Cordgrass Spartina alterniflora acts as a key carbon source to support macrozoobenthos in the salt marsh and nearby mudflat communities

Exotic cordgrass Spartina can influence the trophic interaction and functioning of coastal ecosystems by altering the availability or palatability of carbon sources utilized by macrozoobenthos. Previous studies focused mainly on the trophic dynamics in Spartina-invaded mangroves or marshes, but the effect of Spartina on the diet of macrozoobenthos inhabiting the nearby unvegetated mudflat remains largely unexplored. By analyzing carbon and nitrogen stable isotopes, we compare the trophic contribution of an exotic cordgrass Spartina alterniflora to the macrozoobenthos between the dense S. alterniflora area (DSA) and adjacent tidal bare mudflat (TBM) in the Hepu coast, Guangxi province. Macrozoobenthic species were more abundant in TBM than in DSA, but the difference in carbon and nitrogen stable isotopes was insignificant for the seven conspecifics between two sampling areas, where similar organic carbon sources and trophic levels for macrozoobenthos were found. The MixSIAR mixing model revealed that S. alterniflora acted as the most important carbon source for many mac-rozoobenthic species and all guilds in DSA and TBM. This study showed that S. alterniflora can benefit the adjacent bare mudflat by supplying detritus as the carbon source for macrozoobenthos, which is favorable for maintaining the biodiversity and trophic dynamics of macrozoobenthic communities in coastal ecosystems.

Objective. This investigation probed the prognostic potential for lactate dehydrogenase (LDH), melanoma inhibitory activity protein (MIA), and S-100B protein in cases of malignant melanoma. Methods. 84 cases were segregated into effective cohort (n = 64) and ineffective cohort (n = 20) depending on clinical efficacy. The cases were followed up for three years and segregated into mortality cohort (n = 29) and survival cohort (n = 55) depending upon 3-year survival. Serum LDH, MIA, and S-100B levels were compared across the effective and ineffective cohorts. Serum LDH, MIA, and S-100B levels in cases of different clinical stages were comparatively analyzed, with correlations of these indicators with the clinical stage being evaluated. ROC evaluated the prognostic potential of serum LDH, MIA, and S-100B. Cases were segregated into the high-level and low-level cohorts according to serum LDH, MIA, and S-100B levels, and the survival rates of cases were compared. Results. The levels of LDH, MIA, and S-100B in the effective cohort were significantly lower than those in the ineffective cohort. The AUC value of the composite indicator of serum LDH, MIA, and S-100B for effectiveness evaluation was (0.839). Serum LDH, MIA, and S-100B levels were positively linked to the clinical stage. AUC value of the composite indicator of serum LDH, MIA, and S-100B for prognosis evaluation prediction (0.942) was elevated compared to LDH (0.632), MIA (0.732), or S-100B (0.828) alone. Survival rate of cases of LDH & GE;30.56 mg/L (57.14%, 32/56) was lower than that of cases of LDH < 30.56 mg/L (82.14%, 23/28) (log-rank chi(2) = 4.672, P < 0.05). The survival rate of MIA & GE;5.34 ng/mL cases was lower than that of MIA Conclusion. Serum LDH, MIA, and S-100B protein levels are linked to the clinical stage. The lactate dehydrogenase, melanoma inhibitory protein, and S-100B protein are of good clinical effectiveness and have the prognostic potential for cases of malignant melanoma.

In this work, we report a single crystalline p-Si/(001) beta-Ga2O3 heterojunction diode fabricated using semiconductor grafting technology. The diode showed a breakdown voltage (V-br) of similar to 0.86 kV, which is the highest breakdown voltage reported for Si/(001) Ga2O3 heterojunction diode to date. The average peak electric field (E-m) was calculated to be >2 MV/cm near breakdown, while the specific on resistance (R-on,R-sp) was measured to be 7.7 mQcm(2), corresponding to a power figure of merit of similar to 96 MW/cm(2). Catastrophic breakdown is confirmed by optical microscope inspection. The turn on voltage (Von) of the diode was measured to be around 1.12 V, the on-off ratio was calculated to be 9 x 10(9) at -2 and 2 V, and the ideality factor was extracted to be approximately 1.18. The band structure of the diode was analyzed, and C -V measurements were also performed to understand the trapping behavior at the Si/Ga2O3 interface.

目的观察甲基多巴与拉贝洛尔联合应用于妊娠期高血压治疗的临床效果。方法采用随机数字表法将在我院接受治疗的72例妊娠期高血压患者分为实验组(n=36)和对照组(n=36)。对照组采用硫酸镁治疗,实验组在对照组的基础上加用甲基多巴联合拉贝洛尔治疗。比较两组治疗7d后收缩压(SBP)、24h尿蛋白定量、血流动力学指标水平变化情况,治疗期间不良反应发生率及分娩后新生儿情况。结果治疗后实验组患者SBP、24h尿蛋白定量、血管外周阻力及心脏指数均明显低于对照组,新生儿情况明显优于对照组,差异有统计学意义(P<0.05)。结论甲基多巴与拉贝洛尔联合给药治疗妊娠期高血压有利于提高临床疗效,治疗效果显著。

CPI-203 improves the efficacy of anti-PD-1 therapy by inhibiting the induced PD-L1 overexpression in liver cancer

Hepatocellular carcinoma (HCC) is one of the commonest lethal malignancies worldwide, and often diagnosed at an advanced stage, without any curative therapy. Immune checkpoint blockers targeting the programmed death receptor 1 (PD-1) have shown impressive antitumor activity in patients with advanced-stage HCC, while the response rate is only 30%. Inducible PD-L1 overexpression may result in a lack of response to cancer immunotherapy, which is attributed to a mechanism of adaptive immune resistance. Our study investigated that the overexpression of PD-L1 promoted the invasion and migration of liver cancer cells in vitro, and the induced overexpression of PD-L1 in the tumor microenvironment could weaken the effects of anti-PD-1 immunotherapy in a BALB/c mouse model of liver cancer. CPI-203, a small-molecule bromodomain-containing protein 4 (BRD4) inhibitor, which can potently inhibit PD-L1 expression in vitro and in vivo, combined with PD-1 antibody improved the response to immunotherapy in a liver cancer model. Cell transfection and chromatin immunoprecipitation assay manifested that BRD4 plays a key role in PD-L1 expression;CPI-203 can inhibit PD-L1 expression by inhibiting the BRD4 occupation of the PD-L1 promoter region. This study indicates a potential clinical immunotherapy method to reduce the incidence of clinical resistance to immunotherapy in patients with HCC.

Reinforced soil wall has proved to have excellent impact resistance in engineering, but less attention has paid to their failure process and dynamic response subjected to impact loading. The authors performed model tests and numerical simulations of reinforced soil walls with and without cushion under differential impact loading. The deformation, failure process and impact stress of three retaining walls were mainly analyzed, and the influence mechanism of geosynthetic and cushion on the impact performance of retaining wall were discussed qualitatively. The results showed that the impact deformation of reinforced soil wall with cushion was mainly concentrated on the cushion. The failure process of retaining wall could be divided into two stages: the soil around the impact point of retaining wall was compacted (stage I), a sliding surface formed in retaining wall gradually and retaining wall would failure (stage II). Geosynthetics benefited the sliding surface move back and cushion improved the impact resistance of retaining wall. Furthermore, stress absorption of geosynthetic and cushion was confirmed. The absorption rate of geosynthetic was related to the impact energy positively. The absorption rate of cushion was related to the impact energy positively and related to the thickness of cushion negatively. Engineers can consider the impact energy and the absorption rate of geosynthetic and cushion to design reinforced soil wall with cushion under impact loading.

ObjectiveThis study aimed to investigate the role of cholesterol metabolism-related genes in nonfunctioning pituitary neuroendocrine tumors (NF-PitNETs) invading the cavernous sinus and analyze the differences in immune cell infiltration between invasive and noninvasive ***, a retrospective analysis of single-center clinical data was performed. Second, the immune cell infiltration between invasive and noninvasive NF-PitNETs in the GSE169498 dataset was further analyzed, and statistically different cholesterol metabolism-related gene expression matrices were obtained from the dataset. The hub cholesterol metabolism-related genes in NF-PitNETs were screened by constructing machine learning models. In accordance with the hub gene, 73 cases of NF-PitNETs were clustered into two subtypes, and the functional differences and immune cell infiltration between the two subtypes were further *** clinical data of 146 NF-PitNETs were evaluated, and the results showed that the cholesterol (P = 0.034) between invasive and noninvasive NF-PitNETs significantly differed. After binary logistic analysis, cholesterol was found to be an independent risk factor for cavernous sinus invasion (CSI) in NF-PitNETs. Bioinformatics analysis found three immune cells between invasive and noninvasive NF-PitNETs were statistically significant in the GSE169498 dataset, and 34 cholesterol metabolism-related genes with differences between the two groups were obtained 12 hub genes were selected by crossing the two machine learning algorithm results. Subsequently, cholesterol metabolism-related subgroups, A and B, were obtained by unsupervised hierarchical clustering analysis. The results showed that 12 immune cells infiltrated differentially between the two subgroups. PurposeColon cancer (CC) is a cancer of the large intestine with high prevalence and poor prognosis. enhancer RNAs. Therefore, valuable tools or biomarkers for predicting patient status, directing clinical practice, and reducing overtreatment are needed. Enhancer RNAs (eRNAs), a class of noncoding RNAs transcribed from enhancers, have been shown to function as regulators of oncogene or tumor suppressor gene expression. The aim of our study was to explore the potential roles of eRNAs and their target enhancer-related genes (ERGs) in the prognosis of *** CC cases (stage I-III) from The Cancer Genome Atlas database were used as a training set, and cases from the Gene Expression Omnibus were used as the validation set. ERGs associated with prognosis were screened through three steps: potential, candidate, and prognosis ERGs. Multivariate Cox proportional hazards analysis was used to identify independent prognostic factors, and a nomogram was created. Calibration curves were drawn by comparing predicted and observed survival probability. For validation, the calibration curves and ROC analysis were also applied to two external validation sets. The biological significance and clinical application of the genes obtained were *** on the multiple tiers of strict screening, 11 prognostic ERGs were obtained, which were combined to obtain a prognosis signature. A compound nomogram integrating age, TNM classification, and the prognostic signature was constructed. The effect of secondary capping on the control of phosphorus release from sediment by activated thin-layer capping with Al-PIA

It is well-known that the activated thin-layer capping covering by secondary capping of contaminated sediment poses a threat to the inactivation of activated material. In this study, the static simulation experiment was conducted to study the effect of secondary capping thickness by sediment on the control of TP release from the sediment by aluminum-based P-inactivation agent (Al-PIA), and to propose the phosphorus adsorption pathway of Al-PIA. The results showed that Al-PIA could effectively reduce the release of phosphorus pollutants from the sediment at the capping intensity of 2 kg/m(2). When the secondary capping thickness of sediment were 0, 2, 4, 7, 10, and 15 mm, the average removal rates of TP were 87.57%, 76.39%, 61.22%, 51.32%, 41.93%, and 32.11%, respectively, indicating that the removal efficiency of phosphorus decreased with the increase of the secondary capping thickness of the sediment. The adsorbed phosphorus by Al-PIA was mainly non-apatite inorganic phosphorus (NAIP) in inorganic phosphorus. With the increase of the secondary capping thickness of sediment, the NAIP proportion of phosphorus adsorbed by Al-PIA increased. Meanwhile, the removal rate of phosphorus in the activated capping system showed a first increase and then decrease trend, and the removal rates of total phosphorus (TP), inorganic phosphorus (IP), and organic phosphorus (OP) were obvious except for that of organic phosphorus (OP).

Infection with Influenza A virus (IAV) induces severe inflammatory responses and lung injury, contributing significantly to mortality and morbidity rates. Alterations in the microbial composition of the lungs and intestinal tract resulting from infection could influence disease progression and treatment outcomes. Xiyanping (XYP) injection has demonstrated efficacy in clinical treatment across various viral infections. However, its specific effects and mechanisms against IAV remain unclear. In this study, we established an IAV infection mice model, and utilized 16 S rRNA sequencing, RNA sequencing, protein chips, and molecular docking, to investigate the mechanisms of XYP injection on altering pulmonary and gut microbiota, and identifying its target sites. We revealed that XYP injection significantly reduced mortality, weight loss, lung viral titers, and lung pathology in IAV-infected mice. XYP injection down-regulated the activity of malondialdehyde, and the levels of interleukin (IL)-1 beta, IL-5, IL-6, tumor necrosis factor-alpha, IL-18, IL-15, granulocyte colony-stimulating factor, IL-9, chemokine (C-C motif) ligand-5, and C-X-C motif chemokine ligand 5, while up-regulated the activities of glutathione peroxidase reactive and superoxide dismutase, and the level of interferon-gamma. The diversity of the pulmonary and gut microbiota was altered slightly after XYP injection. The linear discriminant analysis of the gut microbes revealed a higher proportion of potentially beneficial bacteria, including Akkermansia, Parabacteroides goldsteinii, Defluviitaleaceae, Oscillospirales, and Eubacterium_coprostanoligenes_group characterized the XYP group. Peritoneal macrophage RNA sequencing highlighted Serpinb2 as the most significantly regulated gene by XYP injection, along with consistent changes in multiple downstream Th2 structure genes.
