In this work, we use the quantum potential approach to quantum mechanics to show that the Madelung-Bohm trajectories for a particle in a constant gravitational field can be related to those of a free particle by means of a quantum point transformation defined in the extended configuration space. We find that the point transformation also gives a connection between the corresponding quantum Hamiltonians determined by the solutions of the corresponding Schr & ouml;dinger equations for these two problems. We show that the Madelung-Bohm trajectories determined by the stationary solutions to the Schr & ouml;dinger equation for the particle in a constant gravitational field are straight ones, while the corresponding ones for the free particle are parabolic trajectories. The Airy beam is one example of this type of solution. We study the properties of a solution to the Schr & ouml;dinger equation for a free particle with phase singularities (zeroes), and we find that the corresponding solution to the Schr & ouml;dinger equation for the particle in a constant gravitational field also has zeroes at the same spacetime points. However, the Madelung-Bohm trajectories determined by the two solutions are totally different. Furthermore, we remark that similar results can be directly obtained for the paraxial wave equation. Digital Twin-Based Active Learning for Industrial Process Control and Supervision in Industry 4.0

The integration of Digital Twin technology into active learning environments has been established as an innovative strategy to optimize engineering education. This study focuses on the development and evaluation of a learning tool based on Digital Twin technology, enabling interaction and communication between a physical teaching module and a virtualized environment representing the industrial cocoa production process. The system was operated by students using an S7-1200 PLC and an HMI interface developed in WinCC, facilitating real-time process monitoring and control via IoT. To assess its impact, an experimental design was implemented, comparing a pre-test using conventional teaching modules with a post-test incorporating Digital Twin technology. The results indicate a significant improvement in students' accuracy and efficiency, with a 38% reduction in task execution time. This improvement is attributed to the system's immersive and interactive capabilities, which allow for the simulation of actuator and sensor behavior under real-world conditions. These findings highlight the potential of Digital Twins not only as educational tools but also as valuable assets for industrial process optimization in the cocoa sector, aligning with Industry 4.0 principles.

Comparison of RT-qPCR With Branched DNA to Quantify a Lipid Nanoparticle-Encapsulated mRNA Therapeutic in Serum and Liver Tissue Samples From Nonclinical PK Studies

While the branched DNA (bDNA) assay is an established bioanalytical method for measurement of lipid nanoparticle (LNP)-encapsulated messenger RNA (mRNA) pharmacokinetic parameters, reverse transcription-quantitative polymerase chain reaction (RT-qPCR) has been considered as an alternative platform. RT-qPCR and bDNA platforms were compared for sensitivity, specificity, correlation, and overall assay performance using serum and tissue samples from 2 nonclinical mouse studies of a therapeutic mRNA candidate, LNP-PAH-mRNA, which encodes for human phenylalanine hydroxylase enzyme. Pharmacokinetic parameter noncompartmental analysis was completed using Phoenix WinNonlin. The assays were compared using simple linear regression and Bland-Altman analyses. Sensitivity ranged from 0.05 to 6.40 ng/mL for the bDNA assays, from 0.00000761 to 7.61 ng/mL in serum, and from 0.000179 to 179 ng/g in tissue for the RT-qPCR assay. Inter-assay accuracy was within +/- 10%, inter-assay precision was <= 10%, and the total error for both assays was <= 20%. RT-qPCR serum mRNA concentrations were 2- to fourfold lower compared with the bDNA assay, whereas tissue samples were comparable between assays. A linear relationship with - 0.37 to - 0.02 systematic bias demonstrated acceptable concordance. Bland-Altman plots demonstrated close equivalence, with a negative bias of < 0.5, and >= 95% of the data points were within the 95% limits of agreement. The comparison of the RT-qPCR with bDNA assay platforms for quantification of pharmacokinetic properties of an mRNA-LNP therapeutic has demonstrated acceptable concordance. This comparison reinforces the use of the RT-qPCR, a widely accessible strategy, as an alternative platform for the quantification of subsequent mRNA-LNP therapeutics.

After the enormous impact of the Covid-19 crisis, which led to abrupt and drastic changes in our daily lives, it is crucial to focus on urban planning objectives for the coming years. Cities play a vital role in creating wealth and ensuring the well-being of all their inhabitants. Major international efforts to contribute to a fairer, healthier, and more sustainable world have been compromised by the long chain of effects of the pandemic on people in all countries of the world. The current research focused on the application of the Delphi method to 24 experts for sustainable urban planning post-Covid in Cuenca, Ecuador, to identify the most and least important SDG-11 targets to overcome the impacts derived from Covid-19 in the present and post-pandemic future of the city. Only five targets out of twelve were selected, including ¨safe and affordable housing¨, ¨inclusive and sustainable urbanisation¨ and ¨implement policies for inclusion, resource efficiency and disaster risk reduction¨. This reflects the preference of experts for environmental objectives of urban sustainability and social objectives over economic ones. These can form a basis for the implementation of future public policies and sustainable urban planning strategies for the city © 2025, The Authors. All rights reserved.

Change in Youth Mental Health During the COVID-19 Pandemic in a Majority Hispanic/Latinx US Sample

Objective: Children and adolescents, individuals from racial and ethnic minority groups, and those with mental health conditions may be at greater risk for worsened mental health because of the COVID-19 pandemic. This study examined change in mental health from before to during the pandemic among predominantly Hispanic/Latinx adolescents. Method: A total of 322 young adolescents (mean age = 11.99 years, 55% female and 45% male), with a racial/ethnic composition of 72.7% Hispanic/Latinx, 9.3% Black or African American, 5.9% multiple races, 5.0% Asian, 1.6% White, and 1.2% American Indian, completed a mental health screening measure prior to the COVID-19 pandemic and at 3 time-points beginning 1 month after COVID-19 stay-at-home measures were implemented. A subsample also completed a survey about their experience at home during COVID-19. Repeated-measures mixed analysis of covariance was used to evaluate change in each mental health domain, and whether youths who had elevated symptoms at baseline differed in their level of change, controlling for age and gender. Results: For youths who had elevated levels of mental health problems before the pandemic, symptoms were significantly reduced across domains during the pandemic. Reductions in internalizing, externalizing, and total problems were clinically significant. For other youths, there were statistically significant reductions in internalizing and total problems, and no change in attention or externalizing problems. Post hoc analyses revealed that better family functioning was consistently related to lower mental health symptoms in youths during COVID-19 follow-ups. Conclusion: COVID-19 stay-at-home regulations may offer protective effects for youth mental health. Study results may be specific to this population of predominantly Hispanic/Latinx youths from a large city in the southwestern United States.

Serum sample stability in ligand-binding assays: challenges in assessments of long-term, bench-top and multiple freeze-thaw

Chris Macaraeg has been a lead scientist for method development, validation, and study support intended for regulated pre-clinical/clinical studies within the Pharmacokinetics and Drug Metabolism department at Amgen Inc, Thousand Oaks, CA. He joined Amgen in 2006. His expertise also includes automation and method transfer to CROs. Chris received his BS degree in Physiological Science and Neuroscience from the University of California, Los Angeles, CA and MS in Forensic Science from Pace University, New York, NY. Stability of therapeutic proteins in biological matrix is an important parameter to evaluate in bioanalytical support of regulated nonclinical or clinical studies. Despite industry guidance publications, many questions still arise as to how these practices are implemented to establish therapeutic protein stability in bioanalytical method validations. This article presents findings from long-term, bench-top and freeze-thaw stability assessments for three therapeutic monoclonal antibodies using either ELISA or electrochemiluminescent technology. Studies illustrate the principles and challenges in stability tests which represent scenarios that samples will likely encounter during sample analysis. Thoughtful consideration of each study requirements and a fit-for-purpose approach is essential in successful establishment of the sample stability parameters in method validation.

The increased energy demands of the rising global population in a fossil fuel-based energy economy have caused unprecedented anthropogenic CO2 emissions, which presents a potentially irreversible threat to human societies. The imperative transition to carbon-neutral or carbon-negative energy solutions requires the interplay of advanced technologies that can provide electricity generated from renewable sources, including wind, solar, and hydropower, while also providing energy-dense liquid fuels for long-term storage. To this end, electrochemical processes such as CO2 reduction, N2 reduction and hydrogen evolution could provide chemical feedstocks and fuels currently obtained by large-scale emission processes without requiring massive changes to the present energy infrastructures. The economic viability of this approach is highly dependent on the design of catalysts from earth-abundant elements that exhibit high activity and selectivity along with chemical and catalytic stability over long periods of operation. In aqueous solutions, CO2 and N2 reduction must compete with the kinetically more facile hydrogen evolution reaction, which is one of the main challenges in achieving catalytic selectivity. Therefore, understanding hydrogen adsorption interactions on the catalyst surface can simultaneously assist in improving hydrogen evolution activity, as well as increasing selectivity for CO2 and N2 reduction reactions. To this end, compositionally diverse families of materials such as molybdenum chalcogenides (sulfides, selenides, and tellurides) offer an ideal platform to evaluate hydrogen adsorption interactions and develop composition-structure-function relationships. This dissertation details an integrative research approach that includes the (1) synthesis of molybdenum chalcogenides compositions through approaches that allow facile compositional changes as well as control of morphology and dimensionality, (2) characterization of their local and electronic structures,

Understanding centrifugal casting in the manufacture of functionally graded materials

Centrifugal casting is a shaping technique intended for ceramic tubular structure manufacture, where the particle size and density are exploited to produce asymmetrical membranes and functionally graded materials. However, some well-established insights about particle segregation are debatable and remain unclear. For instance, small particles do not necessarily stay in the inner region, and the bigger particles do not accumulate in the outer. Herein several manufacturing parameters were studied through Taguchi's robust design, using discrete element method-based simulations to generate the data. Alumina powder was used as model material and water as the liquid phase, to assess the green cast formation time, the cast thickness, the roundness, the changes in relative density, and particle size distribution along the cast's cross-section. The mean particle diameter and the rotation speed are the most influential parameters for the casting time. The volume fraction of solids on the precursor slurry is decisive regarding the cast structural properties, and particle segregation is negligible, except for size differences above one order of magnitude. When a fraction of denser nickel powder was added, density segregation was also observed, but the size differences can overshadow its effect. In addition, alumina and nickel particles were cast in a lab-scale centrifuge and experimentally validated the segregation of particles. The centrifugal casting method was successfully applied for producing the Al2O3-Ni composites with a gradient distribution of the Ni phase.

Background: Between 32% to 85% of first-generation immigrant Latinx youth experience separation from at least one primary caregiver (Lu et al., 2020; Suarez-Orozco et al., 2002). Research unequivocally associates caregiver separation with detrimental outcomes such as an increased likelihood for emotional and behavioral difficulties (e. g. Lu et al., 2020). Caregiver separation also has the potential to diminish resilience. Previous research suggests strong peer attachments may act as a buffer for negative effects of adversity and promote positive adaptation (e. g. He et al., 2018; Ju & Lee, 2018). Thus, this study sought to examine the relation between caregiver separation and resilience; and investigate the buffering effects of peer attachment in the relationship between caregiver separation and resilience in a sample of recently immigrated adolescents. Method: The sample (n = 60) was composed of adolescents recruited from a high school designed to serve recently immigrated youth. Participant age ranged from 15-25 years old (M=19.71, SD=2.23), and all participants self-identified as Hispanic (100%). Measures for this study included the Migration Experiences Interview (Venta et. al, 2019), the Inventory of Parental and Peer Attachment (Armsden & Greenberg, 1987), and the Resilience Scale (Wagnild & Young, 1993). Results: Analyses indicated a significant effect of separation on resilience scores (F (3, 56) = 4.605, p <.01, η2 =.198). Post-hoc analyses indicated youth who did not experience caregiver separation had significantly higher resilience scores than youth who were separated from both parents. Additionally, analyses revealed moderation effects of peer attachment for youth separated only from their father (b = -.794, p <.05) and separated from both parents (b =.958 p <.05). Mechanical properties of the particle associated with the Laguerre-Gauss beams via the quantum potential point of view

The aim of the present work is to show that the Laguerre-Gauss beams determine a Hamiltonian system with two degrees of freedom for a particle of mass m = 1 under the action of the quantum potential determined by these beams. We show that the integral curves of the Poynting vector constitute a particular subset of solutions to the corresponding Hamilton equations and that the geometrical light rays associated with these twisted beams turn out to be the tangent straight lines to the exact optics energy trajectories at the zeroes of the quantum potential. By using the Hamiltonian formulation, we determine the velocity, the linear momentum, the angular momentum, the torque, and the areal velocity characterizing the particle associated with the Laguerre-Gauss beams.
