NIR-II DNA Nanodevice for Spatiotemporal MicroRNAs Imaging Based on "and" Logic-Gate Strategy

The precise detection of multiple microRNAs (miRNAs) is of great significance to the classification of various cell subtypes. However, miRNA detection is hindered by signal leakage, false-positive signals and low expression of miRNAs. Herein, a DNA nanodevice in the second near infrared region (NIR-II, 1000-1700 nm) (DND-II) is developed for dual miRNAs bioimaging based on "AND" logic-gate signal amplification strategy. The spatiotemporally controlled logic-gate-based DND-II is fabricated by thermosensitive liposomes integrated with the second near-infrared DNA probes. In particular, the "always-on" photoacoustic signal of DNA probes provided an additional imaging tool to monitor the distribution of DND-II in vivo. When both miRNAs are present, the DNA probes generated a "turn on" fluorescent signal through DNA logical analysis and molecular computation, further triggering an entropy-driven dual-cycle fluorescence signal amplification. The spatiotemporal DND-II provides a powerful platform for early diagnosis and photothermal ablation of different subtypes of tumor cells.

Current nanoagents for efficient deep-seated tumor phototheranostics are still facing many barriers, such as limited light penetration depth, insufficient reactive oxygen species production in anaerobic microenvironment, and dissatisfied imaging accuracy. Herein, an A-D-A type small molecule BTP-3V was synthesized. The strong push-pull effect enabled BTP-3V nanoparticles (NPs) to absorb light in the region of 600-1100 nm (extinction coefficient at 980 nm: 3.92 x 104 M-1 cm-1) and emit in the region of 1000-1400 nm (Stokes shift: 283 nm). Upon 980 nm laser excitation, BTP-3V NPs showed efficient hydroxyl radical and superoxide radical production for type-I photodynamic therapy, greatly overcoming the anaerobic disadvantage of malignant tumors. Moreover, the NPs also showed excellent photothermal performance with high photothermal conversion efficiency (39.4 %). In vitro and in vivo investigations corroborated that the biocompatible BTP-3V NPs exhibited remarkable anti-tumor outcome upon 980 nm laser irradiation, benefiting from the combination of type-I photodynamic and photothermal therapy. Significantly, with the short-wave infrared fluorescence emission, BTP-3V NPs could achieve high-resolution imaging of vascular structure, accurate localization of tumor and facile detection of the enrichment and distribution of NPs within the tumor. This study provides valuable insights into nanomaterials for deep-tissue phototheranostics.

Selenocysteine detection and bioimaging in living cells by a colorimetric and near-infrared fluorescent turn-on probe with a large stokes shift

Selenocysteine (Sec) has emerged as an important sensing target in recent years. In this paper, a colorimetric and near-infrared fluorescent turn-on probe for Sec was developed. This probe features a remarkable large Stokes shift (146 nm) and shows a rapid, highly selective detection process for Sec with obvious colorimetric and near-infrared fluorescent (Em 706 nm with Ex 560 nm) turn-on responses. In addition, this probe can be used to quantitatively detect Sec with high sensitivity with a detection limit of 62 nM over a wide linear range (0.2-80 mu M). Moreover, it was further demonstrated that this NIR fluorescent probe can be employed to image both exogenous and endogenous Sec in living cells, indicating that this probe has great potential for applications.

Selenocysteine (Sec) is the 21st naturally occurring amino acid and has emerged as an important sensing target in recent years. However, fluorescent detection of Sec in living systems is challenging. To date, very few fluorescent Sec probes have been reported and most of them respond fluorescence to Sec in the-visible region. In this paper, a very promising near-infrared fluorescent probe for Sec was developed. This probe works in aqueous solution over a wide pH range under mild conditions and can be used for rapid;highly selective and sensitive detection of Sec with significant near-infrared fluorescent turn-on signal changes. In addition, it features a remarkable large Stokes shift (192 nm) and a low detection limit (60 nM) for Sec with a wide linear range (0-70 mu M). Moreover, this probe can be conveniently used to detect Sec in serum samples, living cells, and animals, indicating it holds great promise for biological applications.

Selenocysteine (Sec) is known as the 21st amino acid that is incorporated into selenoproteins (SePs) and plays vital roles in living systems. Accordingly, development of fluorescent detection method for Sec is of significance. Herein, a readily available fluorescent Sec probe is reported. Although this probe has been reported before for detection of biothiols, the studies in this work show that it can be used for rapid, high selective and sensitive detection of Sec over biothiols and many other analytes with dual colorimetric and fluorescent turn-on signal changes simply by slightly changing the detection conditions. The detection limit of this probe for Sec was found to be about 47 nM with a wide linear range (0-80 mu M). In addition, this probe has low cytotoxicity and is able to detect Sec in serum samples and in living cells, indicating it can be used as a potential new molecular tool for Sec detection.

An aza-coumarin-hemicyanine based near-infrared fluorescent probe for rapid, colorimetric and ratiometric detection of bisulfite in food and living cells

A novel aza-coumarin-hemicyanine conjugated hybrid was prepared, which was found to be a promising colorimetric and ratiometric near-infrared (NIR) fluorescent probe for detection of bisulfite/sulfite in aqueous solution. This probe shows a rapid response (within 30 s) and high selectivity and sensitivity for bisulfite/sulfite, giving distinct colorimetric and ratiometric fluorescence changes at 717 and 560 nm. The detection limit for bisulfite was estimated to be 87 nM. In addition, detection of bisulfite in food samples and bioimaging of both exogenous and endogenous bisulfite in living cells with this probe were successfully applied, which indicates that this probe holds great potential for bisulfite detection both in real samples and in living cells. Analgesic properties of chimeric peptide based on morphiceptin and PFRTic-amide

A chimeric opioid peptide (MCRT, YPFPFRTic-NH2) was here designed and synthesized. This peptide was based on morphiceptin (YPFP-NH2) and a neuropeptide FF (NPFF) derivative (PFRTic-NH2) sharing one proline. This peptide is intended to produce potent analgesia. MCRT was found to induce analgesic activity in a dose- and time-dependent manner, as indicated by a tail flick latency test in mice to which it had been intracerebroventricularly administered (5-60 min, 0.025-2.5 nmol/kg (0.5-50 pmol per mouse), ED50 = 1.49 nmol/kg). At 2.5 nmol/kg, MCRT showed significantly higher levels of analgesic activity than morphiceptin or PFR(Tic)amide at 2500 nmol/kg. Naltrindole and cyprodime were found to partially but significantly inhibit this analgesic activity, but naloxone blocked it completely. The kappa opioid receptor antagonist nor-BNI was found to slightly inhibit MCRT and morphiceptin. Pre-injection of BIBP3226 and co-administration of NPFF and MCRT showed that NPFF receptors were involved in the analgesia of MCRT. BIBP3226 was found to weaken the analgesic effects of MCRT, but BIBP3226 could not block the analgesic effects of PFR(Tic)amide. Overall, MCRT was found to have stronger analgesic activity than morphiceptin or PFR(Tic)amide when interacting with mixed mu/delta opioid receptor interactions. MCRT also showed partial interaction with NPFF receptors. (C) 2012 Elsevier B.V. All rights reserved.

The expression profile of vascular endothelial growth factor (VEGF) is highly correlated with the occurrence and development of cancer. This work reports an electrochemiluminescence (ECL) approach for highly sensitive detection of VEGF(165). This approach comprises aptamer-target recognition, T7 exonuclease (17 Exo)-assisted cycling signal amplification and efficient quenching of ECL of CdS:Eu nanocrystals (NCs) by using DNAzyme. In this assay, CdS:Eu NCs were used as the ECL substrate, A guanine (G)-rich single-stranded DNA (ssDNA) sequence and VEGF(165) aptamer were co-immobilized on the surface of the CdS:Eu NCs modified glassy carbon electrode. After recognition and binding to VEGF(165), the aptamer moved away from the electrode surface and induced the proposed cyclic cleavage of the target DNA with T7 Exo. A large amount of G-rich ssDNA was released on the CdS:Eu film and folded into G-quadruplex/hemin DNAzyme in the presence of hemin and K+, consequently decreasing the ECL intensity of CdS:Eu. A good linearity was obtained for VEGF(165) detection within the range of 1 pM to 20 nM with a detection limit of 0.2 pM. This assay could be a universal and promising protocol for detection of various biomarkers for early clinical diagnosis. (C) 2015 Elsevier B.V. All rights reserved.

Preliminary analgesic properties of deltorphin-5-methoxytryptamine chimeric opioid peptides

To further understand the relationship between melatonin (MT) and deltorphins (Dels) in pain modulation, two chimeric peptides (Del I-5-methoxytryptamine and Del II-5-methoxytryptamine) both containing 5-methoxytryptamine at the carboxyl-terminal of Dels mimicking MT were designed, synthesized and characterized by tail-flick assay in mice. Results showed that intracerebroventricular administration of Del I-5-methoxytryptamine (YaFDVVG-X, Xis 5-methoxytryptamine, 5,50 nmol/kg) or Del II-5-methoxytryptamine (YaFEVVG-X, X is 5-methoxytryptamine, 5, 50 nmol/kg) produced stronger analgesia than deltorphins (Dell or Del II alone), and acting even longer and stronger than cocktails containing Del I or Del II (50 nmol/kg) and MT (50 nmol/kg). Naloxone (i.p., 100 nmol/kg) could totally block the analgesic effects induced by the chimeric peptides, while luzindole (specific antagonist of melatonin receptor, i.p., 250 nmol/kg) could only partially inhibit the effects down to that induced by Dels alone. Interestingly, Del I-5-methoxytryptamine and Del II-5-methoxytryptamine act weaker with delta receptor than Dels in vitro but could induce much longer analgesia through co-activating delta opioid receptor and melatonin receptor. The cardiovascular effects of a chimeric opioid peptide based on morphiceptin and PFRTic-NH₂

MCRT (YPFPFRTic-NH2) is a chimeric opioid peptide based on morphiceptin and PFRTic-NH2. In order to assess the cardiovascular effect of MCRT, it was administered by intravenous (i.v.) injection targeting at the peripheral nervous system and by intracerebroventricular (i.c.v.) injection targeting at the central nervous system. Naloxone and L-NAME were injected before MCRT to investigate possible interactions with MCRT. Results show that administration of MCRT by i.v. or i.c.v. injection could induce bradycardia and decrease in mean arterial pressure (MAP) at a greater degree than that with morphiceptin and PFRTic-NH2. When MCRT and NPFF were coinjected, we observed a dose-dependent weakening of these cardiovascular effects by MCRT. Because naloxone completely abolished the cardiovascular effects of MCRT, we conclude that opioid receptors are involved in regulating the MAP of MCRT regardless of modes of injection. The effect of MCRT on heart rate is completely dependent on opioid receptors when MCRT was administered by i.c.v. instead of i.v. The central nitric oxide (NO) pathway is involved in regulating blood pressure by MCRT under both modes of injection, but the peripheral NO pathway had no effect on lowering blood pressure mediated by MCRT when it was administered by i.c.v. Based on the results from different modes of injection, the regulation of heart rate by MCRT mainly involves in the central NO pathway. Lastly, we observed that the cardiovascular effects of MCRT such as bradycardia and decrease of blood pressure, were stronger than that of its parent peptides. Opioid receptors and the NO pathway are involved in the cardiovascular regulation by MCRT, and their degree of involvement differs between intravenous and intracerebroventricular injection.
