Five novel biscembranoids,ximaolides H-L(1-5),along with four known related compounds(6-9) were isolated from the Hainan soft coral Sarcophyton *** structures of the new compounds were determined by extensive spectroscopic analysis,quantum chemical calculations,and/or by comparing their CD spectra with those of the known *** 1 and 2 are the first examples of biscembranoids bearing a 1,35-bridged lactone moiety,4 is the first biscembranoid comprising an uncommon oxetane ring,and 5 represents the first 36-peroxyl *** Ⅰ(2),K(4) and F(9) exhibited interesting anti-inflammatory activity by the inhibition of LPS-induced TNF-α protein release in RAW264.7 macrophages.

为评估香烟烟气中杂环胺的暴露水平,建立了一种基于低共熔溶剂/无机盐双水相萃取-UPLC法测定吸烟者尿液中5种杂环胺(IQ,IQ[4,5-b],Harman,Norharman和Phe-P-1)的方法.制备了7种以氯化胆碱(ChCl)为氢键受体的低共熔溶剂,并对其进行了表征;系统考察了影响萃取效率的主要参数,并将该方法与其它方法进行了对比研究.结果表明,在最佳提取条件下,杂环胺(HAAs)的回收率为81.9%~106.2%,日内和日间精密度分别低于2.9%和3.2%,检出限和定量限分别为0.020~0.097 ng/g和0.19~0.39 ng/g.该方法具有操作简单、环境友好及单样品成本低等特点,为尿液中杂环胺的检测提供了一种新方法,在生物样品检测领域具有良好的开发与应用价值.

Development of fresh solid phase extractant is critical for selective separation and purification of special proteins. Herein, we demonstrated a recombinant Staphylococcal Protein G (rSPG) with a His-tag modified the novel single-metal organic framework (rSPG@Ni-MOF-74). The proposed solid-phase extraction material possessed a uniform spindle-shaped structure, large surface area (709.60 m2 g-1) and pore volume (0.08 m3 g- 1), high metal content (22.57 wt%), which facilitated the interaction between host and guest. As results, the composite displayed outstanding selective recognition and adsorption of IgG, due to synergistic effect of the binding ability of rSPG with the Fc region of IgG, maintained through hydrogen bonding and electrostatic attraction, as well as hydrophobic interaction. The adsorption performance and mechanism of rSPG@Ni-MOF-74 have been fully investigated. Additionally, the rSPG@Ni-MOF-74 composite could effectively separate IgG from serum obtained from healthy humans, with the purity of the separated IgG verified through SDS-PAGE analysis. Furthermore, LCMS/MS analysis identified a high content of IgG (55.3 %) in the eluate from rSPG@Ni-MOF-74, suggesting the great potential of rSPG@Ni-MOF-74 in IgG separation and enrichment from complex matrix.

Since the semiconductor technology has entered the age of 90 nm, the static power consumption of the chip has increased rapidly. For edge computing devices, the waste of power can not be ignored. This paper uses the relationship between power consumption and power supply voltage of FPGA and DSP, analyzes the characteristics of the traditional scheme, and designs a hardware circuit according to the different mechanisms of reducing static power consumption of FPGA and DSP According to the design of FPGA and DSP core power supply voltage compatibility, the system realizes the requirement of FPGA and DSP for power supply voltage amplitude and realizes dynamic control of power supply voltage. After verification and comparison, FPGA and DSP can realize the adjustment of performance power consumption and stable operation under the power system. © 2021 ACM.

Apart from the blood-brain barrier (BBB), the efficacy of immunotherapy for glioblastoma (GBM) is limited by the presence of intrinsic and adaptive immune resistance, implying that co-delivery of various immunotherapeutic agents or simultaneous regulation of different cells is urgently needed. Bacterial outer membrane vesicles (OMVs) offer a unique advantage in the treatment of GBM, owing to their multifunctional properties as carriers and immune adjuvants and their ability to cross the BBB. However, traditional OMVs can lead to toxic side effects and disruption of tight junctions in the BBB. Therefore, to enhance the in vivo safety and targeting capability of OMVs, we introduced engineered OMVs to reduce toxicity and further constructed a modularly assembled nanoplatform by performing simple peptide modifications. This nanoplatform demonstrates satisfactory biosafety and is able to continuously cross the BBB and target GBM with the assistance of Angiopep-2. Subsequently, immunogenic substances on OMVs, along with carried small-interfering RNA (siRNA) and doxorubicin, can promote and enhance the reprogramming and phagocytic abilities of macrophages and microglia, respectively, and increase the immunogenicity of GBM, ultimately overcoming GBM immune resistance to enhance the efficacy of immunotherapy. This OMVs-based nanoplatform provides a new paradigm and insights into the development of immunotherapy for GBM.

No ureteral catheter mini percutaneous nephrolithotomy (NUC-mPCNL) achieves enhanced recovery after surgery (ERAS)

Conventional mini percutaneous nephrolithotomy (mPCNL) typically necessitates the pre-operative placement of ureteral catheters. With the development of enhanced recovery after surgery (ERAS), we introduce a novel approach: no ureteral catheter mini percutaneous nephrolithotomy (NUC-mPCNL). To evaluate the efficacy and safety of this innovative procedure compared to mPCNL, we compared the total operative time, stone clearance rate (SCR), stone-free rate (SFR), and complications among patients undergoing either modality from January to September 2021. Our findings revealed a statistically significant reduction in the mean total operative time for the NUC-mPCNL group (42.6 +/- 2.4 vs. 60.2 +/- 3.4 min, p < 0.001). The SCR, SFR and CSFR were comparable between two groups (98.5% vs. 97.8%, 84.3% vs. 76.0%, 64.7% vs. 60.0%, p = 0.337, 0.295, 0.625). WBC rise, hemoglobin drop and neutrophil percentage rise were similar in both groups. The overall complication rate in the NUC-mPCNL group was lower;however, the difference was not statistically significant (Clavien I, II and III;11 vs. 13, 1 vs. 2 and 1 vs. 1). Consequently, we conclude that NUC-mPCNL effectively shortens the operative time and holds potential for expedited postoperative recovery.

Synthesis and characterization of photo-responsive azobenzene side-chain liquid crystalline polystyrenes with a mesogenic phase of wide temperature range

Polystyrenes (PSs) that provide photochemical stability are commonly used as the chromophore macromolecular carriers. Herein, PS-based alternating copolymers, i.e., alt-P(D-SiH/Ip), alt-P(D-2SiH/Ip) and alt-P(D-SiH/St), are synthesized through anionically copolymerized Si-H functionalized 1,1-diphenylethylene (DPE-SiH/2SiH) with isoprene (Ip)/styrene (St). Anionically polymerized (4-vinylphenyl)-dimethyl-silane is used as the uniform hydrosilylated PS, i.e., Uni.-P(St-SiH). A series of photo-responsive azobenzene side-chain liquid crystalline PSs combine the above PS-based main chains and the mesogenic moiety composed of azobenzene (Azo) and cholesterol (Chol) groups, i.e., Azo-Chol (M), via the hydrosilylation. The polarized optical microscopic (POM) morphologies and thermal properties of Azo-Chol (M) and Azo-Chol-PSs are studied. As desired, they produce the cholesteric phase of wide temperature range due to the PSs and Chol-groups. Moreover, the photo-responsive behavior irradiated under UV-light of different wavelengths are quantitatively evaluated by time-dependent UV-Vis spectroscopy. Azo-Chol-PSs undergo rapid and reversible trans-cis isomerization under the alternating irradiation of UV and visible light. The trans-to -cis isomerization efficiency under 365 nm UV-light is over that under 290-320 nm UV-light, while over 80% of cis-to-trans efficiency can be realized under 400-500 nm Vis-light irradiation This work would provide guidelines for the effective design of photo-responsive anisotropic materials.

Discovery of novel small molecules targeting hepatitis B virus core protein from marine natural products with HiBiT-based high-throughput screening

Due to the limitations of current anti-HBV therapies, the HBV core (HBc or HBcAg) protein assembly modulators (CpAMs) are believed to be potential anti-HBV agents. Therefore, discovering safe and efficient CpAMs is of great value. In this study, we established a HiBiT-based high-throughput screening system targeting HBc and screened novel CpAMs from an in-house marine chemicals library. A novel lead compound 8a, a derivative of the marine natural product naamidine J, has been successfully screened for potential anti-HBV activity. Bioactivity-driven synthesis was then conducted, and the structure‒activity relationship was analyzed, resulting in the discovery of the most effective compound 11a (IC50 = 0.24 μmol/L). Furthermore, 11a was found to significantly inhibit HBV replication in multiple cell models and exhibit a synergistic effect with tenofovir disoproxil fumarate (TDF) and IFNa2 in vitro for anti-HBV activity. Treatment with 11a in a hydrodynamic-injection mouse model demonstrated significant anti-HBV activity without apparent hepatotoxicity. These findings suggest that the naamidine J derivative 11a could be used as the HBV core protein assembly modulator to develop safe and effective anti-HBV therapies.

Microbial Proliferation Deteriorates the Corrosion Inhibition Capability, Lubricity and Stability of Cutting Fluid

The microbial diversity and the effect of microorganism on cutting fluids were analyzed. The surface cutting fluid from the machine tool tank was dominated by aerobic bacteria, while the bottom mainly harbored anaerobic and facultative microorganisms, with Yarrowia lipolytica being the dominant fungus. The obligate anaerobic bacteria were present in the cutting fluid. The secretion of acidic substances and the consumption of triethanolamine and rganophosphate esters led to a significant decrease in the cutting fluid pH and corrosion inhibition capability. Moreover, during the late stage, the number of fungi increased. The resulting fungal mycelium and porous extracellular polymeric substance secreted by microorganisms caused flocculation of the effective components in the cutting fluid, resulting in reduced lubricity and poor stability. © 2024, The Authors. Effects of total saponins from Panacis majoris Rhizoma and its degradation products on myocardial ischemia-reperfusion injury in rats

Ethnopharmacological relevance: Panacis majoris Rhizoma, which is a member of herbal medicine, is known for many years to remove blood stasis, promote blood circulation, and enrich the blood. The active ingredients of this plant are mainly attributed to saponins. Aim of the study: The total saponins from Panacis majoris Rhizoma (TSPJ), and the degradation products of TSPJ (DTSPJ), were designed in this study to compare the protective effects on myocardial ischemia-reperfusion injury, and the aim of this approach is to discover more effective agents for the treatment of ischemic heart diseases. We analyzed the main constituents of TSPJ and DTSPJ, aiming to make clear which saponins played important roles in this protective effect, and also investigated the possible mechanisms. Materials and methods: DTSPJ was prepared by the method of alkaline hydrolysis. High performance liquid chromatography (HPLC) were used to analyze the main chemical constituents of TSPJ and DTSPJ, which were isolated by chromatographic techniques and identified by comparison with the Nuclear Magnetic Resonance (NMR) data in reported literature. Male Wistar rats were randomized to sham-operated group, ischemia-reperfusion group, three TSPJ (50, 100 and 200 mg/kg) groups, three DTSPJ (50, 100 and 200 mg/kg) groups, and isosorbide dinitrate tablet (5.0 mg/kg) group. The rats in all groups were intragastrically administrated once per day for three successive days. The establishment of the model of myocardial ischemia-reperfusion injury was used the following method: firstly, the left coronary artery of experimental rat was ligated for 30 min and then reperfused for 120 min. Then the myocardial infarct size, hemorheological and biochemical parameters, whole blood viscosity, plasma viscosity, platelet adhesion rate, platelet aggregation and histopathology changes were assessed.
