Structural brain characteristics and gene co-expression analysis: A study with outcome label from normal cognition to mild cognitive impairment

Background: Longitudinal studies reported that some elderly people with normal cognition (NC) converted to mild cognitive impairment (MCI), and some remained normal state (NC_S). The underlying factor for this difference conversion of NC is worthy of exploration Methods: Eighty-three NC participants were tracked for eight years. Thirty participants transitioned from NC to MCI (NC_MCI). The remaining 53 participants retained an NC_S. The structural brain features and genetic expression of the 83 NC participants were obtained. We applied weighted gene co-expression network analysis (WGCNA) to inquire into the co-expression network of those. Mediator effect analysis of regulatory roles was conducted to inquire into the associations between brain measures, expression values, and clinical scores. Results: The main results are: 1) 20 brain features and 740 gene expression had significant differences between the two groups, 2) one module including 187 genes had the most correlation with cortical thickness of left superior temporal sulcus (***), 3) NFKBIA and RARA genes were the top two genes that made the greatest contribution to *** thickness, and 4) mediating effect was found between the *** thickness, the NFKBIA and RARA expression levels, and clinical scores. Conclusion: Our results provide a theoretical foundation based on gene expression and brain imaging for the factors of NC with different outcomes.

Reference gene selection for qRT-PCR normalization of gene expression analysis in Melaleuca bracteata F. Muell. under abiotic stresses and hormonal stimuli

Melaleuca bracteata F. Muell. is among the most significant aromatic plants belonging to the Myrtaceae family, and its essential oil has various biological activities. However, the key enzymes involved in essential oil biosynthesis and their expression patterns are not clearly understood. Appropriate reference genes are required for quantitative real-time polymerase chain reaction (qRT-PCR), sensitive for gene expression studies, to obtain reliable results. In this study, 14 candidate genes, namely ACT, 18S, & alpha;-TUB, fl-TUB, GAPDH, EF1-& alpha;, UBQ, UBC, CYP, EIF-4A, DNAJ, PP2A, TIP41, and UBX, were selected from the M. bracteata transcriptome datasets for analysis under abiotic stresses and hormonal stimuli. The stability of these genes was analyzed using three widely used software programs (geNorm, NormFinder, and BestKeeper). Their results were integrated into a compre-hensive ranking by geometric mean. According to the study results, TIP41 and CYP were the most stable genes for abiotic stresses, while TIP41 and UBQ were the most stable genes for hormonal stimulation. Further, TIP41 and CYP were the most stable reference genes across all tested samples, whereas ACT was the least stable. Based on the expression of two target genes (COMT1 and COMT2, which are involved in methyleugenol biosynthesis), the suitability of the selected reference genes for qRT-PCR normalization was further verified. The study results could offer a foundation for gene expression analysis and functional studies in M. bracteata and other Melaleuca species.

Improving the self-healing performance of the water-borne epoxy anticorrosion coating in 3.5 wt% NaCl solution using acid and alkaline dual sensitive organic microspheres

Acid-alkaline double sensitive Eudragit microspheres embedded with corrosion inhibitor 8-hydroxyquinoline (8HQ@Eu) were prepared by a low-cost one-step method, and a novel coating was designed to improve the self-healing performance of the water-borne epoxy resin in 3.5 wt% NaCl solution by using the 8HQ@Eu as smart anti-corrosive carrier. The results of electrochemical impedance spectroscopy (EIS) showed the low-frequency impedance modulus (|Z|(0.01Hz)) of the scratched coating was about 10 times higher than that of the pure epoxy coating in the presence of 8HQ@Eu microspheres after 30 d of immersion, and the oxide film resistance and charge transfer resistance of the 8HQ@Eu sample increased about 10 times and 8 times respectively due to the covering of corrosion inhibitor protective film. While the results of scanning vibrating electrode technique (SVET) indicated the corrosion current of the 8HQ@Eu sample in the scratched area decreased to more than an order of magnitude lower than that of pure epoxy coating within 24 h. Salt spray test indicated that the substrate corrosion and coating delamination near the scratched area were significantly inhibited. XPS analysis directly confirmed that the corrosion inhibitor was delivered to the corroded sites.

Methyleugenol is widely found in essential oils from aromatic plants, attracting insects for pollination and possessing antimicrobial and antioxidant features. Methyleugenol content in the essential oil from Melaleuca bracteata leaves is 90.46 %, making it an excellent material for assessing methyleugenol's biosynthetic pathway. Eugenol synthase (EGS) and eugenol-o-methyltransferase (EOMT) represent key genes in methyleugenol's biosynthetic pathway but their respective functions are unclear. In this study, overexpression and virus-induced gene silencing (VIGS) techniques were applied to investigate the major regulatory gene among MbEGS1, MbEGS2, MbEGS3, and MbEGS4 in the biosynthesis pathway of methyleugenol. We also aimed to examine the coordination mechanism between EGS and EOMT in methyleugenol production by M. bracteata. The data revealed MbEGS4 transcription and methyleugenol content were most significantly increased after overexpression of the MbEGS1-4 gene in M. bracteata and Fragaria vesca. Conversely, MbEGS4 transcription and methyleugenol content were most significantly reduced after MbEGS1-4 gene silencing by VIGS in M. bracteata. These results underscore a major regulatory role for the MbEGS4 gene in the biosynthetic pathway of methyleugenol in M. bracteata. Remarkably, upon overexpression and silencing of MbEOMTs, the transcription levels of MbEGSs were notably upregulated, with MbEGS4 showing the highest increase. Collectively, these findings suggested the presence of a specific transcription factor bound to the promoters of MbEGS and MbEOMT, inducing their expression for the synergistic and efficient production of methyleugenol in M. bracteata.

Since the semiconductor technology has entered the age of 90 nm, the static power consumption of the chip has increased rapidly. For edge computing devices, the waste of power can not be ignored. This paper uses the relationship between power consumption and power supply voltage of FPGA and DSP, analyzes the characteristics of the traditional scheme, and designs a hardware circuit according to the different mechanisms of reducing static power consumption of FPGA and DSP According to the design of FPGA and DSP core power supply voltage compatibility, the system realizes the requirement of FPGA and DSP for power supply voltage amplitude and realizes dynamic control of power supply voltage. After verification and comparison, FPGA and DSP can realize the adjustment of performance power consumption and stable operation under the power system.

A one-pot photocatalytic triazole-based linkerology for PROTACs

Proteolysis-targeting chimeras (PROTACs) are a powerful approach for targeted protein degradation. One of the current bottlenecks for developing PROTACs is the lack of an operationally simple linkerology to rapidly construct PROTACs with various linkers. The classic convergent synthesis strategy by coupling pre-assembled linkers with two ligands stepwise commonly needs at least four steps to give the final target PROTACs, which results in low total yields with long reaction times (several days) and tedious operations. Here, we develop an efficient photocatalytic one-pot linkerology for the rapid coupling of analogs of PROTACs containing triazole-based linkers without any linker-pre-assembled procedure. The reaction was completed within 4 h with up to 95% yields at room temperature. Easily accessible cyclic ethers are directly used as linker precursors to furnish the one-pot fashion, including alkenyl, polyethylene glycol (PEG), ketone, and cyclohexane chains. The study provides a highly efficient, step-economic, operationally simple, and environmentally friendly one-pot linkerology for PROTAC drug discovery.

Expression of a plastid-localized sugar transporter in the suspensor is critical to embryogenesis

Plant growth and development rely on sugar transport between source and sink cells and between different organelles. The plastid-localized sugar transporter GLUCOSE-6-PHOSPHATE TRANSLOCATER1 (GPT1) is an essential gene in Arabidopsis (Arabidopsis thahana). Using a partially rescued gpt1 mutant and cell-specific RNAi suppression of GPT1, we demonstrated that GPT1 is essential to the function of the embryo suspensor and the development of the embryo. GPT1 showed a dynamic expression/accumulation pattern during embryogenesis. Inhibition of GPT1 accumulation via RNA1 using a suspensor-specific promoter resulted in embryos and seedlings with defects similar to auxin mutants. Loss of function of GPT1 in the suspensor also led to abnormal/ectopic cell division in the lower part of the suspensor, which gave rise to ectopic embryo, resulting in twin embryos in some seeds. Furthermore, loss of function of GPT1 resulted in vacuolar locali- zation of PIN-FORMED1 (PIN1) and altered DRS auxin activity. Proper localization of PIN1 on the plasma membrane is essential to polar auxin transport and distribution, a key determinant of pattern formation during embryogenesis. Our findings suggest that the function of GPT1 in the embryo suspensor is linked to sugar and/or hormone distribution between the embryo proper and the maternal tissues, and is important for maintenance of suspensor identity and function during embryogenesis.

Theoretical Analysis and Numerical Simulation of the Motion of RDX Deflagration-Driven Flyer Plate Based on Laser-Initiated Micro-Pyrotechnic Devices

Miniaturized laser-initiated pyrotechnic devices have great application prospects in aerospace and modern weapon systems due to their excellent energy output performance and reliability. In order to develop a low-energy insensitive laser detonation technology based on a two-stage charge structure, it is important to deeply analyze the motion law of a titanium flyer plate driven by the deflagration of the first-stage charge (RDX). The effects of the charge mass of RDX, flyer plate mass, and barrel length on the motion law of flyer plates were studied through a numerical simulation method based on the Powder Burn deflagration model. The consistency between the numerical simulation and the experimental results was analyzed using the paired t confidence interval estimation method. The results show that the Powder Burn deflagration model can effectively describe the motion process of the RDX deflagration-driven flyer plate with a 90% confidence level, and its velocity error is =6.7%. The speed of the flyer plate is proportional to the mass of the RDX charge, inversely proportional to the mass of the flyer plate, and exponentially related to its moving distance. As the moving distance of the flyer plate increases, the RDX deflagration products and air in front of the flyer plate are compressed, which inhibits the motion of the flyer plate. In the optimum state (the mass of the RDX charge is 60 mg, the mass of the flyer is 85 mg, and the length of the barrel is 3 mm), the speed of the titanium flyer reaches 583 m/s, and the peak pressure of the RDX deflagration reaches 2182 MPa. This work will provide a theoretical basis for the refined design of a new generation of miniaturized high-performance laser-initiated pyrotechnic devices.

Screening and Expression Analysis of POD Gene in POD-H2O2 Pathway on Bud Dormancy of Pear (Pyrus pyrifolia)

Bud endodormancy represents a pivotal and intricate biological process influenced by both genetic and epigenetic factors, the exact mechanism of which remains elusive. Hydrogen peroxide (H2O2) functions as a signalling molecule in the regulation of dormancy, with peroxidase (POD) playing a crucial role in governing H2O2 levels. Our prior transcriptomic and metabolomic investigations into diverse pear dormancy phases posited that POD predominantly oversees pear bud dormancy. In this study, we utilised qRT-PCR to screen the most significantly expressed gene, Pyrus pyrifolia POD4-like (PpPOD4-like), from seven POD genes. Subsequently, H2O2 test kits, overexpression methods, and subcellular localisation techniques were employed to assess changes in H2O2 content, POD activity, PpPOD4-like expression, and its cellular positioning during pear bud dormancy. Subcellular localisation experiments revealed that PpPOD4-like is situated on the cell membranes. Notably, H2O2 content exhibited a rapid increase during endodormancy and decreased swiftly after ecodormancy. The fluctuation pattern of POD activity aligned with that of H2O2 content. Additionally, PpPOD4-like expression was markedly upregulated, displaying an overall upward trajectory. Our findings indicate that PpPOD4-like modulates H2O2 levels by regulating POD activity, thereby actively participating in the intricate regulation of pear dormancy processes.
