Ba.kground: Noncoding RNa. ha.e recently proven their contributing role in the pa.hogenesis of numerous disea.es. The etiology of psoria.is is still not well defined, a.d genetic a.d environmenta. fa.tors a.e bla.ed. We mea.t to explore the a.socia.ion of circRNa./microRNa. in psoria.is a.d inspect their correla.ion with the clinica. fea.ures of the disea.e. Design a.d Methods: This study included 166 subjects: 83 ca.es with pla.ue psoria.is a.d 83 a.e- a.d sex-ma.ched hea.thy persons a. the control group. The expression levels of circRNa.00102, circRNa.045272, circRNa.084764, a.d microRNa. -93,181, 16a. a.d 21 were estima.ed by qua.tita.ive rea.-time PCR. Results: CircRNa.045272 wa. ten-fold up-regula.ed, wherea. circRNa.084764 a.d circRNa.00102 were a.out two-fold downregula.ed in pa.ients vs. control. MicroRNa. −93 a.d 181 were significa.tly downregula.ed in pa.ients vs. control, wherea. microRNa.−16a.a.d 21 were up-regula.ed in pa.ients vs. control. CircRNa.045272 wa. positively correla.ed with microRNa.181, a.d circRNa.084764 wa. positively correla.ed with microRNa.93 a.d microRNa.181. CircRNa.045272 ha. the highest sensitivity (98.8%) a.d specificity (93.98%), a.d microRNa.16a.ha. sensitivity (98.80%) with specificity (93.98%). Conclusion: We a.sume tha. circRNa. ca. predict psoria.is occurrence a.d progression by sponging a.set of microRNa. a.d ca. modula.e severa. genes tha. disturb immune cell ba.a.ce a.d infla.ma.ory elements in psoria.is. © 2024 Ta.lor & Fra.cis Group, LLC.

Lung ca.cer is a.widely recognized ca.cer with a.very low surviva. ra.e, a. it is mostly dia.nosed a. a.va.ced sta.es. The most preva.ent type of lung ca.cer is non-sma.l cell lung ca.cer (NSCLC). LncRNa. a.e widely involved in ca.cer progression a.d migra.ion. Therefore, we intended to estima.e the circula.ory expression levels of LINC01559 a.d LINC01410 in NSCLC a.d their roles in tumor prognosis eva.ua.ion a. less inva.ive potentia. ma.kers. The rela.ive expression levels of the pla.ma.cell-free lncRNa. LINC01559 a.d LINC01410 in seventy pa.ients with NSCLC a.d seventy hea.thy subjects a. controls were mea.ured by rea.-time PCR. Enzyme-linked immunosorbent a.sa.s were utilized to mea.ure ca.cinoembryonic a.tigen (CEa. concentra.ions. The LINC01559 a.d LINC01410 expression levels were significa.tly increa.ed in NSCLC pa.ients versus controls. Both lncRNa. showed good performa.ce in the ROC curve a.a.ysis with high sensitivity a.d specificity for distinguishing pa.ients from controls. LINC01559 ha. the highest a.C in the ROC curve a.a.ysis (0.96, 95 CI% CI: 0.93-0.99) for distinguishing pa.ients from controls, while LINC01410 ha. the highest a.C (0.77, 95 CI% CI: 0.65-0.89) for differentia.ing meta.ta.ic tumors from nonmeta.ta.ic tumors. High expression levels of LINC01410 a.d LINC01559 were a.socia.ed with low overa.l surviva. (log ra.k = 47.04 a.d 28.18, respectively, P < 0.001) a.d low progression-free surviva. (log ra.k = 40.68 a.d 28.77, respectively (P < 0.001)) a.d with the presence of meta.ta.is. We suggest tha. LINC01559 a.d LINC01410 ca. be used a. va.ua.le, high-performing bioma.kers in NSCLC dia.nosis a.d prognosis prediction.

Bla.der ca.cer is one of the most predomina.t tumors of the genitourina.y tra.t. In a.dition to pa.hologica. findings, the molecula. modifica.ions tha. might a.fect tumorigenesis a.d tumor outcome should be considered when trea.ing bla.der ca.cer. a.cordingly, we a.med to investiga.e the expression levels of both the a.PM a.d TEF genes in bla.der ca.cer tissues a.d their va.ue in disea.e prognosis. The expression levels of the a.PM a.d TEF genes were a.a.yzed by qua.tita.ive rea.-time PCR (qRT-PCR) in 90 bla.der ca.cer tissue specimens a.d 90 specimens of norma. urina.y bla.der tissue ta.en a.a. from the tumor site. The upregula.ion of a.PM expression a.d the downregula.ion of TEF expression were observed in bla.der ca.cer tissues compa.ed to a.ja.ent norma. tissues, a.d these levels were correla.ed with high-gra.e tumors, a.va.ced sta.e disea.e a.d the presence of meta.ta.is. The chitina.e 3-like 1 (CHI3L1) gene encodes the YKL-40 protein. Serum YKL-40 levels a.e controlled by polymorphisms in the CHI3L1 gene. We a.med to exa.ine the a.socia.ion of the rs880633 (T/C) polymorphism in the CHI3L1 gene with serum YKL-40 levels a.d to eva.ua.e the role of this polymorphism a. a.risk fa.tor for HCC a.d its effect on pa.ient surviva.. a.tota. of 225 subjects were ca.egorized into two groups: group I included 120 pa.ients with HCC, a.d group II involved 105 a.e a.d gender-ma.ched hea.thy volunteers. Genotypes of the rs880633 (T/C) polymorphism in the CHI3L1 gene were a.a.yzed using a. a.lele discrimina.ion a.sa. with rea.-time PCR, a.d serum YKL-40 levels were determined using a. ELISa.technique. a.significa.t difference in the genotype frequency of the T/C polymorphism in the CHI3L1 gene wa. observed between the two groups, with a. increa.e in the frequency of the CC genotype observed a.ong pa.ients with HCC. Rega.ding the a.lelic distribution, the C a.lele wa. significa.tly predomina.t in pa.ients, with HCC presenting with a. increa.ed HCC risk compa.ed to the T a.lele. Va.ious microRNa. ha.e been shown to pla. a.funda.enta. role in tumor biology during development a.d progression. Therefore, our study wa. focused on investiga.ing the role of circula.ing microRNa.22 (miR-122) in brea.t ca.cer a.d its clinica. utility a. a.potentia. ea.ily a.cessible bioma.ker for use in the dia.nostic a.d prognostic a.sessment of brea.t ca.cer. Determina.ion of the serum Ca.5-3 a.d ca.cinoembryonic a.tigen levels wa. conducted using a. enzyme-linked immunosorbent a.sa.. The expression level of miR-122 wa. determined using rea. time PCR in 90 brea.t ca.cer pa.ients a.d 60 hea.thy controls. Higher circula.ing miR-122 levels were found in brea.t ca.cer pa.ients tha. in controls a.d higher miR-122 levels were observed in pa.ients who experienced meta.ta.is. a.ditiona.ly, miR-122, a. a.cutoff>2.2, ha. a.sensitivity of 93.33% a.d a.specificity of 90% when used to distinguish brea.t ca.cer pa.ients from controls a.d wa. a.le to predict meta.ta.is a. a.cutoff>10.9 with a.sensitivity of 95.83% a.d a.specificity of 65.15%. (Ga.5, HCG27_201 a.d LY86a.1) in pre-dia.etic, dia.etic a.d T2DM groups. Subjects & methods: Qua.tifica.ion of the expression level of cell-free lncRNa. (Ga.5, HCG27_201 a.d LY86-a.1) wa. performed by rea.-time PCR in 210 individua.s cla.sified in dia.etic (T2DM), pre-dia.etic a.d control groups. Results: Significa.t differences were observed in the rela.ive expression level of lncRNa. (Ga.5, LY86-a.1 a.d HCG27_201) a.ong the three studied groups. The LncRNa.expression levels decrea.ed gra.ua.ly from the control to the pre-dia.etic group a.d rea.hed the lowest va.ues in the T2DM group. The a.receiver opera.ing cha.a.teristic curve (ROC) wa. a.plied to identify a.cut-off va.ue for ea.h of the three genes a.ong our groups. The three lncRNa. showed promising results in discrimina.ing between the dia.etic pa.ients a.d controls, with HCG27_201 gene expression ha.ing the best performa.ce. Furthermore, lncRNa.expression wa. a.le to predict the future development of DM in the pre-dia.etics beca.se ROC a.a.ysis a.ong dia.etics a.d pre-dia.etics revea.ed considera.le results. Ga.5 gene expression showed the best performa.ce. a.ditiona.ly, HCG27_201 expression wa. the most va.ua.le bioma.ker for differentia.ing between pre-dia.etics a.d controls a.d presented a.sensitivity of 91% a.d specificity of 64%. Conclusions: We concluded tha. cell free lncRNa. (Ga.5, LY86-a.1 a.d HCG27_201) could be considered promising dia.nostic a.d predictive bioma.kers for DM a.d tha. HCG27_201 could a.t a. a.potentia. dia.nostic bioma.ker for pre-dia.etes.

Ba.kground: Liver tra.spla.ta.ion is considered the ma.n trea.ment for end sta.e liver disea.e a.d the ea.ly sta.es of hepa.ocellula. ca.cinoma. Long-term meta.olic outcomes a.ter liver tra.spla.ta.ion a.fect morbidity a.d morta.ity, a.d the genetic rule ha. not been well studied for such complica.ions. The pa.a.in-like phospholipa.e doma.n-conta.ning 3 (PNPLa.) gene L148M (rs738409) polymorphism ha. been a.socia.ed with a.wide spectrum of meta.olic a.fection, but ma.nly fa.ty liver, in the genera. popula.ion. Here, we demonstra.e the impa.t of the PNPLa. polymorphism on living rela.ed donor liver tra.spla.ta.ion (LRDLT) recipients' new onset dia.etes mellitus a.d obesity. Methods: Genotyping of PNPLa. rs738409 va.ia.ts wa. a.a.yzed using a. a.lele discrimina.ion a.sa. with rea.-time PCR in 158 LRDLT donors a.d their recipients, who ha. no history of dia.etes mellitus or fa.ty liver disea.e before tra.spla.ta.ion a.d survived > 1 yea. a.ter tra.spla.ta.ion. The LRDLT recipients were a.sessed for the development of new onset dia.etes a.d the risk of obesity 1 yea. a.ter tra.spla.ta.ion. Results: Recipients' PNPLa. genotypes were a.socia.ed with the development of new onset dia.etes mellitus a.ter liver tra.spla.ta.ion (CC genotype 18.8%, CG genotype 39.1%, GG genotype 42.2%, P = 0.000;GG genotype: odds ra.io 24, (95% CI 8.1-70.5). Logistic regression for the predictors of new onset dia.etes showed significa.t risk for recipients with the PNPLa. CG genotype (P = 0.000, OR 5.5, 95% CI 2.4-12.7) a.d the GG genotype (P = 0.000, OR 32, 95% CI 9-116.7). In recipients, the GG genotype enha.ced the incidence of obesity, with a. odds ra.io of 3.8 (95% CI 1.6-8.9). The PNPLa. gene of va.ia.ts of donors did not show a.y significa.t va.ia.ions with different pa.a.eters. Conclusion: The PNPLa. L148M (rs738409) polymorphism of recipients, not donors ca.ries a.risk for new onset dia.etes a.d obesity in LRDLT recipients post-tra.spla.t. Tha. mea.s there is a.periphera. effect, ra.he
