Decadal Variations in Radiocarbon in Dissolved Inorganic Carbon (DIC) Along a Transect in the Western North Pacific Ocean

We report results for the concentrations and radiocarbon (C-14) compositions of dissolved inorganic carbon (DIC) in water samples collected from a north-south transect in the western North Pacific (NP) Ocean in November 2019 and compare our results with values previously reported for similar transects during the WOCE and CLIVAR projects decades ago. We show that the strong bomb C-14 penetration signal in the upper waters has gradually been reduced and diluted by deep water depleted in C-14-DIC over the last 30 years. The rate of decrease in Delta C-14-DIC values in surface waters was greater from 2005 to 2019 than from 1993 to 2005, and the rates of decrease from 2005 to 2019 were fastest (5.4-7.0 parts per thousand yr(-1)) between 25 degrees and 32 degrees N and slowest (2.1-2.8 parts per thousand yr(-1)) between 13 degrees N and 16 degrees N. The downwelling of the water mass could have accelerated the transport of bomb C-14 in the NP subtropical gyre (25 degrees-32 degrees N). The different decadal changes in Delta C-14-DIC in the North Pacific Intermediate Water (NPIW) from north to south prove that bomb C-14 is transported southward to the southwestern NP and that the bomb C-14 peak likely penetrates the ocean interior gradually. Changes in isopycnic circulation and advection diffusion in deep waters might have resulted in shifts in the baseline Delta C-14-DIC signature.

Selenium-enriched polysaccharides from Pyracantha fortuneana ( SePFP) has many beneficial physiological activities, but how it improves the aging associated abnormal lipid metabolism is still unclear. Therefore, we explored the mechanisms of the regulatory role of SePFP on liver lipid accumulation in aging mice. Methods: 60 naturally aged C57BL/6J male mice were divided into 6 groups: adult group, aging group (21-month-old mice), aging mice treated with low-, medium- and high-doses of SePFP (SePFP-L, SePFP-M, SePFP-H), and aging mice treated with resveratrol (RSV). SePFP and RSV were administrated daily via oral gavage from 16 to 21 months old. The parameters of energy metabolism were measured in all mice before sacrifice, and liver tissues were collected to determine the levels of metabolism-related enzymes by real-time PCR and Western blot. Results: We found that SePFP significantly reduced the body weight, liver to bodyweight ratio, and white fat to body weight ratio in aging mice. SePFP also down-regulated the triglycerides and cholesterol levels in liver and serum, and decreased respiratory quotient in aging mice. The mechanism of SePFP regulating lipid metabolism was mainly through promoting fatty acid transportation to mitochondria and enhancing mitochondrial beta-oxidation and ketone body production. Conclusion: SePFP attenuates liver lipid deposition in aging mice by enhancing hepatic mitochondrial beta-oxidation.

Experimental study on the effect of thermosensitive controlled-release inhibitor on coal spontaneous combustion

Inhibitors are widely used on-site due to their effective prevention. The effect of a novel compound inhibitor composed of thermosensitive hydrogel (TSH) and antioxidants (tea polyphenols (TP) and L-ascorbic acid (VC)) on coal spontaneous combustion (CSC) characteristics was studied. Its inhibitory effect was compared with that of the single component or MgCl2 by macroscopic and microscopic tests. The results show that the release of CO or CO2 from coal treated with inhibitors during temperature-programmed tests was reduced, and the crossing-point temperature of TSH-TP/VC-treated coal increased by 21.4 degrees C. The content of -CH2, -CH3 and oxygen- containing group in inhibitor-treated coal decreased than before, the characteristic temperature of spontaneous combustion increased significantly, and the mass gain values of coal treated with TSH-TP/VC, TSH, TP/VC or MgCl2 decreased by 0.99 %, 0.93 %, 0.87 %, 0.24 %, respectively. The inhibitory efficacy of TSH-TP/VC is superior to the effect of single component or traditional inhibitor MgCl2, and it is effective in inhibiting the entire oxidative decomposition and combustion stage of coal. TSH can effectively cover the coal body and remove the accumulated heat. At the same time, TP/VC hinders the emergence of free radicals and chelates with metal ions, thereby exhibiting a significant suppression of CSC.

The spread of the influenza virus has caused devastating pandemics and huge economic losses worldwide. Antiviral drugs with diverse action modes are urgently required to overcome the challenges of viral mutation and drug resistance, and targeted protein degradation strategies constitute excellent candidates for this purpose. Herein, the first degradation of the influenza virus polymerase acidic (PA) protein using small-molecule degraders developed by hydrophobic tagging (HyT) technology to effectively combat the influenza virus was reported. The SAR results revealed that compound 19b with Boc(2)-(L)-Lys demonstrated excellent inhibitory activity against A/WSN/33/H1N1 (EC50 = 0.015 mu M) and amantadine-resistant strain (A/PR/8/H1N1), low cytotoxicity, high selectivity, substantial degradation ability, and good drug-like properties. Mechanistic studies demonstrated that the proteasome system and autophagic lysosome pathway were the potential drivers of these HyT degraders. Thus, this study provides a powerful tool for investigating the targeted degradation of influenza virus proteins and for antiviral drug development.

Fractions, stability, and influencing factors of soil organic carbon under different land-use in sodic soils

Salt-affected soils have considerable carbon sequestration potential in aboveground biomass and soil organic carbon (SOC). However, organic carbon pool characteristics and the significant important factors affecting the land use of sodic soils have not been clearly elucidated. In this study, 252 samples were collected from 20 typical sodic soil sites across the western Songnen Plain, China. This study investigated the effects of five land-use types, that is, saline wasteland (SWL), degraded grassland (GL), forestland (FL), upland (UL), and paddy fields (PF), on sodic soil SOC fractions and stabilities to find that UL and PF decreased salinity and alkalinity, but increased soil nutrient and water stability aggregate (WSA) contents and stability. Compared with degraded natural land-use types (SWL and GL), FL and croplands (UL and PF) increased the physical protection of SOC and aggregateassociated SOC contents. Furthermore, cropland, especially PF, increased nonlabile organic carbon (NOC) (7.47 g kg- 1), labile organic carbon (LOC) (3.79 g kg- 1), dissolved organic carbon (DOC) (0.16 g kg- 1), and mineralizable organic carbon (MOC) (0.41 g kg-1) fractions. Furthermore, compared with degraded natural lands, PF increased NOC ratios (59.65%) and carbon management index (CMI) (126.1), thus increasing organic carbon content and stability. Structural equation modelling revealed that improvements in soil properties, total nitrogen (TN), WSA, electrical conductivity (EC), and pH were the crucial factors for increasing SOC stock in sodic soils. We concluded that the positive effect that PF had on SOC content and stability were much greater than that of the other land-use types, and that PF was the optimal management practice for increasing SOC sequestration capacity in sodic soils.

Introduction: Podocyte apoptosis is a common mechanism driving progression in Alport syndrome (AS). This study aimed to investigate the mechanism of podocyte apoptosis caused by COL4A3 mutations. Methods: We recruited patients with autosomal dominant AS (ADAS). Patients with minimal change disease (MCD) were recruited as controls. Microarray analysis was carried out on isolated glomeruli from the patients and validated. Then, corresponding mutant human podocytes (p.C1616Y) and 129 mice (p.C1615Y, the murine homolog to the human p.C1616Y) were constructed. The highest differentially expressed genes (DEGs) from microarray analysis were validated in transgenic mice and podocytes before and after administration of MMP-2 inhibitor (SB-3CT) and NOX4 inhibitor (GKT137831). We further validated NOX4/MMP-2/apoptosis pathway by real-time polymerase chain reaction (PCR), immunohistochemistry, and western blot in renal tissues from the ADAS ***: Using microarray analysis, we observed that DEGs, including NOX4/H2O2, MMP-2, and podocyte apoptosis-related genes were significantly upregulated. These genes were validated by real-time PCR, histologic analysis, and western blot in corresponding mutant human podocyte (p.C1616Y) and/or mice models (p.C1615Y). Moreover, we found podocyte apoptosis was abrogated and MMP-2 expression was down-regulated both in vivo and in vitro by NOX4 inhibition, urinary albumin-to-creatinine ratio, 24-hour proteinuria;and renal pathologic lesion was attenuated by NOX4 inhibition in vivo. Furthermore, podocyte apoptosis was attenuated whereas NOX4 expression remained the same by inhibition of MMP-2 both in vivo and in ***: These results indicated that NOX4 might induce podocyte apoptosis through the regulation of MMP-2 in patients with COL4A3 mutations. Our findings provided new insights into the mechanism of ADAS.

Reactive oxygen species-responsive hydrophobic crosslinked chitosan films based on triple-function crosslinkers

Chitosan is widely used in reactive oxygen species (ROS)-responsive films but remains great challenges owing to its weak mechanical strength and strong hydrophilicity. Herein, we synthesized novel hydrophobic crosslinked CS films with ROS-responsive properties and excellent physicochemical properties. A novel crosslinker, 2-((10carboxydecyl)thio)succinic acid, with long-chain alkanes, three carboxyl groups, and sulfhydryl groups was synthesized and then used to produce thioether-containing crosslinked CS membranes. The results suggested that crosslinking could significantly increase the tensile strength of the film from 15.67 MPa to 24.32 MPa. The compact structure of crosslinked chitosan film improved the hydrophobicity and degradability, reduced the thermal stability and swelling rates, exhibited excellent non- cytotoxicity. The in vitro release studies revealed that crosslinked chitosan films could displayed the highest flux about 1.40 mg/ (cm2 h) and significant NR fluorescence change over 80 %. Collectively, our results demonstrate the applicability of these films as ROSresponsive drug delivery systems.

Fractal evolution characteristics of fracture meso-damage in uniaxial compression rock masses using bonded block model

With regard to deep mining in metal mines, an investigation into the failure mode of deep fractured rock masses and their corresponding acoustic emission signal characteristics is conducted via uniaxial compression tests. Subsequently, a fractal damage renormalization group mechanical model is developed to explain the behavior of those fractured rock masses. Employing the bonded block model (BBM) numerical simulation method, fracture process in synthetic rock samples is analyzed, thereby validating the efficacy of the mechanical model. The numerical simulations highlight the critical role of fractures expansion in underlying the deterioration of rock mass strength. As the peak load decreases, the fracture fractal dimension increases, leading to a significant 14.2% reduction in compressive strength accompanied by an approximate 8.7% rise in average fracture fractal dimension. A comparative analysis of tetrahedral and voronoi block synthetic rock samples reveals the tetrahedral block samples exhibit a superior ability to depict the fracture behavior of fractured rock masses. Specifically, they offer a more accurate simulation of acoustic emission characteristics and failure modes. Furthermore, variations in the fracture fractal dimension with respect to the hole defect's position are observed, with the maximum value occurring along the vertical axis of the hole defect. This observation underscores the potential utility of visually monitoring deep rock fracture dynamics as an effective mean for quantitatively evaluating fracture damage and strength degradation in deep rock formations.

Fabrication of nitrogen-doped carbon dots biomass composite hydrogel for adsorption of Cu (II) in wastewater or soil and DFT simulation for adsorption mechanism

With the increase of Cu (II) content, its bioaccumulation becomes a potential pollution to the environment. It is necessary to design an economical and efficient material to remove Cu (II) without causing other environmental hazards. A novel material of alginate composite bead (ALG@NCDs) was synthesized by embedding N-doped carbon dots into pure alginate bead for the adsorption of Cu (II) from wastewater and contaminated soil. The initial concentration, the amount of adsorbent, temperature, adsorption time, and pH value were optimized for the adsorption of Cu (II). According to the Langmuir isothermal adsorption model, the maximum adsorption amount of the material to Cu (II) was 152.44 mg/g. The results of selective adsorption showed that ALG@NCDs had higher affinity to Cu (II) than to Pb (II), Co (II), Ni (II), and Zn (II). After five adsorption-desorption experiment, adsorption capacity of the ALG@NCDs was kept 89% of the initial adsorption capacity. Its Cu (II) adsorption mechanism was studied by density functional theory calculations. In addition, the material could effectively adsorb Cu (II) and release the phytonutrient Ca (II) simultaneously when applied to actual wastewater and soil. The fabricated ALG@NCDs would be a promising material for the adsorption of Cu (II) from wastewater or soil. © 2024 Elsevier Ltd

Millisecond self-heating and quenching synthesis of Fe/carbon nanocomposite for superior reductive remediation

Fe-0-based nanomaterials are extensively applied in environmental remediation, but their passivated oxide shell restricts deep application. However, efforts aimed at revitalizing Fe-oxide shells have shown limited success. Here, we report a "faster win fast" approach by preferential carbon layer deposition in milliseconds to block Feoxide shell growth via carbon-assisted flash Joule heating (C-FJH) reaction. C-FJH induced ultra-high temperature and electric shock promoted reductive Fe formation and subsequently melted to a phase-fusional heterostructure (Fe-0/FeCl2). Therefore, theoretical calculation confirmed that electron delocalization effect of derived heterostructure promoted electron transfer. Synchronously, rapid self-heating/quenching rate (similar to 10(2) K/ms) realized a thin aromatic-carbon layer deposition to sustain both high stability and activity of reductive Fe. The channels of thin aromatic-carbon layer favored inward diffusion of pollutants, which facilitated the subsequent reduction. Accordingly, derived heterostructure and carbon layer jointly contributed to the boosted removal of multiple pollutants (including metal oxyanions, perfluorinated compounds, and disinfection by-products).
