Genome-wide association screening and verification of potential genes associated with defoliation rate induced by defoliant in upland cotton

Defoliant sensitivity is an important trait of machine-picked cotton that directly affects the defoliation efficiency and the quality of machine-picked cotton fibers. Identification of the genetic components underlying defoliant sensitivity in upland cotton (Gossypium hirsutum L.) is highly important. In this study, the defoliation rate (DR) of 328 re-sequenced upland cotton accessions after 5 days of defoliant treatment was evaluated at two locations for two years in four environments;the results showed a great variation among the accessions, and the phenotype were normally distributed. A genome-wide association study (GWAS) was subsequently performed with resequencing data and defoliation rate data. As a result, two quantitative trait loci (QTL) related to defoliation rate were identified, qDS-A05 and qDS-A06, among which, qDS-A06 was identified in three environments. By combining linkage disequilibrium (LD) analysis, haplotype analysis, and expression analysis, a key candidate gene for DR was identified in qDS-A06, which was named GhDS-A06. This gene encodes a carboxylate clamp (CC)-tetratricopeptide repeat (TPR) protein, and is likely involved in the development of the abscission zone according to the reference of the function of TPR proteins in tomato (Solanum lycopersicum) and Arabidopsis (Arabidopsis thaliana L.). Additionally, the favorable haplotype frequency of GhDS-A06 varied among diverse areas of cotton growing in China, and the favorable haplotype frequency in northwestern China was much greater than that in other regions, which suggested that the favorable haplotype GhDS-A06 was artificially selected during breeding for machine-harvested cotton in northwestern China. The findings in the study not only provide genetic insights into defoliant sensitivity in upland cotton but also offer an ideal target for molecular breeding of defoliant sensitivity improved machine-harvested cotton plants.

A model for predicting high BMI of people living with HIV after receiving antiretroviral therapy

Objective: The objective of this study was to evaluate the characteristics of high body mass index (BMI) and normal weight people living with HIV after antiretroviral therapy (ART) and establish a model. Methods: A total of 290 people living with HIV after 1 year of ART treatment were enrolled and divided into two groups based on whether their BMI index was <24 or >= 24 at week 48. The demographic, clinical data were collected and analyzed. Multivariable logistic regression analysis was performed. A model was established and use to predict the occurrence of certain diseases. Results: A total of 290 people living with HIV were included in this study;200 had a normal BMI (BMI < 24) and 90 were high BMI (BMI >= 24) after 1-year ART. Their baseline characteristics were significantly different in relation to age (p = 0.007), sex distribution (p = 0.040), ART regimen (p = 0.040), alanine aminotransferase levels (p < 0.001), and three major serum lipid levels: triglycerides (p < 0.001), cholesterol (p = 0.011), and low-density lipoprotein (p = 0.005). A multivariate logistic regression analysis resulted in the development of a model for the diagnosis of high BMI and hyperlipidemia. The model score is an independent risk factor for hyperlipidemia (odds ratio = 2.674, p = 0.001) and high BMI (p < 0.001). The model score is significantly correlated with the controlled attenuation parameter (CAP) value (r = 0.230, p < 0.001) and can be used to divide the severity of liver steatosis based on CAP value. Conclusions: This study demonstrated a easy-to-use model to detect high BMI, hyperlipidemia, and liver steatosis in people living with HIV without risk factors for BMI changing at baseline after 1 year of ART treatment.

Rapid non-destructive analysis of lignin using NIR spectroscopy and chemo-metrics

Lignin plays an important role in the formation of stone cells in pears. However, the accumulation of lignin had adverse impact on the flavor and quality of the fruit. A rapid and accurate method for measuring the lignin content of pears is therefore required. An improved variables selection method called 'the bootstrapping soft shrinkage combined with frequency and regression coefficient of variables (FRCBOSS)' was therefore developed based on 'the bootstrapping soft shrinkage (BOSS)'technique, to identify the characteristic wavelengths of near-infrared (NIR) spectra for non-destructive and rapid analysis of lignin. Sub-models were generated by weighted bootstrap sampling (WBS) in the FRCBOSS method. For the BOSS method, the new weights of variables were determined only as the absolute values of regression coefficients of variables in each iteration. In contrast, the FRCBOSS method also considers the frequency of variables in variable space. Moreover, the FRCBOSS algorithm overcomes the disadvantage of BOSS in selecting variables, which could incorporate useful wavelengths that would otherwise be removed by the BOSS method. In addition, a range of different pre-treatment methods were used for comparison in the detection of lignin in the Snow pears. These include Savitzky-Golay Smoothing (SG), Normalization (NORM), Standard Normal Variate (SNV), and 1st Derivative (D1), as well as a combination of these methods and the different variables selection method (SiPLS, SiPLS-SPA, SiPLS-CARS, SiPLS-BOSS, and SiPLS-FRCBOSS). The number of variables selected by FRCBOSS was a little larger than that selected by BOSS. The partial least square regression (PLSR) model based on the 19 variables selected by SiPLS-FRCBOSS method had the best prediction ability, with a Rp value of prediction of 0.880 and a RMSEP value of 1.004%. 针对用于室内环境的医疗服务类机器人,提出了一种基于多目视觉传感对其工作面进行位姿精度测试的方法。该方法采用多目视觉传感器测量固定在服务机器人工作面上的靶标,通过非线性最小二乘法对测量坐标系与机器人基坐标系进行对齐,并融合多目图像数据来实现工作面位姿精度测试。最后,将该视觉测试方案在型号为ABIR X8的医疗服务机器人上进行试验验证,依据GB/T 38124 《服务机器人性能测试方法》给出项目和精度指标。该视觉测试系统和方法满足国家标准规定的各项要求。

目的建立快速检测血液中常见致病菌包括大肠埃希菌(EC)、肺炎克雷伯菌(KP)、金黄色葡萄球菌(SA)及耐甲氧西林SA(MRSA)的多重聚合酶链反应(PCR)方法,有利于败血症的及时治疗。方法建立多重PCR方法对EC phoA基因、KP mdh基因、SA femA基因和MRSA mecA基因进行检测,并将16SrDNA作为致病菌感染对照。结果采用建立的多重PCR方法进行检测的特异性为100%,EC检测限为2.75×102CFU/mL,KP为2.43×103 CFU/mL,SA为3.13×102 CFU/mL,MRSA为3.03×102 CFU/mL;用多重PCR方法和传统血培养方法对300个血标本进行检测,传统血培养方法检测出187个血标本内阳性,多重PCR方法有4个血标本为假阴性,未能被检出。结论建立的多重PCR方法可简便、及时地检测血流感染中的EC、KP、SA和MRSA。

The tumor microenvironment of pancreatic ductal adenocarcinoma (PDAC) is the main block for the penetration of chemotherapy. In the tumor microenvironment, a dense matrix composed of fibrin is formed on the exterior, while the interior is featured by high reduction, hypoxia and low pH. How to match the special microenvironment to on-demand drug release is the key to improve chemotherapeutic efficacy. Herein, a microenvironment-responsive micellar system is developed to deepen tumoral penetration. Briefly, the conjugation of a fibrin-targeting peptide to PEG-poly amino acid has been utilized to achieve accumulation of micelles in the tumor stroma. By modification of micelles with hypoxia-reducible nitroimidazole which becomes protonated under acidic conditions, their surface charge is more positive, facilitating deeper penetration into tumors. Paclitaxel was loaded onto the micelles via a disulfide bond to enable glutathione (GSH)-responsive release. Therefore, the immunosuppressive microenvironment is relived through the alleviation of hypoxia and depletion of GSH. Hopefully, this work could establish paradigms by designing sophisticated drug-delivery systems to tactfully employ and retroact the tamed tumoral microenvironment to improve the therapeutic efficacy based on understanding the multiple hallmarks and learning the mutual regulation.

Effects of the seepage capability of overlying and underlying strata of marine hydrate system on depressurization-induced hydrate production behaviors by horizontal well

Marine gas hydrates mostly occur in high-water content and non-diagenetic submarine sediments without tight stratigraphic traps. Therefore, the seepage capability of overlying and underlying strata (OUS) serves as a crucial parameter to analyze hydrate production behaviors. This study focuses on the numerical simulation study on the effects of the seepage capability of OUS on hydrate production behaviors in depressurization-induced production of three-phase hydrate deposits using a horizontal well. As shown by the simulation results, in the early stage of hydrate production, the permeability of the OUS had little effects on gas production rates. In long-term hydrate production, the hydrate deposit with lower permeability OUS has higher cumulative gas production, larger gaswater ratio and hydrate dissociation scope, and less cumulative water production. Moreover, the hydrate production efficiency first decreased rapidly and then tended to be stable as the permeability of OUS increased, and hydrate production behaviors are similar while the permeability of OUS larger than 1.0 mD. Herein, a waterresisting layers were constructed in overlying and/or underlying strata to decrease the seepage capability of OUS. Compared to the original hydrate deposits, the cumulative gas production and gas-water ratio increased by 31% and 147%, respectively but the water production was only 76% on the 5th year of the simulated production in which water-resisting layers were designed. Furthermore, the water-resisting layer in underlying strata produced more significant effects on hydrate production efficiency than the layer in overlying strata. All these indicate that the low-permeability OUS (especially in underlying strata) can restrain fluid invading into hydrate reservoirs, increase the pressure gradient of hydrate reservoirs and significantly improve the hydrate production efficiency during long-term hydrate production. Time-dependent gut microbiota analysis of juvenile Oreochromis niloticus by dietary supplementation of resveratrol

To evaluate the changes in bacterial diversity at various time points under resveratrol supplementation, we aimed to investigate the diversification of gut microbiota and the changes in total genetic diversity. We performed 16S rDNA gene sequencing at different time points (15, 30, and 45 days) to analyze the gut microbiota of tilapia. Fusobacteria, Proteobacteria, and Bacteroidetes (15 days) or Cyanobacteria (30 and 45 days) were found to be the three most abundant phyla. Cyanobacteria (15 and 30 days), Proteobacteria (15 days), Firmicutes and Chlamydiae (30 and 45 days), Planctomycetes (30 days), Bacteroidetes, Actinobacteria, and Fusobacteria (45 days) in the 0.05 g/kg RES group increased as compared to that in the controls. Proteobacteria and Cyanobacteria significantly decreased and increased at 30 and 45 days, respectively, while the reverse pattern was observed at 15 days. The Bacteroidetes:Firmicutes and Proteobacteria:Cyanobacteria ratios were significantly increased (15 and 45 days, P < 0.05) and decreased (30 days, P < 0.05). RES supplementation did not affect the richness and diversity of the gut microbiota in tilapia. Our findings may contribute to the development of strategies for the management of diseases.

Optimized prediction of sugar content in 'Snow' pear using near-infrared diffuse reflectance spectroscopy combined with chemometrics

In near-infrared spectroscopy applications, the original spectra often contain redundant information, which will seriously affect the performance of chemometric models. Therefore, preprocessing, effective wavelengths selection, and appropriate regression models are essential. The objective of this study was to optimize the nondestructive determination multivariate calibration model of sugar content in 'Snow' pears, using near-infrared diffuse reflectance spectroscopy combined with chemometrics. All data (sugar content reference values and spectra data) from three measuring positions (P1, P2, and P3, marked around the pear's equator at angular distances of approximately 120 degrees) were divided into four grouped datasets, namely Set-1 (P1), Set-2 (P2), Set-3 (P3), and Set-4 (average of the three measuring positions). All subsequent optimized processes were performed based on each grouped dataset. First, different preprocessing methods were tested and an optimal method was determined. Then, synergy interval partial least squares and synergy interval partial least squares-competitive adaptive reweighted sampling were applied to select effective regions and effective wavelengths from all wavelengths, respectively, and partial least squares regression models were established and analyzed. In addition, support vector regression models were also established for comparative study. After comprehensive analysis of prediction accuracy and model complexity, the partial least squares regression model based on the 16 selected effective wavelengths for Set-4 was optimal, with the correlation coefficient for prediction, root-mean-square error of prediction, and residual predictive deviation of 0.9701, 0.2311, and 4.12, respectively. The results indicated that with these optimized processes, the multivariate calibration model of sugar content in 'Snow' pears was effectively optimized for each dataset. In this paper, the stochastic-aeroelastic nonlinear response of a three-degree-of-freedom(3-Dof), structural nonlinear airfoil with a control flap is presented. The critical parameter conditions of stochastic P-bifurcation are solved by using the improved average method, the stochastic average method combined with the singularity theory. The results show that the periodic solution produced by Hopf bifurcation has involved a second bifurcation, the nonlinear critical speed of saddle node bifurcation points is advanced, and the airfoil appears bi-stable. The stochastic singularity analysis shows that the increasing in stochastic disturbance intensity will cause an greater of the probability for large amplitude stochastic flutter.
