Stachyose Exerts Anticolitis Efficacy by Re-balancing Treg/Th17 and Activating the Butyrate-Derived PPARγ Signaling Pathway

Ulcerative colitis (UC) is a complex chronic inflammatory disease closely associated with gut homeostasis dysfunction. The previous studies have shown that stachyose, a functional food additive, has the potential to enhance gut health and alleviate UC symptoms. However, the underlying mechanism of its effects remains unknown. In this study, our findings showed that dietary supplements of stachyose had a significant dose-dependent protective effect on colitis symptoms, regulation of gut microbiota, and restoration of the Treg/Th17 cell balance in dextran sulfate sodium (DSS) induced colitis mice. To further validate these findings, we conducted fecal microbiota transplantation (FMT) to treat DSS-induced colitis in mice. The results showed that microbiota from stachyose-treated mice exhibited a superior therapeutic effect against colitis and effectively regulated the Treg/Th17 cell balance in comparison to the control group. Moreover, both stachyose supplementation and FMT resulted in an increase in butyrate production and the activation of PPAR gamma. However, this effect was partially attenuated by PPAR gamma antagonist GW9662. These results suggested that stachyose alleviates UC symptoms by modulating gut microbiota and activating PPAR gamma. In conclusion, our work offers new insights into the benefical effects of stachyose on UC and its potential role in modulating gut microbiota.

Metastasis is one of the main reasons for cancer treatment failure. Unfortunately, most treatment approaches inevitably damage the extracellular matrix (ECM) during tumor cell elimination, thereby augmenting the risk of metastasis. Herein, we proposed a "sieging tumor cells" strategy based on ferric coordination polymers (FeCPs), which involved anchoring tumor cells through ECM consolidation and selectively eliminating them in the tumor regions. Due to the weak coordination interactions and amorphous structure of FeCPs, the acidic tumor microenvironment facilitated their disintegration, releasing salicylic acid (SA), 2,5-dihydroxyterephthalic acid (DHTA) and Fe3+ ions. The released SA inhibited heparinase activity to consolidate the ECM, while Fe-mediated chemodynamic therapy (CDT) was enhanced by DHTA due to its fast electron transport behavior, ultimately inhibiting tumor growth and metastasis. The results from the orthotopic 4T1 breast tumor model indicated that lung metastasis was reduced by about 90%, and the survival rate improved by 70% after FeCP treatment. Overall, this "sieging tumor cells" strategy provides an emerging approach for the treatment of malignant tumors by consolidating the ECM in combination with self-enhanced CDT.

Robust and Regular Micronano Binary Texture on the Complex Curved Surface for Enhanced Reendothelialization and Antithrombotic Performance

Blood-contacting medical devices can easily trigger immune responses, leading to thrombosis and hyperblastosis. Constructing microtexture that provides efficient antithrombotic and rapid reendothelialization performance on complex curved surfaces remains a pressing challenge. In this work, we present a robust and regular micronano binary texture on the titanium surface, characterized by exceptional mechanical strength and precisely controlled wettability to achieve excellent hemocompatibility. Systematic in vitro and in vivo investigations confirmed that the micronano binary texture with superhydrophilic modification effectively suppressed the adhesion and activation of plasma proteins and blood cells, thereby mitigating the subsequent coagulation cascade and thrombosis. Meanwhile, the modified surface significantly upregulated the gene expression involving cell-matrix adhesion, growth factor synthesis, and calcium-mediated cytoskeleton remodeling and then accelerated the formation of a healthy and stable endothelial cell layer. This enhancement of re-endothelialization was not observed with pure titanium and superhydrophobic surfaces. Hence, superhydrophilic micronano binary texture not only significantly inhibits thrombosis but also selectively enhances the integrity and viability of the endothelial cell layer, making it a promising strategy for improving the long-term anticoagulation performance of vascular implants.

Joint Multiscale Cross-Lingual Speaking Style Transfer With Bidirectional Attention Mechanism for Automatic Dubbing

Automatic dubbing, which generates a corresponding version of the input speech in another language, can be widely utilized in many real-world scenarios, such as video and game localization. In addition to synthesizing the translated scripts, automatic dubbing further transfers the speaking style in the original language to the dubbed speeches to give audiences the impression that the characters are speaking in their native tongue. However, state-of-the-art automatic dubbing systems only model the transfer on the duration and speaking rate, disregarding the other aspects of speaking style, such as emotion, intonation and emphasis, which are also crucial to fully understand the characters and speech. In this paper, we propose a joint multiscale cross-lingual speaking style transfer framework to simultaneously model the bidirectional speaking style transfer between two languages at both the global scale (i.e., utterance level) and local scale (i.e., word level). The global and local speaking styles in each language are extracted and utilized to predict the global and local speaking styles in the other language with an encoder-decoder framework for each direction and a shared bidirectional attention mechanism for both directions. A multiscale speaking style-enhanced FastSpeech 2 is then utilized to synthesize the desired speech with the predicted global and local speaking styles for each language. The experimental results demonstrate the effectiveness of our proposed framework, which outperforms a baseline with only duration transfer in objective and subjective evaluations.

Colour associations and consumer product-colour purchase decisions

This article describes a conceptual framework for understanding consumer product-colour purchase decisions. Building on previous work, a new primary factor, colour association, is introduced. The purpose of this study is to test whether consumers' colour-associations connect with their product-colour association, and whether colour-association affects consumers' intended product-colour purchase decisions. The study used two research activities (a laboratory experiment and an interview) and two research methods (multiple choice for 6-alternative-forced-choice and multiple choice for rank-order) to investigate which colour a participant would choose if asked to select a product to purchase when there is a range of colours available. Participants were also asked to provide a reason for their decisions. The study finds that the stronger a participant's preference for a colour, the more likely they are to purchase a product in that colour. The data from interviews suggests that, for some of the products tested, participants are highly influenced by the colour association that they would purchase a product-colour on their one of the colour association than other colours. The updated conceptual framework suggests that the colour association factor could be one of the product-colour decision primary factors that influence consumer product-purchase intentions in the absence of consumer personal colour preferences.

Investigation of designers' colour selection process

Although many design process studies and design theories have been developed and presented, in reality designers have rarely followed standard rules. A design process is often more a form of personal self-expression. The type of approach that designers use in their design work depends on their preference, experience, and education background. Therefore, this work focused on understanding the design process and the colour selection method by collecting data from designers and their design process with the aim to summarize a general colour selection method. This work employs a semi-structured interview approach in a one-to-one interview format to investigate the process of design and collect their colour selection method. A total of 20 designers were interviewed from different design areas, working experience, age groups, cultural background, and workplace around Korea, China, South Africa, Mexico, and the United Kingdom. In the end, a three-step-method of colour selection was summarized and this highlighted the importance of colour association in the colour selection process.

A comparative evaluation of similarity measurement algorithms within a colour palette

Recently, there has been interest in the development of colour palettes from images. Colour palettes have long been used by designers to communicate colours and their relationships but increasingly palettes are being derived automatically from digital images, concepts, or from a plethora of digital design tools online. Methods to predict differences between palettes are growing in popularity. This study is concerned with the prediction of visual self-similarity for colour palettes with large numbers of patches. A psychophysical experiment was carried out to collect the human judgments of similarity and then six different algorithms were introduced and evaluated in terms of their ability to predict the psychophysical data. Two methods to quantify the agreement between the visual data and algorithm predictions were used based on regression analysis with coefficient of determination for the goodness of fit and multidimensional scaling with loss function Kruskal's stress. Of the six algorithms, the Pearson correlation coefficient method was considered to give the best performance.

Evaluation of appropriateness of antibiotic prescribing in primary healthcare institutions in China using proxy indicator

Background Our objectives were to develop a set of proxy indicators (PIs) suited for assessing antibiotic use appropriateness in China ' s primary healthcare institutions (PHIs), and assess performance scores of these PIs while exploring factors that in fl uence the antibiotic appropriateness. Methods We selected potential PIs for the PHIs through a RAND - modi fi ed Delphi procedure, and assessed clinimetric properties, focusing on measurability, applicability, and potential for improvement. PIs with favorable clinimetric properties were used to evaluate antibiotic prescription appropriateness by calculating performance scores of each PI. Institutions were categorized into three clusters representing different levels of appropriateness. We used the chi - square test and an ordinal logistic regression model at PHI level to explore factors in fl uencing antibiotic appropriateness. Findings Eighteen PIs were developed through two rounds of online surveys and one face - to - face meeting involving 20 stakeholders. All PIs met the clinimetric properties criteria and were used to analyze 209,662 antibiotic prescriptions across 269 PHIs. The percentage of PHIs meeting the target ranged from 3.1% to 69.3%, with 6 PIs below 10%. The appropriateness of antibiotic prescriptions was signi fi cantly associated with percentages of patients ' gender of the PHIs. Interpretation The varied and suboptimal performance of the PIs indicated the need for diverse efforts to enhance the rational antibiotic use at PHI level. It was necessary to devise distinct sets of PIs for diverse settings in future endeavors. Depth sensing, as a fundamental prerequisite of autonomous systems, either estimates the depth information for each pixel of a colorful image (i.e., depth prediction) or converts the noisy and sparse point clouds into a dense depth map (i.e., depth completion). Most studies treat depth prediction and depth completion as two independent tasks due to the differences between RGB images and point clouds and overlook the complementarity of two modalities, thus resulting in poor robustness to noise. With the integration of monocular cameras and LiDARs in autonomous systems, a generic framework to unify depth estimation and depth completion for robust depth sensing is essential. In this study, we develop a CAScading Interaction Network (CASIN) to capture the modality interaction to compensate for the deficiency of each modality itself. Specifically, two encoder-decoder architectures are employed to learn modality representation. Their multi-scale interactions are preserved by cascading interaction network under the supervision of one auxiliary task. Finally, the shared representation is integrated to infer dense depth map due to the complementarity of modalities. We evaluated the proposed solution on the KITTI dataset. Our experimental results show that learning the coupled interaction of both modalities under the guidance of an auxiliary task can lead to significant performance improvements with 46.64% gain for depth prediction. Furthermore, CASIN is robust to noisy input signals with less performance degradation, which makes our approach feasible in the wild for robust depth sensing. © 2022 IEEE.

In this article, we are dedicated to coping with the control design for underactuated mechanical system (UMS) with system uncertainties, which is also subject to a set of servo constraints. For the nominal system without any initial condition deviations, this article provides a general model-based servo control with the idea of second-order constraints, which can be derived from both holonomic and nonholonomic ones. For the uncertain system, with the uncertainties being separated into matched portions and mismatched portions, this article provides an adaptive robust control scheme to guarantee the UMS to attain the deterministic performance on the basis of nominal control. The leakage-type adaptive law is to size up the unknown uncertainty bounds. The cart-pole system and the rotational-translational actuator system are used to verify the availability of the proposed adaptive robust control.
