Acanthopanacis Cortex (AC) is a Chinese herbal medicine (Wu-Jia-Pi in PinYin) which is recognized with therapeutic effect of "strengthening bone and sinew", yet its regulating biological network is still obscure. In this study, started from AC's chemical compounds, their direct target proteins were filtered out. Then these targeted proteins were annotated with biological processes under the framework of Gene Ontology (GO) terms' hierarchical networks which were overrepresented and enriched with them with statistic significant p-value set to 0.05. In the biological process hierarchy networks, ossification formed a big one with bone formation associated biological processes. Further protein-protein interaction analysis with these targeted proteins and bone expressed ones formed a clear and concise protein-regulation network for ossification which should be responsible for the therapeutic effect of "strengthening bone and sinew". This study proposed a new approach of analyzing biological functions of a specified Chinese herbal medicine targeting certain therapeutic effect.

Folliculin (FLCN) is the tumor suppressor associated with Birt-Hogg-Dube (BHD) syndrome that predisposes patients to incident of hamartomas and cysts in multiple organs. Its inactivation causes deregulation in the mammalian target of rapamycin complex 1 (mTORC1) signaling pathway. However, the underlying mechanism is poorly defined. In this study, we show that FLCN is a ciliary protein that functions through primary cilia to regulate mTORC1. In response to flow stress, FLCN associates with LKB1 and recruits the kinase to primary cilia for activation of AMPK resided at basal bodies, which causes mTORC1 down-regulation. In cells depleted of FLCN, LKB1 fails to accumulate in primary cilia and AMPK at the basal bodies remains inactive, thus nullifying the inhibitory effect of flow stress on mTORC1 activity. Our results demonstrate that FLCN is part of a flow sensory mechanism that regulates mTORC1 through primary cilia.

Long noncoding RNAs (lncRNAs) play vital roles in tumorigenesis. However, the diagnostic values of most lncRNAs are largely unknown. To investigate whether gastric juice lncRNA-ABHD11-AS1 can be a potential biomarker in the screening of gastric cancer, 173 tissue samples and 130 gastric juice from benign lesion, gastric dysplasia, gastric premalignant lesions, and gastric cancer were collected. ABHD11-AS1 levels were detected by reverse transcription-polymerase chain reaction. Then, the relationships between ABHD11-AS1 levels and clinicopathological factors of patients with gastric cancer were investigated. The results showed that ABHD11-AS1 levels in gastric cancer tissues were significantly higher than those in other tissues. Its levels in gastric juice from gastric cancer patients were not only significantly higher than those from cases of normal mucosa or minimal gastritis, atrophic gastritis, and gastric ulcers but also associated with gender, tumor size, tumor stage, Lauren type, and blood carcinoembryonic antigen (CEA) levels. More importantly, when using gastric juice ABHD11-AS1 as a marker, the positive detection rate of early gastric cancer patients was reached to 71.4 %. Thanks to the special origin of gastric juice, these results indicate that gastric juice ABHD11-AS1 may be a potential biomarker in the screening of gastric cancer.

Syndrome wind-cold-dampness in traditional Chinese medicine (TCM) can be referred to diseases of osteoarthritis, rheumatoid arthritis, cervical spondylitis, and spondylitis and so on. Over thousand years TCM clinical explorations, this syndrome can be treated with a group of herbal medicines in TCM. However, their molecular level biological functions are still not clear. In this study, a set of algorithms e.g., hypergeometric distribution, Jaccard index, and recursive programming for hierarchy network were deployed in the context gene/protein functional similarity and enrichment analysis. The analysis was mainly focused on the associated herbal medicines, their chemical compounds, target proteins' similarity, and gene ontology terms' hierarchical network overrepresentation through the enrichment analysis of hypergeometric distribution with P-value set to 0.05. As a result, these associated herbs share low level (<0.5) of therapeutic similarities on target proteins, yet with common biological functions for TCM syndrome wind-cold-dampness e.g., signal transduction, transportation, chemical homeostatic and metabolic process.
