The analysis of the ubiquitous human-human interactions is pivotal for understanding humans as social beings. Existing human-human interaction datasets typically suffer from inaccurate body motions, lack of hand gestures and fine-grained textual descriptions. To better perceive and generate human-human interactions, we propose Inter-X, a currently largest human-human interaction dataset with accurate body movements and diverse interaction patterns, together with detailed hand gestures. The dataset includes ∼11K interaction sequences and more than 8.1M frames. We also equip Inter-X with versatile annotations of more than 34K fine-grained human part-level textual descrip- tions, semantic interaction categories, interaction order, and the relationship and personality of the subjects. Based on the elaborate annotations, we propose a unified benchmark composed of 4 categories of downstream tasks from both the perceptual and generative directions. Extensive experiments and comprehensive analysis show that Inter-X serves as a testbed for promoting the development of versatile human-human interaction analysis. Identification of dibucaine derivatives as novel potent enterovirus 2C helicase inhibitors: In vitro, in vivo, and combination therapy study

Enterovirus A71 (EV-A71) is a human pathogen causing hand, foot and mouth disease (HFMD) which seriously threatened the safety and lives of infants and young children. However, there are no licensed direct antiviral agents to cure the HFMD. In this study, a series of quinoline formamide analogues as effective enterovirus inhibitors were developed, subsequent systematic structure-activity relationship (SAR) studies demonstrated that these quinoline formamide analogues exhibited good potency to treat EV-A71 infection. As described, the most efficient EV-A71 inhibitor 6i showed good anti-EV-A71 activity (EC50 = 1.238 mu M) in RD cells. Furthermore, compound 6i could effectively prevent death of virus infected mice at dose of 6 mg/kg. When combined with emetine (0.1 mg/kg), this treatment could completely prevent the clinical symptoms and death of virus infected mice. Mechanism study indicated that compound 6i inhibited EV-A71 via targeting 2C helicase, thus impeding RNA remodeling and metabolism. Taken together, these data indicated that 6i is a promising EV-A71 inhibitor and worth extensive preclinical investigation as a lead compound. (C) 2020 Elsevier Masson SAS. All rights reserved.

Reverse genetics reveals contrary effects of two Rdl-homologous GABA receptors of Helicoverpa armigera on the toxicity of cyclodiene insecticides

The resistance to dieldrin gene (Rdl) encodes a subunit of the insect gamma-amino butyric acid (GABA) receptor, and the encoded Rdl subunit is a major target site for cyclodiene and phenylpyrazole insecticides. Since the substitution of a single amino acid (Ala to Ser/Gly at position 302) of the Drosophila melanogaster Rdl gene was first identified to confer high level resistance to dieldrin, mutations at the equivalent positions have been reported to confer resistance to dieldrin and/or fipronil in a wide range of different insects. In the cotton bollworm Helicoverpa armigera, there are two Rdl homologs (Handl-1 and Handl-2) in close proximity on the Z chromosome, which as wild-type sequences, encode alanine and serine respectively at amino acid position 302. In the present study, we used the CRISPR/Cas9 gene editing approach to knock out Handl-1 and Handl-2 and establish two homozygous knockout strains (Delta Rdl-1 and Delta Rdl-2). The Delta Rdl-1 strain showed low levels of resistance (8.0-to 9.3-fold) to three cyclodiene insecticides (endosulfan, aldrin and dieldrin) compared with the background SCD strain. In contrast, toxicity of the three cyclodiene insecticides to the Delta Rdl-2 strain increased significantly (3.6- to 6.3-fold) when compared with the SCD strain. Genetic analysis indicated the obtained resistance to endosulfan and dieldrin in the Delta Rdl-1 strain was sex-linked, which is consistent with the fact that Handl-1 locus is located on the Z chromosome. The above results demonstrate that both Handl-1 and Handl-2 are important determinants for the susceptibility of H. armigera SCD strain to the three cyclodiene insecticides, but have opposite effects. It was also found that Handl-1 and Handl-2 are involved, to some extent, in mediating sensitivity of H. armigera to avermectin and fipronil respectively. In order to realize the accurate, quick, dynamic and intelligent prediction of coal and gas outburst, a prediction method based on BP artificial neural network (BPNN) and case-based reasoning (CBR) is proposed. Aiming at the strong nonlinearity for influencing factors of coal and gas outburst, a method based on BPNN is given to determine case feature weights objectively and scientifically. The case reuse fast method based on weighted average is used to improve the prediction quickness and precision. The proposed method is validated using practical measured data. The simulation example shows that the proposed method has the higher prediction accuracy and stability, the prediction speed of the proposed method is quicker than those of the existing CBR prediction methods, and the proposed method is valid.

为实现对工作面煤与瓦斯突出快速、准确和动态的预测,提出一种基于主成分分析和权重贝叶斯的工作面煤与瓦斯突出预测方法,通过建立工作面煤与瓦斯突出预测的权重贝叶斯模型进行突出危险性等级预测。利用主成分分析确定预测模型中分类变量权重以提高预测准确性。在此基础上,设计基于相似度的训练样本数据更新方式实现对突出预测模型的有效重构。实验结果表明,与朴素贝叶斯模型和权重贝叶斯模型相比,基于主成分分析和权重贝叶斯工作面煤与瓦斯突出预测方法能快速获得高准确度的突出预测结果,为现场指导矿井工作面安全生产提供参考。

A novel label-free hypochlorite amperometric sensor based on target-induced oxidation of benzeneboronic acid pinacol ester

In this work, a novel label-free hypochlorite amperometric sensor based on the direct oxidation of benzeneboronic acid pinacol ester (BAPE) induced by hypochlorite was developed. 4-Aminophenylboronic acid pinacol ester (ABAPE) chemically bonded to functionalized multi-walled carbon nanotubes (f-MWCNTs) was employed to recognize the hypochlorite species. In nearly neutral solutions, the ABAPE probe was able to provide a distinct oxidation signal at approximately 0.65 V vs. Ag/AgCl. When hypochlorite was added, it reacted with ABAPE rapidly to form a stable hydroxyl derivative, resulting in a significant decrease of the current at 0.65 V. Therefore, the hypochlorite target could be detected by recording the reduced oxidation signal of ABAPE. Under optimized conditions, the fabricated sensor offered linear amperometric responses for the target in the concentration ranges of 0.01-0.1 mM and 0.1-1 mM, providing a limit of detection down to 5.3 mu M based on S/N = 3. The presence of common anions, including carbonate, sulfate, nitrate, and halide ions, had no obvious effects on the selective determination of hypochlorite. The amperometric sensor also exhibited excellent accuracy and precision in monitoring the hypochlorite level in tap water. These results suggest that the new sensor has great promise for use in various fields such as water pollution control, environmental monitoring, and food safety.

Enterovirus 71 (EV-A71) infection causes severe hand-foot-and-mouth disease that leads to cardiopulmonary complications and death in young children under 5 years of age. Although there are available vaccines for EV-A71 C⁴, however, there are no efficient drugs for severe cases. Thus, there is an urgent need to find new direct-antiviral agents (DAAs) to control EV-A71 infection. In this study, we report our discovery of the EV-A71 capsid inhibitor PTC-209HBr, a small-molecule Bmi-1 inhibitor and an anticancer agent, and its derivatives that inhibit multiple enteroviruses with an EC₅₀ at a submicromolar efficacy. The mechanism of action of PTC-209HBr was confirmed by time-of-addition, resistance selection and reverse genetics experiments, microscale thermophoresis (MST), viral binding and entry assays, coimmunoprecipitation (Co-IP) and immunofluorescence experiments (IF). Mechanistic studies indicated that PTC-209HBr inhibited EV-A71 infection by impeding the binding between VP1 and the receptor hSCARB2 during the early stage of EV-A71 infection through hindering viral entry into host cells. Collectively, these findings indicated that PCT-209HBr is a novel inhibitor of enteroviruses with a confirmed mechanism of action that can be further developed into EV-A71 DAAs. © 2022 wuhan University

Deep Attentive Interest Collaborative Filtering for Recommender Systems

Collaborative filtering (CF) is a pivotal building block in commercial recommender systems due to its strength and utility in user interest modeling. Recently, many researchers have turned to deep learning as a way to capture richer collaborative signals from user-item feature interactions. However, most deep-based methods only consider nonlinear, high-order interactions while ignoring the explicit collaborative signals in low-order interactions. They also typically ignore the quality of the user and item profiles. These are cornerstones in item recommendation that, we argue, must be considered for high-quality recommendations. Hence, we propose Deep Attentive Interest Collaborative Filtering (DAICF) to overcome these limitations. DAICF profiles users based on their interactive items, i.e., user neighborhood information. Similarly, item profiles are based on users who had interacted with it, i.e., item neighborhood information. Given a user's profile varies over different items, DAICF accurately models his attentive interests based on the specific target item. Low-order collaborative signals are captured by a shallow component, and high-order collaborative signals are captured by a deep component. These two complementary collaborative signals are then fused to provide rich recommendations that cut through today's information overload. By designing a personalized feature extraction method based on bilateral neighborhood information to solve the data sparsity problem in recommender systems, DAICF can dynamically distinguish the importance of a user's historical interaction items for predicting user preferences for a specific target item. A set of experiments against four real-world datasets validate that DAICF outperforms the most recent state-of-the-art recommendation algorithms and justifies the effectiveness and interpretability of our method.

Exact multiplicity, bifurcation curves, and asymptotic profiles of endemic equilibria of a cross-diffusive epidemic model

This study examines the global structure of endemic equilibrium (EE) solutions of a cross-diffusive epidemic model which incorporates the repulsive movement of the susceptible population away from the infected population. We show that the basic reproduction number R0 alone cannot determine the existence of the EEs and the model may have multiple EEs when the repulsive movement rate chi is large. We prove that the set of EEs forms a simple and unbounded curve bifurcating from the curve of disease free equilibria at R0 = 1 as R0 varies from zero to infinity, where the bifurcation curve can be forward or backward. We find conditions under which a forward bifurcation curve is of S-shaped and show that a large chi tends to induce backward bifurcation curves. Results on the asymptotic profiles of the EEs are obtained as the repulsive movement rate is large or the random movement rates are small. Do researchers from prestigious universities deserve advantages in research funding? Evidence from the National Natural Science Foundation of China

Existing literature underscores the unequal distribution of research funding across universities of varying prestige, yet the mechanisms through which university prestige influences funding success remain underexplored. Specifically, do Principal Investigators (PIs) from universities of differing prestige levels face divergent probabilities of funding success? Is this disparity solely attributable to differences in research capabilities, or does university prestige independently confer advantages, even among PIs with comparable qualifications? This study addresses these questions by analyzing unique application data from the National Natural Science Foundation of China, employing a combination of baseline regression models, matching methods, and panel data techniques. The results indicate that university prestige significantly increases the likelihood of funding success, even when controlling for the applicant's research background. Furthermore, among awarded grants, the prestige of the PI's affiliated university positively influences both academic productivity and scientific impact. Early-career affiliation with prestigious universities is also strongly linked to higher academic output and sustained funding acquisition, suggesting a "richer-get-richer" dynamic. By elucidating the mechanisms behind funding decisions, this study contributes to the theoretical understanding of research funding allocation and provides empirical insights for optimizing funding policy.
