Ru/Pt@BSA nanoparticles for efficient photo-catalytic oxidation of NAD(P)H and targeted cancer treatment under hypoxic conditions

Photo-catalytic oxidation of intracellular nicotinamide adenine dinucleotide(2'-phosphate)(NAD(P)H)has attracted much attention for cancer ***,the general oxygen-dependent mechanism heavily depresses the efficacy in hypoxic *** solve this problem,herein platinum nanoparticles(Pt NPs)with catalase-like(CAT-like)and catalytic H_(2)evolution activities were introduced as a powerful assistant to enhance the photo-catalytic NAD(P)H oxidation of Ru1([Ru(phen)2(PIP-OCH_(3))]^(2+),phen=1,10-phenanthroline,PIP-OCH_(3)=2-(4-methoxy phenyl)-1H-imidazo[4,5-f][1,10]phenanthroline)under hypoxic and even oxygen-free ***,Pt NPs can transform the original and in situ formed H_(2)O_(2)once again into O_(2)by the CAT-like activity,thus relieving tumor hypoxia and realizing cyclic utilization(at least in part)of the precious oxygen in ***,Pt NPs can also be served as H_(2)evolution catalysts while using Ru1 as the photosensitizer and NAD(P)H as the electron and proton *** this process,NAD(P)H is oxidized without the participation of oxygen,which can provide an effective way even under oxygen-free *** co-encapsulation of Rul and Pt NPs in bovine serum albumin(BSA)with tumor targeting ability,the resultant Ru/Pt@BSA could photo-catalyze intracellular NAD(P)H oxidation under hypoxic conditions(3%O_(2)),and exhibited an efficient and selective anticancer activity both in vitro and in *** results may provide new sights for efficient and targeted cancer treatment underhypoxic conditions.

A highly positively charged Ru(II) complex with photo-labile ligands for selective and efficient photo-inactivation of intracellular aureus

Due to the protection afforded by host cells, intracellular Staphylococcus aureus (S. aureus), particularly methicillin-resistant S. aureus (MRSA), poses a significantly greater challenge to eliminate compared to the extracellular counterparts. It is highly desirable to develop novel antibacterial agents which are capable of selectively and efficiently eradicating intracellular bacteria, including drug-resistant strains, while being less prone to induce bacterial resistance. In this work, two Ru(II) complexes (Ru1 and Ru2) with photo-labile ligands were designed and synthesized. Both Ru1 and Ru2 could covalently bind to DNA after photo-induced ligand dissociation. Compared to Ru1, the incorporation of a triphenylamine group adorned with two positively charged cationic pyridine units significantly boosts the DNA binding constant, bacterial binding/uptake level, and subsequently, the antibacterial activity of Ru2. Ru2 could selectively photo-inactivate intracellular S. aureus and MRSA, being more efficient than vancomycin both in vitro and in vivo. Interestingly, after 20 days' treatment at sublethal concentrations, S. aureus cells exhibited no obvious drug resistance towards Ru2 upon irradiation. Such appealing results may provide new sights for developing novel antibacterial agents against intractable intracellular pathogens and also prevalent drug resistance.

An efficient strategy based on two-stage chromatography and in vitro evaluation for rapid screening and isolation of acetylcholinesterase inhibitors from Scutellaria baicalensis Georgi

The extraction of Scutellaria baicalensis Georgi was investigated using the response surface methodology-genetic algorithm mathematical regression model, and the extraction variables were optimized to maximize the flavonoid yield. Furthermore, a simple and efficient ultrafiltration-liquid chromatography-mass spectrometry and molecular docking methods were developed for the rapid screening and identification of acetylcholinesterase inhibitors present in Scutellaria baicalensis Georgi. Subsequently, four major chemical constituents, namely baicalein, norwogonin, wogonin, and oroxylin A, were identified as potent acetylcholinesterase inhibitors. This novel approach, involving the use of ultrafiltration-liquid chromatography-mass spectrometry and molecular docking methods combined with stepwise flow rate counter-current chromatography and semi-preparative high-performance liquid chromatography, could potentially provide a powerful tool for the screening and extraction of acetylcholinesterase inhibitors from complex matrices and be a useful platform for the production of bioactive and nutraceutical ingredients.

We developed a highly regioselective base-induced synthesis of N-2-substituted 1,2,3-triazoles from N-sulfonyl-1,2,3-triazoles and alkyl bromides/alkyl iodides at room temperature. We propose an S(N)2-like mechanistic pathway to explain the high N-2-regioselectivity. The protocol features a broad substrate scope and generates products in good to excellent yields (72-90%).

As new variants of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) continue to emerge, they raise increasing concerns about the efficacy of neutralizing antibodies and vaccines. This situation underscores the urgent need for specific drugs against the coronavirus disease 2019 (COVID-19). Given that COVID-19 is particularly associated with substantial inflammation, the development of novel, effective antiviral and antiinflammatory agents represents a promising research direction. In this study, we virtually screened a library consisting of 2900 anti-inflammatory small molecules for their inhibitory effects on the 3-chymotrypsin-like protease (3CLpro) of SARS-CoV-2 and selected 23 promising candidates for further testing using a fluorescence resonance energy transfer (FRET) assay. The results indicated that Gnetol had the most potent inhibitory effect against SARS-CoV-2 3CLpro. Further structural modifications led to the identification of compounds 38 and 39, which displayed superior inhibitory activity. Compound 39 showed good selectivity for host proteases. Subsequently, Gnetol and its structural analogs, which demonstrated SARS-CoV-2 3CLpro inhibitory activity, were tested for their anti-inflammatory effects. Among these, Piceatannol and compound 39 exhibited enhanced antiinflammatory effects, with compound 39 alone showing the most potent antiviral and anti-inflammatory activity. Thus, our study has explored a new research strategy for discovering antiviral and anti-inflammatory bifunctional molecules. The discovery of Gnetol and its structural analogs has provided new lead candidates for the development of COVID-19 therapeutics.

Rep-MCA-former: An efficient multi-scale convolution attention encoder for text-independent speaker verification

In many speaker verification tasks, the quality of speaker embedding is an important factor in affecting speaker verification systems. Advanced speaker embedding extraction networks aim to capture richer speaker features through the multi-branch network architecture. Recently, speaker verification systems based on transformer encoders have received much attention, and many satisfactory results have been achieved because transformer encoders can efficiently extract the global features of the speaker (e.g., MFA-Conformer). However, the large number of model parameters and computational latency are common problems faced by the above approaches, which make them difficult to apply to resource-constrained edge terminals. To address this issue, this paper proposes an effective, lightweight transformer model (MCA-former) with multi-scale convolutional self-attention (MCA), which can perform multi-scale modeling and channel modeling in the temporal direction of the input with low computational cost. In addition, in the inference phase of the model, we further develop a systematic re-parameterization method to convert the multi-branch network structure into the single-path topology, effectively improving the inference speed. We investigate the performance of the MCA-former for speaker verification under the VoxCeleb1 test set. The results show that the MCA-based transformer model is more advantageous in terms of the number of parameters and inference efficiency. By applying the re-parameterization, the inference speed of the model is increased by about 30%, and the memory consumption is significantly improved.

Coreactant-free and Near-Infrared Electrochemiluminescence Immunoassay with n-Type Au Nanocrystals as Luminophores

The electrochemiluminescence (ECL) bioassay is prominently carried out with the involvement of the coreactant. To remove the detrimental effects of the coreactant on the ECL of luminophores, herein, a promising ECL immunoassay strategy with biocompatible nanoparticles as the luminophore is proposed, which involves directly and electrochemically oxidizing the luminophore methionine-capped Au (Met@Au) nanocrystals (NCs) without the participation of any coreactant. Met@Au NCs are a kind of n-type nanoparticles, and they can be electrochemically injected with valence band (VB) holes around +0.80 and +1.10 V (vs Ag/AgCl). The electrochemically injected exogenous VB hole can recombine with the endogenous conduction band electron of Met@Au NCs and eventually bring out two coreactant-free and near-infrared ECL processes around 0.80 V (ECL-1) and 1.10 V (ECL-2). The intensity of coreactant-free ECL is primarily determined by the electrochemical oxidation-induced hole-injection process. ECL-2 is considerably stronger than ECL-1 and can be exploited for determining the carcinoembryonic antigen (CEA) in a sandwich immunoassay procedure with a linear range from 0.1 to 50 pg/mL as well as a limit of detection of 0.03 pg/mL (S/N = 3).

This paper designs a Signal Source Board based on PXIe standard. The Signal Source Board adopts XCKU060-2FFVA1157I of Xilinx company as the main control chip of the FPGA, AD9144 of ADI company as the DAC, the Host Computer sends the analog signal waveform code table to be generated to the Signal Source Board through the PCIe Bus, and then the FPGA of the Source Board based sends the analog signal waveform code table to two pieces of AD9144 through the JESD204B interface. Finally, the generated analog signal is filtered through the low-pass filter HMC1023, and completes the output of 8-channel analog signal. It achieves the high-quality generation of any modulated signal from baseband to mid-frequency, and meets the synchronous output requirements of 8-channel analog signal. © 2022 ACM.

With the increasing complexity of circuit board function, both the performance index of circuit board and the number of circuit test points are increasing day by day. It is far from satisfying the test demand of large-scale board production manually. In order to realize automatic test of electrical signal on board, automatic configuration of test sequence and automatic generation of test report, this paper designed an automatic test software management platform based on Labview and TestStand, and proposed a solution of automatic generation of test report according to template. And this paper provides a method of parameter configuration and management of instrument and result threshold of automatic test sequence, which further improves the test efficiency of automatic test and realizes unified management of test parameters, compared with gradually configuring instrument parameters in each test step of test sequence. © 2022 ACM.

HClO-responsive dinuclear Ru(II) complexes for selective imaging and efficient photo-inactivation of intracellular bacteria

Intracellular bacteria (ICB), cloaked by the protective barriers of host cells, pose a formidable challenge to selective and efficient eradication. The employment of activatable photosensitizers based antibacterial photodynamic therapy (aPDT) holds significant potential for selective imaging and photo-inactivation of ICB while minimizing side effects on normal cells. Drawing inspiration from the elevated hypochlorous acid (HClO) levels in ICB infected phagocytes, herein we firstly designed and synthesized a series of HClOresponsive dinuclear Ru(II) complexes (Ru1-Ru3) to achieve such a goal. Initially, the luminescence, 1 O2 generation and aPDT activity of these Ru(II) complexes were suppressed due to the quenching effect of the azo group, but were recovered after reaction with HClO in solutions or within ICB infected phagocytes. The detailed results revealed that Ru1 and Ru3 could not only selectively visualize ICB, but also demonstrated remarkable aPDT activity against ICB, surpassing vancomycin both in vitro and in vivo .
