaints included common cold, flu, hearing and vision problems. Psychosocial ones included feeling lonely, feeling depressed, problems at home and work, and high level of anger expression, smoking cigarettes and using drugs. The study’s implications for health, education, counseling, and research interventions were discussed.

The psychosocial impact of child labour in Jordan: A national study

The purpose of this study was to identify the psychosocial impact of child labour in Jordan, distinguishing between the impact on working school children, nonworking school children and working and nonschooled children. More than 351 million children across the world work. Emotional, behavioral and psychiatric problems in working children are a considerable public health problem in developing countries. A descriptive, comparative, cross-sectional design was used in this study. All data were collected in 2010. A total of 4008 children aged six to 16 years were interviewed. Of these, 2093 (52.2%) were nonworking school children, 896 (22.4%) working school children, and 1019 (25.4%) working and nonschooled children. Working school children reported psychosocial problems more often than working and nonschooled children. Child labour has a negative impact on children's psychosocial health and development. These findings indicate that psychoeducational support and problem-solving programs help children to cope better with the consequences of their work and study. More effort is needed to reduce children's involvement in labour. Understanding the impact of work on child mental and social health is essential to inform policy and future research as well as to improve the lives of children.
