NSUN3 Aggravates Sepsis-Associated Acute Kidney Injury by Stabilising TIFA mRNA Through m5C

BackgroundAcute kidney injury (AKI) is a common complication of sepsis and also a risk factor for progression of chronic kidney disease. NOP2/Sun RNA methyltransferase 3 (NSUN3) is involved in the regulation of sepsis progression. However, the mechanism by which NSUN3 regulates sepsis-associated AKI (SA-AKI) remains unclear. MethodsSA-AKI mouse model and lipopolysaccharide (LPS)-induced injury model in HK-2 cells were constructed. Haematoxylin-eosin staining, quantitative polymerase chain reaction (qPCR), western blotting, cell counting kit 8, flow cytometry, 2 ',7 '-dichlorofluorescein diacetate, enzyme-linked immunosorbent assay, methylation RNA immunoprecipitation-qPCR, actinomycin D and TdT-mediated dUTP Nick-End Labelling staining assays were utilised to explore the expression and related mechanism of NSUN3 in the SA-AKI models. ResultsThe expression of NSUN3 and tumour necrosis factor receptor-associated factor (TRAF)-interacting protein with forkhead-associated domain (TIFA) was upregulated in mice with SA-AKI and LPS-induced HK-2 cells. Knockdown of NSUN3 inhibited LPS-induced injury in HK-2 cells. Mechanically, NSUN3 increased TIFA mRNA stability and upregulated its expression through m5C modification. Moreover, knockdown of NSUN3 was found to alleviate LPS-induced HK-2 cell injury and SA-AKI in mice by reducing TIFA expression. ConclusionNSUN3 aggravates SA-AKI by stabilising TIFA mRNA through m5C, indicating that NSUN3 may be a biomarker for SA-AKI.

Hemostatic Interventions and All-Cause Mortality in Hemodynamically Unstable Pelvic Fractures: A Systematic Review and Meta-Analysis

Objective. To conduct a systematic review and meta-analysis of the all-cause mortality associated with the most commonly used hemostatic treatments in patients with hemodynamically unstable pelvic fractures. Methods. Up to April 30, 2023, we searched PubMed, Embase, Web of Science, and Cochrane, including the references to qualified papers. A meta-analysis was performed on studies that reported odds ratios (ORs) or the number of events needed to calculate them. The PROSPERO registration number was CRD42023421137. Results. Of the 3452 titles identified in our original search, 29 met our criteria. Extraperitoneal packing (EPP) (OR = 0.626 and 95% CI = 0.413-0.949), external fixation (EF) (OR = 0.649 and 95% CI = 0.518-0.814), and arterial embolism (AE) (OR = 0.459 and 95% CI = 0.291-0.724) were associated with decreased mortality. Resuscitative endovascular balloon occlusion of the aorta (REBOA) (OR = 2.824 and 95% CI = 1.594-5.005) was associated with increased mortality. A random effect model meta-analysis of eight articles showed no difference in mortality between patients with AE and patients with EPP for the initial treatments for controlling blood loss (OR = 0.910 and 95% CI = 0.623-1.328). Conclusion. This meta-analysis collectively suggested EF, AE, or EPP as life-saving procedures for patients with hemodynamically unstable pelvic fractures.

Amphiphilic ionic liquid hydrogel electrolytes with high ionic conductivity towards dendrite-free ultra-stable aqueous zinc ion batteries

Aqueous zinc ion batteries (AZIBs) are highly promising as large-scale energy storage devices with high capacity, safety, and low cost. However, the parasitic reaction on the of Zn anode and the growth of Zn dendrites greatly prevent the practical application of AZIBs. Here, we propose an effective strategy with hydrogel electrolytes by adding dual amphiphilic ionic liquids into acrylamide, allowing for the occurrence of the following phenomena related to the Zn anode: 1) Zn2+ preferential adsorption by the amphiphilic ionic groups to regulate the electrodeposition behavior of Zn2+ on the Zn(002) crystal surface and inhibit the growth of Zn dendrites;2) water removal from the Zn2+ solvation sheath, reducing water activity and inhibiting side reactions;3) improved environmental adaptability of the hydrogel electrolyte through enhancing its mechanical properties and adhesion. The obtained hydrogel electrolyte has a high ionic conductivity of 41.47 mS/cm, a highly reversible Zn plating/stripping cycle of 1400 h in Zn||Zn symmetric batteries and an enhanced electrochemical performance in Zn||NVO full batteries (94 % capacity retention at 5.0 A g- 1/3000 cycles). Even under harsh conditions (10 mA cm -2/10 mAh cm -2), these cells maintained excellent cycling stability. This study provides new ideas for the practical applications of AZIBs.

Central Retinal Artery Occlusion Visual Outcomes from a Large Northern California Cohort

Purpose: To assess visual acuity (VA) outcomes in a large cohort of patients diagnosed with nonarteritic central retinal artery occlusion (CRAO), and to ascertain whether time from symptom onset to presentation, presenting VA, or conservative treatment delivery (anterior chamber paracentesis, ocular massage, intraocular pressure lowering drugs, hyperventilation, or some combination of those) impacted ultimate VA outcomes. Design: Retrospective cohort study. Subjects: The study included 794 patients who presented with CRAO between 2011 and 2020. Within this cohort, 484 individuals presented within 30 days of symptom onset and had comprehensive documentation regarding the details of their presentation, management, and follow-up >= 90 days postdiagnosis. Methods: Retrospective chart review was conducted for all patients with a diagnosis of CRAO initially identified via International Classification of Diseases coding, followed by confirmation of diagnosis by 2 retina specialists. Cases of arteritic CRAO were excluded. Main Outcome Measures: Visual acuity recovery, defined as improvement from <= 20/200 or worse at presentation to >= 20/100 >= 90 days after diagnosis. Results: Of the 794 identified patients, 712 (89.7%) presented with VA of <= 20/200. Similarly, 447 (92.4%) of the 484-patient subset that presented within 30 days and had comprehensive documentation presented with VA <= 20/200. Of the 441 of those patients with documented follow-up, 380 (86.2%) remained at that level. Of the 244 patients who presented within 4.5 hours of symptom onset, 227 (93%) presented <= 20/200 and 201 (92.6%) of the 217 of those with follow-up data did not improve beyond that threshold. There was no significant difference (P P <0.05) in final VA between patients presenting before versus after 4.5 hours from time of vision loss. Severe Traumatic Brain Injury Outcomes in Patients with Premorbid Psychiatric Illness

OBJECTIVE: Individuals with psychiatric illnesses (PIs) have increased rates of traumatic brain injury (TBI). Nonetheless, the influence of underlying PI on TBI outcomes is poorly understood. METHODS: We analyzed the medical records of 633 adult-severe TBI patients admitted to our institution between 2010 and 2021. We identified patients with pre- morbid PI (Psych (+) group, n = 129) and a subset with only a substance use disorder (SUD (+) group, n = 60) and compared them to patients without PI (Psych (-) group, n = 480). Outcome measures included discharge Glasgow Coma Scale (GCS), length of stay, in-hospital survival, and Glasgow Outcome Scale-Extended (GOS-E). RESULTS: The Psych (+) group had increased in- hospital survival (69.8% vs. 55.0%, P = 0.003) and fewer patients with severe (3-8) discharge-GCS (28.7% vs. 46.0%, P < 0.001). The SUD (+) group had increased in-hospital survival (70.0% vs. 55.0%, P = 0.028) and fewer patients with severe discharge-GCS (28.3% vs. 46.0%, P = 0.009). However, the Psych (+) (21.0 vs. 10.0 days, P < 0.001) and SUD (+) (16.0 v. 10.0 days, P = 0.011) groups had longer length of stay. The Psych (+) group had a higher mean GOS-E at discharge (2.7 vs. 2.4, P = 0.004), 6-months (3.8 vs. 3.0, P = 0.006) and 1-year (3.4 vs. 2.3, P = 0.027). The SUD (+) group also had a higher mean GOS-E at discharge (2.8 vs. 2.4, P = 0.034), six months (3.8 vs. 3.0, P = 0.035), and one year (3.5 vs. 2.3, P = 0.008). Additionally, there were no significant differences in injury severity or computed tomography scan findings. CONCLUSIONS: Individuals with PI and SUD appeared to have better outcomes but more complicated hospital stays following severe TBI. Future studies should investigate the mechanisms underlying these outcomes.

The purpose of this research is to study and conduct an experimental test for investigating the increase of efficiency of a solar panel when cooling it with a backside cooling system. There were 3 experiment test-rigs by using 3 different refrigerants: water, ice, and dry ice. By containing a type of refrigerant into its identical box that was designed by wrapping insulation around the box while drilling the holes for the box that did not contain any refrigerant. Then covered the box with a solar panel. To determine which refrigerant could be the best helped increase the efficiency of a solar panel. The research team conducted the tests with 2 time periods: a short time for 1 hour operation when collecting experimental results for every 2 minutes and a long time for 6 hours when collecting experimental results every 10 minutes. It is found from the experimental tests that significantly different capacity in power generated for different types of refrigerants could be obviously noticed even for the short time. For the long-time test, ice was the best refrigerant that continuously enhanced the efficiency for the solar panel with 25.2 watts compared with 24.0 watts for no refrigerant cooling, which was 6.25% increase. © 2024 IEEE.

Materiality, Experience and the Body: The Catholic Pilgrimage of Sheshan in Shanghai, China

This paper foregrounds the Turnerian experiences of pilgrims themselves, for whom pilgrimage is perhaps first and foremost the process of experiencing faith with their whole body and mind. At the Chinese Catholic pilgrimage site of Sheshan, located in western Shanghai, multiple meanings and possibilities are written onto the body of the pilgrim as it interacts with sacred materialities. In the process, the pilgrim materially orients themselves towards the transcendent other and to people and events throughout time. The boundary between subject and object is increasingly blurred in the pilgrim's imagination, and pilgrimage becomes a 'porous' mind-body experience for them. In the process, as pilgrims repeatedly physically enact doctrine and doctrinal texts in the course of pilgrimage-while simultaneously rooting them in their own personal lives-Sheshan is, through the concrete actions of worshippers layered up over time, continually being re-made as sacred.

Efficient removal and recovery of phosphorus from industrial wastewater in the form of vivianite

With the shortage of phosphorus resources, the concept of phosphorus recovery from wastewater is generally proposed. Recently, phosphorus recovery from wastewater in the form of vivianite has been widely reported, which could be used as a slow-release fertilizer as well as the production of lithium iron phosphate for Li-ion batteries. In this study, chemical precipitation thermodynamic modeling was applied to evaluate the effect of solution factors on vivianite crystallization with actual phosphorus containing industrial wastewater. The modeling results showed that the solution pH influences the concentration of diverse ions, and the initial Fe2+ concentration affects the formation area of vivianite. The saturation index (SI) of vivianite increased with the initial Fe2+ concentration and Fe:P molar ratio. pH 7.0, initial Fe2+ concentration 500 mg/L and Fe:P molar ratio 1.50 were the optimal conditions for phosphorus recovery. Mineral Liberation Analyzer (MLA) accurately determined the purity of vivianite was 24.13%, indicating the feasibility of recovering vivianite from industrial wastewater. In addition, the cost analysis showed that the cost of recovering phosphorus by the vivianite process was 0.925 USD/kg P, which can produce high-value vivianite products and realize "turn waste into treasure".

Nitrite accumulation stability evaluation for low-strength ammonium wastewater by adsorption and biological desorption of zeolite under different operational temperature

How to achieve stable nitrite accumulation was still a huge challenge for low-carbon and energy-saving biological nitrogen removal of low-strength ammonium wastewater. This study proposed a new way to solve this problem with zeolite biological fixed bed (ZBFB) by cycle operation of adsorption and biological desorption. In order to evaluate nitritation performance of this reactor, the influence of operational temperature on nitrite accumulation stability was investigated by 126 cycles operation in four parallel ZBFB reactors for low-strength ammonium wastewater (50 mg/L NH4+-N). It was found that higher operational temperature (i.e., 36.0 degrees C), rather than other temperature (i.e., 27.0 degrees C, 30.0 degrees C, 33.0 degrees C), could maintain stable nitrite accumulation with nitrite production rate of 0.312 kg NO2--N.m(-3) ***(-1) and nitrite accumulation ratio higher than 95.0% after biological desorption. High-throughput sequencing analysis results showed that bacterial structure significantly changed in ZBFB under different operational temperature, and obvious enrichment of genus Nitrosomonas (AOB) and gradually enhanced free ammonia (FA) inhibition on genus Nitrospira and Nitrobacter (NOB) were found by elevation of operational temperature, leading to different nitrite accumulation performance in ZBFB reactors. The mechanism for stable nitrite accumulation performance by ZBFB might be attributed to overwhelming growth rate of AOB than NOB, faster ammonium desorption and enhanced FA inhibition on NOB under operational temperature (i.e., 36.0 degrees C). All in all, keeping high temperature for biological desorption step should be extremely crucial for stable nitrite accumulation by ZBFB, which could facilitate further low-carbon and energy-saving biological nitrogen removal for low-strength ammonium wastewater treatment. BACKGROUND The completely autotrophic nitrogen removal over nitrite (CANON) system has excellent performance in treating ammonium-rich wastewater and it depends on ammonia oxidizing bacteria (AOB) working together with anammox bacteria in a reactor. Higher ammonium nitrogen loading requires higher aeration to provide sufficient dissolved oxygen (DO) for nitritation, whereas high-strength aeration in theCANON will destroy the sludge structure and inhibit the activity of anammox bacteria. The present study uses a stable pre-nitritation system to reduce a part of nitritation and also decrease aeration intensity in the CANON. RESULTS Continuous aeration to maintain the DO concentration between 1.5 and 4.3 mg L(-1)in the CANON achieved a stable nitrogen removal rate (1.22 kg N m(-3)d(-1)) from iron oxide red wastewater with 400-1016 mg N L(-1)after 183 days of operation. Using a mixed alkalinity donor [sodium carbonate (Na2CO3) and bicarbonate (NaHCO3)] in the combined system can save approximate to 14.1-28.2% of the alkalinity cost compared with using NaHCO(3)only. CONCLUSIONS A zeolite biofilm aeration filter (ZBAF) used in a pre-nitritation process can tolerate high DO concentration of <= 6.5 mg L(-1)for nitritation and avoid all of the nitritation taking place in the CANON system. This can protect the sludge structure and anammox bacteria from high-strength aeration erosion. (c) 2020 Society of Chemical Industry
