MB based RT-qPCR increase the clinical application of cfEBV DNA for NPC in non-endemic area of China

To compare the performance of magnetic bead (MB) and the concentrated precipitation (CP) based RT-qPCR to qualify cell free EBV DNA (cfEBV DNA) for nasopharyngeal carcinoma (NPC) in non-endemic area of China. From January 2014 to June 2024, a retrospective analysis of 2 cohort studies on cfEBV DNA in NPC patients was conducted to assess the diagnostic value, positive detection rate and clinical application. cfEBV DNA detection with CP based RT-qPCR in cohort 1 and MB based RT-qPCR method in cohort 2. The MB based RT-qPCR for the quantitative measurement of cfEBV DNA load was higher than the CP based RT-qPCR in the same plasma samples from NPC patients (P < 0.001). CP based RT-qPCR measured cfEBV DNA in 1405 NPC and 244 healthy controls in cohort 1 with 40.8% sensitivity (AUC = 0.704, 95% CI: 0.676-0.731). In cohort 2(683 naive NPC and 303 controls), cfEBV DNA had a sensitivity of 75.84% (AUC = 0.879, 95% CI: 0.86-0.90). There were no significant differences in TNM stage among NPC between the two cohorts (P > 0.05). The MB method considerably increased the positive detection rate of cfEBV DNA in NPCs at stages III-IV, T2-T4, N1-N3, and M0 (P < 0.001). At the end of treatment, 97.51% of patients had no detectable EBV and just 2.49% had detectable cfEBV DNA. Those who received <= 2 or >= 3 cycles of NAC had a median t(1/2) clearance rate of 9.8 days and 12.6 days, respectively. MB based RT-qPCR increased the quantity of cfEBV DNA. MB based RT-qPCR demonstrated superior sensitivity and positive detection rates for cfEBV DNA. cfEBV DNA can be more positively noticed, with a higher diagnostic value and a broader variety of clinical applications among NPC in non-endemic areas.

Adherence to dietary guidelines and liver cancer risk: Results from two prospective cohort studies

Background and aims: Although dietary factors have been extensively investigated as potential risk factors for liver cancer, the evidence is inconclusive. Our study systematically assessed the associations between ten dietary guidelines compliance scores and liver cancer risk among adult people, and found out the dietary patterns for liver cancer prevention. Methods: Participants of 59,844 men and 72,680 women, aged 40-74 years and living in urban Shanghai, were included in two prospective cohort studies conducted between 2002-2006 and 1996-2000, respectively. Dietary intakes were assessed during baseline in-person interviews using validated food-frequency questionnaires, and dietary guideline compliance scores were calculated by adjusting for total energy intake and adapting existing dietary recommendations. Hazards ratios (HRs) with 95 % confidence intervals (CIs) were evaluated by both tertile categories and per standard deviation (SD) increment using Cox proportional hazard regression models to assess the associations between ten dietary guideline compliance scores and liver cancer risk. Results: In the two cohorts, 431 male and 256 female incident liver cancer cases were identified during a mean follow-up of 11.90 and 17.44 years, respectively. There were no statistically significant associations between these ten dietary guidelines and male liver cancer risk (P > 0.05). In contrast, only the modified reversed Empirical Dietary Inflammation Pattern (rEDIP) tended to be associated with the low risk of female liver cancer (HR T3 vs. T1 = 0.67, 95 % CI: 0.48-0.92, P trend = 0.016, HR per SD = 0.94, 95 % CI: 0.85-1.03). The inverse associations appeared stronger between rEDIP and liver cancer risk at younger ages (<55 years) in women (HR per SD = 0.91, 95 % CI: 0.84-0.99) compared to the older women (>55 years). Seismic performance of steel frame with a self-centering beam

Self-centering moment-resisting frames are capable of returning to their original plumb position after a severe earthquake, having sustained no or only slight structural damage. In this paper, a steel frame composed of a self-centering I-beam is proposed, with its capacities for self-centering and inelastic deformation provided by post-tensioned (PT) strands and energy dissipating (ED) elements, respectively. Based on an analysis of its mechanical properties, an experiment involving this self-centering frame under cyclic loading was conducted to investigate its load transfer mechanism, bearing capacity, hysteretic behaviour, ductility, energy dissipating capacity and self-centering capacity. The results agree well with the theoretical analysis, and it can be concluded that its force-displacement hysteretic curve has a typical flag shape. The plastic deformation of the specimen is concentrated on the ED elements, which can be replaced easily after an earthquake. As the gap-opening of the anchorage plate occurs, the PT strands and ED elements provide the bending stiffness for the structure. When the story drift is loaded to 4%, the skeleton curve does not have a descending section, which means that the structure still has sufficient bearing and deformation capacities. The analysis of the residual story drift and the equivalent viscous damping coefficient shows that the frame has good energy dissipating and self-centering capabilities. A finite element model is established with OpenSees to simulate the seismic behaviour of the self-centering frame. Using the model, the seismic performance of the frame is analysed in terms of its structural parameters, including the section area of the PT strands, initial stress on the PT strands and section area of the ED elements. Tough synthetic spider-silk fibers obtained by titanium dioxide incorporation and formaldehyde cross-linking in a simple wet-spinning process

Due to its unique mechanical properties, spider silk shows great promise as a strong super-thin fiber in many fields. Although progress has been made in the field of synthesizing spider-silk fiber from recombinant spidroin (spider silk protein) in the last few decades, methods to obtain synthetic spider-silk fibers as tough as natural silk from small-sized recombinant protein with a simple spinning process have eluded scientists. In this paper, a recombinant spidroin (MW: 93.4 kDa) was used to spin tough synthetic spider-silk fibers with a simple wet-spinning process. Titanium oxide incorporation and formaldehyde cross-linking were used to improve the mechanical properties of synthetic spider-silk fibers. Fibers treated with incorporation or/and cross-linking varied in microstructure, strength and extensibility while all exhibited enhanced strength and toughness. In particular, one fiber possessed a toughness of 249 +/- 22 MJ/m(3). This paper presents a new method to successfully spin tough spider-silk fibers in a simple way. Takeout Service Automation With Trained Robots in the Pandemic-Transformed Catering Business

Under strict social-distancing directives during the COVID-19 pandemic, one of the most impacted businesses is probably the catering industry, which has been forced to earn their revenue mainly from delivery and takeout services. However, traditional takeouts require patrons to wait in line, order and pick up their meals, incurring unnecessary human contact and service inefficiency. We observe these drawbacks and propose a contactless meal order and takeout service (Mots) automated system realized by AI-assisted smart robots to address the issue. In our Mots system, we develop a bump-free schedule based on the Welsh-Powell coloring algorithm for grouping robots into several non-colliding moving batches. Simulation results show that our Mots solution can effectively improve takeout efficiency and promote service accuracy, boosting business profits up to 95.4% under simulated cases for various cafeteria scales and shop popularity differences, compared to the traditional takeout method. Our experiments suggest that Mots is also capable of accommodating a sudden surge of arriving patrons within a short period of time. Furthermore, we have implemented a proof-of-concept prototype to demonstrate our Mots automated operations.

Biotransformation of rutin to isoquercitrin using recombinant rhamnosidase from Bifidobacterium breve

To biotransform rutin into isoquercitrin. A alpha-l-rhamnosidase from Bifidobacterium breve was produced by using Escherichia coli BL21 for biotransformation of rutin to isoquercitrin. The enzyme was purified by Ni2+-NTA chromatography to yield a soluble protein with a specific activity of 56 U protein mg(-1). The maximum enzyme activities were at pH 6.5, 55 A degrees C, 20 mM rutin, and 1.2 U enzyme ml(-1). Under optimal conditions, the half-life of the enzyme was 96 h. The K (m) and V (max) values were 2.2 mM, 56.4 mu mol mg(-1) min(-1) and 2.1 mM, 57.5 mu mol mg(-1) min(-1) using pNP-Rha and rutin as substrates, respectively. The kinetic behavior indicated that the recombinant alpha-l-rhamnosidase has good catalytic performance for producing isoquercitrin. 20 mM rutin was biotransformed into 18.25 and 19.87 mM isoquercitrin after 60 and 240 min. The specific biotransformation of rutin to isoquercitrin using recombinant alpha-l-rhamnosidase from B. breve is a feasible method for use in industrial processes.

Effect of annealing temperature on the microstructure and mechanical properties of laves phase reinforced FeCrAl alloy thin-walled tubes

FeCrAl alloy has great application potential to replace traditional zirconium alloy as an accident-tolerant fuel (ATF) candidate material. In this study, Fe-13Cr-4Al-2Mo-0.65Nb-0.4Ta alloy was prepared into a thin-walled tube with a diameter of 9.5 mm and a wall thickness of 0.35 mm by cold rolling. The effects of different annealing temperatures on the microstructure morphology, texture growth direction, and me-chanical properties of FeCrAl alloy were studied., alloy underwent recovery and recrystallization with increasing annealing temperature, and the grain size increased. The theoretical calculation showed that the pinning action of Fe2Mo4C and Fe2Nb increased the dislocation density, and the interaction with the dislocation increased the room temperature strength. The addition of Ta led to the formation of more second-phase precipitates at the grain boundary, which hindered the growth and refined grains. The high-temperature stable second phase provided sufficient temperature strength for the alloy.(c) 2022 Elsevier B.V. All rights reserved.

Impact of 5-Fu/oxaliplatin on mouse dendritic cells and synergetic effect with a colon cancer vaccine

Objective： The aim of the present study was to investigate the effects of 5-fluorouracil（5-Fu） and oxaliplatin on the function and activation pathways of mouse dendritic cells（DCs）, and to clarify whether 5-Fu/oxaliplatin combined with the CD1d-MC38/α-galactosylceramide（α-GC） tumor vaccine exhibits synergistic effects on the treatment of colon cancer in ***： The combination of the Toll like receptor（TLR） ligands and/or 5-Fu/oxaliplatin was added into myeloid-derived DCs in vitro culture. DC phenotypic changes were detected by flow cytometry, and the secretion of DC cytokines was detected by cytometric bead array（CBA）. A MC38 mouse colon cancer model was constructed and the DCs were isolated from the spleen, tumor tissue and lymph nodes following intraperitoneal injection of 5-Fu/oxaliplatin. The cell phenotypes were detected by flow cytometry. The tumor infiltrating leukocytes,splenocytes and lymph node cells were co-cultured with the dead MC38 tumor cells, and the secretion levels of interferon-γ（IFN-γ） were detected. 5-Fu/oxaliplatin combined with our previously developed CD1d-MC38/α-GC tumor vaccine was used to inhibit the growth of MC38 colon cancer in mice, and the tumor growth rate and survival time were ***： 5-Fu/oxaliplatin exerted no significant effect on the expression of the stimulating phenotypes of DCs in vitro, while it could reduce the expression of programmed death ligand 1/2（PD-L1/L2） and promote interleukin-12（IL-12） secretion by DCs. Furthermore 5-Fu/oxaliplatin was beneficial to the differentiation of T-helper 1（Th1） cells. 5-Fu/oxaliplatin further enhanced the stimulating phenotypic expression of DCs in tumor bearing mice, decreased PD-L1/L2 expression, and specifically activated the lymphocytes. The CD1d-MC38/α-GC tumor vaccine combined with 5-Fu/oxaliplatin could exert a synergistic role that resulted in a significant delay of the tumor growth rate, and an increase in the survival time of tumor bearing ***： 5

Pore-forming technology and performance of MEA for direct methanol fuel cells

BACKGROUND: The commercialization of DMFCs is seriously restricted by its relatively low power density. Lots of work has been concentrated on catalysts with high activity, the optimization of flow path design, development of new kinds of proton exchange membrane and modification of Nafion membrane. Meanwhile, very few reports have involved the structure optimization of the membrane electrode assembly (MEA). To improve the performance of direct methanol fuel cells (DMFCs), the catalyst layer (CL) structures of anode and cathode were optimized by utilizing ammonium carbonate as pore forming agent. RESULTS: The polarization curves showed that in catalyst slurry the optimal content of ammonium carbonate was 50 wt%, and the DMFC performance was enhanced from 75.65 mW cm2 to 167.42 mW cm2 at 55 degrees C and 0.2 MPa O2. Electrochemical impedance spectroscopy and electrochemical active surface area (EASA) testing revealed that the improved performance of optimized MEAs could be mainly attributed to the increasing EASA and the enhanced mass transfer rate of CLs. But poor methanol crossover limited the performance enhancement of MEAs with porous anodes. CONCLUSION: With regard to improving cell performance, this pore-forming technology is better applied to the cathode catalyst layer to improve its structure rather than the anode catalyst layer.
