Multi-Granularity Relational Attention Network for Audio-Visual Question Answering

Recent methods for video question answering (VideoQA), aiming to generate answers based on given questions and video content, have made significant progress in cross-modal interaction. From the perspective of video understating, these existing frameworks concentrate on the various levels of visual content, partially assisted by subtitles. However, audio information is also instrumental in helping get correct answers, especially in videos with real-life scenarios. Indeed, in some cases, both audio and visual contents are required and complement each other to answer questions, which is defined as audio-visual question answering (AVQA). In this paper, we focus on importing raw audio for AVQA and contribute in three ways. Firstly, due to no dataset annotating QA pairs for raw audio, we introduce E-AVQA, a manually annotated and large-scale dataset involving multiple modalities. E-AVQA consists of 34,033 QA pairs on 33,340 clips of 18,786 videos from the e-commerce scenarios. Secondly, we propose a multi-granularity relational attention method with contrastive constraints between audio and visual features after the interaction, named MGN, which captures local sequential representation by leveraging the pairwise potential attention mechanism and obtains global multi-modal representation via designing the novel ternary potential attention mechanism. Thirdly, our proposed MGN outperforms the baseline on dataset E-AVQA, achieving 20.73% on WUPS@0.0 and 19.81% on BLEU@1, demonstrating its superiority with at least 1.02 improvement on WUPS@0.0 and about 10% on timing complexity over the baseline.

Green synthesis of epigallocatechin gallate-ferric complex nanoparticles for photothermal enhanced antibacterial and wound healing

Bacterial infections are a significant global health concern, particularly in the context of skin infections and chronic wounds, which was further exacerbated by the emerging of antibiotic resistance. Therefore, there are urgent needs to develop alternative antibacterial strategies without inducing significant resistance. Photothermal therapy (PTT) is a promising alternative approach but usually faces limitations such as the need for stable and environmental-friendly PTT agents and ensuring biocompatibility with living tissues, necessitating ongoing research for its clinical advancement. Herein, in this study, with the aim to develop a green synthesized PTT agent for photothermal enhanced antibacterial and wound healing, we proposed a facile one-pot method to prepare epigallocatechin gallate-ferric (EGCG-Fe) complex nanoparticles. The obtained nanoparticles showed improved good size distribution and stability with high reproducibility. More importantly, EGCG-Fe complex nanoparticles have additional photothermal conversion ability which can give photothermal enhanced antibacterial effect on various pathogens, including Gram-positive Staphylococcus aureus (S. aureus) and Gramnegative Escherichia coli (E. coli) strains. EGCG-Fe complex nanoparticles also showed powerful biofilm prevention and destruction effects with promoted antibacterial and wound healing on mice model. In conclusion, EGCG-Fe complex nanoparticles can be a robust green material with effective and novel light controllable antibacterial properties for photothermal enhanced antibacterial and wound healing applications.

Angiogenesis inhibition has become a promising therapeutical strategy for cancer treatment. Current clin-ical anti-angiogenesis treatment includes antibodies against vascular endothelial growth factor (VEGF) or VEGF receptor, fusion proteins with high affinity to VEGF receptor, and tyrosine kinase inhibitors of VEGF receptor. However, current treatments are prone to systemic toxicity or acquiring drug resistance. A nat-ural bioactive lipid 1,2-dipalmitoyl-sn-glycero-3-phosphate (dipalmitoyl phosphatidic acid, DPPA) was re-ported to exhibit anti-angiogenic and anti-tumoral activity. However, the hydrophobic property of DPPA largely restricted its clinical use, while systemic infusion of free DPPA could result in undesirable side effects. Herein, we successfully developed DPPA-based lipid-nanoparticles (DPPA-LNPs) which turns the "therapeutic payload into nanocarrier". This strategy could improve on DPPA's hydrophiliciy, thereby fa-cilitating its systemic administration. . DPPA-LNPs not only retained the therapeutic anti-angiogenic and anti-tumoral bioactivity of parental DPPA, but also greatly improved its tumor targeting ability via en-hanced permeability and retention (EPR) effect. This strategy not only eliminates the limitation of drug encapsulation rate, toxicity of the delivery vehicle;but also enhances DPPA bioacvtity in vitro and in vivo. Systemic administration of DPPA-LNPs significantly suppressed the blood vessel formation and tu-mor growth of triple negative breast cancer and liver cancer growth on both xenograft tumor *** of significance This is the first-in-kind self-therapeutic inherent lipid to be made into a nanocarrier, with inherent anti-angiogenic and anti-tumor properties. DPPA nanocarrier is fully natural, fully compatible with min-imal systemic toxicity. Adjustment of the energy transfer pathway in nanoplatelet-organic molecule hybrids through spectral overlap

Two-dimensional (2D) CsPbBr3 nanoplatelets (NPLs) have garnered significant attention because of their promising applications in optoelectronics and photo- catalysis. Despite substantial progress in the development of these materials, a critical gap remains. This gap is particularly evident in the understanding of singlet and triplet energy transfer mechanisms within hybrid systems that incorporate organic molecules, such as Rose Bengal (RoseB). Using a combination of steady-state and transient absorption spectroscopy, we demonstrate that both singlet and triplet energy transfer are feasible within this hybrid system. Our findings reveal that the efficiency of these processes highly depends on the CsPbBr3 NPLs and RoseB spectral overlap, which is in turn influenced by the NPLs thickness. The energy transfer rate of the system increases with increasing spectral overlap, as does the energy transfer efficiency. As the NPLs thickness increases, a higher proportion of singlet energy transfer and a corresponding decrease in triplet energy transfer occur. Our results offer new strategies for controlling energy transfer pathways, which can enhance the design and optimization of high-performance optical devices and improve the efficiency of photocatalytic systems.

Influenza, driven by highly mutable RNA viruses, poses a major global health threat, exacerbated by the frequent emergence of antiviral resistance. To tackle this challenge, we developed hydrophobic tag tethering degradation (HyTTD) technology to enhance the efficacy of oseltamivir against resistant strains. Among these, compound L12, featuring a 1-adamantylamine hydrophobic tag attached via a nine-carbon linker, demonstrated exceptional antiviral activity, with a 157-fold improvement in potency (EC50 = 0.68 mu M) against the oseltamivir-resistant H1N1-H274Y strain compared to oseltamivir (EC50 = 106.8 mu M), and minimal cytotoxicity at 50 mu M. Western blot and immunofluorescence analyses demonstrated that L12 selectively induced the degradation of viral neuraminidase (NA) and inhibited nucleoprotein (NP) expression in a dose-dependent manner. Mechanistic studies revealed that L12 did not interfere with NA synthesis but promotes NA protein degradation through ubiquitination. These results highlight the pioneering application of HyTTD in overcoming antiviral resistance, showcasing it as a powerful platform for future drug development.

Risk assessment and spatial analysis of paralytic shellfish toxin (PST) concentrations and acute dietary exposure of commercially available shellfish in coastal areas in China

Paralytic shellfish toxins (PSTs) are produced by marine dinoflagellates and can be accumulated in filter feeding shellfish. In this study, we assessed acute dietary exposure to PSTs among residents of Chinese coastal provinces and cities, utilizing point and probabilistic estimates. Additionally, we performed global and local spatial autocorrelation analyses to evaluate PSTs contamination and dietary exposure levels in the coastal regions of China. The point estimate revealed that Hebei province exhibited the highest level of PSTs dietary exposure, measuring 39.57 mu g STX eq./kg bw. In all regions, except for Guangxi, Jiangsu, Shandong, and Shanghai, the ratio between acute dietary exposure and the acute reference dose (ARfD) exceeded 1. Probabilistic estimates indicated that P99.9 exceeded the ARfD value exclusively in Guangdong, Hainan, and Liaoning. In comparison, the point estimate was significantly higher than the P99.9 value of the probabilistic estimate. The spatial analysis results showed "low-high" aggregation of PSTs concentration and point estimate dietary exposure levels in Liaoning and Shandong, while probabilistic estimate dietary exposure level exhibited "low-high" aggregation in Guangxi. This implied the presence of regional variations in PSTs contamination and dietary exposure within the coastal regions of China, along with specific localized clustering features. Therefore, it is advisable to develop regionalized precision monitoring and dietary interventions to ensure the safety of shellfish consumption, particularly in response to the regional distribution characteristics of PSTs contamination and dietary exposure.

Background: Intrahepatic cholangiocarcinoma (ICC) ranks second among primary liver cancers in terms of prevalence, characterized by poor prognosis and scarce therapeutic interventions. Fufang-Biejia-Ruangan tablet (BJRG), a traditional Chinese herbal remedy, has been widely used for liver diseases. However, its therapeutic efficacy and underlying mechanisms in ICC remain poorly understood. Aim of the study: This study aims to systematically investigate the anti-ICC effects of BJRG, focusing on tumor progression and microenvironment modulation, through experimental and transcriptomic analyses. Methods: The chemical composition of BJRG was analyzed employing ultra-performance liquid chromatographymass spectrometry (UPLC-MS). In vitro assays were performed with QBC939 and LX-2 cell lines. Two primary ICC models (AKT/YAP and sgP53/KRAS) were established via hydrodynamic tail-vein injection of corresponding plasmids. A co-culture system for subcutaneous tumor formation was developed using cancer-associated fibroblasts (CAFs) derived from the AKT/YAP model and primary tumor cells derived from the sgP53/KRAS model. Results: UPLC-MS analysis identified 1091 chemical components, primarily terpenoids, sugars, glycosides, and phenylpropanoids. The therapeutic efficacy of BJRG was evaluated for the treatment of ICC. BJRG treatment slowed down the growth of both human ICC cell lines and AKT/YAP ICC mouse model. Mechanistically, BJRG inhibited HIPPO-PI3K/AKT signaling pathway in ICC tumor cells. Importantly, BJRG significantly inhibited the growth of CAFs via HIPPO-PI3K/AKT cascades. Of note, co-culture CAFs with ICC cell lines substantially sensitized the efficacy of BJRG in sgP53/KRAS syngeneic tumor model. Furthermore, BJRG therapy not only affected CAFs but also induced alterations in vascular structures and hypoxic conditions within lesions in the AKT/YAP model. This letter deals with the knowledge-aided adaptive detection problem of radar targets in nonhomogeneous correlated compound Gaussian sea clutter. The lognormal distribution is used as the prior distribution of the clutter texture to match the non-Gaussianity of sea clutter. In addition, in order to ensure the estimation accuracy of the covariance matrix structure of sea clutter, a convex combination estimator (CCE) is proposed by jointly exploiting the prior information and the current secondary data. Then, a knowledge-aided adaptive detector is designed on the basis of the complex parameter Gradient test and the CCE. Numerical experiments verify the effectiveness of the proposed CCE and adaptive detector in comparison with their counterparts, respectively.

Synthesis and anti-influenza virus activity of substituted dibenzoxepine-based baloxavir derivatives

Seasonal influenza poses a significant threat to global public health, driving the need for effective anti-influenza agents. The PA protein, which captures the pre-mRNA cap structure, is crucial for the replication of the influenza virus and serves as an important target for developing such agents. Baloxavir, a PA inhibitor, has shown excellent activity against influenza A and B viruses. In this study, its structure was optimized using bioisosteric replacement to develop novel dibenzoxepine-based derivatives for combating influenza. As the lead compounds, ATV03 (EC50 = 0.78 f 0.10 nM, SI > 64103) and ATV07 (EC50 = 0.78 f 0.01 nM, SI = 31603) demonstrated excellent anti-influenza A (H3N2) activity and SI, and possessed favorable anti-influenza B activity, with 2.02 f 0.40 nM and 2.32 f 0.29 nM of EC50 respectively. They showed improved bioavailability and metabolic stability. Mechanism studies revealed that ATV03 and ATV07 both possessed significant activity in inhibiting PA and RdRp as well as disturbing NP. Consequently, ATV03 was selected for further investigation in the fight against seasonal and pandemic influenza due to its superior bioavailability, metabolic stability, and efficacy against multiple influenza A viruses.
