Transgenic expression of SeCYP9A186 and PxFMO2 confers resistance to emamectin benzoate in Plutella xylostella

BACKGROUNDThe overexpression of metabolic enzymes constitutes a crucial mechanism for insects to detoxify xenobiotics and metabolic pesticides. A flavin-containing monooxygenase gene (PxFMO2) from Plutella xylostella and a P450 gene (SeCYP9A186, F116V mutant allele) from Spodoptera exigua have been reported to be involved in insecticide resistance. In this study, we aim to utilize transgenic technology to validate their in vivo detoxification functions in Plutella *** established two transgenic strains of Plutella xylostella with expressing an endogenous PxFMO2 gene from Plutella xylostella and an exogenous SeCYP9A186 gene from S. exigua, respectively. Bioassays demonstrated that the transgenic Plutella xylostella strain (IPP-FMO2) expressing PxFMO2 exhibited a 12-fold resistance to emamectin benzoate and a 6.4-fold resistance to chlorantraniliprole compared to the background strain (IPP-S). In contrast, the transgenic Plutella xylostella strain (IPP-9A186) expressing SeCYP9A186 displayed a 235-fold resistance to emamectin benzoate and a 115-fold resistance to abamectin. Moreover, resistance to emamectin benzoate in the IPP-9A186 strain of Plutella xylostella was inherited as an incompletely dominant trait and was genetically linked to the transgene *** results not only elucidated the in vivo contribution of the PxFMO2 and SeCYP9A186 to the insecticide resistance phenotype in Plutella xylostella, but also provided a genetic engineering toolkit to manipulate resistance pathways. Influenza A virus(IAV)binds sialic acid receptors on the cell surface to enter the host cells,which is the key step in initiating infection,transmission and *** the factors that contribute to the highly efficient entry of IAV into human cells will help elucidate the mechanism of viral entry and pathogenicity,and provide new targets for *** the present study,we reported a novel membrane protein,C1QTNF5,which binds to the hemagglutinin protein of IAV and promotes IAV infection in vitro and in *** found that the HA1 region of IAV hemagglutinin is critical for the interaction with C1QTNF5 protein,and C1QTNF5 interacts with hemagglutinin mainly through its N-terminus(1–103 aa).In addition,we further demonstrated that overexpression of C1QTNF5 promotes IAV entry,while blocking the interaction between C1QTNF5 and IAV hemagglutinin greatly inhibits viral ***,C1QTNF5 does not function as a receptor to mediate IAV infection in sialic acid-deficient CHO-Lec2 cells,but promotes IAV to attach to these cells,suggesting that C1QTNF5 is an important attachment factor for *** work reveals C1QTNF5 as a novel IAV attachment factor and provides a new perspective for antiviral strategies.

Living therapeutics is an emerging antitumor modality by living microorganisms capable of selective tropism and effective therapeutics. Nevertheless, primitive microbes could only present limited therapeutic functionalities against tumors. Hybridization of the microbes with multifunctional nanocatalysts is of great significance to achieve enhanced tumor catalytic therapy. In the present work, nitric oxide synthase (NOS)-engineered Escherichia coli strain MG1655 (NOBac) was used to hybridize with the sonopiezocatalytic BaTiO3 nanoparticles (BTO NPs) for efficient tumor-targeted accumulation and antitumor therapy. Under ultrasound irradiation, superoxide anions created by the piezocatalytic reaction of BTO NPs could immediately react with nitric oxide (NO) generated from NOBac to produce highly oxidative peroxynitrite ONOO- species in cascade, resulting in robust tumor piezocatalytic therapeutic efficacy, prompting prominent and sustained antitumoral immunoactivation simultaneously. The present work presents a promising cancer immunotherapy based on the engineered and hybridized microbes for highly selective and sonopiezo-controllable tumor catalytic therapy.

Rerouting charge transfer for pharmaceutical wastewater electrochemical treatment via interfacial cocatalyst modification

Electrochemical oxidation stands as a pivotal technology for refractory wastewater treatment. However, the high cost and low elemental abundance of commercial electrodes limit its widespread application. This work tries to address this by introducing a charge-transfer rerouting strategy via cocatalyst modification using earth-abundant elements. Here, we uncover the role of the cocatalyst in enhancing electrode performance. The in-situ reconstructed cocatalyst induces a substantial rerouting of the charge transfer pathway, facilitating the mass/charge transfer of organics while concurrently suppressing the oxygen evolution side reaction. The Ti-Fe2O3 electrode, loaded with the cocatalyst PbO2, exhibits both high current efficiency (similar to 45.4 %) and low energy requirement (similar to 31.8 kW h kg(-1) COD), surpassing other reported electrodes and displaying great versatility in various scenarios with good stability and reusability. Moreover, this charge-transfer rerouting strategy holds promise for synergy with other methodologies, such as nanostructure engineering and molecular imprinting, to further enhance the reactivity and selectivity of electrocatalysts in environment and energy-related domains.

Household catastrophic health expenditure and depressive mood among Chinese adults, children, and adolescents: a population-based panel study

BackgroundNumerous studies have suggested that catastrophic health expenditure (CHE) is associated with depressive mood. However, most published studies have examined the relationship between CHE and depressive mood only among middle-aged and older people who are already susceptible to depressive mood. The objective of our analysis was to determine the associations between household CHE and depressive mood among adults and children/*** study population consisted of Chinese residents who participated in the 2016 CFPS, 2018 CFPS, and 2020 CFPS. Our analytical sample was restricted to children/adolescents aged 10-17 years and adults aged 18 years and older. We utilized multilevel random effects multivariate logistic regression models to investigate the associations between CHE and depressive mood among both adults and children/*** study revealed that 15% of adults and 12.61% of children/adolescents had experienced CHE and that CHE was positively associated with depressive mood among adults (OR = 1.34, 95% CI: 1.21, 1.50) and among children/adolescents (OR = 1.48, 95% CI: 1.12, 1.96) after adjustment for potential confounding factors. This positive association persisted in different subgroup analyses. In addition, we found that being insured with either urban or rural health insurance was associated with decreased odds of depressive *** study indicated that CHE is common in Chinese families and may increase the risk of depressive mood for both adults and children/adolescents. These findings emphasize the need to focus on expanding health insurance coverage, as well as implementing family-based mental health resources and financial literacy programs to reduce the psychological impact of CHE across all age groups.

Two-in-one additive-engineering strategy for improved air stability of planar perovskite solar cells

The stability issue of organic-inorganic halide perovskite solar cells (PSCs) has become obstruction for their practical applications, although they have attained rapid growth in power conversion efficiency (PCE). Here, we demonstrate a two-in-one additive-engineering strategy by doping organic additives benzoquinone (BQ) into MAPbI3 film and 2,3,5,6-tetrafluoro-7,7,8,8-tetracyanoquinodimethane (F4TCNQ) into hole transport layer (HTL) in a single device, resulting in efficient PSCs with enhanced air stability. The BQ and the F4TCNQ can respectively improve the crystal quality of the MAPbI3 film and increase the moisture tolerance of HTL. The two additives suppress the charge recombination and take a synergistic effect on reduction of traps, contributing to the long-term air stability of unencapsulated planar PSCs. The resulting PSCs with a PCE over 16% can maintain 95% of their initial efficiency after 1400 h in ambient air environment. This strategy provides a new way for the air stability enhancement of PSCs by engineering the perovskite layer and HTL with organic additives in a meanwhile.

Myricetin (3, 5, 7, 3', 4', 5'-hexahydroxyflavone) is a natural flavonol compound found in a large variety of plants, including berries, oranges, grapes, herbs, teas, and wine. In the last decade, a convergence of evidence has demonstrated that myricetin has good biological activity as an anti-tumor, anti-inflammatory, and anti-oxidation agent. In studies involving various types of cancer cells, myricetin has been shown to suppress cancer cell invasion and metastasis, to induce cell cycle arrest and apoptosis of cancer cells, and to inhibit their proliferation. These findings have raised interest in myricetin as a potential tumor inhibitor in human patients. In this review, evidence of myricetin's anti-tumor activity and its underlying molecular mechanisms published in the last decade are summarized.

Bacterial imaging in tumour diagnosis

Some bacteria, such as Escherichia coli (E. coli) and Salmonella typhimurium (S. typhimurium), have an inherent ability to locate solid tumours, making them a versatile platform that can be combined with other tools to improve the tumour diagnosis and treatment. In anti-cancer therapy, bacteria function by carrying drugs directly or expressing exogenous therapeutic genes. The application of bacterial imaging in tumour diagnosis, a novel and promising research area, can indeed provide dynamic and real-time monitoring in both pre-treatment assessment and post-treatment detection. Different imaging techniques, including optical technology, acoustic imaging, magnetic resonance imaging (MRI) and nuclear medicine imaging, allow us to observe and track tumour-associated bacteria. Optical imaging, including bioluminescence and fluorescence, provides high-sensitivity and high-resolution imaging. Acoustic imaging is a real-time and non-invasive imaging technique with good penetration depth and spatial resolution. MRI provides high spatial resolution and radiation-free imaging. Nuclear medicine imaging, including positron emission tomography (PET) and single photon emission computed tomography (SPECT) can provide information on the distribution and dynamics of bacterial population. Moreover, strategies of synthetic biology modification and nanomaterial engineering modification can improve the viability and localization ability of bacteria while maintaining their autonomy and vitality, thus aiding the visualization of gut bacteria. However, there are some challenges, such as the relatively low bacterial abundance and heterogeneously distribution within the tumour, the high dimensionality of spatial datasets and the limitations of imaging labeling tools. In summary, with the continuous development of imaging technology and nanotechnology, it is expected to further make in-depth study on tumour-associated bacteria and develop new bacterial imaging methods for tumour diagnosis. Formation of Highly Negatively Charged Supported Lipid Bilayers on a Silica Surface: Effects of Ionic Strength and Osmotic Stress

Solid supported lipid bilayers (SLBs) serve as an excellent platform for biophysical studies. However, the formation of highly negatively charged SLBs on negatively charged surfaces remains a challenge due to electrostatic repulsion. Here, we study the effects of ionic strength and osmotic stress on the formation of highly negatively charged SLBs on the silica surface. We used quartz crystal microbalance-dissipation to study the adsorption and rupture of highly negatively charged small unilamellar vesicles on the silica surface in different concentrations of NaCl and under different osmotic stresses. It was demonstrated that an increase in the ionic strength of the solution enhances SLB formation. Both hypertonic and moderate hypotonic osmotic stress can promote the formation of SLBs. However, the SLB cannot be formed under high hypotonic osmotic stress. Importantly, osmotic stress alone without a change in ionic strength is insufficient to promote SLB formation. Moreover, the topographical images obtained by atomic force microscopy showed that complete bilayers were formed under hypertonic osmotic stress and high ionic strength, whereas defects were noticed in the bilayers formed under hypotonic osmotic stress. Furthermore, the fluidity of the lipid bilayers was studied by fluorescence recovery after photobleaching. A higher membrane fluidity was observed for the complete lipid bilayers compared to that of the lipid bilayers with defects. Our findings further the understanding of how ionic strength and osmotic stress affect the formation of highly negatively charged SLBs on negatively charged surfaces, providing insights for preparing model biological membranes.

Salt stress-induced changes in soil metabolites promote cadmium transport into wheat tissues
