Interferon Regulatory Factor 4 (IRF-4) Targets IRF-5 to Regulate Epstein-Barr Virus Transformation

The cellular interferon regulatory factor-4 (IRF-4), which is a member of IRF family, is involved in the development of multiple myeloma and Epstein-Barr virus (EBV)-mediated transformation of B lymphocytes. However, the molecular mechanism of IRF-4 in cellular transformation is unknown. We have found that knockdown of IRF-4 leads to high expression of IRF-5, a pro-apoptotic member in the IRF family. Overexpression of IRF-4 represses IRF-5 expression. Reduction of IRF-4 leads to growth inhibition, and the restoration of IRF-4 by exogenous plasmids correlates with the growth recovery and reduces IRF-5 expression. In addition, IRF-4 negatively regulates IRF-5 promoter reporter activities and binds to IRF-5 promoters in vivo and in vitro. Knockdown of IRF-5 rescues IRF-4 knockdown-mediated growth inhibition, and IRF-5 overexpression alone is sufficient to induce cellular growth inhibition of EBV-transformed cells. Therefore, IRF-5 is one of the targets of IRF-4, and IRF-4 regulates the growth of EBV-transformed cells partially through IRF-5. This work provides insight on how IRFs interact with one another to participate in viral pathogenesis and transformation.

Thermodynamic analysis of an ammonia-water solution cross-type absorption-resorption heat pump system

Absorption-resorption heat pump (ARHP) can realize the efficient utilization of low-grade heat under lower operating pressure than traditional absorption heat pump (AHP), but the application is limited by the low coefficient of performance (COP). In this paper, an ammonia-water solution cross-type absorption-resorption heat pump is proposed to improve the performance, and the rectifier can be eliminated because the ammonia component can be balanced with the adjustment of mass flow rate at the resorber inlet. A thermodynamic model is established to investigate the influences of different parameters on the coefficient of performance. The results reveal that: When the high/low pressure pair is 1.5/0.44 MPa and the temperatures of absorber, resorber and desorber are 40 degrees C, 40 degrees C and 5 degrees C, the COP of the proposed system can reach 1.564, which is 8.1% higher than traditional ARHP. When the temperatures of generator, absorber, resorber and desorber are 115 degrees C, 35 degrees C, 40 degrees C and 5 degrees C, the COP is improved up to 6.4% compared with the balanced-type ARHP. Besides, the proposed system has wider working temperature ranges than the traditional system and the temperature ranges of absorber and desorber are increased by 1 degrees C and 2 degrees C, respectively. The advantages of the proposed system also include the lower optimal low-pressure values. Therefore, the ammonia-water solution cross-type absorption-resorption heat pump is appropriate for working in larger pressure differences and makes waste heat recovery more economical and efficient.

A three-dimensional two-phase non-adiabatic model is proposed for air-breathing micro direct methanol fuel cells (DMFC). The coupled heat and mass transport, along with the electrochemical reactions occurring in the air-breathing DMFC are modeled. Based on this new non-adiabatic model, the reactants distribution and concentration distribution, the pressure and flow velocity distribution, the saturation distribution, the potential and current density distribution, the temperature distribution and their effects on the cell performance are carefully examined.

An improvement of convective precipitation nowcasting through lightning data dynamic nudging in a cloud-resolving scale forecasting system

A lightning data dynamic nudging (LDN) method was designed to adjust the dynamic field in convective clouds based on the physical relationship between lightning and vertical velocity. Under this framework, vertical velocity data retrieved from the Guangdong-Hongkong-Macau lightning location system were assimilated by a nudging approach using the Global/Regional Assimilation and Prediction System in mesoscale model (GRAPES-Meso). The effect of the assimilation on short-term ( < 6 h) precipitation forecast was evaluated by simulating the squall line event on 7 May and a continuous experiment from May to August 2018. Firstly, a series of sensitivity experiments was carried out to adjust the configuration of the assimilation system based on the squall line event. The results show that LDN expanded the spatial distribution of the positive vertical velocity ( > 1 m s(-1)) at 700 hPa by about 2%, and thus enlarged the spatial distribution of severe rainfall ( > 40 mm h (-1)). It should be noted that LDN did not significantly change the intensity of the updraft (i.e., the maximum vertical velocity) of the squall line relative to the simulation without assimilation. Overall, the equitable threat score (ETS) and fractions skill score (FSS) were increased by 0.04 and 0.13, respectively, and the positive effect lasted for 2-3 h after the data were assimilated. In view of the data assimilation frequency, using two successive nudging procedures with an interval of 12 min had the best effect on the forecast of severe precipitation. For the assimilation of both lightning and radar observations, the asynchronous assimilation of lightning and radar measurements performed slightly better than the synchronous assimilation. LDN improved the accuracy of severe precipitation forecast ( > 40 mm h(-1) and > 20 mm h(-1) for the squall line case and the continuous run, respectively), whereas assimilating radar data improved weak precipitation forecast (1-20 mm h(-1) in the squall line case an

The cellular interferon (IFN) regulatory factor-7 (IRF7) is closely associated with the Epstein-Barr virus (EBV)-mediated transformation of B lymphocytes in vitro and in vivo. However, the exact role of IRF7 in viral transformation is not clear. We have found that knockdown of IRF7 leads to growth inhibition Of EBV-transformed cells, and restoration of IRF7 by exogenous plasmid correlates with growth recovery of the viral transformed cells. In addition, IRF7-knockdown cells have a lower proliferation but a higher apoptotic rate than control cells. Moreover, reduction of IRF7 leads to reduction of major viral oncoprotein, latent membrane protein 1 (LMP1). Our data suggest that IRF7 may be a factor in EBV transformation and a useful target in the therapy of EBV-mediated neoplasia. (C) 2014 Elsevier B.V. All rights reserved.

This paper researches the manually controlled site reselection focusing on the approaching phase and landing phase of soft landing of manned lunar landing. First, this paper introduces the phase division of manned lunar landing, establishes the kinetic model for lunar module and then makes research on the manual control site selection scheme under TV guidance. In the TV guidance scheme, the astronaut can directly select new lunar landing point within the field of view. The target guidance is achieved by automatic control system. Meanwhile, the impact of response delay of the astronaut on tasks in actual execution is considered. According to the simulation results of the guidance scheme, as the TV guidance is not restricted by the angle of sight, the adjustable range of the landing point is relatively great. Meanwhile, TV guidance has relatively great requirements on the attitude maneuver. In addition, the earlier the site reselection made by the astronaut, the greater the adjustable range of the landing point is. © 2016 IEEE.

Association between hs-CRP/HDL-C ratio and three-month unfavorable outcomes in patients with acute ischemic stroke: a second analysis based on a prospective cohort study

Objective The associations between the ratio of blood high-sensitivity C-reactive protein (hs-CRP) to high-density lipoprotein cholesterol (HDL-C) (hs-CRP/HDL-C ratio) and outcomes in patients with acute ischemic stroke (AIS) have yet to be established. This study is the first to examine the relationship between the hs-CRP/HDL-C ratio and three-month unfavorable outcomes in patients with AIS. Methods This secondary analysis utilized data from a prospective cohort study involving 1559 AIS patients recruited at a South Korean hospital between January 2010 and December 2016. We constructed a binary logistic regression model to explore the association between the hs-CRP/HDL-C ratio and unfavorable outcomes in patients with AIS. An attempt was made to use a generalized additive model (GAM) with smooth curve fitting to elucidate potential nonlinear interactions. Furthermore, inflection points were identified via a recursive method, and binary logistic regression models were developed for each side of these inflection points. Ultimately, a log-likelihood ratio test was used to identify the most appropriate model for explaining the connection between the hs-CRP/HDL-C ratio and unfavorable outcomes in patients with AIS. Results The incidence of unfavorable outcomes was 24.5%, with a median hs-CRP/HDL-C ratio of 3.64. After accounting for other factors, the binary logistic regression model revealed a statistically significant positive association between the hs-CRP/HDL-C ratio and the likelihood of poor outcomes in AIS patients (OR = 1.013, 95% CI: 1.005-1.022;P = 0.002). A nonlinear relationship was observed, with the first inflection point of the hs-CRP/HDL-C ratio at 42.74. Each 1-unit increase in the hs-CRP/HDL-C ratio was associated with a 2.4% greater risk of unfavorable outcomes (OR = 1.024, 95% CI: 1.011-1.038, P < 0.001). Three-dimensional propagation characteristics of the upward connecting leaders in six negative tall-object flashes in Guangzhou

Six downward negative flashes terminated on tall structures in Guangzhou are analyzed. The three-dimensional (3-D) lightning channels are reconstructed from dual-station optical observations. For each reconstructed 3-D upward connecting leader (UCL) channel, its 3-D length and speed are calculated. The 3-D length values of the six positive UCLs range from 180 to 818 m. There are 38 3-D speed values which are calculated combining the 3-D UCL channel and the high-speed images for the six UCLs. The 3-D speed values range from 0.8 to 143 x 10(5) m s(-1) and four of them (11%, 4/38) are on the order of 10(6) m s(-1). For comparison, the corresponding two-dimensional (2-D) parameters are calculated using the single-station high-speed images. The values of the 2-D length and 2-D speed range from 147 to 610 m and 0.3 to 10.6 x 10(5) m s(-1), respectively. From the statistical analysis, we determine that the average value of the 3-D speed is 1.3 times that of the 2-D speed. When the time is approaching the return stroke (RS), the propagation speed of the UCL is increasing. All of the four 3-D speed values on the order of 10(6) m s(-1) occur less than 0.2 ms prior to the RS. When the 3-D length is shorter than 300 m, 77% (20/26) of the corresponding 3-D speed values are smaller than 5 x 10(5) m s(-1). When the 3-D length is longer than 300 m or the UCL tip height is higher than 650 m, all of the corresponding 3-D speed values are faster than 5 x 10(5) m s(-1). (C) 2014 Elsevier B.V. Identification and engineering a C4-dicarboxylate transporter for improvement of malic acid production inAspergillus niger

Modification of C-4-dicarboxylate transport processes is an important strategy for the development of efficient malic acid producing cell factory inAspergillus niger. However, there is a lack of identification and functional research of malic acid transport proteins, which seriously hinders the construction of high-yield malic acid metabolic engineering strains. A C-4-dicarboxylate transport protein (DCT) DCT1 is identified as major malic acid transport protein and exhibits significant elevation in malic acid production when overexpressed. DCT1 is found by homology searches and domain analyses withSpMAE1 fromSchizosaccharomyces pombeas the template. Phylogenetic and domain analyses show that DCTs belong to voltage-dependent slow-anion channel transporter (SLAC1) family and are members of Tellurite-resistance/Dicarboxylate Transporter (TDT) Family. DCT1 disruption dramatically decreases malic acid titer by about 85.6% and 96.2% at 3 days and 5 days compared with the parent strain, respectively. Meanwhile, the citric acid titers increase by 36.4% and 13.7% at 3 days and 5 days upon DCT1 deficiency. These results suggest that DCT1 is the major malic acid transporter inA. niger. Overexpression ofdct1with its native promoter significantly improves malic acid production yielding up to 13.86 g/L and 30.79 g/L at 3 days and 5 days, respectively, which is 36.8% and 22.8% higher than those in the parent strain. However, the citric acid has no significant change during the 5-day fermentation. These results demonstrate the importance of C-4-dicarboxylate transporters for the efficient production of malic acid. Furthermore, enhancement of malic acid transport process is a feasible approach of efficient malic acid production in this citric acid producingA. nigerstrain.

Observation of Residual Voltage in Low-Voltage Surge Protective Devices Due to Nearby M-Components

This paper presents experimental results obtained from the Guangdong comprehensive observation experiment on lightning discharge during summer 2014. Residual voltages and current flows in a surge protective device (SPD) connected to an overhead distribution line were observed due to nearby M-components of a triggered lightning flash. Residual voltages with varying characteristics caused by different M-components are analyzed at two terminals of the SPD, and corresponding formation mechanisms for the different characteristics are also discussed. Our results show that the residual voltages at two terminals of the SPD caused by three types of M-component are determined from both the line induction and the ground potential rise (GPR) in a nearby triggered lightning at 40 m distance. The M-component with a large peak current would produce large surge energies in the power line. Under a given peak current, the M-component has greater surge energy than the return stroke. The ground terminal of the SPD is still affected by the GPR, and our results also show that the continuing current superimposed with M-components is an important factor in determining the magnitude of line coupling in power distribution lines when a lightning flash occurs nearby.
