Prognostic Health Management (PHM) is designed to assess and monitor the health status of systems, anticipate the onset of potential failure, and prevent unplanned downtime. In recent decades, collecting massive amounts of real-time sensor data enabled condition monitoring (CM) and consequently, detection of abnormalities to support maintenance decision-making. Additionally, the utilization of PHM techniques can support energy sustainability efforts by optimizing energy usage and identifying opportunities for energy-saving measures. Escalators are efficient machines for transporting people and goods, and measuring energy consumption in time can facilitate PHM of escalators. Fixed loss energy, or no-load energy, of escalators denotes the energy consumption by an unloaded escalator. Fixed loss energy varies over time indicating varying operating conditions. In this paper, we propose to use escalators’ fixed loss energy for PHM. We propose an approach to compute daily fixed loss energy based on energy consumption sensor data. The proposed approach is validated using a set of experimental data. The advantages and disadvantages of each approach are also presented, and recommendations are given. Finally, to illustrate PHM, we set up an EWMA chart for monitoring the fixed loss over time and demonstrate the potential in reducing energy costs associated with escalator operation. © 2023, CC BY-NC-SA.

Phenotypic plasticity and genetic variation of cotton yield and its related traits under water-limited conditions

Global warming is limiting availability of water resources in arid and semi-arid regions,and so understanding water use efficiency(WUE)is increasingly important for agricultural production in those *** China is the largest cotton producing area,the problem of balancing WUE and efficient cotton production is a major *** this study,we used a natural population of 517 Upland cotton accessions to conduct a water-controlled trial in south and north of Xinjiang over two years.A total of 18 traits including agronomic traits,fiber yield indices and fiber quality indices,were investigated for broad-sense heritability and coefficient of *** water limitation was found to promote the establishment of favorable agronomic traits in cotton,associated with an increased cotton yield of 8.46%in Xinjiang,at the expense of a certain degree of fiber quality,such as decreased fiber length and an over-higher micronaire *** detected 33 QTL related to response to water limitation using a drought resistance coefficient(DRC),and 6 QTL were found using a comprehensive indicator of CIDT(comprehensive index of drought tolerance)at the genetic level by integrating resequencing *** novel QTL-hotspots including six differentially expressed genes(DEGs)were further identified related to the drought response of *** findings not only suggested a new approach to irrigation of cotton fields in Xinjiang,but also provided abundant genetic evidence for genetic breeders to study drought improvement of crops.

BACKGROUND: The risk factors of hypertensive disorders in pregnancy (HDP) could be summarized into three categories: clinical epidemiological factors, hemodynamic factors and biochemical factors. OBJECTIVE: To establish models for early prediction and intervention of HDP. METHODS: This study used the three types of risk factors and support vector machine (SVM) to establish prediction models of HDP at different gestational weeks. RESULTS: The average accuracy of the model was gradually increased when the pregnancy progressed, especially in the late pregnancy 28-34 weeks and >= 35 weeks, it reached more than 92%. CONCLUSION: Multi-risk factors combined with dynamic gestational weeks' prediction of HDP based on machine learning was superior to static and single-class conventional prediction methods. Multiple continuous tests could be performed from early pregnancy to late pregnancy.

Comparison and development of a metagenomic next-generation sequencing protocol for combined detection of DNA and RNA pathogens in cerebrospinal fluid

Background: The purpose of this study was to evaluate different pretreatment, extraction, amplification, and library generation methods for metagenomic next-generation sequencing (mNGS) of cerebrospinal fluid (CSF) and to develop an efficient procedure for the simultaneous detection of DNA and RNA pathogens. Methods: We generated thirteen mock CSF samples with four representative pathogens of encephalitis. Each sample was subjected to ten different methods by varying sample pretreatment/nucleic acid extraction (microbial DNA, total DNA, total NA, total RNA, Whole Transcriptome Amplification (WTA)) and library generation (Illumina or NEB). Negative extraction controls (NECs) were used for each method variation. Results: We found that the quality of mNGS sequencing reads was higher from the NEB kit for library generation. Microbial DNA and total RNA increased microbial deposition by depleting the host DNA. Methods total NA and total RNA can detect gram-positive, gram-negative, RNA and DNA pathogens. We applied mNGS, including total NA and NEB library generation, to CSF samples from five patients diagnosed with infectious encephalitis and correctly determined all pathogens identified in clinical etiological tests. Conclusions: Our findings suggested that total nucleic acid extraction combined with NEB library generation is the most effective mNGS procedure in CSF pathogen detection. The optimization of positive criteria and databases can improve the specificity and sensitivity of mNGS diagnosis.

Medical Data Mining Course Development in Postgraduate Medical Education: Web-Based Survey and Case Study

Background: Medical postgraduates' demand for data capabilities is growing, as biomedical research becomes more data driven, integrative, and computational. In the context of the application of big data in health and medicine, the integration of data mining skills into postgraduate medical education becomes important. Objective: This study aimed to demonstrate the design and implementation of a medical data mining course for medical postgraduates with diverse backgrounds in a medical school. Methods: We developed a medical data mining course called "Practical Techniques of Medical Data Mining" for postgraduate medical education and taught the course online at Peking Union Medical College (PUMC). To identify the background knowledge, programming skills, and expectations of targeted learners, we conducted a web-based questionnaire survey. After determining the instructional methods to be used in the course, three technical platforms-Rain Classroom, Tencent Meeting, and WeChat-were chosen for online teaching. A medical data mining platform called Medical Data Mining - R Programming Hub (MedHub) was developed for self-learning, which could support the development and comprehensive testing of data mining algorithms. Finally, we carried out a postcourse survey and a case study to demonstrate that our online course could accommodate a diverse group of medical students with a wide range of academic backgrounds and programming experience. Results: In total, 200 postgraduates from 30 disciplines participated in the precourse survey. Based on the analysis of students' characteristics and expectations, we designed an optimized course structured into nine logical teaching units (one 4-hour unit per week for 9 weeks). The course covered basic knowledge of R programming, machine learning models, clinical data mining, and omics data mining, among other topics, as well as diversified health care analysis scenarios. Effect of a closed-loop medication order executive system on safe medication administration at a tertiary hospital: a quasi-experimental study

Background: Closed-loop electronic medication management systems are effective measures for preventing medication errors (MEs). However, there is limited evidence supporting this, and few studies have evaluated the long-term effects of these systems on safe medication. Objective: To evaluate the long-term effects of implementing a closed-loop medication order executive system on the safe clinical use of medications. Design: A quasi-experimental design. Method: Data from 2017 to 2023 were extracted and retrospectively analyzed. The primary outcome indicator was the ME rate. Secondary outcome indicators were the accuracy of order verification and patient identification and the implementation rate of fresh medicine dispensing. The autoregressive integrated moving average (ARIMA) model in time-series analysis was used to evaluate the level and trend changes in ME rates using SPSS 25.0 before and after system implementation. Root cause analysis and descriptive statistics were used to assess changes in types, stages, and causes of ME rates. The independent samples t-test was used to analyze secondary outcomes. Results: Overall, 295 MEs were reported with a mean of 0.26 +/- 0.26 ME rates per month during 2017-2023. The ARIMA model showed a decrease in the average level of ME rates after system implementation, with no statistically significant decrease in the long term, and a significant drop in the ME rate in the short and medium term (p < 0.01). Nurses' administration accounted for more than 60% of errors post-implementation, and lack of communication was a prominent factor. The accuracy of order verification and patient identification and the implementation rate of fresh medicine dispensing all increased after implementation. Conclusion: Adopting a closed-loop executive system is beneficial for ensuring patient medication safety, but a single integrated system does not completely eliminate MEs. The model updating based on near infrared spectroscopy for the sex identification of silkworm pupae from different varieties by a semi-supervised learning with pre-labeling method

It is effective to accurately discriminate the sex of silkworm pupae with the same varieties based on near infrared spectroscopy. However, when the model is promoted to classify new varieties of silkworm pupae, the model's performance becomes worse, due to the cultivation environment and varieties changing. In the aims of improving the generalization ability and accuracy of the model, this paper proposed a model updating strategy based on semi-supervised learning. First, support vector machine identification model was built after the original spectra was pretreated by Savitzky-Golay convolution smoothing operation, which could effectively reduce spectra noise. Then, the support vector machine model gave the pre-labelings of unlabeled silkworm pupae in the updated set, which were divided into male samples and female samples. According to the correlation coefficients that calculated by Pearson correlation coefficient and Euclidean distance, a total of 8 reliable samples were selected from the male and female samples, respectively. The reliable samples were added to the original training set to update the original model. Finally, the updated model was used to test the test sets from the varieties of silkworm pupae that were the same with updated *** results showed the performance of the non-updated model for silkworm pupae from the three new varieties just reached 54.55%, 68.52%, 86.84%, respectively. The support vector machine model updated by using Pearson correlation coefficient improved the accuracy to 100%, 96.30%, 97.37%, and the model updated by Euclidean distance increased the identification accuracy of the three varieties that were not involved in the modeling to 100%, 75.93%, 92.10% respectively. The results showed that the performance of the model updated by Pearson correlation coefficient was better than Euclidean distance. Inflammatory resolution and vascular barrier restoration after retinal ischemia reperfusion injury

Background: Several retinal pathologies exhibit both inflammation and breakdown of the inner blood-retinal barrier (iBRB) resulting in vascular permeability, suggesting that treatments that trigger resolution of inflammation may also promote iBRB restoration. Methods: Using the mouse retinal ischemia-reperfusion (IR) injury model, we followed the time course of neurodegeneration, inflammation, and iBRB disruption and repair to examine the relationship between resolution of inflammation and iBRB restoration and to determine if minocycline, a tetracycline derivative shown to reverse microglial activation, can hasten these processes. Results: A 90-min ischemic insult followed by reperfusion in the retina induced cell apoptosis and inner retina thinning that progressed for approximately 2 weeks. IR increased vascular permeability within hours, which resolved between 3 and 4 weeks after injury. Increased vascular permeability coincided with alteration and loss of endothelial cell tight junction (TJ) protein content and disorganization of TJ protein complexes. Shunting of blood flow away from leaky vessels and dropout of leaky capillaries were eliminated as possible mechanisms for restoring the iBRB. Repletion of TJ protein contents occurred within 2 days after injury, long before restoration of the iBRB. In contrast, the eventual re-organization of TJ complexes at the cell border coincided with restoration of the barrier. A robust inflammatory response was evident a 1 day after IR and progressed to resolution over the 4-week time course. The inflammatory response included a rapid and transient infiltration of granulocytes and Ly6C(+) classical inflammatory monocytes, a slow accumulation of Ly6C(neg) monocyte/macrophages, and activation, proliferation, and mobilization of resident microglia. Extravasation of the majority of CD45(+) leukocytes occurred from the superficial plexus. The presence of monocyte/macrophages and increased numbers of microglia were sustained until

The response of temperature and pressure of hydrate reservoirs in the first gas hydrate production test in South China Sea

The first offshore natural gas hydrate production test of China in 2017 has proved the feasibility of hydrate exploitation from clayey-silt reservoirs, which possesses the highest reservoirs than other types of hydrate resources. However, owing to the absence of monitoring wells in this production test, the hydrate dissociation behavior cannot be analyzed through pressure and temperature changes of hydrate reservoirs. This paper focuses on the simulation study on the detailed response of the temperature and pore pressure of hydrate reservoirs of Well SHSC-4 during the gas production by depressurization. Meanwhile, it highlights the analysis of favorable areas for the formation of secondary hydrates and the influence of the secondary hydrates on pressure and temperature field of hydrate reservoirs. The simulation results indicate that in the first 60 days, the hydrate reservoirs feature a dissociation radius of about 5 m, and the gas production from hydrate dissociation accounts for about 85%. After 1 year, 2 years and 5 years of hydrate exploitation, the influence radius of low-pressure area (<10 MPa) is 15 m, 16 m and 17 m, respectively, suggesting that the hydrate reservoirs have higher gas production efficiency in the first year. Furthermore, the temperature and pressure of hydrate reservoirs are not favorable to the formation of secondary hydrates in the first 60 days. In long-term production, secondary hydrates are mainly formed at hydrate dissociation front. This can increase the pore pressure and further decrease the effective stress in the local areas of hydrate reservoirs, thus affecting mechanical stability of the local hydrate reservoirs.

In recent years, pregnant women in China generally face problems such as unbalanced and excessive nutrition, lack of proper exercise during pregnancy, which shows a significant increase in weight during pregnancy, leading to an increasing trend of perinatal complications. At present, there is less sports health management system for pregnant women throughout pregnancy. In view of this, based on the existing research in related fields, this study deeply explores the appropriate monitoring methods of pregnant women's sports in China. In this study, effective and convenient testing methods and evaluation criteria were proposed for pregnant women's sports and psychology. The research idea was a method based on the pulse wave to detect pregnant women's cardiac reserve capacity, and grading the Diastolic/Systolic value of pregnant women as a reference for grading individualized target heart rate range of moderate intensity exercise. The effective time and energy expenditure of pregnant women were assessed by monitoring the exercise process. The purpose is to help and guide pregnant women in the whole process of pregnancy self-movement management, and thus improve the quality of maternal health care services in China.
