Folliculin (FLCN) is the tumor suppressor associated with Birt-Hogg-Dube (BHD) syndrome that predisposes patients to incident of hamartomas and cysts in multiple organs. Its inactivation causes deregulation in the mammalian target of rapamycin complex 1 (mTORC1) signaling pathway. However, the underlying mechanism is poorly defined. In this study, we show that FLCN is a ciliary protein that functions through primary cilia to regulate mTORC1. In response to flow stress, FLCN associates with LKB1 and recruits the kinase to primary cilia for activation of AMPK resided at basal bodies, which causes mTORC1 down-regulation. In cells depleted of FLCN, LKB1 fails to accumulate in primary cilia and AMPK at the basal bodies remains inactive, thus nullifying the inhibitory effect of flow stress on mTORC1 activity. Our results demonstrate that FLCN is part of a flow sensory mechanism that regulates mTORC1 through primary cilia.

The rapid postoperative recurrence and short survival time of glioblastoma (GBM) patients necessitate immediate and effective postoperative treatment. Herein, an immediate and mild postoperative local treatment strategy is developed that regulates the postoperative microenvironment and delays GBM recurrence. Briefly, an injectable hydrogel system (imGEL) loaded with Zn(II)(2)-AMD3100 (AMD-Zn) and CpG oligonucleotide nanoparticles (CpG NPs) is injected into the operation cavity, with long-term function to block the recruitment of microglia/ macrophages and activate cytotoxic T cells. The finding indicated that the imGEL can regulate the immune microenvironment, inhibit GBM recurrence, and gain valuable time for subsequent adjuvant clinical chemotherapy.

The fruits of Citrus are popular all over the world. It was reported that the extracts of fruits possessed antiallergic activities. In China, Citri Reticulatae Pericarpium (CRP) is a production of peels. In this study, flavonoids were extracted by solvent method, and enriched by polyamide. Then antiallergic activity of flavonoids was investigated in vivo and in vitro. Under optimal parameters, most abundant compounds of extracts were hesperidin and narirutin, their contents were 232.0325 and 98.7946 mg/g, respectively. In vitro, RBL-2H3 cell line was used to investigate the antiallergic activity. As the results shown, -hexosaminidase release of antigen-induced degranulation was decreased by flavonoids. An ovalbumin-induced allergic asthmatic model was used to test the activity of the flavonoids in vivo. After treated with different concentrations of flavonoids, the levels of IgE, IL-4, and IL-13 decreased significantly. These findings provide evidence that flavonoids extracted from CRP have potential using in antiallergic therapeutic area. Practical applicationsOranges are popular all over the world as fruit, besides a little percent of peels are applied as traditional medicine in China, the others are thrown away as rubbish. To explore the new way to extend the application of peels, we extracted the total flavonoids from Citri Reticulatae Pericarpium (CRP) and evaluated the antiallergic activity of it. Fortunately, the total flavonoids have antiallergic activity in vitro and vivo. The findings provide evidence that extracts from CRP have potential using in antiallergic therapeutic area. It can also afford a theoretical basis for the new application of orange peel.
