Functional characterization and comparison of lycopene epsilon-cyclase genes in Nicotiana tabacum

Background: Lycopene epsilon-cyclase (epsilon-LCY) is a key enzyme in the carotenoid biosynthetic pathway (CBP) of higher plants. In previous work, we cloned two Nt epsilon-LCY genes from allotetraploid tobacco (Nicotiana tabacum), Nt epsilon-LCY2 and Nt epsilon-LCY1, and demonstrated the overall effect of Nt epsilon-LCY genes on carotenoid biosynthesis and stress resistance. However, their genetic and functional characteristics require further research in polyploid plants. Results: Here, we used CRISPR/Cas9 to obtain Nt epsilon-LCY2 and Nt epsilon-LCY1 mutants in allotetraploid *** K326. Nt epsilon-LCY2 and NtE-LCY1 had similar promoter cis-acting elements, including light-responsive elements. The Nt epsilon-LCY genes were expressed in roots, stems, leaves, flowers, and young fruit, and their highest expression levels were found in leaves. Nt epsilon-LCY2 and Nt epsilon-LCY1 genes responded differently to normal light and high light stress. Both the Nt epsilon-LCY2 and the Nt epsilon-LCY1 mutants had a more rapid leaf growth rate, especially nt epsilon-Icy2-1. The expression levels of CBP genes were increased in the nt epsilon-Icy mutants, and their total carotenoid content was higher. Under both normal light and high light stress, the nt epsilon-Icy mutants had higher photosynthetic capacities and heat dissipation levels than the wild type, and this was especially true of nt epsilon-Icy2-1. The reactive oxygen species content was lower in leaves of the nt epsilon-Icy mutants. Conclusion: In summary, the expression patterns and biological functions of the Nt epsilon-LCY genes Nt epsilon-LCY1 and Nt epsilon-LCY2 differed in several respects. The mutation of Nt epsilon-LCY2 was associated with a greater increase in the content of chlorophyll and various carotenoid components, and it enhanced the stress resistance of tobacco plants under high light.

Training with One2MultiSeq: CopyBART for social media keyphrase generation

Keyphrase generation, which can help people obtain key information from a long document (social media posts or scientific articles) in a short time, has made significant progress in recent years, especially for training by concatenating keyphrases with a predefined order. However, when using beam search for keyphrase generation, models tend to repeatedly generate the highest priority keyphrase type in each beam branch, which causes the model to weaken the generation performance on the underdog keyphrase type. To tackle this, we introduce the One2MultiSeq paradigm, which allows the model to train with two sets of keyphrases that have completely opposite connection orders. Moreover, given that social media content is often colloquial, informal, and multimodal (comprising not just text but also images), these properties necessitate the incorporation of a priori knowledge for models to effectively process such information. However, contemporary models lack this requisite capacity, thereby limiting their ability to proficiently handle these discrete data elements. To overcome this, we incorporate the pretrained model BART as our backbone architecture and employ a copy mechanism to further augment its keyphrase generation capabilities. Experimental results show that our method outperformed relatively advanced models, with gains of 3.51, 1.55, and 2.47 percentage points in F1@1, F1@3, and MAP@5, respectively, on the unimodal Twitter dataset;3.23, 2.68, and 4.07 on the multimodal Tweet dataset;and increases of 4.32, 0.32, and 7.07 in F1@3, F1@5, and MAP@5, respectively, on the StackExchange dataset.

Characterization and formation mechanism of the catastrophic flash flood-debris flow hazard triggered by the July 2023 extreme rainstorm in Hantai Gully of Beijing, China

Recent extreme rainfall events have led to frequent flash flood-debris flow hazards in the mountainous regions of western and northern Beijing, China, resulting in significant loss of life and property. To decipher the formation mechanism and hazard characteristics of these local disasters, the July 2023 flash flood-debris flow in Hantai Village (HTV) and Hantai Gully (HTG) of Changping District, Beijing, was selected as the focus of this study. This study acquired topographic and geological data of the study area through regional disaster investigation, field surveys in severely affected regions, and indoor assessments and analyses, utilizing 1: 1000 topographic maps and 1: 20,000 geological maps sourced from unmanned aerial vehicle (UAV) remote sensing images. Then, the characteristics of rainfall and the associated hazard processes (flash flood and debris flow) were systematically investigated and analyzed. By further exploring the formation mechanism of these events, relevant hazard prevention and mitigation suggestions were proposed. The results indicate that the total flood volume in the upstream channel of HTG is approximately 6870 m3 within 1 h, demonstrating significant destructive power. The leading edge of the flash flood/debris flow exits the gully (the southern entrance of HTV) at an instantaneous speed of 74.47 km/h and maintained a speed of 16.42 km/h by the time it reached the northern exit of HTV (outlet of the HTG), with a movement time of 1.83 min. This brief event duration presents significant challenges for timely evacuation. This catastrophic flash flood-debris flow event is identified as a compound hazard triggered by extreme rainfall from the remnant circulation of Typhoon Doksuri, with the sudden increase in surface runoff amount due to short-term heavy rainfall being the primary cause. Hydrothermal Synthesis of Ni-MOF Vulcanized Derivatives for High-Performance Supercapacitors

A series of nickel sulfides derived from the hexagonal Ni-MOF are vulcanized through adjusting the hydrothermal time and the thiourea concentration. Among all the obtained nickel sulfides, one sample Ni-S2-3, which inherits the Ni-MOF's morphology, shows the best electrochemical performance with a remarkable specific capacitance of 1128 Fg(-1) at 1 Ag-1, a rate capacitance of 50% and a long cycle life of 74% retention after 5000 cycles. Furthermore, the asymmetrical supercapacitors (SCs) based on Ni-S2-3//AC exhibit a good supercapacitive performance with a maximum power density of 16.3 Wh kg(-1) at a power density of 800 Wkg(-1). All these results indicate that vulcanizing Ni-MOF is an effective way to fabricate a superior electrode material with excellent electrochemical performance for SCs.

Acquisition of rhotic sounds has been shown to be challenging for children in many languages. However, less is known about the acquisition of syllable-final rhotic sounds, known as the r-suffix or erhua in Mandarin Chinese. The current study examines the acquisition of Mandarin Chinese r-suffix among preschool children. Words with r-suffix were extracted from the CASS-CHILD corpus, a child language corpus, and compared with adult production. Results showed that r-suffix production can emerge as early as one and a half years old, but children do not achieve the same level of production as adults at the age of 6. Accuracy in its usage increased to 50% by the age of four and remained steady thereafter. Common errors observed were the omission and misarticulation of the r-suffix, as well as the incorrect production of the preceding vowels. Acoustic analysis showed that children's r-suffix had higher F3 and F2 compared to those of adults, with no significant differences in F3 and F2 observed among children of different ages. Our results suggest that although the production of the r-suffix begins early, complete mastery of it may be achieved relatively late in the developmental process.

Sarcopenia and COVID-19 Outcomes

Coronavirus disease 2019 (COVID-19) spread rapidly and became a severe global public health threat. Older adults have a high risk of COVID-19 and its associated mortality. Sarcopenia has emerged as a predictor of poor outcomes in COVID-19 patients, including lengthy hospital stays, mortality, intensive care unit admission, need for invasive mechanical ventilation, and poor rehabilitation outcomes. Chronic inflammation, immune dysfunction, respiratory muscle dysfunction, and swallowing dysfunction may underlie the association between sarcopenia and the poor outcomes of COVID-19 patients. Interleukin 6 receptor blockers (tocilizumab or sarilumab) are recommended for treating patients with severe COVID-19, and their therapeutic effects on sarcopenia are of great interest. This review aimed to analyze the current reports on the association between sarcopenia and COVID-19 and provide an update on the contribution of sarcopenia to the severity and adverse outcomes of COVID-19 and its underlying mechanisms. We also aimed to explore the different screening tools for sarcopenia concurrent with COVID-19, and advocate for early diagnosis and treatment of sarcopenia. Given that the fight against the COVID-19 pandemic may be long-term, further research into understanding the effects of sarcopenia in patients infected with the Omicron variant is necessary.

Peculiar Effect of Water on Tribological Properties of Natural Deep Eutectic Solvent

Deep eutectic solvents (DESs) have emerged as promising green liquid lubricants for solving tribological problems due to their economic and excellent physicochemical and lubrication properties. However, a trace amount of water would affect their structure, physicochemical properties, and tribological properties. The effect of water on the tribological properties of DES is still unclear and needs further investigate. Herein, we carried out a systematic investigation into the chemical structure, rheological properties, and tribological performance of DES-water (DES-W) binary systems constructed by combining DES with varying contents of water. The results revealed that low levels of water in DES had a minimal impact on its chemical structure but affected its fluidity and viscosity. Frictional experiments demonstrated that DES-W binary systems displayed a reduced coefficient of friction from 0.094 to 0.025 compared to pure DESs and manifested outstanding antiwear properties under a high-load condition. This was attributed to the formation of hydration layers, adsorption layers, and tribochemical films at the tribointerface through physicochemical adsorption and tribochemical reactions. Our findings not only foster the design and development of green lubricating materials but also expand the engineering applications of DESs to solve wear-related mechanical failures in practical application.

Macromolecular crowding facilitates rapid fabrication of intact, robust cell sheets

Objectives To develop a rapid and simple method to fabricate intact, robust cell sheets from common cell culture dishes by combination of a macromolecular crowding (MMC) reagent and vitamin *** It was found that 3T3 fibroblasts or human bone marrow mesenchymal stem cells (hBMSCs) and their secreted cell derived extracellular matrices could be easily detached as intact cell sheets under gently pipetting after treated by MMC and vitamin C for 4 days. This method also allowed fabrication of functional multi-layered hepatic cell sheets by culturing 10 x 10(4) cells/cm(2) HepG2 cells on top of confluent 3T3 fibroblast layers. What's more, MMC induced hBMSC cell sheets demonstrated 1.9 times larger area and 1.6 times greater cell number than that of cell sheets harvested from temperature-responsive cell culture *** MMC based method make it possible to fabricate various types of cell sheets more conveniently, economically, and thus may facilitate wide application of cell sheet technology.

Engineering cellular microenvironments with biomaterials is an effective strategy for endothelial cell expansion and functionality in vascular tissue engineering. The basement membrane (BM) is a natural vascular endothelium microenvironment that plays an important role in promoting rapid expansion and function of endothelial cells. However, mimicking the crucial function of BM with an ideal biomaterial remains challenging. In this study, we developed a cell-derived decellularized extracellular matrix (c-dECM) paper to mimic the role of BM in endothelial cell expansion and function. The results showed that c-dECM paper was a stable, biocompatible, and biodegradable scaffold that significantly promoted endothelial cell expansion by modulating cell migration, adhesion, and proliferation both in vivo and in vitro. Moreover, the biomimetic c-dECM paper can profoundly promote endothelial cell function by increasing the synthesis and release of nitric oxide (NO) and prostaglandin I2 (PGI2) and upregulating the expression of anticoagulant and vascularized genes, including thrombomodulin (THBD), tissue factor pathway inhibitor (TFPI), endothelial growth factor (VEGF) and endoglin (CD105). These data indicate that the c-dECM is a potential biomaterial for constructing vascular tissue engineering scaffolds or developing in vitro models to study the functional mechanisms of endothelial cells.
