With the continuous expansion of China's power grid scale and the increasing number of power users year by year, it is necessary to ensure the normal power supply of users. When a power failure occurs, it is particularly critical whether the emergency repair task can be completed quickly and scientifically. In this paper, an intelligent repair model of "unmanned" distribution network based on deep reinforcement learning is proposed, which adopts speech recognition technology and deep reinforcement learning algorithm to achieve the "unmanned" of the whole system. Users can transmit the emergency repair information to the voice recognition module of the power supply emergency repair center by voice, SMS and IMS, and the module will get the emergency repair position and the amount of emergency repair tasks. Then, the resource allocation module is used to learn the emergency repair resource allocation strategy online, and the intelligent control of emergency repair in distribution network is realized. To verify the proposed algorithm, it is compared with two typical allocation strategies under the same settings. The results of the experiments demonstrate that the method based on deep reinforcement learning performs better in terms of emergency repair delay and intelligent emergency repair of the power supply in distribution networks.

Steroidal saponins from the roots of Polygonatum odoratum and their inhibitory effects against pancreatic lipase

Twenty steroidal saponins were isolated from the water extract of fresh fibrous roots of Polygonatum odoratum (Mill.) Druce, of which seven were new compounds (1-7) along with thirteen known compounds (8-20). Their chemical structures were elucidated by extensive spectroscopic and chemical techniques. All isolated compounds were evaluated for porcine pancreatic lipase inhibitory activity. Compounds 9 and 10 showed inhibitory effects with IC50 values of 201.90 +/- 0.036 mu M and 151.20 +/- 0.058 mu M (positive control orlistat;2.82 +/- 0.060 mu M).

Hemostatic Interventions and All-Cause Mortality in Hemodynamically Unstable Pelvic Fractures: A Systematic Review and Meta-Analysis

Objective. To conduct a systematic review and meta-analysis of the all-cause mortality associated with the most commonly used hemostatic treatments in patients with hemodynamically unstable pelvic fractures. Methods. Up to April 30, 2023, we searched PubMed, Embase, Web of Science, and Cochrane, including the references to qualified papers. A meta-analysis was performed on studies that reported odds ratios (ORs) or the number of events needed to calculate them. The PROSPERO registration number was CRD42023421137. Results. Of the 3452 titles identified in our original search, 29 met our criteria. Extraperitoneal packing (EPP) (OR = 0.626 and 95% CI = 0.413-0.949), external fixation (EF) (OR = 0.649 and 95% CI = 0.518-0.814), and arterial embolism (AE) (OR = 0.459 and 95% CI = 0.291-0.724) were associated with decreased mortality. Resuscitative endovascular balloon occlusion of the aorta (REBOA) (OR = 2.824 and 95% CI = 1.594-5.005) was associated with increased mortality. A random effect model meta-analysis of eight articles showed no difference in mortality between patients with AE and patients with EPP for the initial treatments for controlling blood loss (OR = 0.910 and 95% CI = 0.623-1.328). Conclusion. This meta-analysis collectively suggested EF, AE, or EPP as life-saving procedures for patients with hemodynamically unstable pelvic fractures.

Hypothermia blacks β-catenin degradation after focal ischemia in rats

Dephosphorylated and activated glycogen synthase kinase (GSK) 3 beta hyperphosphorylates beta-catenin, leading to its ubiquitin-proteosome-mediated degradation. beta-catenin-knockdown increases while beta-catenin overexpression prevents neuronal death in vitro;in addition, protein levels of beta-catenin are reduced in the brain of Alzheimer's patients. However, whether beta-catenin degradation is involved in stroke-induced brain injury is unknown. Here we studied activities of GSK 3 and beta-catenin, and the protective effect of moderate hypothermia (30 degrees C) on these activities after focal ischemia in rats. The results of Western blot showed that GSK 3 beta was dephosphorylated at 5 and 24 h after stroke in the normothermic (37 degrees C) brain;hypothermia augmented GSK 3 beta dephosphorylation. Because hypothermia. reduces infarction, these results contradict with previous studies showing that GSK 3 dephosphorylation worsens neuronal death. Nevertheless, hypothermia blocked degradation of total GSK 3 beta protein. Corresponding to GSK 3 activity in normothermic rats, beta-catenin phosphorylation transiently increased at 5 h in both the ischemic penumbra and core, and the total protein level of-catenin degraded after normothermic stroke. Hypothermia did not inhibit beta-catenin phosphorylation, but it blocked beta-catenin degradation in the ischemic penumbra. In conclusion, moderate hypothermia can stabilize beta-catenin, which may contribute to the protective effect of moderate hypothermia. (c) 2008 Elsevier B.V. All rights reserved.

The Akt signaling pathway contributes to postconditioning's protection against stroke;the protection is associated with the MAPK and PKC pathways

We previously reported that ischemic postconditioning with a series of mechanical interruptions of reperfusion reduced infarct volume 2 days after focal ischemia in rats. Here, we extend this data by examining long-term protection and exploring underlying mechanisms involving the Akt, mitogen-activated protein kinase (MAPK) and protein kinase C (PKC) signaling pathways. Post-conditioning reduced infarct and improved behavioral function assessed 30 days after stroke. Additionally, postconditioning increased levels of phosphorylated Akt (Ser473) as measured by western blot and Akt activity as measured by an in vitro kinase assay. Inhibiting Akt activity by a phosphoinositide 3-kinase inhibitor, LY294002, enlarged infarct in postconditioned rats. Postconditioning did not affect protein levels of phosphorylated-phosphatase and tensin homologue deleted on chromosome 10 or -phosphoinositide-dependent protein kinase-1 (molecules upstream of Akt) but did inhibit an increase in phosphorylated-glycogen synthase kinase 3 beta, an Akt effector. In addition, postconditioning blocked beta-catenin phosphorylation subsequent to glycogen synthase kinase, but had no effect on total or non-phosphorylated active beta-catenin protein levels. Furthermore, postconditioning inhibited increases in the amount of phosphorylated-c-Jun N-terminal kinase and extracellular signal-regulated kinase 1/2 in the MAPK pathway. Finally, postconditioning blocked death-promoting delta PKC cleavage and attenuated reduction in phosphorylation of survival-promoting epsilon PKC. In conclusion, our data suggest that postconditioning provides long-term protection against stroke in rats. Additionally, we found that Akt activity contributes to postconditioning's protection;furthermore, increases in epsilon PKC activity, a survival-promoting pathway, and reductions in MAPK and delta PKC activity;two putative death-promoting pathways correlate with postconditioning's protection.

A novel Povarov-type reaction for straightforward synthesis of novel spiro bi-tetrahydroquinolines with readily available 1,2-DHQs (1,2-dihydroquinolines) and aromatic imines was developed. The reaction could be selectively stopped at the first stage under a Bronsted acid catalyst to afford the corresponding functionalized 1,2-DHQs conveniently.

Steroidal saponins from Allium chinense and their inhibitory effects on pancreatic lipase

Allium chinense *** is an important medicinal and edible homologous plant and indigenous to China and grown in other countries. Phytochemical studies have shown that it is rich in biologically active natural products. Herein, five new and twelve known steroidal saponins were isolated from the bulbs of Allium chinense ***. These compounds were structurally determined by analysis of their NMR, IR and HR-ESI-MS data. All isolates were evaluated for inhibition of porcine pancreatic lipase activity. Compounds 4, 7 and 10 exhibited potent inhibition of porcine pancreatic lipase. The relationship between structure and activity of these compounds is also investigated in this paper.

Surface interaction mechanisms of air bubbles, asphaltenes and oil drops in aqueous solutions with implications for interfacial engineering processes

Hypothesis: Surface interactions of bubbles and oil with interface-active species like asphaltenes influence many interfacial phenomena in various engineering processes. It holds both fundamental and practical significance to quantitatively characterize these ***: The surface forces of air bubbles, asphaltenes and asphaltenes-toluene droplets in various aqueous solutions have been quantified using an integrated thin film drainage apparatus and an atomic force microscope coupled with bubble probe. The effects of asphaltenes concentration, pH, salinity, Ca2+ ions and surfactants have been ***: Hydrophobic interaction drives attachment of bubbles and asphaltenes surfaces or oil droplets under high salinity condition. Increasing asphaltenes concentration in oil droplets enhances their hydrophobic attraction with bubbles due to strengthened asphaltenes adsorption and aggregation at oil-water interface. Increasing pH weakens the hydrophobic interaction as oil surfaces become more negatively charged and less hydrophobic. Under low salinity condition, strong electrical double layer and van der Waals repulsion inhibits the bubble-oil droplet contact. Introducing Ca2+ ions and surfactants leads to strong steric repulsion, preventing bubble-oil contact. This research has advanced our mechanistic understanding of how bubbles and oil droplets interact in aqueous systems and offers useful insights to modulate such interactions in oil production, water treatment and other interfacial processes.

GhEXL3 participates in brassinosteroids regulation of fiber elongation in Gossypium hirsutum

Cotton fiber (Gossypium hirsutum) serves as an ideal model for investigating the molecular mechanisms of plant cell elongation at the single-cell level. Brassinosteroids (BRs) play a crucial role in regulating plant growth and development. However, the mechanism by which BR influences cotton fiber elongation remains incompletely understood. In this study, we identified EXORDIUM-like (GhEXL3) through transcriptome analysis of fibers from BR-deficient cotton mutant pagoda 1 (pag1) and BRI1-EMS-SUPPRESSOR 1 (GhBES1.4, encoding a central transcription factor of BR signaling) overexpression cotton lines. Knockout of GhEXL3 using CRISPR/Cas9 was found to impede cotton fiber elongation, while its overexpression promoted fiber elongation, suggesting a positive regulatory function for GhEXL3 in fiber elongation. Furthermore, in vitro ovule culture experiments revealed that the overexpression of GhEXL3 partially counteracted the inhibitory effects of brassinazole (BRZ) on cotton fiber elongation, providing additional evidence of GhEXL3 involvement in BR signaling pathways. Moreover, our findings demonstrate that GhBES1.4 directly binds to the E-box (CACGTG) motif in the GhEXL3 promoter region and enhances its transcription. RNA-seq analysis revealed that overexpression of GhEXL3 upregulated the expression of EXPs, XTHs, and other genes associated with fiber cell elongation. Overall, our study contributes to understanding the mechanism by which BR regulates the elongation of cotton fibers through the direct modulation of GhEXL3 expression by GhBES1.4.

Optimising maize threshing process with temporal proximity soft actor-critic deep reinforcement learning algorithm

Maize threshing is a crucial process in grain production, and optimising it is essential for reducing post-harvest losses. This study proposes a model-based temporal proximity soft actor-critic (TP-SAC) algorithm to optimise the maize threshing process in the threshing drum. The proposed approach employs an LSTM model as a real-time predictor of threshing quality, achieving an R2 of 97.17% and 98.43% for damage and unthreshed rates on the validation set. In actual threshing experiments, the LSTM model demonstrates an average error of 5.45% and 3.83% for damage and unthreshed rates. The LSTM model is integrated with the TP-SAC algorithm, acting as the environment with which the TP-SAC interacts, enabling efficient training with limited real-world data. The TPSAC algorithm addresses the temporal correlation in the threshing process by incorporating temporal proximity sampling into the SAC algorithm's experience replay mechanism. TP-SAC outperforms the standard SAC algorithm in the simulated environment, demonstrating better sample efficiency and faster convergence. When deployed in actual threshing operations, the TP-SAC algorithm reduces the damage rate by an average of 0.91% across different feed rates compared to constant control. The proposed TP-SAC algorithm offers a novel and practical approach to optimising the maize threshing process, enhancing threshing quality.
