Over the past three decades, enzymatic polymerization has dramatically developed and gradually broadened as a creative methodology in the construction of polymeric materials with tailor-made structures and properties. Compared with transition metal catalyst polymerizations, enzymatic polymerization is more attractive in the biomedicine field due to the metal-free residue, good biocompatibility, and few by-products. Meanwhile, enzymatic polymerization has far more activity towards macrolides. In this review, the synthesis of lipase-catalyzed polymer materials is systematically summarized, focusing on the synthesis of the complex and well-defined polymers. The enzymatic polyester synthesis was then discussed concerning the different reaction types, including ring-opening polymerization, polycondensation, a combination of ring-opening polymerization with polycondensation, and chemoenzymatic polymerization. Besides, exploration of novel biocatalysts and reaction media was also described, with particular emphasis on the enzymes obtainedviaimmobilization or protein engineering strategies, green solvents, and reactors. Finally, recent developments in catalytic kinetics and mechanistic studies through the use of spectroscopy, mathematics, and computer techniques have been introduced. Besides, we addressed the remaining central issues in enzymatic polymerization and discussed current studies aimed at providing answers.

Context: It is an efficient strategy to apply inhibition of mast cell degranulation for evaluating anti-allergic effects of compounds. Previous works confirmed that narirutin had anti-allergic activity in OVA induced allergic asthma murine model. However, the mechanism is not clear. Objective: Here, inhibitory mechanism of narirutin on RBL-2H3 cells degranulation was investigated. Materials and methods: Cell viability was analyzed by CCK-8 kits, cell degranulation was analyzed by EliSA methods, morphology and ultrastructure of cells was observed by atomic force microscopy, intracellular Ca (2+) concentration was measured by fluorescence microscopre, mRNA expression were measured by PCR, and signaling pathways were measured by WB. Results: The results showed that narirutin have no direct effects on mRNA expression of Fc epsilon RI subunit. However, it inhibited Ca2+ influx by suppressing the phosphorylation of Syk, LAT and PLC gamma 1 signaling pathway transduction. Subsequently, the inhibition of Ca2+ influx directly leads to NF-kappa B signaling pathway transduction decreased. Narirutin can also suppress the phosphorylation of MAPK signaling pathways by decreasing the expression of P-p38, P-ERK and P-JNK, inhibit the synergistic effect for Ca2+ influx, and then reduce the release of IL-4, TNF-alpha, histamine and beta-HEX. Conclusion: Our study suggested that the inhibitory mechanism of narirutin on RBL-2H3 cells degranulation could be related to regulate MAPK, NF-kappa B and Tyrosine kinase signaling pathway.

Ginseng(Panax ginseng C. A. Mey.) is a traditional medicinal herb in Asia. Studies have shown that ginsenosides significantly affect immnnoregulation and rare ginsenosides have anti-allergic effects. In this research, a high temperature and high pressure method was utilized to increase the contents of rare ginsenosides in the ginseng extract(GE). The anti-allergic effects of this extract were investigated in vivo. Water was used as the extraction solvent in extracting the rare ginsenosides via the high temperature and high pressure method. Extraction time and temperature were investigated in order to increase the contents of rare ginsenosides. Rare ginsenosides were qualitatively analyzed by HPLC-ESI-MS and quantitatively analyzed by HPLC-UV. Anti-allergic effects of the extracts were assessed using the ovalbumin(OVA)-induced allergic asthma model in vivo. An extraction temperature of 145 ℃ and extraction time of 2.0 h were chosen as the optimal conditions. Compared with traditional method, the contents of total rare ginsenosides extracted were considerably higher using the new method, that is, 14.74 times that extracted by the traditional method. In our in vivo experiments, treatment with high concentration GE may have anti-allergic erects in decreasing the total amount of IgE in serum and IL-4 in bronchoalveolar lavage fluid(BALF), aud in improving the ratio of CD4^+ to CD8^+ T cells. The high temperature and high pressure method was an effective method to obtain GE containing more rare ginsenosides, which maybe become anti-allergic agents.

A series of oxidized products have been systematically semisynthesized from 20(S)-ginsenoside Rg(3), Rh-2, 20(S)-protopanaxadiol (PPD) and their 20(R)-epimers and the majority of these products were evaluated for their cytotoxic activity against HeLa cells and HepG2 cells by MIT assay for the first time. Twenty-two products were obtained and elucidated based on comprehensive H-1 NMR, C-13 NMR, two-dimensional (2D) NMR, and mass spectral data and the results reported in previous literature. All the four ocotillol type saponins (203,24R(delta 86, delta 85);205,24S(delta 87, delta 88);20R,24R(delta 86, delta 86);20R,245(delta 86, delta 87) were obtained. In addition, eight compounds (3, 8, 9, 10, 15, 16, 19 and 22) with the cyclized side chain were firstly identified. Most of the tested compounds possessed cytotoxicity to a certain degree against the two types of cells which implied these oxidized products could play a certain role on anti-cancer functions of the raw materials in vivo. Meanwhile, the results proved that the configurations at C-20 or C-24 and the number of glycosyl at C-3 have important influence on the cytotoxicity. The products 1, 2,11-17, 20 and 22 should possess great activities and deserved further investigation as potential cytotoxic agents. (C) 2015 Elsevier Inc. All rights reserved.
