Objective: To investigate the effect of mifepristone, an oral contraceptive, on apoptosis in human ovarian luteinized granulosa cells. Study design: Human ovarian luteinized granulosa cells were treated in vitro with 1.25, 2.5, 5.0, 10.0, or 20.0 mu M of mifepristone. Nuclear morphology, apoptosis ratio, and level of caspase-3 expression were determined with immunofluorescence microscopy, the terminal deoxynucleotidyl transferase mediated nick end labeling (TUNEL) assay, and flow cytometry. Results: We found that mifepristone-treated cells contained single condensed chromatin with multiple nuclear fragments, which is morphologic evidence of apoptosis. A significant difference was observed in the TUNEL assay between mifepristone-treated cells and control cells (P < 0.05). Consistent with the results of the TUNEL assay, the fluorescence intensity of caspase-3 in drug-treated cells was also significantly different (P < 0.05) from control cells;specifically, the difference between cells treated with different doses of drugs was much smaller and negligible. Conclusion: From these results, we propose that mifepristone induces human ovarian luteinized granulosa cells to undergo apoptosis by activating caspase-3. (C) 2008 Elsevier Ireland Ltd. Objectives. To clarify the change of peripheral CD4+CD25+ regulatory T lymphocytes in normal pregnancy and pre-eclampsia and to explore its role in the pathogenesis of pre-eclampsia. Methods. Twenty-seven women with pre-eclampsia, 27 with normal third trimester pregnancy and 27 healthy non-pregnant women were recruited. Blood samples were taken and surface antigen CD4 and CD25 were labelled with fluorescence-conjugated antibodies. CD4+CD25+ regulatory T cells were analysed by flow cytometry, and the proportion in T cells and the amount of regulatory T lymphocytes were calculated. Results. The amount and the proportion of regulatory T cells were not significantly different among non-pregnant, normal pregnancy and pre-eclampsia groups (p > 0.05 for both). There were no significant differences in the proportion and the amount of CD4+CD25+ regulatory T cells between mild and severe pre-eclampsia subgroups (p > 0.05 for both). Conclusions. No significant change of peripheral regulatory T cells was observed in the current investigation, suggesting other mechanisms rather than CD4+CD25+ regulatory T cells were involved in the pathogenesis of pre-eclampsia.

Because of aeroengine properties such as the strong nonlinearity and time-varying uncertainty, a new identification algorithm of aeroengine model based on improved LS-SVM was brought forward. In the method, LS-SVM robustness was improved by adding weighed values to errors and its sparseness was improved by clipping algorithm. In terms of the recorded flight data on some turbofan engine, improved LS-SVM identification model of aeroengine was set up. Through the identification of the recorded flight data, the results show that the improved LS-SVM identification model has the advantages of high identification precision, good self-adaptability and strong robustness. It is effective that the improved LS-SVM identification model is used in aeroengine.

Laffey, Perturbing non-real eigenvalues of nonnegative real matrices, Electron. J. Linear Algebra 12 (2005) 73-76] has shown that (7 = (rho + 2t, b - t + ic, b - t - ic, lambda(4),...,lambda(n)) is also the spectrum of some non-negative matrix. Laffey (2005) has used a rank one perturbation for small t and then used a compactness argument to extend the result to all non-negative t.
