Purpose The paper aims to investigate the biological role of microRNAs secreted by preimplantation embryo into the blastocoel fluid and to detect a distinctive molecular signature for identifying embryos with the highest implantation potential. Methods We carried on a multicenter retrospective study involving five European IVF centers. We collected 112 blastocoel fluid samples from embryos on day 5 post-fertilization, cultured individually, along with data on blastocyst grade and embryo transfer outcomes. Using a custom TLDA Array, we compared the expression levels of 89 miRNAs between 33 fluids from high-quality implanted embryos and 30 fluids from high-quality not-implanted embryos. Expression differences were assessed using SAM and t-test. Additionally, correlation and function enrichment analysis and network construction were conducted to identify the biological roles of deregulated microRNAs. Results We identified six up-regulated microRNAs in the blastocoel fluid from implanted embryos, significantly and positively correlated across all samples (r >= 0.7;P <= 0.05). They could take part in pluripotency circuits, regulating and being regulated by transcription factors associated with stemness, cell growth, and embryo development. The ROC curve analysis confirmed the potential of these miRNAs as implantation classifiers. Conclusion The six miRNAs up-regulated in blastocoel fluid from implanted embryos may represent a functional molecular signature for evaluating blastocyst quality and identifying the most competent embryos. Their evaluation associated with non-invasive preimplantation genetic testing, integrating epigenetic and genomic analyses, could enhance implantation grade and allow for identification of the euploid embryo not able to implant.

The purpose of this thesis is to discuss the use of liquid metals in next-generation nuclear fission and fusion reactors. Liquid metals can be employed as coolant (fis- sion machines) or as coolant/breeder/carrier for the tritium exploitation (in future fusion rectors), since they present good thermo-physical and nuclear properties with respect to water. Due to the complexity of the systems where liquid metals have to be used, suitable multi-physics tools must be developed to correctly retrieve the inherent physical aspects of their employment. Several European initiatives are today supporting the research and development of this tools, as for instance, the EUROfusion consortium and the PASCAL project (where the FALCON consortium is heavily involved). Thus, the work here presented aims to verify, test and develop tools and new models for the multi-physics assessment of advanced nuclear system based on liquid metals. For this reason, two areas of activity are discussed: mod- elling of Liquid Metal-Cooled Reactors for future Generation IV reactors within the context of reactor physics and thermal-hydraulics assessments, and modelling of radioisotope transport in liquid metals in fusion systems. The first part of this thesis regards the use of liquid metals in future fission nuclear reactors. The activities have been carried on with the scope to pursue the verification activities of the FRENETIC code, a multi-physics tool that has been being developed in the last years at Politecnico di Torino. Currently, the FALCON consortium and the PASCAL project are supporting the design of the ALFRED reactor in order to exploit Lead Fast Reactors technology. As a conse- quence, the benchmark of FRENETIC has been performed on the ALFRED core, analysing both steady state and transient coupled scenarios. Capsules are long-chain carbohydrate polymers that envelop the surfaces of many bacteria, protecting them from host immune responses. Capsule biosynthesis enzymes are potential drug targets and valuable biotechnological tools for generating vaccine antigens. Despite their importance, it remains unknown how structurally variable capsule polymers of Gram-negative pathogens are linked to the conserved glycolipid anchoring these virulence factors to the bacterial membrane. Using Actinobacillus pleuropneumoniae as an example, we demonstrate that CpsA and CpsC generate a poly(glycerol-3-phosphate) linker to connect the glycolipid with capsules containing poly(galactosylglycerol-phosphate) backbones. We reconstruct the entire capsule biosynthesis pathway in A. pleuropneumoniae serotypes 3 and 7, solve the X-ray crystal structure of the capsule polymerase CpsD, identify its tetratricopeptide repeat domain as essential for elongating poly(glycerol-3-phosphate) and show that CpsA and CpsC stimulate CpsD to produce longer polymers. We identify the CpsA and CpsC product as a wall teichoic acid homolog, demonstrating similarity between the biosynthesis of Gram-positive wall teichoic acid and Gram-negative capsules.

A second hypothesis, related to the suh,3scts' estimate or the decision made by people in general before and after group discussion, predicted that when groups vslusd rish, subjects rslstivcly cautious in their initial choices would visw people in general as more risky after group discussion, also, when groups valued caution, subjects rslstivsly risky in their initial choices would View people in general as more cautious error group discussion. The study involved two phases. The first phase was conducted in class and consisted of administering an sight-item risx-tssing questionnaire to deter- mine which psrsons valued risk and caution on each of the items. In the sc- cond laboratory phase or the study, 10 rsmsls and 9 male groups discussed the risk-taking items and arrived at a unanimous decision on each of them. Although hypothesis 1 was not supported, Hypothesis 2 did receive partial support. Three possibilities were offered to account for the failure to sup- port hypothesis 1: ths nature of the items used in the study; the small number of groups discussing each item; and possibly the fact. that the perceived rela- tive riskincss instrument is o poor measure of value.

Prenatal diagnosis of a partial 6q trisomy: A case report

Objective To present a case of partial 6q trisomy diagnosed prenatally. Method A 28-year-old woman underwent genetic amniocentesis at 23 weeks of gestation on the detection of an enlarged nuchal fold (8.5 mm), which was the only clinical abnormality oh routine ultrasound examination. Fetal karyotyping revealed a partial trisomy 6q (q21-q22) caused by a balanced maternal chromosomal insertion. Results A female infant was delivered at 38 weeks of gestation. At birth, minor dysmorphisms were recorded, which included low set ears, macrocephaly and a webbed neck. At 17 months of age, neurological developmental assessment was normal. Conclusions The appearance of phenotypic expression and clinical outcome of partial 6q trisomy depends on the specific chromosomal region involved in the segmental aneusomy. Objectives: The aim of this study was to evaluate the diagnostic accuracy of a specific protocol of computed tomography-colonography with intravenous contrast medium and urographic phase, which combined simultaneously the study of the intestinal and urinary tract, in the preoperative evaluation of women with deep infiltrating endometriosis (DIE) of anterior and posterior pelvic compartments. Methods: We retrospectively analyzed 73 women who underwent 74 computed tomography-colonography with intravenous contrast medium and urographic phase examinations for strong clinical suspicion of DIE. All the women had surgical confirmation. Results: Computed tomography-colonography with intravenous contrast medium and urographic phase in detecting DIE rectosigmoid involvement had a sensitivity of 82.3% and a specificity of 66.7%, while in detecting DIE urinary tract involvement had a sensitivity of 45.9% and a specificity of 78.4%. Conclusions: Computed tomography-colonography with intravenous contrast medium and urographic phase is a useful technique for the preoperative planning of selected women with DIE, in particular, for the detection of sigmoid colon and bladder lesions especially when performed with a dose reduction protocol.

The Fast REactor NEutronics/Thermal-hydraulICs (FRENETIC) code has been developed during the last years at Politecnico di Torino, implementing a full-core coupled neutronic/thermal-hydraulics model for steady-state and transient analysis of liquid-metal cooled fast breeder reactor (LMFBR). In the framework of the validation activities for the code, an analysis of the sodium-cooled reactor EBR-II, previously carried out in the frame of a IAEA Coordinated Research Project, is performed with FRENETIC including the most recent physics models. In particular, photon transport and heat deposition are taken into account, a feature which has been proved in previous studies to be relevant to the correct study of the EBR-II core. To this purpose, a set of nuclear data for photons has been generated by means of the Monte Carlo code Serpent-2, and it is demonstrated that the code is able to take into account the photon heat deposition in the EBR-II. © The Authors, published by EDP Sciences. To compare the diagnostic accuracy of transvaginal sonography (TVS) and computed tomography-colonography with contrast media and urographic phase (CTCU) in the preoperative detection of deep infiltrating endometriosis (DIE). Forty-seven patients with clinical suspicion of DIE underwent preoperative TVS and CTCU. Imaging data were compared with histopathologic analysis. Sensitivity, specificity, positive and negative predictive values and test accuracies of the two modalities were calculated. For diagnosing intestinal DIE, TVS and CTCU had a sensitivity of 98 and 71%, specificity of 33 and 50%, positive predictive value of 91 and 91%, negative predictive value of 67 and 20%, accuracy of 89 and 68%, respectively. For diagnosing ureteral DIE, TVS and CTCU had a sensitivity of 10 and 60%, specificity of 94.8 and 70.2% on the right;sensitivity of 28.5 and 57.1%, specificity of 96.3 and 76.9% on the left, respectively. TVS should be regarded as an accurate, radiation-free first-line diagnostic modality for patients with suspicion of posterior endometriosis. CTCU should be regarded as a complementary imaging modality, particularly for sigmoid or ureteral endometriosis.

Soft Human-Robot Handover Using a Vision-Based Pipeline

Handing over objects is an essential task in human-robot collaborative scenarios. Previous studies have predominantly employed rigid grippers to perform the handover, focusing on generating grasps that avoid physical contact with people. In this paper, we present a vision-based open-palm handover solution where a soft robotic hand exploits contact with the human hand for improved grasp success and robustness. The human-robot physical interaction allows the robotic hand to slide over the human palm and firmly cage the object. The identification of the human hand plane and object pose is achieved through a versatile perception pipeline that exploits a single RGB-D camera. Through experimental trials, we show that the system achieves successful grasps over multiple objects with different geometries and textures. A comparative analysis assesses the robustness of the proposed soft handover method against a baseline approach. A study with 30 participants evaluates users' perception of human-robot interaction during the handover, highlighting the effectiveness and preference for the proposed pipeline.

Characterization of fetal haematopoietic progenitors circulating in maternal blood of seven aneuploid pregnancies

A retrospective study was carried out in order to investigate the phenotype of fetal haematopoietic progenitors circulating in the maternal blood of seven aneuploid pregnancies. Five of the blood samples were taken during pregnancies affected by various fetal aneuploidies, while the other two were collected after therapeutic abortion due to prenatal cytogenetic diagnosis of trisomies 21 and 18. Haematopoietic progenitor cells, isolated by labelling the erythropoietin receptors with the biotinylated ligand before magnetic sorting and/or fibronectin cell adhesion assay, were cultured in a suitable semisolid medium. Single- or dual-colour fluorescence in situ hybridization (FISH) was utilized to identify and enumerate fetal cells amplified in culture. Fetal trisomies were confirmed in the FISH analysis with chromosome-specific probes in all the cases analysed. The fetal purity rate ranged from 16 to 26 per cent. Haematopoietic progenitors of fetal origin were found to include CFU-E, CFU-GEMM, and possibly also M-BFU-E. Interestingly, a more immature progenitor with high self-renewal capacity (CFU-blast cell) isolated by fibronectin sorting was shown to have a relatively high frequency in one case of Down syndrome. In general, the results of this study demonstrate the feasibility of diagnosing the major fetal chromosomopathies by culturing fetal cells taken from maternal blood. Furthermore, our initial data on the sequential sorting for fibronectin and erythropoietin receptors lead us to believe that this approach may broaden the range of fetal haematopoietic progenitors retrievable from the maternal circulation. © 1997 John Wiley & Sons, Ltd.
