目的:探讨和分析氨基葡萄糖对膝骨关节炎(osteoarthritis,DOA)的治疗方法在临床中应用中的疗效体会,为广大骨关节炎病患者的治疗提供参考.方法:选取本院2010年10月-2012年10月期间采用盐酸氨基葡萄糖治疗90例膝D O A患者.男性患者36例、女性患者54例;年龄均在45岁~75岁,平均年龄60.5岁,病程2个月到6年不等.口服盐酸氨基葡萄糖片、每日三次、每次480毫克,6周为一疗程,4~6个月重复一个疗程.所有患者均经过2个疗程的治疗后,(采用国际DOA的评分标准:Leuesne指数评定膝关节休息疼痛、运动疼痛、压痛、关节活动度、晨僵和活动能力).结果:有85例患者膝关节疼痛、活动度、晨僵均有不同程度的好转,有10例患者在第一疗程后膝关节疼痛消失,有20例患者在第一疗程结束后疼痛、晨僵明显缓减、膝关节活动度明显改善,有15例患者在第二疗程结束后疼痛消失,有52例患者在第二疗程治疗结束后、膝关节疼痛、晨僵等女明显好转、活动度明显改善,有5例患者无效,有3例患者出现胃部不适(既往患者有慢性胃炎史,患者自服奥美拉唑缓减).结论:盐酸氨基葡糖治疗退行性骨性关节炎,可延缓骨关节炎的病理过程,延缓病情的进展、缓减疼痛、肿胀等症状,改善关节功能,延缓关节变性形成,适合在临床上进行推广与应用.

Camptothecin (CPT) has been shown to block disassembly of the topoisomerase I (Topo I)/DNA cleavable complex. However, the poor aqueous solubility, intrinsic instability, and severe toxicity of CPTs have limited their clinical applications. Herein, we report the design and synthesis of H2O-soluble and orally bioavailable hexacyclic CPT derivatives. By analysis of a virtual chemical library and cytotoxicity screening in vitro, 9 and 11 were identified as potential prodrugs and chosen for further characterization in vivo. Both compounds exhibited remarkable anticancer and anti-inflammation efficacies in animals and improved drug-like profiles.

Thallium (Tl) is a highly toxic rare element. Severe Tl poisoning can cause neurological brain damage or even death. The present study was designed to investigate contents of Tl and other associated heavy metals in arable soils and twelve common vegetables cultivated around a steel plant in South China, a newly-found initiator of Tl pollution. Potential health risks of these metals to exposed population via consumption of vegetables were examined by calculating hazard quotients (HQ). The soils showed a significant contamination with Tl at a mean concentration of 1.34 mg/kg. The Tl levels in most vegetables (such as leaf lettuce, chard and pak choy) surpassed the maximum permissible level (0.5 mg/kg) according to the environmental quality standards for food in Germany. Vegetables like leaf lettuce, chard, pak choy, romaine lettuce and Indian beans all exhibited bioconcentration factors (BCF) and transfer factors (TF) for Tl higher than 1, indicating a hyperaccumulation of Tl in these plants. Although the elevated Tl levels in the vegetables at present will not immediately pose significant non-carcinogenic health risks to residents, it highlights the necessity of a permanent monitoring of Tl contamination in the steel-making areas. The cholera toxin B subunit (CTB) is a nontoxic portion of the cholera toxin that retains mucosal adjuvant properties. Expression of CTB in Escherichia coli is difficult as CTB aggregates and accumulates as insoluble inclusion bodies. To remedy this problem, the periplasmic chaperone, SKP, was investigated as possible co-expression partner to increase the solubility of recombinant CTB (rCTB) in E. coil. The result showed co-expression of SKP enhanced the soluble expression of rCTB in E. coli. Moreover, soluble rCTB was successfully expressed and secreted into the periplasmic space through the direction of the LTB leader signal. rCTB in periplasm was purified using an immobilized D-galactose resin;GM1-ELISA ex-periments showed that rCTB retains strong GM1 ganglioside-binding activity. Intranasal administration of ovalbumin (OVA) with rCTB significantly induced both mucosal and humoral immune responses specific to OVA. These data indicate that co-expression of the molecular chaperone SKP with CTB increased the solubility of rCTB while maintaining its function. (c) 2017 Elsevier Inc. All rights reserved.

Dietary resveratrol (RES) supplementation may have some pharmacological effects including anti inflammation. Previous studies have shown that Kupffer cell activation and apoptosis induction increases the transcription of pro- and anti-inflammatory cytokines. The main purpose of this study was to investigate the pro- and anti-inflammatory activities of 0.1 or 0.3 g/kg RES as a dietary supplement in juvenile freshwater tilapia (Oreochromis niloticus). The results showed that hepatic and serum immunoglobulin M (IgM) significantly decreased and increased while anti- and pro-inflammatory cytokines significantly increased and decreased, respectively, in the RES-treated groups. The expression of serum and hepatic IgM and anti-inflammatory cytokines [interleukin (IL)-10] and its inverse inhibitor interferon (IFN)-gamma significantly increased while pro-inflammatory cytokine transcription significantly decreased. Hematoxylin-eosin staining and scanning electron microscopy revealed intestinal deformation, irregular goblet cells, and apoptotic cells in the 03 g/kg RES groups. RES (0.3 g/kg) also induced necrosis, apoptosis, reduction in Kupffer cell number, compressed sinusoids, and deformation of epidermal cells in the liver of the treated groups. In conclusion, the results of the present study show that high doses of RES were absorbed in the gut and then damaged the liver and intestinal tissue. (C) 2017 Published by Elsevier Ltd.
