Influences of Community Coalescence on the Assembly of Bacterial Communities of the Small-Scale Complex Aquatic System from the Perspective of Bacterial Transmission, Core Taxa, and Co-occurrence Patterns

Recirculating aquaculture and aquaponics are considered sustainable aquaculture models playing important roles in animal-derived protein supply. In these aquaculture systems, microorganisms are crucial for the system stability. The community coalescence by mixing substances and microorganisms from various microhabitats under hydraulic forces is important for shaping the bacterial communities in these small-scale complex systems. However, the influences of community coalescence on bacterial communities remain rarely revealed in these systems. In this study, aquaponics (APS) and recirculating aquaculture (RAS) systems were set up to explore the bacterial community coalescence across different microhabitats, including water, fish feces, biofilter biofilms, and plant rhizosphere environment. Our results showed that diversity and compositions varied across different microhabitats in both systems. However, bacterial transmissions across these microhabitats differed between systems. The core microbiome of the RAS and APS were formed under community coalescence with the highest contribution of bacterial taxa derived from the fish feces. Nevertheless, the plant rhizosphere bacterial community also contributed to the core microbiome of the APS. Furthermore, the core taxa showed a higher average degree than the other nodes in the bacterial community networks in all microhabitats except for the plant rhizosphere environment, implying the important roles of core taxa in maintaining these bacterial community networks. Our results provide new insights into the assembly of bacterial communities under community coalescence in the artificial aquatic ecosystems comprising complex microhabitats, which is vital for developing microbial solutions for regulating the microbial communities to improve system performance in the future.

In coastal engineering, predicting seabed erosion and deposition can help understand hydrodynamic changes of the coastal area. The present work aims to address the prevalent challenge that there is often a shortage of measured data in coastal areas. In this paper, a new seabed erosion and deposition formula with four forms is proposed and combined with 2D hydrodynamic harbor engineering simulation of Daishan Island situated in Zhejiang province, China. The simulation shows that (1) in the open channel, the proposed new formula considers the friction coefficient before and after trenching, thus providing a more accurate prediction result;(2) in the sheltered channels, the proposed new formula can roughly estimate the deposition intensity;(3) in the basin entrance, the proposed new formula provides an estimation better aligned with reality than existing formulae. Meanwhile, the recovery saturation coefficient is not a constant;it can be set at 0.31 in the obtained formulas for calculations during the back silting, and at 0.11 during scouring. Therefore, the proposed new formula provides an improved solution to prediction of seabed erosion and deposition and is applicable to Daishan coastal area in different situations.

In the traditional project cost management, due to the influence of many factors, its calculation accuracy is not high enough, which brings great difficulties to the construction project cost management. Using BIM technology to build a cost management model can provide very intuitive analysis results for cost managers, thus providing strong technical support for cost management of construction projects. Epigenetic dysregulation is prevalent in human cancers, affecting gene expression and metabolic patterns to meet the demands of malignant evolution and abnormal epigenetic processes, and resulting in a protumor immune microenvironment. Tumors require a steady supply of methionine for maintaining epigenetic flexibility, which is the only exogenous precursor of methyl donor S-adenosylmethionine for methylation, crucial for their resistance to therapies and survival in a nutrient-deficient microenvironment. Thus, tumor cells upregulate the Lat4 transporter to compete and deprive methionine in the microenvironment, sustaining their malignant phenotypes and also impairing immune cell functions. Addressing this methionine addiction is the key to overcoming drug resistance and improving immune response. Despite the challenge of lacking specific Lat4 inhibitors, an oxaliplatin prodrug crosslinked fluorinated polycation/anti-Lat4 small interfering RNA complex nanoregulator (AS-F-NP) has been designed and developed here. This nanoregulator restricted the greedy methionine uptake of tumor cells by knocking down Lat4, which in turn inhibited the malignant evolution of the tumor while restoring the viability and function of tumor-infiltrating immune cells.

股东的权利,从享有公司收益、参与重大决策,到提起诉讼要求损害赔偿,被广泛认为是一个经济体中有效配置资本的重要影响因素。康美药业案作为中国第一起实质性集体诉讼,其判决结果深刻影响了资本市场主体的行为。采用事件研究法,以2018—2022年中国A股上市公司为样本,检验了2021年11月12日康美药业证券虚假陈述集体诉讼案一审判决所带来的强化股东保护的公司治理效应。结果发现,盈余管理程度高、被交易所问询多以及存在关联交易的企业,在康美药业案一审判决前后的市场反应更差。此外,相较于其他重要事件日,一审判决对潜在违规企业的威慑更为明显,且威慑效应具有持续性,公司更倾向于守法经营,带来公司治理的优化。为保障中小股东的合法权益,建议政府进一步降低集体诉讼门槛和维权成本,为投资者提供便捷有效的诉讼维权渠道;上市公司增强财务信息质量和透明度,守法经营,依法披露,提升内部治理水平。

Information, migration, and the value of clean air

Using a variant of the Rosen-Roback model of inter-city migration that incorporates public access to air quality information, we demonstrate that information constraints create a wedge between revealed and true hedonic prices for pollution that depends upon individuals' perception biases. We empirically test our theoretical pre-dictions by leveraging the unexpected disclosure of PM2.5 data in China. We find that migration decisions become much more responsive to pollution and that the hedonic price of avoiding PM2.5 exposure nearly doubles - from 171 to 336 Chinese Yuan - in response to the information shock. Our results highlight the role of imperfect information in migration decisions and its impact on non-market valuation in countries where public access to information is restricted.

Cryptococcus is a pathogenic fungus that poses a threat to human health. Conventional detection methods have limited the rapid and accurate qualitative and quantitative analysis of Cryptococcus, affecting early diagnosis and treatment. In this study, we developed a Point-of-Care Testing (POCT) platform that integrates lateral flow immunoassay (LFIA) with smartphones, enabling both rapid qualitative and quantitative detection of Cryptococcus. The LFIA strip utilizes latex microspheres (LMs) as labeling probes, achieving a detection limit of 3000 CFU/mL and presenting higher sensitivity than the Colloidal Gold Nanoparticles Lateral Flow Immunoassay (AuNPs-LFIA) strip, and approximately eight times that of the AuNPs-LFIA strip. Additionally, it exhibiting no cross-reactivity with over 24 common pathogens and validated in clinical samples. For quantitative analysis, artificial intelligence algorithms were employed to convert smartphone-captured images into grayscale values. Eleven feature values were utilized as a dataset for machine learning to construct a linear regression model, with Mean Squared Error (MSE) and R2 reaching 0.45 and 0.91, respectively. Moreover, the recovery rates in the serum samples ranged from 90.0 % to 108 %, indicating a good practicability. This research presents a rapid diagnostic technology for Cryptococcus and lays the theoretical and technical groundwork for detecting other pathogens.

Solid electrochemiluminescence sensor by immobilization luminol in Zn-Co-ZIF CNFs for sensitive detection of procymidone in vegetables

A solid-state electrochemiluminescence (ECL) sensor was fabricated by immobilizing luminol, a classical luminescent reagent, on a Zn-Co-ZIF carbon fiber-modified electrode for the rapid and sensitive detection of procymidone (PCM) in vegetable samples. The sensor was created by sequentially modifying the glassy carbon electrode with Zn-Co-ZIF carbon fiber (Zn-Co-ZIF CNFs), Pt@Au NPs, and luminol. Zn-Co-ZIF CNFs, prepared through electrospinning and high-temperature pyrolysis, possessed a large specific surface area and porosity, making it suitable as carrier and electron transfer accelerator in the system. Pt@Au NPs demonstrated excellent catalytic activity, effectively enhancing the generation of active substances. The ECL signal was significantly amplified by the combination of Zn-Co-ZIF CNFs and Pt@Au NPs, which can subsequently be diminished by procymidone. The ECL intensity decreased proportionally with the addition of procymidone, displaying a linear relationship within the concentration range 1.0 x 10-13 to 1.0 x 10-6 mol L-1 (R2 = 0.993). The sensor exhibited a detection limit of 3.3 x 10-14 mol L-1 (S/N = 3) and demonstrated outstanding reproducibility and stability, making it well-suited for the detection of procymidone in vegetable samples.

Due to the limitations of current anti-HBV therapies, the HBV core (HBc or HBcAg) protein assembly modulators (CpAMs) are believed to be potential anti-HBV agents. Therefore, discovering safe and efficient CpAMs is of great value. In this study, we established a HiBiT-based high-throughput screening system targeting HBc and screened novel CpAMs from an in-house marine chemicals library. A novel lead compound 8a, a derivative of the marine natural product naamidine J, has been successfully screened for potential anti-HBV activity. Bioactivity-driven synthesis was then conducted, and the structure‒activity relationship was analyzed, resulting in the discovery of the most effective compound 11a (IC50 = 0.24 μmol/L). Furthermore, 11a was found to significantly inhibit HBV replication in multiple cell models and exhibit a synergistic effect with tenofovir disoproxil fumarate (TDF) and IFNa2 in vitro for anti-HBV activity. Treatment with 11a in a hydrodynamic-injection mouse model demonstrated significant anti-HBV activity without apparent hepatotoxicity. These findings suggest that the naamidine J derivative 11a could be used as the HBV core protein assembly modulator to develop safe and effective anti-HBV therapies.
